    SummaryObjectiveC–C chemokine receptor type 5 (CCR5) has been implicated in rheumatoid arthritis and several inflammatory diseases, where its blockade resulted in reduced joint destruction. However, its role in modulating cartilage and bone changes in post-traumatic osteoarthritis (OA) has not yet been investigated. In this study, we investigated changes in articular cartilage, synovium and bone in a post-traumatic OA model using CCR5-deficient (CCR5−/−) mice.MethodDestabilization of the medial meniscus (DMM) was performed on the right knee of 10-week old CCR5−/− and C57BL/6J wild-type (WT) mice to induce post-traumatic OA. The contralateral left knee served as sham-operated control. Knee joints were analyzed at 4-, 8- and 12-weeks after surgery to evaluate cartilage degeneration and synovitis by histology, and bone changes via micro-CT.ResultsOur findings showed that CCR5−/− mice exhibited significantly less cartilage degeneration than WT mice at 8- and 12-weeks post-surgery. CCR5−/− mice showed some altered bone parameters 18- and 22-weeks of age, but body size and weight were not affected. The effect of CCR5-ablation was insignificant at all time points post-surgery for synovitis and for bone parameters such as bone volume/total volume, connectivity density index (CDI), structure model index (SMI), subchondral bone plate thickness, and trabecular bone number, thickness and spacing.ConclusionThese findings suggest that CCR5−/− mice developed less cartilage degeneration, which may indicate a potential protective role of CCR5-ablation in cartilage homeostasis. There were no differences in bone or synovial response to surgery suggesting that CCR5 functions primarily in cartilage during the development of post-traumatic OA

    A total of 39 organic faba bean and 59 pea seed lots obtained during 2009-2011 from 32 organic farms throughout Germany were assessed for seed-borne fungi. Seed samples collected from the harvested fields grown from these seed lots were also assessed. The focus in the pathogen identification was on the ascochyta blight complex pathogens Ascochyta pisi, Mycosphaerella pinodes and Phoma medicaginis in pea and A. fabae and P. medicaginis in faba bean and on Fusarium spp. Fusarium spp. were found only rarely on peas with 40 of the seed samples and 44 of the harvested samples free of Fusarium spp. and the maximum infestation rate of 12 % in one harvested sample. Eighteen of the sown and 40 of the harvested lots had infestation rates of > 10 % with the Ascochyta complex pathogens. Several seed samples had infection levels over 50 %. Only 9 out of 39 faba bean seed lots and 4 out of the 36 harvested samples were free of Ascochyta blight and Fusarium spp. pathogens. Ten and 20 % of the sown and harvested seed samples had infection levels > 10 % with a given pathogen. In most cases, the faba bean seeds were infected with one pathogen only, however, mixed infections occurred. Overall, highly infested seed lots resulted in highly infested harvest lots. However, harvests were often also infested when they originated from clean seed thus pointing to the importance of local soil borne and air borne infections

    Permutable subnormal subgroups of finite groups

    The aim of this paper is to prove certain characterization theorems for groups in which permutability is a transitive relation, the so called PT -groups. In particular, it is shown that the finite solvable PT -groups, the finite solvable groups in which every subnormal subgroup of defect two is permutable, the finite solvable groups in which every normal subgroup is permutable sensitive, and the finite solvable groups in which conjugate-permutability and permutability coincide are all one and the same class. This follows from our main result which says that the finite modular p-groups, p a prime, are those p-groups in which every subnormal subgroup of defect two is permutable or, equivalently, in which every normal subgroup is permutable sensitive. However, there exist finite insolvable groups which are not PT -groups but all subnormal subgroups of defect two are permutable

    The nuclear factor of activated T cells (NFAT) family of transcription factors controls calcium signaling in T lymphocytes. In this study, we have identified a crucial regulatory role of the transcription factor NFATc2 in T cell-dependent experimental colitis. Similar to ulcerative colitis in humans, the expression of NFATc2 was up-regulated in oxazolone-induced chronic intestinal inflammation. Furthermore, NFATc2 deficiency suppressed colitis induced by oxazolone administration. This finding was associated with enhanced T cell apoptosis in the lamina propria and strikingly reduced production of IL-6, -13, and -17 by mucosal T lymphocytes. Further studies using knockout mice showed that IL-6, rather than IL-23 and -17, are essential for oxazolone colitis induction. Administration of hyper-IL-6 blocked the protective effects of NFATc2 deficiency in experimental colitis, suggesting that IL-6 signal transduction plays a major pathogenic role in vivo. Finally, adoptive transfer of IL-6 and wild-type T cells demonstrated that oxazolone colitis is critically dependent on IL-6 production by T cells. Collectively, these results define a unique regulatory role for NFATc2 in colitis by controlling mucosal T cell activation in an IL-6-dependent manner. NFATc2 in T cells thus emerges as a potentially new therapeutic target for inflammatory bowel diseases

    Gauge Orbit Types for Theories with Classical Compact Gauge Group

    We determine the orbit types of the action of the group of local gauge transformations on the space of connections in a principal bundle with structure group O(n), SO(n) or Sp(n)Sp(n) over a closed, simply connected manifold of dimension 4. Complemented with earlier results on U(n) and SU(n) this completes the classification of the orbit types for all classical compact gauge groups over such space-time manifolds. On the way we derive the classification of principal bundles with structure group SO(n) over these manifolds and the Howe subgroups of SO(n).Comment: 57 page

    CCD BV and 2MASS photometric study of the open cluster NGC 1513

    We present CCD BV and JHKs_{s} 2MASS photometric data for the open cluster NGC 1513. We observed 609 stars in the direction of the cluster up to a limiting magnitude of V19V\sim19 mag. The star count method shows that the centre of the cluster lies at α2000=04h09m36s\alpha_{2000}=04^{h}09^{m}36^{s}, δ2000=492843\delta_{2000}=49^{\circ}28^{'}43^{''} and its angular size is r=10r=10 arcmin. The optical and near-infrared two-colour diagrams reveal the colour excesses in the direction of the cluster as E(BV)=0.68±0.06E(B-V)=0.68\pm0.06, E(JH)=0.21±0.02E(J-H)=0.21\pm0.02 and E(JKs)=0.33±0.04E(J-K_{s})=0.33\pm0.04 mag. These results are consistent with normal interstellar extinction values. Optical and near-infrared Zero Age Main-Sequences (ZAMS) provided an average distance modulus of (mM)0=10.80±0.13(m-M)_{0}=10.80\pm0.13 mag, which can be translated into a distance of 1440±801440\pm80 pc. Finally, using Padova isochrones we determined the metallicity and age of the cluster as Z=0.015±0.004Z=0.015\pm 0.004 ([M/H]=0.10±0.10[M/H]=-0.10 \pm 0.10 dex) and log(t/yr)=8.40±0.04\log (t/yr) = 8.40\pm0.04, respectively.Comment: 15 pages, 12 figures and 4 tables, accepted for publication in Astrophysics & Space Scienc

    Prototyping the Semantics of a DSL using ASF+SDF: Link to Formal Verification of DSL Models

    A formal definition of the semantics of a domain-specific language (DSL) is a key prerequisite for the verification of the correctness of models specified using such a DSL and of transformations applied to these models. For this reason, we implemented a prototype of the semantics of a DSL for the specification of systems consisting of concurrent, communicating objects. Using this prototype, models specified in the DSL can be transformed to labeled transition systems (LTS). This approach of transforming models to LTSs allows us to apply existing tools for visualization and verification to models with little or no further effort. The prototype is implemented using the ASF+SDF Meta-Environment, an IDE for the algebraic specification language ASF+SDF, which offers efficient execution of the transformation as well as the ability to read models and produce LTSs without any additional pre or post processing.Comment: In Proceedings AMMSE 2011, arXiv:1106.596

    Finite element simulation of three-dimensional free-surface flow problems

    An adaptive finite element algorithm is described for the stable solution of three-dimensional free-surface-flow problems based primarily on the use of node movement. The algorithm also includes a discrete remeshing procedure which enhances its accuracy and robustness. The spatial discretisation allows an isoparametric piecewise-quadratic approximation of the domain geometry for accurate resolution of the curved free surface. The technique is illustrated through an implementation for surface-tension-dominated viscous flows modelled in terms of the Stokes equations with suitable boundary conditions on the deforming free surface. Two three-dimensional test problems are used to demonstrate the performance of the method: a liquid bridge problem and the formation of a fluid droplet

    Graded Poisson-Sigma Models and Dilaton-Deformed 2D Supergravity Algebra

    Fermionic extensions of generic 2d gravity theories obtained from the graded Poisson-Sigma model (gPSM) approach show a large degree of ambiguity. In addition, obstructions may reduce the allowed range of fields as given by the bosonic theory, or even prohibit any extension in certain cases. In our present work we relate the finite W-algebras inherent in the gPSM algebra of constraints to algebras which can be interpreted as supergravities in the usual sense (Neuveu-Schwarz or Ramond algebras resp.), deformed by the presence of the dilaton field. With very straightforward and natural assumptions on them --like demanding rigid supersymmetry in a certain flat limit, or linking the anti-commutator of certain fermionic charges to the Hamiltonian constraint-- in the ``genuine'' supergravity obtained in this way the ambiguities disappear, as well as the obstructions referred to above. Thus all especially interesting bosonic models (spherically reduced gravity, the Jackiw-Teitelboim model etc.)\ under these conditions possess a unique fermionic extension and are free from new singularities. The superspace supergravity model of Howe is found as a special case of this supergravity action.
