    Using Texture Synthesis for Non-Photorealistic Shading from Paint Samples

    This paper presents several methods for shading meshes from scanned paint samples that represent dark to light transitions. Our techniques emphasize artistic control of brush stroke texture and color. We ïŹrst demonstrate how the texture of the paint sample can be separated from its color gradient. We demonstrate three methods, two real-time and one off-line for producing rendered, shaded images from the texture samples. All three techniques use texture synthesis to generate additional paint samples. Finally, we develop metrics for evaluating how well each method achieves our goal in terms of texture similarity, shading correctness and temporal coherence

    The cell adhesion molecule L1 regulates the expression of choline acetyltransferase and the development of septal cholinergic neurons

    Mutations in the L1 gene cause severe brain malformations and mental retardation. We investigated the potential roles of L1 in the regulation of choline acetyltransferase (ChAT) and in the development of septal cholinergic neurons, which are known to project to the hippocampus and play key roles in cognitive functions. Using stereological approaches, we detected significantly fewer ChAT-positive cholinergic neurons in the medial septum and vertical limb of the diagonal band of Broca (MS/VDB) of 2-week-old L1-deficient mice compared to wild-type littermates (1644 ± 137 vs. 2051 ± 165, P = 0.038). ChAT protein levels in the septum were 53% lower in 2-week-old L1-deficient mice compared to wild-type littermates. ChAT activity in the septum was significantly reduced in L1-deficient mice compared to wild-type littermates at 1 (34%) and 2 (40%) weeks of age. In vitro, increasing doses of L1-Fc induced ChAT activity in septal neurons with a significant linear trend (*P = 0.0065). At 4 weeks of age in the septum and at all time points investigated in the caudate-putamen (CPu), the number of ChAT-positive neurons and the levels of ChAT activity were not statistically different between L1-deficient mice and wild-type littermates. The total number of cells positive for the neuronal nuclear antigen (NeuN) in the MS/VDB and CPu was not statistically different in L1-deficient mice compared to wild-type littermates, and comparable expression of the cell cycle marker Ki67 was observed. Our results indicate that L1 is required for the timely maturation of septal cholinergic neurons and that L1 promotes the expression and activity of ChAT in septal neurons

    Therapeutic limitations in tumor-specific CD8+ memory T cell engraftment

    BACKGROUND: Adoptive immunotherapy with cytotoxic T lymphocytes (CTL) represents an alternative approach to treating solid tumors. Ideally, this would confer long-term protection against tumor. We previously demonstrated that in vitro-generated tumor-specific CTL from the ovalbumin (OVA)-specific OT-I T cell receptor transgenic mouse persisted long after adoptive transfer as memory T cells. When recipient mice were challenged with the OVA-expressing E.G7 thymoma, tumor growth was delayed and sometimes prevented. The reasons for therapeutic failures were not clear. METHODS: OT-I CTL were adoptively transferred to C57BL/6 mice 21 – 28 days prior to tumor challenge. At this time, the donor cells had the phenotypical and functional characteristics of memory CD8+ T cells. Recipients which developed tumor despite adoptive immunotherapy were analyzed to evaluate the reason(s) for therapeutic failure. RESULTS: Dose-response studies demonstrated that the degree of tumor protection was directly proportional to the number of OT-I CTL adoptively transferred. At a low dose of OT-I CTL, therapeutic failure was attributed to insufficient numbers of OT-I T cells that persisted in vivo, rather than mechanisms that actively suppressed or anergized the OT-I T cells. In recipients of high numbers of OT-I CTL, the E.G7 tumor that developed was shown to be resistant to fresh OT-I CTL when examined ex vivo. Furthermore, these same tumor cells no longer secreted a detectable level of OVA. In this case, resistance to immunotherapy was secondary to selection of clones of E.G7 that expressed a lower level of tumor antigen. CONCLUSIONS: Memory engraftment with tumor-specific CTL provides long-term protection against tumor. However, there are several limitations to this immunotherapeutic strategy, especially when targeting a single antigen. This study illustrates the importance of administering large numbers of effectors to engraft sufficiently efficacious immunologic memory.     Forest biodiversity, carbon sequestration, and wood production: modeling synergies and trade-offs for ten forest landscapes across Europe

    Original ResearchEurope’s forests provide vital habitat for biodiversity and essential ecosystem services whose provision must be sustained or enhanced over the coming century. However, the potential to secure or increase forest ecosystem services, while securing the habitat requirements of taxa remains unclear, especially within the context of uncertain climate and socio-economic developments. To tease out the associated trade-offs and synergies, we used 10 case study landscapes within nine countries throughout Europe. Starting with the current status of the forests in the case study landscapes, we simulated forest development 100 years into the future. Simulations were embedded in three combined climate and socio-economic frame scenarios based on global and European policies which varied in their climate change mitigation efficiency. Scenarios were translated into country specific projections of climate variables, and resultant demands for wood products. Forest management regimes were projected to vary in response to these scenarios at local scales. The specific combinations of alternative forest management practices were based on parallel research and input from local forest stakeholders. For each case study, a specific forest growth simulator was used. In general, the climate scenarios applied did not cause fundamentally different ecosystem service outputs at the case study level. Our results revealed almost no reduction in outcomes for biodiversity indicators with an increase in wood production, and in some cases synergistic results occurred when diversity was actively promoted as part of the management concept. Net carbon uptake was not strongly correlated with biodiversity, indicating that biodiversity-friendly forest management doesn’t need to curtail carbon sequestration. Notably, we obtained heterogeneous results for the relation between sustainable wood production and net carbon uptake. Most scenarios resulted in a more or less reduced net carbon uptake over the long term, often due to stand age class distribution shifts. Levels of sustainable wood production varied widely during the simulation period, from significant increases (Sweden, Lithuania) to minor changes (Slovakia, Turkey) and slight decreases (Ireland, Netherlands). We place our results within the larger context of European forest policy and the challenges of simulating and contrasting forest biodiversity and the ecosystem services that societies depend on outcomes for biodiversity indicators with an increase in wood production, and in some cases synergistic results occurred when diversity was actively promoted as part of the management concept.     Further progress in the study of epsilon iron oxide in archaeological baked clays

    The occurrence of Δ-Fe2O3 in archaeological samples that have been subjected to high temperatures is gradually being detected by the use of micrometric structural characterization techniques. This work provides new information by revealing that the Δ-Fe2O3 is formed as a response to temperature, the aggregation state and the position within the baked clay with respect to the nearest heat source. In addition, depending mainly on the atmospheric environment, the temperature reached by the combustion structure, the distance from the heating source and the particle aggregation, other iron oxide magnetic phases are produced. In the baked clay studied here, hematite is found over the whole range of samples studied but its magnetic contribution is negligible. Magnetite is observed at the sample surface, probably due to local atmospheric environment closest to the combustion source. Maghemite is found at all depths up to 6 cm below the sample surface. Δ-Fe2O3 has a limited distribution, found within 2–3 cm of the sample surface. Furthermore, the viability of this compound as a palaeofield marker has been evaluated in both archaeological and synthetic samples. The results indicate that Δ-Fe2O3 is able to register the direction of the magnetic field. Linear palaeointensity plots have been obtained in synthetic samples, although the value of the palaeofield could be, sometimes, overestimated

    The epithelial cholinergic system of the airways

    Acetylcholine (ACh), a classical transmitter of parasympathetic nerve fibres in the airways, is also synthesized by a large number of non-neuronal cells, including airway surface epithelial cells. Strongest expression of cholinergic traits is observed in neuroendocrine and brush cells but other epithelial cell types—ciliated, basal and secretory—are cholinergic as well. There is cell type-specific expression of the molecular pathways of ACh release, including both the vesicular storage and exocytotic release known from neurons, and transmembrane release from the cytosol via organic cation transporters. The subcellular distribution of the ACh release machineries suggests luminal release from ciliated and secretory cells, and basolateral release from neuroendocrine cells. The scenario as known so far strongly suggests a local auto-/paracrine role of epithelial ACh in regulating various aspects on the innate mucosal defence mechanisms, including mucociliary clearance, regulation of macrophage function and modulation of sensory nerve fibre activity. The proliferative effects of ACh gain importance in recently identified ACh receptor disorders conferring susceptibility to lung cancer. The cell type-specific molecular diversity of the epithelial ACh synthesis and release machinery implies that it is differently regulated than neuronal ACh release and can be specifically targeted by appropriate drugs

    Development of Safety Design Criteria for the Lead-cooled Fast Reactor

    The Lead-cooled Fast Reactor (LFR) provisional System Steering Committee (pSSC) of the Generation-IV International Forum (GIF) has proposed a set of Safety Design Criteria (SDC) dedicated to LFRs. The objective of the LFR SDC is to prescribe a set of reference criteria for the design of LFR systems, structures, and components with the aim of achieving the safety goals of the Generation-IV reactor system. The LFR SDC has been derived from the already existing Safety Design Criteria for the Sodium-cooled Fast Reactor (SFR), since the GIF LFR and SFR systems share a number of characteristics, design features and some corresponding safety-related phenomenology. For the development of the LFR SDC it was also found useful to use the same structure and methodology of the GIF SFR SDC. A set of reference safety design criteria for LFRs is systematically and comprehensively laid out in the SDC to facilitate the development, safety assessment and licensing of LFRs, including BREST-OD-300, ALFRED, SSTAR, SVBR-100, CLEAR-I, and MYRRHA. The paper summarises results of the steps taken to draft the present set of LFR SDC and provides outlook for further review and development activities, in particular towards individual sets of detailed Safety Design Guidelines. Naval Postgraduate Schoo
