    Glial-Restricted Precursors Are Derived from Multipotent Neuroepithelial Stem Cells

    AbstractNeuroepithelial cells in the developing ventricular zone differentiate into neurons, astrocytes, and oligodendrocytes. It is not known, however, whether this differentiation occurs in a single step or is a pathway utilizing intermediate more restricted precursor cells. To characterize the generation of glial cells from multipotent stem cells we have cultured neuroepithelial (NEP) cells from E10.5 rat embryos. Cultured NEP cells do not express any glial differentiation markers when grown on fibronectin/laminin under nondifferentiation conditions. NEP cells, however, differentiate into A2B5 immunoreactive cells which can subsequently give rise to oligodendrocytes and astrocytes. Clonal analysis of NEP cells demonstrates that the A2B5 immunoreactive cells arise in clones that contain neurons and astrocytes, indicating that A2B5+cells arise from multipotent NEP precursor cells. A2B5+cells, maintained as undifferentiated cells over multiple passages, can subsequently give rise to both oligodendrocytes and astrocytes. A2B5+cells, however, do not generate neurons. Thus A2B5+cells represent a restricted progenitor cell population that differentiates from a multipotent NEP cell. Based on our results we propose that differentiation of the multipotential NEP cells to terminally differentiated glial cells occurs via intermediate restricted precursors

    Vibrio vulnificus MO6-24/O Lipopolysaccharide Stimulates Superoxide Anion, Thromboxane B\u3csub\u3e2\u3c/sub\u3e, Matrix Metalloproteinase-9, Cytokine and Chemokine Release by Rat Brain Microglia in Vitro

    Although human exposure to Gram-negative Vibrio vulnificus (V. vulnificus) lipopolysaccharide (LPS) has been reported to result in septic shock, its impact on the central nervous system’s innate immunity remains undetermined. The purpose of this study was to determine whether V. vulnificus MO6-24/O LPS might activate rat microglia in vitro and stimulate the release of superoxide anion (O2−), a reactive oxygen species known to cause oxidative stress and neuronal injury in vivo. Brain microglia were isolated from neonatal rats, and then treated with either V. vulnificus MO6-24/O LPS or Escherichia coli O26:B6 LPS for 17 hours in vitro. O2− was determined by cytochrome C reduction, and matrix metalloproteinase-2 (MMP-2) and MMP-9 by gelatinase zymography. Generation of cytokines tumor necrosis factor alpha (TNF-α), interleukin-1 alpha (IL-1α), IL-6, and transforming growth factor-beta 1 (TGF-ÎČ1), chemokines macrophage inflammatory protein (MIP-1α)/chemokine (C-C motif) ligand 3 (CCL3), MIP-2/chemokine (C-X-C motif) ligand 2 (CXCL2), monocyte chemotactic protein-1 (MCP-1)/CCL2, and cytokine-induced neutrophil chemoattractant-2alpha/beta (CINC-2α/ÎČ)/CXCL3, and brain-derived neurotrophic factor (BDNF), were determined by specific immunoassays. Priming of rat microglia by V. vulnificus MO6-24/O LPS in vitro yielded a bell-shaped dose-response curve for PMA (phorbol 12-myristate 13-acetate)-stimulated O2− generation: (1) 0.1–1 ng/mL V. vulnificus LPS enhanced O2− generation significantly but with limited inflammatory mediator generation; (2) 10–100 ng/mL V. vulnificus LPS maximized O2− generation with concomitant release of thromboxane B2 (TXB2), matrix metalloproteinase-9 (MMP-9), and several cytokines and chemokines; (3) 1000–100,000 ng/mL V. vulnificus LPS, with the exception of TXB2, yielded both attenuated O2− production, and a progressive decrease in MMP-9, cytokines and chemokines investigated. Thus concentration-dependent treatment of neonatal brain microglia with V. vulnificus MO6-24/O LPS resulted in a significant rise in O2− production, followed by a progressive decrease in O2− release, with concomitant release of lactic dehydrogenase (LDH), and generation of TXB2, MMP-9, cytokines and chemokines. We hypothesize that the inflammatory mediators investigated may be cytotoxic to microglia in vitro, by an as yet undetermined autocrine mechanism. Although V. vulnificus LPS was less potent than E. coli LPS in vitro, inflammatory mediator release by the former was clearly more efficacious. Finally, we hypothesize that should V. vulnificus LPS gain entry into the CNS, it would be possible that microglia might become activated, resulting in high levels of O2− as well as neuroinflammatory TXB2, MMP-9, cytokines and chemokines

    The rapidly shifting ideological terrain of computing has a profound impact on Social Informatics's critical and empirical analysis of computerization movements. As these movements incorporate many of the past critiques concerning social fit and situational context leveled against them by Social Informatics research, more subtle and more deeply ingrained modes of ideological practice have risen to support movements of computerization. Among these, the current emphasis on the promises of data and data analytics presents the most obvious ideological challenge. In order to reorient Social Informatics in relation to these new ideological challenges, Louis Althusser's theory of ideology is discussed, with its implications for Social Informatics considered. Among these implications, a changed relationship between Social Informatics's critical stance and its reliance on empirical methods is advanced. Addressed at a fundamental level, the practice of Social Informatics comes to be reoriented in a more distinctly reflective and ethical direction

    Diffuse inverse Compton and synchrotron emission from dark matter annihilations in galactic satellites

    Annihilating dark matter particles produce roughly as much power in electrons and positrons as in gamma ray photons. The charged particles lose essentially all of their energy to inverse Compton and synchrotron processes in the galactic environment. We discuss the diffuse signature of dark matter annihilations in satellites of the Milky Way (which may be optically dark with few or no stars), providing a tail of emission trailing the satellite in its orbit. Inverse Compton processes provide X-rays and gamma rays, and synchrotron emission at radio wavelengths might be seen. We discuss the possibility of detecting these signals with current and future observations, in particular EGRET and GLAST for the gamma rays.Comment: 13 pages, 5 figure

    Non-vacuum Solutions of Bianchi Type VI_0 Universe in f(R) Gravity

    In this paper, we solve the field equations in metric f(R) gravity for Bianchi type VI_0 spacetime and discuss evolution of the expanding universe. We find two types of non-vacuum solutions by taking isotropic and anisotropic fluids as the source of matter and dark energy. The physical behavior of these solutions is analyzed and compared in the future evolution with the help of some physical and geometrical parameters. It is concluded that in the presence of isotropic fluid, the model has singularity at t~=0\tilde{t}=0 and represents continuously expanding shearing universe currently entering into phantom phase. In anisotropic fluid, the model has no initial singularity and exhibits the uniform accelerating expansion.     We document the absence of carriage of Neisseria meningitidis W-135 of the sequence type 11 in returning pilgrims after the Hajj 2002. This finding contrasts with the 15% carriage rate we previously reported in pilgrims returning from the Hajj 2001. The epidemiology of carriage may be changing or may have been controlled by vaccination and a policy of administering antibiotics to pilgrims from countries with a high incidence of meningococcal disease

    Dark matter and Colliders searches in the MSSM

    We study the complementarity between dark matter experiments (direct detection and indirect detections) and accelerator facilities (the CERN LHC and a s=1\sqrt{s}= 1 TeV e+e−e^+e^- Linear Collider) in the framework of the constrained Minimal Supersymmetric Standard Model (MSSM). We show how non--universality in the scalar and gaugino sectors can affect the experimental prospects to discover the supersymmetric particles. The future experiments will cover a large part of the parameter space of the MSSM favored by WMAP constraint on the relic density, but there still exist some regions beyond reach for some extreme (fine tuned) values of the supersymmetric parameters. Whereas the Focus Point region characterized by heavy scalars will be easily probed by experiments searching for dark matter, the regions with heavy gauginos and light sfermions will be accessible more easily by collider experiments.     Using the large acceptance apparatus FOPI, we study pion emission in the reactions (energies in GeV/nucleon are given in parentheses): 40Ca+40Ca (0.4, 0.6, 0.8, 1.0, 1.5, 1.93), 96Ru+96Ru (0.4, 1.0, 1.5), 96Zr+96Zr (0.4, 1.0, 1.5), 197Au+197Au (0.4, 0.6, 0.8, 1.0, 1.2, 1.5). The observables include longitudinal and transverse rapidity distributions and stopping, polar anisotropies, pion multiplicities, transverse momentum spectra, ratios for positively and negatively charged pions of average transverse momenta and of yields, directed flow, elliptic flow.
