    In whey cheese manufacture, whey, plain or added with milk, is heated by direct fire, bubbling steam or alternatively in jacketed vats. In some cases, salt s or organic acids are previously added. At 80-85 OC, the first particles of curd form; at 85-95 'C, the curd may be cooked for a few minutes to reduce moisture content and/or to obtain the desirable level of browning. After drainage at room temperature during molding for ca. 4 h, whey cheese is stored at ca. 4 'C. The typical mass yield is 6%, but addition of milk, calcium salts and preliminary concentration of protein (by condensation or ultrafiltration techniques) may increase yield considerably. Some types of whey cheeses are supposed to be consumed within a short time upon manufacture (e.g., Ricotta, Requeijdo and Manouri), whereas others bear a longer shelf life (e.g., Gjetost, Mysost and Myzithra). Whey cheeses are significantly different from one another in terms of chemical composition, which is mainly due to variations in the source and type of whey, as well as to the processing practices followed. Moisture content and pH of whey cheeses are usually high and favor microorganism growth (molds, yeasts, lactic acid bacteria and Enterobacteriaceae account for the dominant microflora in these cheeses). Adequate packaging of whey cheeses should be provided, and legislation should be prepared to fix standard characteristics of each type of whey cheese, and hence protect typical products from adulteration and fakes. Marketing efforts should also be aimed at increasing whey cheese consumption, either directly or incorporated in desserts, snack dips and pasta-type dishes

    The effects of a plant proteinase inhibitor from Enterolobium contortisiliquum on human tumor cell lines

    Supplementary to the efficient inhibition of trypsin, chymotrypsin, plasma kallikrein, and plasmin already described by the EcTI inhibitor from Enterolobium contortisiliquum, it also blocks human neutrophil elastase (K(iapp)=4.3 nM) and prevents phorbol ester (PMA)-stimulated activation of matrix metalloproteinase (MMP)-2 probably via interference with membrane-type 1 (MT1)-MMP. Moreover, plasminogen-induced activation of proMMP-9 and processing of active MMP-2 was also inhibited. Furthermore, the effect of EcTI on the human cancer cell lines HCT116 and HT29 (colorectal), SkBr-3 and MCF-7 (breast), K562 and THP-1 (leukemia), as well as on human primary fibroblasts and human mesenchymal stem cells (hMSCs) was studied. EcTI inhibited in a concentration range of 1.0-2.5 mu M rather specifically tumor cell viability without targeting primary fibroblasts and hMSCs. Taken together, our data indicate that the polyspecific proteinase inhibitor EcTI prevents proMMP activation and is cytotoxic against tumor cells without affecting normal tissue remodeling fibroblasts or regenerative hMSCs being an important tool in the studies of tumor cell development and dissemination

    Uso potencial da brita e argila na redução da toxicidade de efluentes da aquicultura contendo diflubenzuron.

    Resumo: O crescimento acelerado da aquicultura no Brasil e no mundo vem a atender a uma demanda crescente por proteína animal. No entanto, este crescimento deve vir acompanhado da medidas de conservação dos recursos naturais, bem como a minimização dos impactos sobre a qualidade da água dos viveiros de criação e dos efluentes gerados por esta atividade. O presente estudo tem por objetivo avaliar a toxicidade do inseticida diflubenzuron (usado na piscicultura) sobre o organismo Artemia salina, após a filtragem com um sistema natural de tratamento de efluentes, utilizando como meios filtrantes a brita e a argila expandida. Os resultados obtidos indicam que a argila expandida foi mais eficiente na redução da toxicidade do que a brita. Embora ainda sejam necessários testes com outros organismos não alvo, sugere-se o uso da argila como filtro na saída dos viveiros para melhoria da qualidade da água do efluente

    Improving light harvesting in polymer photodetector devices through nanoindented metal mask films

    To enhance light harvesting in organic photovoltaic devices, we propose the incorporation of a metal (aluminum) mask film in the system’s usual layout. We fabricate devices in a sandwich geometry, where the mask (nanoindented with a periodic array of holes of sizes d and spacing s) is added between the transparent electrode and the active layer formed by a blend of the semiconducting polymer P3HT and substituted fullerene. Its function is to promote trapping of the incident light into the device’s cavity (the region corresponding to the active layer). For d, we set a value that allows light diffraction through the holes in the relevant absorption range of the polymer. To optimize the mask structure, we consider a very simple model to determine the s leading to trapped fields that are relatively intense and homogeneous within the device. From measurements of the action spectra, we show that, indeed, such architecture can considerably improve the resulting photocurrent efficiencies—one order of magnitude in the best situation studied.

    Effectiveness factor for immobilized biocatalysts: two substratestwo products reactions

    Immobilized enzymes are being increasingly used as biocatalysts in numerous processes to obtain high-value products for the pharmaceutical, flavour and fragrance industries (Gandhi et al., 2000). The major advantages of immobilization include the increase in enzyme stability, the possibility of enzyme reutilization and the easy separation of the biocatalysts from the reaction mixture. However, it is necessary to account for mass transfer limitations that, under some conditions, may arise in these systems (Gómez et al., 2003; Jeison et al., 2003). These resistances comprise the effects of intraparticle diffusion and external mass-transfer. Given the complexity of the kinetics of multisubstrate enzyme reactions, reactor modelling studies that account for mass-transfer phenomena are so far limited to single-substrate ones (Gómez et al., 2003). To compare the observed reaction rate with the reaction rate in the absence of mass-transfer limitations, an overall effectiveness factor is usually calculated (Gómez et al., 2003; Jeison et al., 2003). In this work, a model is developed to calculate the overall effectiveness factor for immobilized enzymes that carry out irreversible two substrates-two products reactions following kinetic mechanisms such as the Ternary Complex or the Ping-Pong Bi-Bi with inhibition by the second substrate. The model has two dimensionless parameters for each substrate – Thiele modulus (reaction/intraparticle diffusion), Biot number (film diffusion/intraparticle diffusion) – and one related to the reaction kinetics. Their influence on the effectiveness factor is analysed. The results obtained can be applied in the design and simulation of enzymatic reactors

    Removing data and using metafounders alleviates biases for all traits in Lacaune dairy sheep predictions

    Bias in dairy genetic evaluations, when it exists, has to be understood and properly addressed. The origin of biases is not always clear. We analyzed 40 yr of records from the Lacaune dairy sheep breeding program to evaluate the extent of bias, assess possible corrections, and emit hypotheses on its origin. The data set included 7 traits (milk yield, fat and protein contents, somatic cell score, teat angle, udder cleft, and udder depth) with records from 600,000 to 5 million depending on the trait,-1,900,000 animals, and-5,900 genotyped elite artificial insemination rams. For the-8% animals with missing sire, we fit 25 unknown parent groups. We used the linear regression method to compare "partial" and "whole" predictions of young rams before and after progeny testing, with 7 cut-off points, and we obtained estimates of their bias, (over)dispersion, and accuracy in early proofs. We tried (1) several scenarios as follows: multiple or single trait, the "official" (routine) evalua-tion, which is a mixture of both single and multiple trait, and "deletion" of data before 1990; and (2) sev-eral models as follows: BLUP and single-step genomic (SSG)BLUP with fixed unknown parent groups or metafounders, where, for metafounders, their relation-ship matrix gamma was estimated using either a model for inbreeding trend, or base allele frequencies esti-mated by peeling. The estimate of gamma obtained by modeling the inbreeding trend resulted in an estimated increase of inbreeding, based on markers, faster than the pedigree-based one. The estimated genetic trends were similar for most models and scenarios across all traits, but were shrunken when gamma was estimated by peeling. This was due to shrinking of the estimates of metafounders in the latter case. Across scenarios, all traits showed bias, generally as an overestimate of genetic trend for milk yield and an underestimate for the other traits. As for the slope, it showed overdisper-sion of estimated breeding values for all traits. Using multiple-trait models slightly reduced the overestimate of genetic trend and the overdispersion, as did including genomic information (i.e., SSGBLUP) when the gam-ma matrix was estimated by the model for inbreeding trend. However, only deletion of historical data before 1990 resulted in elimination of both kind of biases. The SSGBLUP resulted in more accurate early proofs than BLUP for all traits. We considered that a snowball ef-fect of small errors in each genetic evaluation, combined with selection, may have resulted in biased evaluations. Improving statistical methods reduced some bias but not all, and a simple solution for this data set was to remove historical records
