    Three-dimensional solutions for the geostrophic flow in the Earth's core

    In his seminal work, Taylor (1963) argued that the geophysically relevant limit for dynamo action within the outer core is one of negligibly small inertia and viscosity in the magnetohydrodynamic equations. Within this limit, he showed the existence of a necessary condition, now well known as Taylor's constraint, which requires that the cylindrically-averaged Lorentz torque must everywhere vanish; magnetic fields that satisfy this condition are termed Taylor states. Taylor further showed that the requirement of this constraint being continuously satisfied through time prescribes the evolution of the geostrophic flow, the cylindrically-averaged azimuthal flow. We show that Taylor's original prescription for the geostrophic flow, as satisfying a given second order ordinary differential equation, is only valid for a small subset of Taylor states. An incomplete treatment of the boundary conditions renders his equation generally incorrect. Here, by taking proper account of the boundaries, we describe a generalisation of Taylor's method that enables correct evaluation of the instantaneous geostrophic flow for any 3D Taylor state. We present the first full-sphere examples of geostrophic flows driven by non-axisymmetric Taylor states. Although in axisymmetry the geostrophic flow admits a mild logarithmic singularity on the rotation axis, in the fully 3D case we show that this is absent and indeed the geostrophic flow appears to be everywhere regular.Comment: 29 Pages, 8 figure

    New insights into the regulatory pathways associated with the activation of the stringent response in bacterial resistance to the PBP2-targeted antibiotics, mecillinam and OP0595/RG6080

    Objectives: The diazabicyclooctane β-lactamase inhibitor OP0595 (RG6080) also acts as an antibiotic, targeting PBP2 in Enterobacteriaceae, but this activity is vulnerable to mutational resistance. We used WGS to investigate the basis of this resistance. Methods: Twenty OP0595-selected mutants, comprising four derived from each of five different Escherichia coli strains, were sequenced on Illumina HiSeq. Reads from each mutant were mapped to the assembled genome of the corresponding parent. A variant-calling file generated with Samtools was parsed to determine genetic alterations. Results: Besides OP0595, the mutants consistently showed decreased susceptibility to mecillinam, which likewise targets PBP2, and grew as stable round forms in the presence of subinhibitory concentrations of OP0595. Among the 20 mutants, 18 had alterations in genes encoding tRNA synthase and modification functions liable to induce expression of the RpoS sigma factor through activation of the stringent response or had mutations suppressing inactivators of RpoS or the stringent response signal-degrading enzyme, SpoT. TolB was inactivated in one mutant: this activates RcsBC regulation and was previously associated with mecillinam resistance. The mechanism of resistance remained unidentified in one mutant. Both the RpoS and RcsBC systems regulate genes of cell division, including ftsAQZ that can compensate for loss or inhibition of PBP2, allowing survival of the challenged bacteria as stable round forms, as seen. Conclusions: WGS identified the global stringent response signal, entailing induction of RpoS, as the main mediator of mutational resistance to OP0595 in E. coli

    Cephalosporinases associated with outer membrane vesicles released by Bacteroides spp. protect gut pathogens and commensals against beta-lactam antibiotics

    Objectives: To identify β-lactamase genes in gut commensal Bacteroides species and to assess the impact of these enzymes, when carried by outer membrane vesicles (OMVs), in protecting enteric pathogens and commensals. Methods: A deletion mutant of the putative class A β-lactamase gene (locus tag BT_4507) found in the genome of the human commensal Bacteroides thetaiotaomicron was constructed and a phenotypic analysis performed. A phylogenetic tree was built from an alignment of nine Bacteroides cephalosporinase protein sequences, using the maximum likelihood method. The rate of cefotaxime degradation after incubation with OMVs produced by different Bacteroides species was quantified using a disc susceptibility test. The resistance of Salmonella Typhimurium and Bifidobacterium breve to cefotaxime in liquid culture in the presence of B. thetaiotaomicron OMVs was evaluated by measuring bacterial growth. Results: The B. thetaiotaomicron BT_4507 gene encodes a β-lactamase related to the CepA cephalosporinase of Bacteroides fragilis. OMVs produced by B. thetaiotaomicron and several other Bacteroides species, except Bacteroides ovatus, carried surface-associated β-lactamases that could degrade cefotaxime. β-Lactamase-harbouring OMVs from B. thetaiotaomicron protected Salmonella Typhimurium and B. breve from an otherwise lethal dose of cefotaxime. Conclusions: The production of membrane vesicles carrying surface-associated β-lactamases by Bacteroides species, which constitute a major part of the human colonic microbiota, may protect commensal bacteria and enteric pathogens, such as Salmonella Typhimurium, against β-lactam antibiotics

    Recent north magnetic pole acceleration towards Siberia caused by flux lobe elongation

    The wandering of Earth’s north magnetic pole, the location where the magnetic field points vertically downwards, has long been a topic of scientific fascination. Since the first in situ measurements in 1831 of its location in the Canadian arctic, the pole has drifted inexorably towards Siberia, accelerating between 1990 and 2005 from its historic speed of 0–15 km yr−1 to its present speed of 50–60 km yr−1. In late October 2017 the north magnetic pole crossed the international date line, passing within 390 km of the geographic pole, and is now moving southwards. Here we show that over the last two decades the position of the north magnetic pole has been largely determined by two large-scale lobes of negative magnetic flux on the core–mantle boundary under Canada and Siberia. Localized modelling shows that elongation of the Canadian lobe, probably caused by an alteration in the pattern of core flow between 1970 and 1999, substantially weakened its signature on Earth’s surface, causing the pole to accelerate towards Siberia. A range of simple models that capture this process indicate that over the next decade the north magnetic pole will continue on its current trajectory, travelling a further 390–660 km towards Siberia

    HST followup observations of two bright z ~ 8 candidate galaxies from the BoRG pure-parallel survey

    We present followup imaging of two bright (L > L*) galaxy candidates at z > 8 from the Brightest of Reionizing Galaxies (BoRG) survey with the F098M filter on HST/WFC3. The F098M filter provides an additional constraint on the flux blueward of the spectral break, and the observations are designed to discriminate between low- and high-z photometric redshift solutions for these galaxies. Our results confirm one galaxy, BoRG 0116+1425 747, as a highly probable z ~ 8 source, but reveal that BoRG 0116+1425 630 - previously the brightest known z > 8 candidate (mAB = 24.5) - is likely to be a z ~ 2 interloper. As this source was substantially brighter than any other z > 8 candidate, removing it from the sample has a significant impact on the derived UV luminosity function in this epoch. We show that while previous BoRG results favored a shallow power-law decline in the bright end of the luminosity function prior to reionization, there is now no evidence for departure from a Schechter function form and therefore no evidence for a difference in galaxy formation processes before and after reionization.Comment: Accepted by ApJL, 7 pages, 4 figure
