    Tibial or hip BMD predict clinical fracture risk equally well: results from a prospective study in 700 elderly Swiss women

    Summary: In a randomly selected cohort of Swiss community-dwelling elderly women prospectively followed up for 2.8 ± 0.6years, clinical fractures were assessed twice yearly. Bone mineral density (BMD) measured at tibial diaphysis (T-DIA) and tibial epiphysis (T-EPI) using dual-energy X-ray absorptiometry (DXA) was shown to be a valid alternative to lumbar spine or hip BMD in predicting fractures. Introduction: A study was carried out to determine whether BMD measurement at the distal tibia sites of T-EPI and T-DIA is predictive of clinical fracture risk. Methods: In a predefined representative cohort of Swiss community-dwelling elderly women aged 70-80years included in the prospective, multi-centre Swiss Evaluation of the Methods of Measurement of Osteoporotic Fracture risk (SEMOF) study, fracture risk profile was assessed and BMD measured at the lumbar spine (LS), hip (HIP) and tibia (T-DIA and T-EPI) using DXA. Thereafter, clinical fractures were reported in a bi-yearly questionnaire. Results: During 1,786 women-years of follow-up, 68 clinical fragility fractures occurred in 61 women. Older age and previous fracture were identified as risk factors for the present fractures. A decrease of 1 standard deviation in BMD values yielded a 1.5-fold (HIP) to 1.8-fold (T-EPI) significant increase in clinical fragility fracture hazard ratio (adjusted for age and previous fracture). All measured sites had comparable performance for fracture prediction (area under the curve range from 0.63 [LS] to 0.68 [T-EPI]). Conclusion: Fracture risk prediction with BMD measurements at T-DIA and T-EPI is a valid alternative to BMD measurements at LS or HIP for patients in whom these sites cannot be accessed for clinical, technical or practical reason

    Brønstedsäure‐Katalyse – Kontrolle der Konkurrenz zwischen monomerem und dimerem Reaktionsweg erhöht Stereoselektivität

    Chirale Phosphorsäuren (CPS) sind inzwischen ein bevorzugter Katalysatortyp in der Organokatalyse, jedoch bleibt die Auswahl der optimalen Katalysatorstruktur weiterhin eine Herausforderung. Zusätzlich können unbekannte konkurrierende Reaktionswege die maximale Stereoselektivität und das Potenzial von Vorhersagemodellen einschränken. Bei der CPS-katalysierten Transferhydrierung von Iminen haben wir für viele Systeme zwei Reaktionswege mit inverser Stereoselektivität gefunden, bei denen entweder eine monomere CPS oder ein Wasserstoffbrücken-verknüpftes Dimer als Katalysator fungiert. NMR-Messungen und DFT-Berechnungen offenbarten ein dimeres Intermediat mit einer stärkeren Substrataktivierung durch Kooperativitätseffekte. Beide Wege können separiert werden: Niedrige Temperaturen und hohe Katalysatoranteile begünstigen den dimeren Reaktionsweg (ee bis zu −98 %), während niedrige Temperaturen und reduzierte Katalysatoranteil den monomeren Reaktionsweg fördern und zu einem signifikant verbesserten ee führen (92–99 % ee; vorher 68–86 % bei höheren Temperaturen). Insgesamt wird eine große Auswirkung auf die CPS-Katalyse in Bezug auf Reaktionsoptimierung und Vorhersage erwartet

    Characterisation of proteins in excretory/secretory products collected from salmon lice, Lepeophtheirus salmonis

    Background  The salmon louse, Lepeophtheirus salmonis, is an ectoparasitic copepod which feeds on the mucus, skin and blood of salmonid fish species. The parasite can persist on the surface of the fish without any effective control being exerted by the host immune system. Other ectoparasitic invertebrates produce compounds in their saliva, excretions and/or secretions which modulate the host immune responses allowing them to remain on or in the host during development. Similarly, compounds are produced in secretions of L. salmonis which are thought to be responsible for immunomodulation of the host responses as well as other aspects of crucial host-parasite interactions.  Methods  In this study we have identified and characterised the proteins in the excretory/secretory (E/S) products of L. salmonis using LC-ESI-MS/MS.  Results  In total 187 individual proteins were identified in the E/S collected from adult lice and pre-adult sea lice. Fifty-three proteins, including 13 serine-type endopeptidases, 1 peroxidase and 5 vitellogenin-like proteins were common to both adult and pre-adult E/S products. One hundred and seven proteins were identified in the adult E/S but not in the pre-adult E/S and these included serine and cysteine-type endopeptidases, vitellogenins, sphingomyelinase and calreticulin. A total of 27 proteins were identified in pre-adult E/S products but not in adult E/S.  Conclusions  The assigned functions of these E/S products and the potential roles they play in host-parasite interaction is discussed

    Atomic force microscopy analysis of nanoparticles in non-ideal conditions

    Nanoparticles are often measured using atomic force microscopy or other scanning probe microscopy methods. For isolated nanoparticles on flat substrates, this is a relatively easy task. However, in real situations, we often need to analyze nanoparticles on rough substrates or nanoparticles that are not isolated. In this article, we present a simple model for realistic simulations of nanoparticle deposition and we employ this model for modeling nanoparticles on rough substrates. Different modeling conditions (coverage, relaxation after deposition) and convolution with different tip shapes are used to obtain a wide spectrum of virtual AFM nanoparticle images similar to those known from practice. Statistical parameters of nanoparticles are then analyzed using different data processing algorithms in order to show their systematic errors and to estimate uncertainties for atomic force microscopy analysis of nanoparticles under non-ideal conditions. It is shown that the elimination of user influence on the data processing algorithm is a key step for obtaining accurate results while analyzing nanoparticles measured in non-ideal conditions

    Proteases of haematophagous arthropod vectors are involved in blood-feeding, yolk formation and immunity : a review

    Ticks, triatomines, mosquitoes and sand flies comprise a large number of haematophagous arthropods considered vectors of human infectious diseases. While consuming blood to obtain the nutrients necessary to carry on life functions, these insects can transmit pathogenic microorganisms to the vertebrate host. Among the molecules related to the blood-feeding habit, proteases play an essential role. In this review, we provide a panorama of proteases from arthropod vectors involved in haematophagy, in digestion, in egg development and in immunity. As these molecules act in central biological processes, proteases from haematophagous vectors of infectious diseases may influence vector competence to transmit pathogens to their prey, and thus could be valuable targets for vectorial control
