A randomised open-label study of tiagabine given two or three times daily in refractory epilepsy
SummaryEfficacy and tolerability of tiagabine was evaluated in patients with non-controlled partial seizures in a multicentre, open-label, parallel group study. Tiagabine was administered either two (b.i.d.) or three times daily (t.i.d.) as adjunctive therapy and titrated stepwise to a target of 40mg/day during a 12-week, fixed-schedule titration period; this was followed by a 12-week flexible continuation period. The primary efficacy endpoint was the proportion of patients completing the fixed-schedule titration period. A total of 243 patients were randomised and received treatment, 123 to b.i.d. and 120 to t.i.d. dosing. Fewer patients in the b.i.d. (76 and 62%) than in the t.i.d. (87 and 72%) group completed the fixed-schedule titration period (OR: 0.562; 95% CI: 0.309â1.008; P=0.0532). The median percentage decrease in all types of seizure (excluding status epilepticus) during the fixed schedule titration period was 33.4% for the b.i.d. and 23.8% for the t.i.d. groups (P=0.9634; Van Elteren's test). The proportion of responders was similar for the b.i.d. and t.i.d. groups. There were no significant differences between dosage regimens in the change in median seizure rates from baseline. Adverse events were more frequent during the titration than the continuation period. Most events were mild and related to the central nervous system. Although their incidence was similar between treatment groups, severity was more frequent in the b.i.d. group. Our results suggest that during titration tiagabine is better tolerated with t.i.d. dosing, but during long-term maintenance, a t.i.d. schedule is as effective and well tolerated as b.i.d
Detecting metal-rich intermediate-age globular clusters in NGC4570 using K-band photometry

DOI: 10.1007/s10509-009-0093-8Globular cluster systems (GCSs) of most early-type galaxies feature two peaks in their optical colour distributions. Blue-peak globular clusters (GCs) are believed to be old and metal-poor, whereas the ages, metallicities, and the origin of the red-peak GCs are still being debated. We obtained deep K-band photometry and combined it with Hubble Space Telescope observations in g and z to yield a full spectral energy distribution from the optical to the near-infrared. This now allows us to break the ageâmetallicity degeneracy. We used our evolutionary synthesis models galev for star clusters to compute a large grid of models with different metallicities and a wide range of ages. Comparing these models to our observations revealed a large population of intermediate-age (1â3 Gyr) and metal-rich (âsolar-metallicity) GCs, that will give us further insights into the formation history of this galaxy.Peer reviewe
Toward a predictive understanding of earthâs microbiomes to address 21st century challenges
Microorganisms have shaped our planet and its inhabitants for over 3.5 billion years. Humankind has had a profound influence on the biosphere, manifested as global climate and land use changes, and extensive urbanization in response to a growing population. The challenges we face to supply food, energy, and clean water while maintaining and improving the health of our population and ecosystems are significant. Given the extensive influence of microorganisms across our biosphere, we propose that a coordinated, cross-disciplinary effort is required to understand, predict, and harness microbiome function. From the parallelization of gene function testing to precision manipulation of genes, communities, and model ecosystems and development of novel analytical and simulation approaches, we outline strategies to move microbiome research into an era of causality. These efforts will improve prediction of ecosystem response and enable the development of new, responsible, microbiome-based solutions to significant challenges of our time
The PL calibration for Milky Way Cepheids and its implications for the distance scale
The rationale behind recent calibrations of the Cepheid PL relation using the
Wesenheit formulation is reviewed and reanalyzed, and it is shown that recent
conclusions regarding a possible change in slope of the PL relation for
short-period and long-period Cepheids are tied to a pathological distribution
of HST calibrators within the instability strip. A recalibration of the
period-luminosity relation is obtained using Galactic Cepheids in open clusters
and groups, the resulting relationship, described by log L/L_sun =
2.415(+-0.035) + 1.148(+-0.044)log P, exhibiting only the moderate scatter
expected from color spread within the instability strip. The relationship is
confirmed by Cepheids with HST parallaxes, although without the need for
Lutz-Kelker corrections, and in general by Cepheids with revised Hipparcos
parallaxes, albeit with concerns about the cited precisions of the latter. A
Wesenheit formulation of Wv = -2.259(+-0.083) - 4.185(+-0.103)log P for
Galactic Cepheids is tested successfully using Cepheids in the inner regions of
the galaxy NGC 4258, confirming the independent geometrical distance
established for the galaxy from OH masers. Differences between the extinction
properties of interstellar and extragalactic dust may yet play an important
role in the further calibration of the Cepheid PL relation and its application

Climatic effects on sugarcane ripening under the influence of cultivars and crop age
The lack of information about the effects of cultivars, crop age and climate on the sugarcane (Saccharum ssp.) crop yield and quality has been the primary factor impacting the sugar-ethanol sector in Brazil. One of the processes about which we do not have a satisfactory understanding is sugarcane ripening and the effects of cultivars, crop age and climate on that. Sugarcane ripening is the process of sucrose accumulation in stalks, which is heavily influenced by several factors, mainly by climatic conditions such as air temperature and water deficits. Because it is a complex process, studies of the variables involved in sugarcane ripening can provide important information, resulting in a better use of commercial cultivars, bringing advantages to growers, processing units, breeding programs and scientific community. In this review, we discuss the available knowledge of the interaction between climate conditions and sugarcane ripening, under the influence of genotypic characteristics and crop age. In several studies, the main conclusion is that sugarcane ripening depends on a complex combination of climate variables, the genetic potential of cultivars and crop management. Soil moisture and air temperature are the primary variables involved in sugarcane ripening, and their combination stimulates the intensity of the process. In addition, the need for studies integrating the effects of climate on plant physiological processes and on the use of chemical agents to stimulate sugarcane ripening is highlighted
Rootâlesion nematodes of potato: current status of diagnostics, pathogenicity and management
Rootâlesion nematodes of the genus Pratylenchus are migratory endoparasites with worldwide economic impact on several important crops including potato, where certain species like P. penetrans, P. neglectus and P. scribneri reduce the yield and quality of potato tubers. Morphological identification of Pratylenchus spp. is challenging, and recent advancements in molecular techniques provide robust and rapid diagnostics to differentiate species without need of specialist skills. However, the fact that molecular diagnostics are not available for all Pratylenchus species means that there are limitations in worldwide application. In general, rootâlesion nematodes are difficult to manage once introduced into agricultural land and damage can be related to pathogenicity and population densities. In addition, rootâlesion nematodes interact with fungi such as V. dahliae, resulting in disease complexes that enhance the damage inflicted on the potato crop. Management interventions are often focused on limiting nematode reproduction before planting crops and include the application of nematicides, and cultural practices such as crop rotation, cover crops, biofumigation, and biological control. Understanding the limitations of the available crop protection strategies is important and there are many gaps for further study. This review discusses the status of the diagnosis, distribution, pathogenicity and management of the main species of rootâlesion nematodes, reported to infect potatoes worldwide, and highlights areas for potential future research
Star clusters near and far; tracing star formation across cosmic time

In this review, we will first focus on their constituents, i.e.\ detailed insight into their stellar populations and their surrounding ionised, warm, neutral, and molecular gas. We, then, move beyond the Local Group to review star cluster populations at various evolutionary stages, and in diverse galactic environmental conditions accessible in the local Universe. At high redshift, where conditions for cluster formation and evolution are more extreme, we are only able to observe the integrated light of a handful of objects that we believe will become globular clusters. We therefore discuss how numerical and analytical methods, informed by the observed properties of cluster populations in the local Universe, are used to develop sophisticated simulations potentially capable of disentangling the genetic map of galaxy formation and assembly that is carried by globular cluster populations.Peer reviewedFinal Accepted Versio
