    Association of increased levels of homocysteine and peripheral arterial disease in a Japanese-Brazilian population

    Objectives. Our aim was to evaluate the possible association between homocysteine levels and peripheral arterial disease (PAD) in a population-based study of Japanese-Brazilians.Materials and methods. This cross-sectional study was derived from a population-based survey on the prevalence of diabetes and associated diseases conducted in Japanese-Brazilians. A total of 1330 male and female subjects aged >= 30 years were submitted to clinical examination and laboratory procedures including homocysteine measurement. the ankle-brachial index (ABI) was calculated, subjects with ABI values <0.9 were diagnosed with PAD. the evaluable population included 1008 subjects. Logistic regression was used taking PAD as the dependent variable.Results. Mean age of the population was 56.5 years and overall prevalence of PAD was 20%. A worse cardiovascular profile was found in male patients, including significantly higher homocysteine levels (11.9 +/- 1.8 vs. 9.1 +/- 1.1 mu mol/L, p < 0.001). Men with PAD had higher prevalence rates of hyperhomocysteinemia compared to women (22.7% vs 7.6%). Univariate analysis showed an odds ratio of hyperhomocysteinemia for PAD of 1.51 [1.02-2.25] in men and 1. 69 [1.06-2.68] in women. After adjustment for other cardiovascular risk factors, higher levels of homocysteine were only significantly related to PAD in men.Conclusion. in a Japanese-Brazilian population, elevated levels of homocysteine are associated with PAD in men.     Life Satisfaction in Retirees Who are still Working 1

    Retirement does not always represent the final exit from the labor market. Work is one of the most important aspects in people’s lives, and a relationship seems to exist between continuity of work during retirement and life satisfaction. This study evaluated the perception of life satisfaction among 230 retirees who were still working. Participants completed a questionnaire containing scales for Life Satisfaction, Job Perception and Reasons for Returning to Work. Correlations and multiple linear regression indicated that perception of the work, satisfaction with income and intrinsic reasons (such as the feeling of productivity) influenced life satisfaction of retirees who returned to work. Our results reinforce the adoption of public and organizational policies to maintain retirees who wish to continue working.Nem sempre a aposentadoria significa a saída definitiva do mercado de trabalho. O trabalho é um dos aspectos mais relevantes na vida das pessoas adultas e parece haver uma relação entre a continuidade do trabalho durante a aposentadoria e a satisfação com a vida. Este estudo avaliou a percepção de 230 aposentados que ainda estavam trabalhando e a satisfação com suas vidas. Os participantes responderam a um questionário contendo escalas de Satisfação com a Vida, Percepção do Trabalho e Motivos para o Retorno ao Trabalho. As correlações e a regressão linear múltipla apontaram que a percepção do trabalho, a satisfação com a renda e os motivos intrínsecos, como o sentimento de produtividade, influenciaram a satisfação com a vida desses aposentados que retornaram ao trabalho. Tais resultados apontam para a necessidade de políticas públicas e organizacionais visando a retenção de aposentados que desejam permanecer trabalhando.La jubilación no siempre representa la salida definitiva del mercado laboral. El trabajo es uno de los aspectos más importantes en la vida de las personas adultas y parece haber una relación entre la continuidad del trabajo durante la jubilación y la satisfacción con la vida. Este estudio evaluó la percepción de 230 jubilados que aun trabajan y su satisfacción con la vida. Los participantes respondieron a un cuestionario que contiene las escalas de Satisfacción con la Vida, Percepción del Trabajo y Razones para el Regreso al Trabajo. Las correlaciones y la regresión linear múltiple, demostraron que la percepción del trabajo, la satisfacción con la renta y las razones intrínsecas - como la sensación de la productividad - influyeron en la satisfacción con la vida de estos jubilados. Estos resultados refuerzan la necesidad de políticas públicas y organizacionales que permitan la permanencia de jubilados que deseen seguir trabajando

    Hepatitis C Virus Infection as a Traumatic Experience

    Objective The purpose of this study was to evaluate whether individuals consider their HCV infection to be a potentially traumatic experience. Additionally, we investigated its association with Post-Traumatic Stress Disorder (PTSD) and the impact of PTSD diagnosis on health-related quality of life (HRQoL) in HCV infected subjects. Methods We conducted a cross-sectional survey of 127 HCV-infected outpatients recruited at a University Hospital in Salvador, Brazil. All subjects answered an orally-administered questionnaire to gather clinical and socio-demographic data. We investigated traumatic experiences and the subject's perception of the disease using the Trauma History Questionnaire. PTSD and other psychiatric diagnoses were assessed through the Mini International Neuropsychiatric Interview-Brazilian Version 5.0.0 (M.I.N.I. PLUS). HRQoL was assessed using Short-Form 36 (SF-36). Results Approximately 38.6% of the patients considered hepatitis C to be a traumatic experience. Of these, 60.7% had a PTSD diagnosis. PTSD was associated with significant impairment in quality of life for individuals in seven SF-36 domains as shown bymultivariate analysis: Role-Physical (β: −24.85; 95% CI: −42.08; −7.61), Bodily Pain (β: −19.36; 95% CI: −31.28; −7.45), General Health (β: −20.79; 95% CI: −29.65; −11.92), Vitality (β: −11.92; 95% CI: −20.74; −3.1), Social Functioning (β: −34.73; 95% CI: −46.79; −22.68), Role-Emotional (β: −26.07; 95% CI: −44.61; −7.53), Mental Health (β: −17.46; 95% CI: −24.38; −10.54). Conclusion HCV is frequently a traumatic experience and it is strongly associated with PTSD diagnosis. PTSD significantly impaired HRQoL

    Preliminary biocompatibility investigation of magnetic albumin nanosphere designed as a potential versatile drug delivery system

    Background: The magnetic albumin nanosphere (MAN), encapsulating maghemite nanoparticles, was designed as a magnetic drug delivery system (MDDS) able to perform a variety of biomedical applications. It is noteworthy that MAN was efficient in treating Ehrlich’s tumors by the magnetohyperthermia procedure. Methods and materials: In this study, several nanotoxicity tests were systematically carried out in mice from 30 minutes until 30 days after MAN injection to investigate their biocompatibility status. Cytometry analysis, viability tests, micronucleus assay, and histological analysis were performed. Results: Cytometry analysis and viability tests revealed MAN promotes only slight and temporary alterations in the frequency of both leukocyte populations and viable peritoneal cells, respectively. Micronucleus assay showed absolutely no genotoxicity or cytotoxicity effects and histological analysis showed no alterations or even nanoparticle clusters in several investigated organs but, interestingly, revealed the presence of MAN clusters in the central nervous system (CNS). Conclusion: The results showed that MAN has desirable in vivo biocompatibility, presenting potential for use as a MDDS, especially in CNS disease therapy

    Pulmonary Inflammation Is Regulated by the Levels of the Vesicular Acetylcholine Transporter

    Acetylcholine (ACh) plays a crucial role in physiological responses of both the central and the peripheral nervous system. Moreover, ACh was described as an anti-inflammatory mediator involved in the suppression of exacerbated innate response and cytokine release in various organs. However, the specific contributions of endogenous release ACh for inflammatory responses in the lung are not well understood. To address this question we have used mice with reduced levels of the vesicular acetylcholine transporter (VAChT), a protein required for ACh storage in secretory vesicles. VAChT deficiency induced airway inflammation with enhanced TNF-alpha and IL-4 content, but not IL-6, IL-13 and IL-10 quantified by ELISA. Mice with decreased levels of VAChT presented increased collagen and elastic fibers deposition in airway walls which was consistent with an increase in inflammatory cells positive to MMP-9 and TIMP-1 in the lung. In vivo lung function evaluation showed airway hyperresponsiveness to methacholine in mutant mice. The expression of nuclear factor-kappa B (p65-NF-kappa B) in lung of VAChT-deficient mice were higher than in wild-type mice, whereas a decreased expression of janus-kinase 2 (JAK2) was observed in the lung of mutant animals. Our findings show the first evidence that cholinergic deficiency impaired lung function and produce local inflammation. Our data supports the notion that cholinergic system modulates airway inflammation by modulation of JAK2 and NF-kappa B pathway. We proposed that intact cholinergic pathway is necessary to maintain the lung homeostasis

    The Durie/Salmon staging system continues to be used worldwide in patients with multiple myeloma. However, in recent years, new systems have been proposed. The International Myeloma Working Group performed a retrospective study with 11,179 patients and proposed an "International Staging System" utilizing serum levels of â2 microglobulin and albumin. In addition, current research has focused on the usefulness of cytogenetic and molecular data as prognostic factors. These data suggest that these parameters are powerful discriminators of a poor prognostic group of myeloma patients. Indeed, these prognostic indexes have been utilized in clinical trials, with interesting and encouraging results.O esquema de Durie / Salmon continua a ser utilizado para estadiar os pacientes com mieloma múltiplo.     AC Biosusceptometry (ACB) was previously employed towards recording gastrointestinal motility. Our data show a reliable and successful evaluation of gastrointestinal transit of liquid and solid meals in rats, considering the methods scarcity and number of experiments needed to endorsement of drugs and medicinal plants.     A review of Cushing’s disease treatment by the Department of Neuroendocrinology of the Brazilian Society of Endocrinology and Metabolism

    The treatment objectives for a patient with Cushing’s disease (CD) are remission of hypercortisolism, adequate management of co-morbidities, restoration of the hypothalamic-pituitary-adrenal axis, preservation of fertility and pituitary function, and improvement of visual defects in cases of macroadenomas with suprasellar extension. Transsphenoidal pituitary surgery is the main treatment option for the majority of cases, even in macroadenomas with low probability of remission. In cases of surgical failure, another subsequent pituitary surgery might be indicated in cases with persistent tumor imaging at post surgical magnetic resonance imaging (MRI) and/or pathology analysis of adrenocorticotropic hormone-positive (ACTH+) positive pituitary adenoma in the first procedure. Medical treatment, radiotherapy and adrenalectomy are the other options when transsphenoidal pituitary surgery fails.There are several options of medical treatment, although cabergoline and ketoconazole are the most commonly used alone or in combination. Novel treatments are also addressed in this review. Different therapeutic approaches are frequently needed on an individual basis, both before and, particularly, after surgery, and they should be individualized. The objective of the present review is to provide the necessary information to achieve a more effective treatment for CD. It is recommended that patients with CD be followed at tertiary care centers with experience in treating this condition
