    Platelet-rich plasma as a chance of recovery for liver cirrhosis patient

    The search for pathogenetic methods for treating cirrhosis of the liver continues. One of the promising areas of regenerative medicine is the use of platelet-rich plasma (PRP). Due to numerous publications on the efficacy of PRP in conditions of abnormal liver damage in the experiment, the work is devoted to the study of the possibility of manufacturing PRP from the blood of patients with liver cirrhosis (LC) for the purpose of proposals for further clinical use. The study included 12 men suffering from CP-class B-C (average grade by ChildPugh classification - 8.8) at the age of 45 ± 2.2 years. The comparison group consisted of 5 healthy men at the age of 44 ± 1.8 years. All men measured blood platelets in accordance with the usual method. The manufacture of PRP was carried out according to the scheme, which includes the collection of blood from the elbow vein in a 9 ml syringe-flask, which contains an anticoagulant. The collected blood in the same test tube is centrifuged in two stages: on the first - within 12` at a speed of 4000 rpm. Then the formed plasma, namely, from the supernatant layer, is recruited into another tube without an anticoagulant and treated in a centrifuge for 15` at a speed of 3600 rpm. After the removal of poor plasma platelets, the final product - a PRP aggregate in the volume of 0.3 ml - is obtained, which is used for the next determination of platelet count using an automated hemo-analyzer. In patients with LC, both thrombocytopenia and thrombocytosis were observed. In the PRP, the increase in the total platelet count was 2.2-2.4 times, both in baseline thrombocytopenia and in baseline thrombocytosis, due to the absence of the mean platelet count and the platelet distribution width. In the control group male, the average increase in the number of platelets was 3.37 times, in the absence of significant changes in the mean platelet count and platelet distribution width. The fact is that the use of autologous PRP in patients with LC is likely to be ineffective. In the presence of initial thrombocytopenia - there is no possibility to reach a "critical" quantity of 1 million G/L, which is necessary for the implementation of regenerative potential of PRP. Concomitant presence of morpho-functional defects of these platelets practically makes it impossible for such a method of protection/regeneration of liver parenchyma in patients with lC. In patients with LC there is a moderate increase in the concentration of platelets in the manufacture of PRP by standard method; therefore, it is necessary to find new methods for obtaining PRP for autologous use

    Disturbance can slow down litter decomposition, depending on severity of disturbance and season: an example from Mount Kilimanjaro

    Deforestation and land-use change affect ecosystem processes such as carbon cycling. Here, we present results from a litter decomposition experiment in six natural and six disturbed vegetation types along an elevation gradient of 3600 m on the southern slopes of Mount Kilimanjaro, Tanzania. We exposed litter bags with a standard material for up to 12 weeks each in two seasons. In the cold wet season we sampled the full elevation gradient and in the warm wet season we repeated the sampling in the lower part of the elevation gradient. Though we found significantly negative effects of disturbance in forest ecosystems, this was only due to differences between natural and burned Podocarpus forests. Disturbance characterized by a more open vegetation structure in many of the studied vegetation types had no general effect when we studied the full elevation gradient; this also included non-forest vegetation types. Land-use intensity had a significant negative effect on decomposition rates but only in the warm wet season, not in the cold wet season. Temperature and humidity were the most important drivers of decomposition overall and for all subsets of vegetation types and seasons. Our study shows that negative effects of disturbance or land-use intensity on decomposition depended on the severity of disturbance and on the season. Nevertheless, climate was generally the most relevant driver of decomposition. Therefore, vegetation types with moderate levels of disturbance can retain high functionality in regards to carbon cycling over short periods of time. More and longer decomposition studies are necessary to better predict consequences of land-use change for carbon cycling in the Afrotropics.</p

    Evaluation of a Brief Sodium Screener in Two Samples

    The Sodium Screener© (SS©), as developed by NutritionQuest (Berkeley, CA, USA), was designed to reduce the burden of repeated dietary or urinary sodium measurements, but the accuracy of daily sodium intake estimates has not been reported. Associations were examined between sodium intakes derived from the SS© scores and repeated 24-h recalls (24DR) in two studies with different administration modes. In one study, 102 registered dietitians (RD) completed three Automated Self-Administered 24DRs (ASA24©), version 2014, followed by the SS©; both were self-administered and web-based. In the second sample, (the Study of Household Purchasing Patterns, Eating, and Recreation or SHoPPER), trained dietitians conducted 24DR interviews with 69 community-dwelling adults in their homes; all the community adults then completed a paper-based SS© at the final visit. In the RD study, SS©-predicted sodium intakes were 2604 ± 990 (mean ± Standard deviation (SD)), and ASA24© sodium intakes were 3193 ± 907 mg/day. In the SHoPPER sample, corresponding values were 3338 ± 1310 mg/day and 2939 ± 1231 mg/day, respectively. SS© -predicted and recall sodium estimates were correlated in the RD study (r = 0.381, p = 0.0001) and in the SHoPPER (r = 0.430, p = 0.0002). Agreement between the SS© and 24-h recalls was poor when classifying individuals as meeting the dietary sodium guidelines of 2300 mg/day or not (RD study: kappa = 0.080, p = 0.32; SHoPPER: kappa = 0.207, p = 0.08). Based on repeated 24DR either in person or self-reported online as the criterion for estimating daily sodium intakes, the SS© may require additional modifications

    The study of individual differences in emotional responding can provide considerable insight into interpersonal dynamics and the etiology of psychopathology. Heart rate variability (HRV) analysis is emerging as an objective measure of regulated emotional responding (generating emotional responses of appropriate timing and magnitude). This review provides a theoretical and empirical rationale for the use of HRV as an index of individual differences in regulated emotional responding. Two major theoretical frameworks that articulate the role of HRV in emotional responding are presented, and relevant empirical literature is reviewed. The case is made that HRV is an accessible research tool that can increase the understanding of emotion in social and psychopathological processes

    Induction of apoptosis in human cancer cells by targeting mitochondria with gold nanoparticles

    A major challenge in designing cancer therapies is the induction of cancer cell apoptosis, although activation of intrinsic apoptotic pathways by targeting gold nanoparticles to mitochondria is promising. We report an in vitro procedure targeting mitochondria with conjugated gold nanoparticles and investigating effects on apoptosis induction in the human breast cancer cell line Jimt-1. Gold nanoparticles were conjugated to a variant of turbo green fluorescent protein (mitoTGFP) harbouring an amino-terminal mitochondrial localization signal. Au nanoparticle conjugates were further complexed with cationic maltotriose-modified poly(propylene imine) third generation dendrimers. Fluorescence and transmission electron microscopy revealed that Au nanoparticle conjugates were directed to mitochondria upon transfection, causing partial rupture of the outer mitochondrial membrane, triggering cell death. The ability to target Au nanoparticles into mitochondria of breast cancer cells and induce apoptosis reveals an alternative application of Au nanoparticles in photothermal therapy of cancer

    Availability of Food Preparation Supplies among Pregnant Women: Preliminary Results from the Decision Making, Eating, and Weight Gain during Pregnancy (DEW) Study

    Background: Lack of cooking supplies may be a potential barrier to preparing healthy meals at home. We examined the availability of food preparation supplies among pregnant women in relation to sociodemographic characteristics. Methods: We used preliminary data (N=59) from an ongoing study which enrolled English-speaking women aged 18+ years, pregnant with singleton gestation \u3c36\u3eweeks, pre-pregnancy BMI 18.5-40 kg/m2, and planning to deliver at UMMHC. Women completed the Food Preparation Checklist (FPC) at home. The FPC asks women if 41 specific food preparation items; scores reflect number of items present in the home. Other variables were self-reported. Pearson’s correlation, t-tests, and ANOVAs provided comparisons. We constructed an adjusted linear regression model to explore FPC by sociodemographic characteristics. Results: Women were aged 30.3 (SD=4.1) years, 64.4% were non-Hispanic White, 84.8% were married or living with a partner, and 30.5% reported difficulty paying for basic expenses. Women were enrolled at 22.7 (SD=5.6) weeks gestation; 30.5% were primigravid. Mean pre-pregnancy BMI was 25.0 (SD=4.6) kg/m2; 25.4% were overweight and 17.0% obese. Average FPC score was 32.3 (SD=6.1; range:14-39). FPC scores were higher among Non-Hispanic White women (34.6±3.5 vs. 28.1±7.5, p\u3c0.0001), those with higher education (28.3±7.0 high school/GED or less, 31.0±6.2 some/college degree, vs. 34.7±4.6 some/degree graduate, p\u3c0.01), those married or living with a partner (33.3±5.7 vs. 26.9±5.7, p\u3c0.01), with lower pre-pregnancy BMI (r=-0.38, p\u3c0.01), and who had no difficulty paying for basic expenses (34.0±5.0 vs. 28.4±6.6, p\u3c0.001). In a model that additionally adjusted for pre-pregnancy BMI, non-Hispanic White women had on average 5.7 more food preparation items (95% CI: 3.2, 8.3) and those reporting difficulty paying for basic expenses 3.8 fewer items (95% CI: -6.8, -0.9). Conclusions: Understanding the food preparation supplies available to pregnant women may be useful when designing interventions to improve diet quality and promote healthy weight gain during pregnancy

    Early-season plant-to-plant spatial uniformity can affect soybean yields

    Increased soybean (Glycine max L. Merril) seed costs have motivated interest in reduced seeding rates to improve profitability while maintaining or increasing yield. However, little is known about the effect of early-season plant-to-plant spatial uniformity on the yield of modern soybean varieties planted at reduced seeding rates. The objectives of this study were to (i) investigate traditional and devise new metrics for characterizing early-season plant-to-plant spatial uniformity, (ii) identify the best metrics correlating plant-to-plant spatial uniformity and soybean yield, and (iii) evaluate those metrics at different seeding rate (and achieved plant density) levels and yield environments. Soybean trials planted in 2019 and 2020 compared seeding rates of 160, 215, 270, and 321 thousand seeds ha−1 planted with two different planters, Max Emerge and Exact Emerge, in rainfed and irrigated conditions in the United States (US). In addition, trials comparing seeding rates of 100, 230, 360, and 550 thousand seeds ha−1 were conducted in Argentina (Arg) in 2019 and 2020. Achieved plant density, grain yield, and early-season plant-to-plant spacing (and calculated metrics) were measured in all trials. All site-years were separated into low- (2.7 Mg ha−1), medium- (3 Mg ha−1), and high- (4.3 Mg ha−1) yielding environments, and the tested seeding rates were separated into low ( 300 seeds m−2) levels. Out of the 13 metrics of spatial uniformity, standard deviation (sd) of spacing and of achieved versus targeted evenness index (herein termed as ATEI, observed to theoretical ratio of plant spacing) showed the greatest correlation with soybean yield in US trials (R2 = 0.26 and 0.32, respectively). However, only the ATEI sd, with increases denoting less uniform spacing, exhibited a consistent relationship with yield in both US and Arg trials. The effect of spatial uniformity (ATEI sd) on soybean yield differed by yield environment. Increases in ATEI sd (values > 1) negatively impacted soybean yields in both low- and medium-yield environments, and in achieved plant densities below 200 thousand plants ha−1. High-yielding environments were unaffected by variations in spatial uniformity and plant density levels. Our study provides new insights into the effect of early-season plant-to-plant spatial uniformity on soybean yields, as influenced by yield environments and reduced plant densities.Fil: Pereyra, Valentina M..
