    Interplay of seismic and aseismic deformations during earthquake swarms: An experimental approach

    Observations of earthquake swarms and slow propagating ruptures on related faults suggest a close relation between the two phenomena. Earthquakes are the signature of fast unstable ruptures initiated on localized asperities while slow aseismic deformations are experienced on large stable segments of the fault plane. The spatial proximity and the temporal coincidence of both fault mechanical responses highlight the variability of fault rheology. However, the mechanism relating earthquakes and aseismic processes is still elusive due to the difficulty of imaging these phenomena of large spatiotemporal variability at depth. Here we present laboratory experiments that explore, in great detail, the deformation processes of heterogeneous interfaces in the brittle-creep regime. We track the evolution of an interfacial crack over 7 orders of magnitude in time and 5 orders of magnitude in space using optical and acoustic sensors. We explore the response of the system to slow transient loads and show that slow deformation episodes are systematically accompanied by acoustic emissions due to local fracture energy disorder. Features of acoustic emission activities and deformation rate distributions of our experimental system are similar to those in natural faults. On the basis of an activation energy model, we link our results to the Rate and State friction model and suggest an active role of local creep deformation in driving the seismic activity of earthquake swarms

    A phosphorylation map of the bovine papillomavirus E1 helicase

    BACKGROUND: Papillomaviruses undergo a complex life cycle requiring regulated DNA replication. The papillomavirus E1 helicase is essential for viral DNA replication and plays a key role in controlling viral genome copy number. The E1 helicase is regulated at least in part by protein phosphorylation, however no systematic approach to phosphate site mapping has been attempted. We have utilized mass spectrometry of purified bovine papillomavirus E1 protein to identify and characterize new sites of phosphorylation. RESULTS: Mass spectrometry and in silico sequence analysis were used to identify phosphate sites on the BPV E1 protein and kinases that may recognize these sites. Five new and two previously known phosphorylation sites were identified. A phosphate site map was created and used to develop a general model for the role of phosphorylation in E1 function. CONCLUSION: Mass spectrometric analysis identified seven phosphorylated amino acids on the BPV E1 protein. Taken with three previously identified sites, there are at least ten phosphoamino acids on BPV E1. A number of kinases were identified by sequence analysis that could potentially phosphorylate E1 at the identified positions. Several of these kinases have known roles in regulating cell cycle progression. A BPV E1 phosphate map and a discussion of the possible role of phosphorylation in E1 function are presented

    Development of novel chitosan / guar gum inks for extrusion-based 3D bioprinting: process, printability and properties

    The major limitation of 3D bioprinting is the availability of inks. In order to develop new ink formulations, both their rheological behavior to obtain the best printability and the target bio-printed objects conformities must be studied. In this paper, for the first time in our knowledge, the preparation and the characterization of novel ink formulations based on two natural biocompatible polysaccharides, chitosan (CH) and guar gum (GG), are presented. Five ink formulations containing different proportions of CH and GG were prepared and characterized in terms of rheological properties and solvent evaporation. Their printability was assessed (by varying the nozzle diameter, pressure and speed) using an extrusion-based 3D bioprinting process performed directly in air at 37 °C. Results showed that the incorporation of GG improved both the printability of the pure chitosan ink by increasing the viscosity of the solution and the shape fidelity by accelerating the solvent evaporation. We showed that the ink containing 15% (w/w) of GG and 85% (w/w) of CH had the best printability. This formulation was therefore used for the preparation of membranes that were characterized by infrared spectroscopy (FTIR) and X-Ray Diffraction (XRD) before and after gelation as well as for their mechanical properties (Young modulus, strength and strain at break). The optimal process printing parameters were determined to be: 27 G micronozzle, extrusion pressure below 2 bars and robot head speed between 20 and 25 mm/s.     Heterogeneities in leishmania infantum infection : using skin parasite burdens to identify highly infectious dogs

    Background: The relationships between heterogeneities in host infection and infectiousness (transmission to arthropod vectors) can provide important insights for disease management. Here, we quantify heterogeneities in Leishmania infantum parasite numbers in reservoir and non-reservoir host populations, and relate this to their infectiousness during natural infection. Tissue parasite number was evaluated as a potential surrogate marker of host transmission potential. Methods: Parasite numbers were measured by qPCR in bone marrow and ear skin biopsies of 82 dogs and 34 crab-eating foxes collected during a longitudinal study in Amazon Brazil, for which previous data was available on infectiousness (by xenodiagnosis) and severity of infection. Results: Parasite numbers were highly aggregated both between samples and between individuals. In dogs, total parasite abundance and relative numbers in ear skin compared to bone marrow increased with the duration and severity of infection. Infectiousness to the sandfly vector was associated with high parasite numbers; parasite number in skin was the best predictor of being infectious. Crab-eating foxes, which typically present asymptomatic infection and are non-infectious, had parasite numbers comparable to those of non-infectious dogs. Conclusions: Skin parasite number provides an indirect marker of infectiousness, and could allow targeted control particularly of highly infectious dogs

    Disturbance of deep-sea environments induced by the M9.0 Tohoku Earthquake

    The impacts of the M9.0 Tohoku Earthquake on deep-sea environment were investigated 36 and 98 days after the event. The light transmission anomaly in the deep-sea water after 36 days became atypically greater (∼35%) and more extensive (thickness ∼1500 m) near the trench axis owing to the turbulent diffusion of fresh seafloor sediment, coordinated with potential seafloor displacement. In addition to the chemical influx associated with sediment diffusion, an influx of 13C-enriched methane from the deep sub-seafloor reservoirs was estimated. This isotopically unusual methane influx was possibly triggered by the earthquake and its aftershocks that subsequently induced changes in the sub-seafloor hydrogeologic structures. The whole prokaryotic biomass and the development of specific phylotypes in the deep-sea microbial communities could rise and fall at 36 and 98 days, respectively, after the event. We may capture the snap shots of post-earthquake disturbance in deep-sea chemistry and microbial community responses

    Early Clinical Manifestations Associated with Death from Visceral Leishmaniasis

    The visceral leishmaniasis (VL) is a disease potentially fatal if not diagnosed and treated opportunely. This article presents the results of the study on the manifestations identified at the time of the clinical suspicion of the VL cases. This study was conducted in Belo Horizonte, the capital of the State of Minas Gerais, located in southeastern Brazil. This study is both timely and substantive because the Belo Horizonte is an area of transmission of VL, with one of the highest VL-death proportions of Brazil. The patients with higher risk of death had at least one of the following characteristics: ≥60 years, weakness, HIV co-infection, bleeding, jaundice and other associated infections. During the period 2002–2009, 8% to 22% of the patients with VL progressed to death in Belo Horizonte, whilst the proportion in the country was much lower and varied between 5% and 9%. This study has identified vulnerable patients who are at higher risk of death from VL and who would benefit from early predictive evaluation of the prognostic. Hence, the knowledge regarding the factors associated with death may contribute for clinical management and for reduction of deaths from VL
