    The Sm-Nd isotopic method in the geochronology laboratory of the University of Brasília

    Nd isotopes represent one of the best tools to investigate the processes involved in the evolution of the continental crust and mantle. This is due mainly to the similar geochemical behaviour of Sm and Nd, both light rare earth elements, which inhibits their fractionation during most varied geological processes. In order to carry out crustal evolution studies in central Brazil, the Sm-Nd isotopic method was implanted at the Geochronology Laboratory of the University of Brasília. The Sm-Nd separation methodology is basically that described in Richard et al. (1976), with the addition of some improvements. In this study we describe in detail the methodology used in Brasília. Precision and accuracy were checked with the international standards such as JB-3, BCR-1, BHVO-1 and La Jolla, and the following results were found: JB-3 (Nd=15.74 ppm and Sm=4.28 ppm), BCR-1 (143Nd/144Nd=0.512647±8, Nd=28.73 ppm and Sm=6.66 ppm), BHVO-1 (Nd=24.83 ppm and Sm=6.2 ppm) and La Jolla (143Nd/144Nd=0.511835±14)

    Report from the multinational irritable bowel syndrome initiative 2012

    Q1Q1In 2012, a group of 29 internationally recognized experts in the pathophysiology, diagnosis, and treatment of irritable bowel syndrome (IBS) convened to audit the current state of IBS research. The meeting was preceded by a comprehensive online survey that focused on research needs for IBS diagnosis (particularly the strengths and shortcomings of current criteria), definitions used in clinical trials for IBS patients and “healthy controls,” potential biomarkers for IBS, and outcome measures in drug trials. While the purpose of the meeting was not to make binding recommendations, participants developed a framework for future questions and research needs in IBS. First, participants indicated the need for revised criteria for the diagnosis of IBS; in particular, inclusion of bloating and de-emphasis of pain as criteria were considered critical needs. Second, participants noted that definitions of normal, healthy controls varied widely among clinical trials; these definitions need to be standardized not only to improve the reliability of results, but also to better facilitate inter-trial comparisons and data synthesis. Third, participants highlighted the need for accurate biomarkers of disease. Fourth and finally, participants noted that further defining outcome measures, so that they are functionally relevant and reflect normalization of bowel function, is a critical need.     High density polyethylene and polystyrene microplastics as vectors of Triclosan towards marine invertebrates: signals of reduced bioreactivity

    Triclosan (TCS) is an emerging contaminant used as a preservative for its antifungal and bactericidal properties. Previous studies have showed different capacity of TCS sorption depending on the MP type, which was related to their physicochemical properties. As a part of the general objective to evaluate the capacity of polyethylene(PE) and polystyrene (PS) microplastics as TCS vectors in marine environments, the marine bivalve Mytilus galloprovincialis was used as a model organism to evaluate effects on immune function after in vivo co-exposure. Acute TCS spill scenario in the presence/absence of microplastics in the surrounding water was simulated under laboratory conditions. Mussels were simultaneously exposed to one type of weathered microplastics (PE or PS) and a high TCS concentration during 72 hours. In general, a decrease in lysosomal membrane stability was observed with all treatments compared to the control after three days, with most treatments resulting in moderate cell stress in mussels. A overall decrease in phagocytic efficiency was also observed in all treated mussels compared to the control mussels, the only exception being mussels individually exposed to PE, where a slight but not statistically significant increase was detected. No differences were observed between treatments in extracellular lysozyme activity and oxyradicals production. In general, mussels co-exposed to TCS+PE and TCS+PS showed effects on immune function that were lower than or comparable to those observed with individual exposures to TCS, PE and PS. Based on the results of this study, an additive effect on immune function derived from the co-exposure of TCS+PE and TCS+PS microplastics can be discarded. However, a weak antagonistic effect is suggested, potentially due to a reduction in their bioreactivity as a consequence of the adsorption of TCS to the surface of the ingested microplastics. The comparison of the results of bioaccumulation of TCS in mussels between the individual treatments and treatments with co-exposure to TCS, which are currently being analyzed, will help to strengthen or refute this hypothesis

    Complexity measures of heart-rate variability in amyotrophic lateral sclerosis with alternative pulmonary capacities

    Objective: the complexity of heart-rate variability (HRV) in amyotrophic lateral sclerosis (ALS) patients with different pulmonary capacities was evaluated. Methods: We set these according to their pulmonary capacity, and specifically forced vital capacity (FVC). We split the groups according to FVC (FVC > 50% (n = 29) and FVC < 50% (n = 28)). In ALS, the presence of an FVC below 50% is indicative of noninvasive ventilation with two pressure levels and with the absence of other respiratory symptoms. As the number of subjects per group was different, we applied the unbalanced one-way analysis of variance (uANOVA1) test after three tests of normality, and effect size by Cohen’s d to assess parameter significance. Results: with regard to chaotic global analysis, CFP4 (p < 0.001; d = 0.91), CFP5 (p = 0.0022; d = 0.85), and CFP6 (p = 0.0009; d = 0.92) were enlarged. All entropies significantly increased. Shannon (p = 0.0005; d = 0.98), Renyi (p = 0.0002; d = 1.02), Tsallis (p = 0.0004; d = 0.99), approximate (p = 0.0005; d = 0.97), and sample (p < 0.0001; d = 1.22). Detrended fluctuation analysis (DFA) (p = 0.0358) and Higuchi fractal dimension (HFD) (p = 0.15) were statistically inconsequential between the two groups. Conclusions: HRV complexity in ALS subjects with different pulmonary capacities increased via chaotic global analysis, especially CFP5 and 3 out of 5 entropies

    Decreased ATP production and myocardial contractile reserve in metabolic heart disease

    Available online 01 February 2018Metabolic syndrome is a cluster of obesity-related metabolic abnormalities that lead to metabolic heart disease (MHD) with left ventricular pump dysfunction. Although MHD is thought to be associated with myocardial energetic deficiency, two key questions have not been answered. First, it is not known whether there is a sufficient energy deficit to contribute to pump dysfunction. Second, the basis for the energy deficit is not clear. To address these questions, mice were fed a high fat, high sucrose (HFHS) 'Western' diet to recapitulate the MHD phenotype. In isolated beating hearts, we used 31P NMR spectroscopy with magnetization transfer to determine a) the concentrations of high energy phosphates ([ATP], [ADP], [PCr]), b) the free energy of ATP hydrolysis (∆G~ATP), c) the rate of ATP production and d) flux through the creatine kinase (CK) reaction. At the lowest workload, the diastolic pressure-volume relationship was shifted upward in HFHS hearts, indicative of diastolic dysfunction, whereas systolic function was preserved. At this workload, the rate of ATP synthesis was decreased in HFHS hearts, and was associated with decreases in both [PCr] and ∆G~ATP. Higher work demands unmasked the inability of HFHS hearts to increase systolic function and led to a further decrease in ∆G~ATP to a level that is not sufficient to maintain normal function of sarcoplasmic Ca2+-ATPase (SERCA). While [ATP] was preserved at all work demands in HFHS hearts, the progressive increase in [ADP] led to a decrease in ∆G~ATP with increased work demands. Surprisingly, CK flux, CK activity and total creatine were normal in HFHS hearts. These findings differ from dilated cardiomyopathy, in which the energetic deficiency is associated with decreases in CK flux, CK activity and total creatine. Thus, in HFHS-fed mice with MHD there is a distinct metabolic phenotype of the heart characterized by a decrease in ATP production that leads to a functionally-important energetic deficiency and an elevation of [ADP], with preservation of CK flux.Ivan Luptak, Aaron L.
