This publication was originally a product of The Regional Initiative, a 1992-1995special project cosponsored by the Council on Foundations and twenty-four of the nation's regional associations of grantmakers (RAGs). The purpose of the Initiative was to enhance the capacity of regional associations to meet their members' needs, by building both management and program effectiveness. The Initiative's long-term goal was to strengthen regional associations as agents of organized philanthropy in American life

Accuracy of the Omron HBF-500 Body Composition Monitor
The Omron HBF-500 is a relatively new and inexpensive body composition monitor that incorporates both hand-to-hand and foot-to-foot electrical impedance technology. At this time, the authors are not aware of any studies examining the accuracy of this monitor. PURPOSE: To assess the accuracy of the Omron HBF-500 body composition monitor using the BOD POD as a criterion. METHODS: Sixteen men and 23 women signed an informed consent and participated in the study (22.7±3.7 years, 168.8±9.1 cm, 73.8±17.8 kg, 25.7±5.6 kg.m-2). Participants were asked to refrain from exercise and caffeine on the day of testing, and were asked not eat a heavy meal three hours prior and to remain normally hydrated. Participants removed all jewelry and garments down to tight fitting clothing (swim suits or running tights) and were assessed on the BOD POD and Omron according to manufacturer’s guidelines. RESULTS: The Omron HBF-500 significantly overestimated percent body fat (%BF) by 3.5% compared to the BOD POD (31.7±9.6% and 28.6±9.6%, respectively), t(38) = -6.05, p = 0.001. The Omron HBF-500 also measured weight significantly higher than the BODPOD (74.4±17.7kg and 73.8±17.8kg, respectively), t(38) = -11.2, p = 0.001. Finally, the Omron HBF-500 was significantly correlated with the BOD POD when assessing body fat, r = .93. CONCLUSIONS: Caution should be taken when using the Omron HBF-500 as a measure of body fat. Those who choose to use this instrument should be aware the 3.5% higher reading compared to the BOD POD
Relation of own growth of sires of bulls to sons in progeny test stations
Abstract: Live weights and daily gains of 8 243 performance tested young sires of Czech Fleckvieh (dual purpose) till the age 400 days were analysed using Random Regression (RR) and Single Trait Animal Models. Evaluations were for entire period and for daily gains in various consecutive monthly intervals. Systematic environmental effects explained a higher proportion of variability in the RR for gains in short consecutive intervals (GSCI) than for other definitions of growth. The expected average reliability of estimated Breeding Values (BV) of young animals was similar for all methods from 0.42 to 0.46, but the rankings of animals differed. Determination (r 2 ) of BV between methods ranged from 0.64 to 0.94. Within-method correlations of BV of parents with progeny according the data from performance-test stations were highest for the GSCI. Correlations of BV of sires for own growth at performance-test stations with the BV of net gain of groups of progeny at progeny-test stations with the final live weight around 600 kg and age 500 days were according the method of sires evaluation from 0.26 to 0.38. Preferred method was according GSCI. This method also allows include more data in the evaluations, and separate the growth curve into genetic and non-genetic parts
Predicting Phenotypic Diversity and the Underlying Quantitative Molecular Transitions
During development, signaling networks control the formation of multicellular patterns. To what extent quantitative fluctuations in these complex networks may affect multicellular phenotype remains unclear. Here, we describe a computational approach to predict and analyze the phenotypic diversity that is accessible to a developmental signaling network. Applying this framework to vulval development in C. elegans, we demonstrate that quantitative changes in the regulatory network can render ~500 multicellular phenotypes. This phenotypic capacity is an order-of-magnitude below the theoretical upper limit for this system but yet is large enough to demonstrate that the system is not restricted to a select few outcomes. Using metrics to gauge the robustness of these phenotypes to parameter perturbations, we identify a select subset of novel phenotypes that are the most promising for experimental validation. In addition, our model calculations provide a layout of these phenotypes in network parameter space. Analyzing this landscape of multicellular phenotypes yielded two significant insights. First, we show that experimentally well-established mutant phenotypes may be rendered using non-canonical network perturbations. Second, we show that the predicted multicellular patterns include not only those observed in C. elegans, but also those occurring exclusively in other species of the Caenorhabditis genus. This result demonstrates that quantitative diversification of a common regulatory network is indeed demonstrably sufficient to generate the phenotypic differences observed across three major species within the Caenorhabditis genus. Using our computational framework, we systematically identify the quantitative changes that may have occurred in the regulatory network during the evolution of these species. Our model predictions show that significant phenotypic diversity may be sampled through quantitative variations in the regulatory network without overhauling the core network architecture. Furthermore, by comparing the predicted landscape of phenotypes to multicellular patterns that have been experimentally observed across multiple species, we systematically trace the quantitative regulatory changes that may have occurred during the evolution of the Caenorhabditis genus
Molecular architecture of Streptococcus pneumoniae surface thioredoxin-fold lipoproteins crucial for extracellular oxidative stress resistance and maintenance of virulence.
19 pags, 8 figs, tabsThe respiratory pathogen Streptococcus pneumoniae has evolved efficient mechanisms to resist oxidative stress conditions and to displace other bacteria in the nasopharynx. Here we character ize at physiological, functional and structural levels two novel surface-exposed thioredoxin-family lipoproteins, Etrx1 and Etrx2. The impact of both Etrx proteins and their r edox partner methionine sulfoxide reductase SpMsrAB2 on pneumococcal pathogenesis was assessed in mouse virulence studies and phagocytosis assays. The results demonstrate that loss of function of either both Etrx proteins or SpMsrAB2 dramatically attenuated pneumococcal virulence in the acute mouse pneumonia model and that Etrx proteins compensate each other. The deficiency of Etrx proteins or SpMsrAB2 further enhanced bacterial uptake by macrophages, and accelerated pneumococcal killing by H2O2 or free methionine sulfoxides (MetSO). Moreover, the absence of both Etrx redox pathways provokes an accumulation of oxidized SpMsrAB2 in vivo. Taken together our results reveal insights into the role of two extracellular electron pathways required for reduction of SpMsrAB2 and surface-exposed MetSO. Identification of biomarkers indicative of barotrauma and recovery in black rockfish Sebastes melanops
A Sebastes-specific complementary DNA (cDNA) microarray was developed to identify potential biomarkers involved in the capture stress and recovery of Sebastes species if they are assisted in returning to their original depth of capture following barotrauma. Black rockfish Sebastes melanops were exposed to simulated decompression from 450 kPa (c. 35 m depth) (which resulted in barotrauma) and subsequent recompression. Sebastes melanops were sampled for liver tissue at days 3, 15 and 31 post-barotrauma. Potential candidate genes were identified from the microarray and then quantitative real-time PCR (qrt-PCR) was used to validate expression levels in biological replicates. Six potential biomarkers associated with the innate immune system were identified that were up-regulated in liver tissue at 3 days post-barotrauma: complement C1q-like protein 2, complement component C3, complement regulatory plasma protein, serum amyloid A-5, c-type lysozyme and hepcidin precursor type I. In addition, complement c1q was correlated to the presence of a ruptured swimbladder, providing further support that this gene may be a good biomarker of injury and recovery. Immune genes were no longer up-regulated at day 31 post-barotrauma, a good indication of recovery in S. melanops.Keywords: microarray, gene expression, non-model organism, fis
Impurity generation during intense lower hybrid heating experiments on the Alcator C tokamak
and disposal, in whole or in part by or for the United States govern-ment is permitted. Experiments are underway on the Alcator C Tokamak with over 1 MW of RF power injected into the plasma at a frequency of 4.6 GHz to study both heating and current drive effects. During these studies, impurity genera-tion from limiter structures has been observed. The RF induced impurity influx is a strongly nonlinear function of net injected power. For Prf < 500 kW, only small effects are seen. As Prf approaches 1 MW, however, sharp increases in impurity influxes and Zeff are observed. Three different lim-iter materials have been used during these studies: molybdenum, graphite, and silicon-carbide coated graphite. In each case, the materials of the limiter structure are seen to dominate the increased impurity influx. In a typical case, with Prf = 1.0 MW e = 1.3 x 1014 cm- 3, and the SiC coated limiters, Zeff is seen to increase from 1.5 before the RF pulse to about 4 during the heating. At the same time, central Te increases from 2000 eV to 3000 eV and central Ti from 1200 eV to 1800 eV. Similar effects are seen in both H2 and D2 working gas discharges. The contribution to impurity genera-tion of nonthermal electrons, which are produced by the RF, is under investi-gation. Changes in edge plasma temperature and density, as well as the possibility that the particle transport is affected by the RF, are also being examined. Results of the experiments with the three different limiter materials are compared, and contributions of impurity radiation to the overall power balance are estimated

Recovery potential of black rockfish, Sebastes melanops Girard, recompressed following barotrauma
Overfished species of rockfish, Sebastes spp., from the Northeast Pacific experience high bycatch mortality because of ‘barotrauma’, a condition induced from the rapid change in pressure during capture. Field experiments show that it may be possible for rockfish to recover from barotrauma if quickly recompressed; however, no work has followed the physiological recovery of rockfish after recompression or determined whether it is possible for rockfish to survive barotrauma in the long term. Barotrauma was induced in adult black rockfish, Sebastes melanops Girard, from a simulated depth of 35 m, followed by recompression. Blood and selected tissues (eye, heart ventricle, head kidney, liver, rete mirabile and gonad) were sampled at days 3, 15 and 31 post-recompression to evaluate the tissue- and physiologic-level response during recovery. No mortality from barotrauma occurred during the experiments, and feeding resumed in 80% of both treatment and control fish. The primary injury in treatment fish was the presence of a ruptured swimbladder and/or a ruptured tunica externa (outer layer of swimbladder), which was slow to heal. Blood plasma was analysed for glucose, sodium, chloride, potassium, calcium, phosphorus, insulin-like growth factor-1 and cortisol. Plasma analyses indicated no strong effects because of barotrauma, suggesting overall handling stress outweighed any effect from barotrauma. Rockfish with ruptured swimbladders may face compromised competency in the wild; however, it appears the majority of black rockfish decompressed from 35 m have a high potential for recovery if recompressed immediately after capture. This research suggests recompression could be a valuable bycatch mortality reduction tool for rockfish in recreational fisheries.Keywords: recompression, barotrauma, histology, rockfish, decompressio
