    Ellagic Acid Recovery by Solid State Fermentation of Pomegranate Wastes by Aspergillus niger and Saccharomyces cerevisiae : a Comparison

    Abstract: Fermentation in solid state culture (SSC) has been the focus of increasing interest because of its potential for industrial applications. In previous studies SSC of pomegranate wastes by Aspergillus niger has been extensively developed and optimized for the recovery of ellagic acid (EA), a high value bioactive. In this study we comparatively investigated the SSC of powdered pomegranate husks by A. niger and Saccharomyces cerevisiae and evaluated the recovery yields of EA by an ultrasound and microwave-assisted 7:3 water/ethanol extraction. Surprisingly enough, the yields obtained by S. cerevisiae fermentation (4% w/w) were found 5-fold higher than those of the A. niger fermented material, with a 10-fold increase with respect to the unfermented material. The EA origin was traced by HPLC analysis that showed a significant decrease in the levels of punicalagin isomers and granatin B and formation of punicalin following fermentation. Other extraction conditions that could warrant a complete solubilization of EA were evaluated. Using a 1:100 solid to solvent ratio and DMSO as the solvent, EA was obtained in 4% yields from S. cerevisiae fermented husks at a high purity degree. Hydrolytic treatment of S. cerevisiae fermented pomegranate husks aorded a material freed of the polysaccharides components that gave recovery yields of EA up to 12% w/w

    Open questions and research needs in the adoption of conservation agriculture in the mediterranean area

    This article aims to provide a review of major challenges and research needs for the diffusion of conservation agriculture (CA) and the improvement of crop–soil–water conditions in Southern Europe and Northern Africa. A multidisciplinary study and a participatory approach are at the basis of an international project of research and innovation action, “Research-based participatory approaches for adopting conservation agriculture in the Mediterranean Area-CAMA”. It aims to understand the reasons and the research needs that limit a large CA diffusion in the Mediterranean countries. CAMA aims to provide significant advances to CA through multidisciplinary research at the field and farm scales (with main emphasis on smallholder), encompassing a socio-economic analysis of the reasons that obstacle the CA diffusion, legume crop improvement as a component of improved CA cropping systems, and a network of long-term experiments on CA and soil characteristic modification.     Exploiting the Ref-1-APE1 node in cancer signaling and other diseases: from bench to clinic

    Reduction-oxidation factor 1-apurinic/apyrimidinic endonuclease (Ref-1/APE1) is a critical node in tumor cells, both as a redox regulator of transcription factor activation and as part of the DNA damage response. As a redox signaling protein, Ref-1/APE1 enhances the transcriptional activity of STAT3, HIF-1α, nuclear factor kappa B, and other transcription factors to promote growth, migration, and survival in tumor cells as well as inflammation and angiogenesis in the tumor microenvironment. Ref-1/APE1 is activated in a variety of cancers, including prostate, colon, pancreatic, ovarian, lung and leukemias, leading to increased aggressiveness. Transcription factors downstream of Ref-1/APE1 are key contributors to many cancers, and Ref-1/APE1 redox signaling inhibition slows growth and progression in a number of tumor types. Ref-1/APE1 inhibition is also highly effective when paired with other drugs, including standard-of-care therapies and therapies targeting pathways affected by Ref-1/APE1 redox signaling. Additionally, Ref-1/APE1 plays a role in a variety of other indications, such as retinopathy, inflammation, and neuropathy. In this review, we discuss the functional consequences of activation of the Ref-1/APE1 node in cancer and other diseases, as well as potential therapies targeting Ref-1/APE1 and related pathways in relevant diseases. APX3330, a novel oral anticancer agent and the first drug to target Ref-1/APE1 for cancer is entering clinical trials and will be explored in various cancers and other diseases bringing bench discoveries to the clinic

    Automatic model - based test case generation for uml diagrams using tree travelsal algorithm

    The foundation of any model-based testing (MBT) with Unified Modelling Language (UML) diagrams is test case generation (TCG) which predicts the expected functionalities of a system under test (SUT). However, problems associated with existing test case generation methods are lack of integration with various UML diagrams and tools, inability to cover all the model elements of UML diagrams, failure to generate comprehensive test cases based on adequate coverage criteria and lack of support tools for automatic generation of test cases. To address these challenges, efficient mapping strategies for model elements that engenders effective artefacts extraction and test case generation processes were proposed. The methodology employed in this research comprised constructing relevant models and algorithms as well as implementing with the use of Java programming language. Specifically, an enhanced elements mapper, artefacts extractor (parser) and test case generator were developed and integrated to produce the support tool. The elements mapper yielded an accuracy result of 99.31%. The artefacts extractor recorded 99.64% accuracy while the test case generator recorded 100% accuracy. The improved methods proved to be more robust and efficiently generated quality test cases with eliminated redundancies based on all the descriptive attributes of UML diagrams. Limitations of existing the methods were addressed in the proposed method which is able to integrate more diagrams to generate quality test cases
