Rice (Oryza sativa L.) can be divided into two major categories, which is upland rice and lowland rice. Apart from being the staple food for more than half of the world population, it is also known as model plant for functional genomics study. However, it possess high amount of starch and polysaccharide that makes the isolation of good quality RNA for downstream purposes often a difficult task. While there are many studies being carried out for lowland rice extraction, none has been reported for upland rice. This study is the first to report on evaluation of three RNA extraction methods for three Malaysian upland rice cultivars in order to determine the best method to isolate high grade RNA. The result obtained demonstrated that good quality RNA in terms of integrity, purity and quality can be isolated from young leaves of these cultivars by using guanidine isothiocyanate based extraction method that is fast, simple and efficient and had been proven suitable for further downstream applications

Rice is one of the most important species in the family of Poaceae. As one of the major crop that is consumed by world population, it is cultivated commercially in many parts of the world. Hence, the phylogeny study of this crop is crucial as a step for improvement of its breeding programs. Phylogenetic relationship among 12 rice cultivars that originated from two common sub-species; Indica and Japonica were inferred by comparing protein sequence data sets derived from its flowering time gene, namely RFT1 and analyzed using maximum parsimony (MP) method. The predicted structure of RFT1 protein was generated by I-TASSER server and analyzed using YASARA software. The result showed that the cultivars were classified into two major groups, where the first group (Japonica) evolved first followed by the second group (Indica). The findings suggested that some cultivars had a close relationship with each other even it is originates from different varieties. The relationships among these cultivars provide useful information for better understanding of molecular evolution process and designing good breeding program in order to generate new cultivar

Investigation of Non-linear Chirp Coding for Improved Second Harmonic Pulse Compression

    Get PDF
Non-linear frequency-modulated (NLFM) chirp coding was investigated to improve the pulse compression of the second harmonic chirp signal by reducing the range side lobe level. The problem of spectral overlap between the fundamental component and second harmonic component (SHC) was also investigated. Therefore, two methods were proposed: method I for the non-overlap condition and method II with the pulse inversion technique for the overlap harmonic condition. In both methods, the performance of the NLFM chirp was compared with that of the reference LFM chirp signals. Experiments were performed using a 2.25 MHz transducer mounted coaxially at a distance of 5 cm with a 1 mm hydrophone in a water tank, and the peak negative pressure of 300 kPa was set at the receiver. Both simulations and experimental results revealed that the peak side lobe level (PSL) of the compressed SHC of the NLFM chirp was improved by at least 13 dB in method I and 5 dB in method II when compared with the PSL of LFM chirps. Similarly, the integrated side lobe level (ISL) of the compressed SHC of the NLFM chirp was improved by at least 8 dB when compared with the ISL of LFM chirps. In both methods, the axial main lobe width of the compressed NLFM chirp was comparable to that of the LFM signals. The signal-to-noise ratio of the SHC of NLFM was improved by as much as 0.8 dB, when compared with the SHC of the LFM signal having the same energy level. The results also revealed the robustness of the NLFM chirp under a frequency-dependent attenuation of 0.5 dB/cm·MHz up to a penetration depth of 5 cm and a Doppler shift up to 12 kHz

Inhibiting ERK Activation with CI-1040 Leads to Compensatory Upregulation of Alternate MAPKs and Plasminogen Activator Inhibitor-1 following Subtotal Nephrectomy with No Impact on Kidney Fibrosis

Extracellular-signal regulated kinase (ERK) activation by MEK plays a key role in many of the cellular processes that underlie progressive kidney fibrosis including cell proliferation, apoptosis and transforming growth factor β1-mediated epithelial to mesenchymal transition. We therefore assessed the therapeutic impact of ERK1/2 inhibition using a MEK inhibitor in the rat 5/6 subtotal nephrectomy (SNx) model of kidney fibrosis. There was a twentyfold upregulation in phospho-ERK1/2 expression in the kidney after SNx in Male Wistar rats. Rats undergoing SNx became hypertensive, proteinuric and developed progressive kidney failure with reduced creatinine clearance. Treatment with the MEK inhibitor, CI-1040 abolished phospho- ERK1/2 expression in kidney tissue and prevented phospho-ERK1/2 expression in peripheral lymphocytes during the entire course of therapy. CI-1040 had no impact on creatinine clearance, proteinuria, glomerular and tubular fibrosis, and α-smooth muscle actin expression. However, inhibition of ERK1/2 activation led to significant compensatory upregulation of the MAP kinases, p38 and JNK in kidney tissue. CI-1040 also increased the expression of plasminogen activator inhibitor-1 (PAI-1), a key inhibitor of plasmin-dependent matrix metalloproteinases. Thus inhibition of ERK1/2 activation has no therapeutic effect on kidney fibrosis in SNx possibly due to increased compensatory activation of the p38 and JNK signalling pathways with subsequent upregulation of PAI-1

Plastic waste is one type of waste that would disturb the environment if it is not handled properly. Alternative treatment to solve this problem is by using pyrolysis technology that enable to convert plastic waste into fuel oil, charcoal, and non-condensable gas. The success of pyrolysis technology is influenced by several factors, including the type of reactor, the particle size, the processing time, and the temperature of the pyrolysis process. The purpose of this study was to determine the effect of temperature on the yield and the calorific value of plastic pyrolysis oil. Based on the experiments, increasing the temperature, the yield of the oil produced and calorific value will be higher. The yield and the highest calorific value generated for the pyrolysis process at a temperature of 400 0C, are 44% and 10,292 cal/ g respectively
