    Study of the stability of a paramagnetic label linked to mesoporous silica surface in contact with rat mesothelial cells in culture.

    Stable radicals detectable by electron paramagnetic resonance (EPR) may be use in the investigation of early events in cell-particle toxicity. Piperidine-N-oxyl derivatives (nitroxides), covalently linked to the surface of a high surface area silica (used as model solid for the technique), served as probes in the investigation of the effects of incubation of silica particles with mesothelial cells. A mesoporous silica (MCM-41), prepared by precipitation from a micellar solution, was the most appropriate silica-based particle for this purpose, as its channels allow direct contact with small molecules but not with macromolecules. The cytotoxicity of this amorphous silica is very low, allowing relatively high particle loading in the cell cultures. Both the high surface area of the sample and the large amount of inorganic material extracted from the cell culture provide enough material to run reasonably intense EPR spectra. Computer-aided analysis of the EPR spectra of silica-bound nitroxides provided information on the sensitivity of the labeled silica monitoring different environments, e.g., to follow the path of particles in a mammalian cell culture. Upon contact of the particles with mesothelial cells, the mean distance among the labels at the silica surface decreased as a consequence of the release of oxidizing and/or radical moieties from the cells

    The onset and solidification path of a basaltic melt by in situ differential scanning calorimetry (DSC) and ex situ investigations

    The in situ differential scanning calorimetry (DSC) technique has been applied to investigate the solidification paths of a basaltic liquid. The starting glass was heated up to 1300◦C, kept at this superliquidus temperature for 2 h and cooled at rates (∆T/∆t) of 7, 60, 180, 1000, and 1800◦C/h, down to 800 and 600◦C. Glass transition temperature (Tg), crystallization temperature (Tx_HR) and melting temperature (Tm) were measured by in situ DSC spectra on heating. Tx measured along the cooling paths (Tx_CR) shows exothermic peaks that change from a single symmetric shape (7 and 60◦C/h) to multi-component patterns (180, 1000, and 1800◦C/h). The recovered products characterized by field emission gun source of the scanning electron microscopy and electron probe micro-analyzer-wavelength dispersive spectrometers show a phase assemblage of spinel (sp), clinopyroxene (cpx), melilite (mel), plagioclase (plg), and glass. Moreover, crystal size distributions (CSDs) and growth rates (Gmax and GCSD) were also determined. The crystal content slightly increases from 7 to 1800◦C/h. Faceted sp are present in all the run products with an amount always <2 area%. Cpx increases from 7 to 1800◦C/h, changing its texture from almost faceted to dendritic between 60 and 180◦C/h. The area% of mel follows an asymmetric Gaussian trend, while plg nucleates only at 7◦C/h with a content <2 area%. The coupling of DSC and SEM outcomes indicate that sp nucleate first, followed by cpx and mel (and/or plg). The increment of ∆T/∆t causes an increase of the CSD slope (m) and crystal population density per size (n0 ), as well as a decrease of the crystal size, for both cpx and sp. The log-linear CSD segments with different slopes at 7 and 60◦C/h suggest multiple nucleation events and crystal growth by coarsening. Gmax and GCSD for cpx and sp directly measured on the actual crystallization time by DSC spectra, both increase with the increasing of ∆T/∆t. The onset temperature of crystallization (Txi ) decreases as ∆T/∆t increases, following an exponential trend that defines the uppermost portion of a time-transformation-temperature-like curve. This analytical model allows us to quantitatively model the kinetic crystallization paths of dry basalts

    Preliminary observations of caulerpin accumulation from the invasive Caulerpa cylindracea in native Mediterranean fish species

    Recent studies have shown that the Mediterranean white sea bream Diplodus sargus includes the invasive green alga Caulerpa cylindracea in its diet, with consequent metabolic and enzymatic alterations. As a result of this novel alimentary habit, the bioactive algal red pigment caulerpin has been detected in its tissues. However, this may not be an isolated case: other fish species have also been reported to feed on C. cylindracea, although the possible accumulation of caulerpin in their tissues has not yet been investigated. In this report, we analysed stomach contents and caulerpin levels in the native sparid species Spondyliosoma cantharus, Sarpa salpa, and Diplodus vulgaris, and in the scarid Sparisoma cretense, along with the Lessepsian siganid Siganus luridus. C. cylindracea was found in the stomachs of all but one fish species, the exception being S. cretense, in which prey items could not be determined due to the high degree of digestion. Chemical analysis of fish tissues revealed that only S. cantharus and S. salpa accumulated caulerpin, while no traces of the compound were detected in the other species. Despite intense research efforts on natural products obtained from C. cylindracea, a complete picture of the impacts caused by fish including this alga in their diet has not been elucidated. The identification of caulerpin in other Mediterranean native fish suggests a need for further research in order to assess the possible transfer of such molecules to humans through seafood consumption

    Fault structure and slip localization in carbonate-bearing normal faults: An example from the Northern Apennines of Italy

    Carbonate-bearing normal faults are important structures for controlling fluid flow and seismogenesis within the brittle upper crust. Numerous studies have tried to characterize fault zone structure and earthquake slip processes along carbonate-bearing faults. However, due to the different scales of investigation, these studies are not often integrated to provide a comprehensive fault image. Here we present a multi-scale investigation of a normal fault exhumed from seismogenic depths. The fault extends for a length of 10 km with a maximum width of about 1.5 km and consists of 5 sub-parallel and interacting segments. The maximum displacement (370e650 m) of each fault segment is partitioned along sub-parallel slipping zones extending for a total width of about 50 m. Each slipping zone is characterized by slipping surfaces exhibiting different slip plane phenomena. Fault rock development is controlled by the protolith lithology. In massive limestone, moving away from the slip surface, we observe a thin layer (<2 cm) of ultracataclasite, cataclasite (2e10 cm) and fault breccia. In marly limestone, the fault rock consists of a cataclasite with hydrofractures and smectite-rich pressure solution seams. At the micro-nanoscale, the slip surface consists of a continuous and thin (<300 mm) layer composed of coarse calcite grains (~5e20 mm in size) associated with sub-micrometer grains showing fading grain boundaries, voids and/or vesicles, and suggesting thermal decomposition processes. Micrometer-sized calcite crystals show nanoscale polysynthetic twinning affected by the occurrence of subgrain boundaries and polygonalized nanostructures. Investigations at the kilometres-tens of meter scale provide fault images that can be directly compared with high-resolution seismological data and when combined can be used to develop a comprehensive characterization of seismically active fault structures in carbonate lithologies. Micro and nanoscale investigations along the principal slipping zone suggest that different deformation processes, including plastic deformation and thermal decomposition, were active during seismic slip

    L’identité professionnelle des enseignants face aux changement. Analyses empiriques dans le contexte italienne

    La finalité de cet article est de proposer, à partir d’un certain nombre des donnés empiriques récoltées dans le contexte éducatif italien, une explication psychosociale de l’élaboration de l’identité professionnelle des enseignants en situation de changement. Une telle explication sera orientée à mettre en évidence le rôle essentiel (et l’articulation) des éléments biographiques, des représentations sociales et des pratiques éducatives dans le processus d’adaptation identitaire sollicité par les changements qui caractérisent le contexte éducatif au point de vue institutionnel, social et individuel (nouveaux processus de migration, introduction des nouvelles technologies à l’école, plusieurs reformes de la loi scolaire et de l’organisation de l’enseignement). Deux recherches ont été conduites dans une région de l’Italie du Sud (Campanie) en utilisant un entretien narratif semi structuré (Bruner, 1992; Vermersch, 1994) avec deux groupes d’enseignants (52 et 43 respectivement). Les résultats permettent de décrire un certain nombre d’éléments identitaires du « métier » enseignant (représentations, biographies, pratiques) qui semblent jouer un rôle important dans le processus d’adaptation aux changements considérés

    Interaction of the NRF2 and p63 transcription factors promotes keratinocyte proliferation in the epidermis

    Epigenetic regulation of cell and tissue function requires the coordinated action of transcription factors. However, their combinatorial activities during regeneration remain largely unexplored. Here, we discover an unexpected interaction between the cytoprotective transcription factor NRF2 and p63-a key player in epithelial morphogenesis. Chromatin immunoprecipitation combined with sequencing and reporter assays identifies enhancers and promoters that are simultaneously activated by NRF2 and p63 in human keratinocytes. Modeling of p63 and NRF2 binding to nucleosomal DNA suggests their chromatin-assisted interaction. Pharmacological and genetic activation of NRF2 increases NRF2-p63 binding to enhancers and promotes keratinocyte proliferation, which involves the common NRF2-p63 target cyclin-dependent kinase 12. These results unravel a collaborative function of NRF2 and p63 in the control of epidermal renewal and suggest their combined activation as a strategy to promote repair of human skin and other stratified epithelia

    Sonographic Assessment of Uterine Biometry for the Diagnosis of Diffuse Adenomyosis in a Tertiary Outpatient Clinic

    Background: to compare several uterine biometric parameters at transvaginal ultrasound (TVUS) between adenomyosis and non-adenomyosis uteri and evaluate their role for the diagnosis of diffuse adenomyosis. Methods: prospective observational study conducted between the 1 February 2022 and the 30 April 2022. In this case, 56 patients with TVUS diagnosis of adenomyosis were included. A 1:1 ratio age and parity-matched group of non-adenomyosis patients was selected. We compared sonographic uterine biometric parameters (longitudinal (LD), anteroposterior (APD) and transverse (TD) diameters, volume, simple and complex diameter ratios) and investigated their diagnostic performance. Results: all sonographic parameters were significantly different between the study groups, except for TD/(LD+APD). Optimal cut-off values of APD and LD/APD showed the best sensitivity and specificity. APD diameter equal or superior to 39.5 mm (95% CI, 36.2–42.8) had sensitivity of 0.70 (95% CI, 0.57–0.80), specificity of 0.71 (95% CI, 0.59–0.82) and accuracy of 0.75 (95% CI, 0.66–0.84). LD/APD equal or inferior to 2.05 (95% CI, 1.96–2.13) showed sensitivity and specificity of 0.70 (95% CI, 0.57–0.80) each and accuracy of 0.72 (95% CI, 0.62–0.81). Conclusions: several biometric uterine parameters at TVUS in fertile-aged women were statistically different between adenomyosis and non-adenomyosis uteri, though their optimal cut-off values showed low accuracy in diagnosing adenomyosis

    Chemoecological studies on marine natural products: terpene chemistry from marine mollusks

    Some species of nudibranchs (Mollusca) protect themselves from predatory attacks by storing defensive terpene chemicals acquired from dietary sponges (Porifera) in specialized body parts called MDFs (mantle dermal formations), often advertising their unpalatability to potential predators by means of bright coloration patterns. Consequently, the survival of these trophic specialist species is closely related to the possibility of obtaining the defensive tools from sponges that live in their immediate vicinity; therefore, it is important to determine as precisely as possible the chemical composition of nudibranch extracts prior to any ecological studies addressing issues that involve their alimentary behavior and their defensive strategies, including the significance of their color patterns. Some of our recent studies on the chemical composition of terpene extracts from nudibranchs belonging to the genera Chromodoris and Hypselodoris are summarized. We also report the development of a method to assay extracts and purified metabolites for their feeding deterrent activity against co-occurring generalist predators. In a recent chemoecological study, showing that repugnant terpene chemicals are accumulated at extremely high concentrations in exposed parts of the nudibranchs' bodies, the feeding deterrence assays were carried out on the generalist marine shrimp Palaemon elegans, very common in the Mediterranean. We have modified this assay for use with the Australian shrimp species P. serenus, and confirmed the ecological validity of the assay by analysis of extracts from species of sponges and mollusks that live in the same habitat as P. serenus. The deterrent properties of haliclonacyclamine alkaloids isolated from the sponge Haliclona sp. were demonstrated, with the alkaloid mixture demonstrating palatability deterrence at concentrations as low as 0.05 mg/mL, and complete deterrence at 0.75 mg/mL. In contrast, the diterpene thuridillin metabolites from the sacoglossan mollusk Thuridilla splendens did not deter feeding by P. serenus

    Uterine carcinosarcoma vs endometrial serous and clear cell carcinoma: a systematic review and meta-analysis of survival

    Background: It is unclear whether uterine carcinosarcoma (UCS) is more aggressive than endometrial serous carcinoma (SC) and clear cell carcinoma (CCC). Objectives: To compare the prognosis of UCS to that of endometrial SC and CCC, through a systematic review and meta-analysis. Methods: Four electronic databases were searched from January 2000 to October 2020. All studies assessing hazard ratio (HR) for death in UCS vs SC and/or CCC. HRs for death with 95% confidence interval were extracted and pooled by using a random-effect model. A significant P-value &lt;0.05 was adopted. Results: Six studies with 11 029 patients (4995 with UCS, 4634 with SC, 1346 with CCC and 54 with either SC or CCC) were included. UCS showed a significantly worse prognosis than SC/CCC both overall (HR = 1.51; P = 0.008) and at early stage (HR = 1.58; P &lt; 0.001). Similar results were found for UCS vs SC (HR = 1.53; P &lt; 0.001) and UCS vs CCC (HR = 1.60; P &lt; 0.001). Conclusions: Compared to SC and CCC, UCS has a significantly worse prognosis, with a 1.5–1.6-fold increased risk of death. This might justify a more aggressive treatment for UCS compared to SC and CCC. Further studies are necessary to define the prognostic impact of different molecular subgroups

    Insulin secretion in patients with latent autoimmune diabetes (LADA): half way between type 1 and type 2 diabetes: action LADA 9

    Background: The study of endogenous insulin secretion may provide relevant insight into the comparison of the natural history of adult onset latent autoimmune diabetes (LADA) with types 1 and 2 diabetes mellitus. The aim of this study was to compare the results of the C-peptide response to mixed-meal stimulation in LADA patients with different disease durations and subjects with type 2 and adult-onset type 1 diabetes. Methods: Stimulated C-peptide secretion was assessed using the mixed-meal tolerance test in patients with LADA (n = 32), type 1 diabetes mellitus (n = 33) and type 2 diabetes mellitus (n = 30). All patients were 30 to 70 years old at disease onset. The duration of diabetes in all groups ranged from 6 months to 10 years. The recruitment strategy was predefined to include at least 10 subjects in the following 3 disease onset categories for each group: 6 to 18 months, 19 months to 5 years and 5 to 10 years. Results: At all time-points of the mixed-meal tolerance test, patients with LADA had a lower stimulated C-peptide response than the type 2 diabetes group and a higher response than the type 1 diabetes group. The same results were found when the peak or area under the C-peptide curve was measured. When the results were stratified by time since disease onset, a similar pattern of residual insulin secretory capacity was observed. Conclusions: The present study shows that the magnitude of stimulated insulin secretion in LADA is intermediate between that of type 1 and type 2 diabetes mellitus
