Untersuchung zum Einfluss von Bluthochdruck auf die Immunreaktion nach experimentellem Schlaganfall
Einleitung: IschĂ€mische SchlaganfĂ€lle ziehen ausgeprĂ€gte EntzĂŒndungsprozesse im Gehirn sowie Immunreaktionen in der Körperperipherie nach sich, welche den Erkrankungsverlauf und die Regeneration maĂgeblich beeinflussen. Die Modulation dieser Immunantwort stellt folglich einen vielversprechenden experimentellen Ansatz in der Schlaganfalltherapie dar. Der ischĂ€mische Schlaganfall ist auĂerdem mit verschiedenen KomorbiditĂ€ten und Risikofaktoren assoziiert, deren Auswirkungen auf die komplexen postischĂ€mischen pathophysiologischen Prozesse nur in Teilen aufgeklĂ€rt sind. So hat eine arterielle Hypertonie (Bluthochdruck) als wichtigster modifizierbarer Risikofaktor durch die Induktion von GefĂ€ĂschĂ€den eine zentrale Bedeutung fĂŒr die Pathogenese von ischĂ€mischen SchlaganfĂ€llen und geht auĂerdem mit einer Aktivierung des Immunsystems einher. Der konkrete Einfluss der Hypertonie auf die postischĂ€mische EntzĂŒndungsreaktion wurde bislang nicht hinreichend untersucht. Die Einbeziehung wichtiger Begleiterkrankungen, wie Bluthochdruck, in die prĂ€klinische Schlaganfallforschung gewinnt zunehmend an Bedeutung, da ein erweitertes VerstĂ€ndnis der pathophysiologischen ZusammenhĂ€nge auch eine bessere Ăbertragbarkeit neuer immunmodulatorischer Therapiekonzepte auf den Menschen in Aussicht stellt.
Zielstellung: Das Hauptziel dieser Arbeit ist die Identifizierung pathofunktioneller ZusammenhĂ€nge zwischen der Immunantwort nach Schlaganfall und manifestem Bluthochdruck. DafĂŒr wurde die zentrale und periphere EntzĂŒndungsreaktion nach experimentellem Schlaganfall in einem prĂ€morbiden hypertensiven Tiermodell (spontan-hypertensive Ratte, SHR) im Vergleich mit normotensiven Tieren mithilfe von vorwiegend durchflusszytometrischen, histologischen und molekularbiologischen Methoden analysiert. Daneben sollte das verwendete hypertensive Tiermodell fĂŒr die Untersuchung immunologischer Aspekte der translationalen Schlaganfallforschung evaluiert werden.
Tiere, Material und Methoden: FĂŒr alle Tierversuche und Organentnahmen wurden ausschlieĂlich mĂ€nnliche Ratten der StĂ€mme Wistar-Kyoto und SHR im Alter von 12 bis 14 Wochen verwendet. Die Induktion des ischĂ€mischen Infarkts erfolgte mithilfe eines permanenten, transkraniellen Schlaganfallmodells oder mittels photochemischer Thrombose. In AbhĂ€ngigkeit von der Untersuchungsgruppe wurden die Tiere 1 oder 4 Tage nach Infarktinduktion bzw. Sham-Operation schmerzfrei getötet. Neben wenigen neuroanatomischen und neurofunktionellen Ausleseparametern wurde als HauptzielgröĂe die Immunzellverteilung im Gehirn und im Blut erfasst. DafĂŒr wurden Hirnzellisolate und Vollblutproben mit maximal 8 fluoreszenzgekoppelten Antikörpern in verschiedenen Kombinationen markiert und in einem 3-Laser-Durchflusszytometer (FACSCanto II) gemessen und ausgewertet. Zudem wurden in kryokonservierten Hirnschnitten relevante Immunzellpopulationen und AdhĂ€sionsmolekĂŒle mittels Immunfluorezenztechniken markiert und fĂŒr die Darstellung der rĂ€umlichen Verteilung mit einem Konfokal-Mikroskop (LSM710, Zeiss) analysiert. ZusĂ€tzlich wurde die Gen-und Proteinexpression selektiver Zytokine und AdhĂ€sionsmolekĂŒle in dissoziiertem Hirngewebe ermittelt. Die statistische Auswertung wurde je nach erfasster ZielgröĂe mittels zweiseitigem t-Test, Wilcoxon Rangsummentest, Pearson-Korrelation oder Varianzanalyse-Verfahren durchgefĂŒhrt. Ein Signifikanzniveau von p<0,05 wurde fĂŒr alle statistischen Verfahren festgelegt.
Ergebnisse: Neben einer vergröĂerten LĂ€sion konnte in hypertensiven Tieren insbesondere eine gesteigerte Infiltration von Zellen des angeborenen Immunsystems in das ischĂ€mische Hirn gezeigt werden. Eine Verschiebung des Makrophagen-Granulozyten-VerhĂ€ltnisses wies darĂŒber hinaus auf eine verĂ€nderte EntzĂŒndungskinetik bei Vorliegen von Bluthochdruck hin. Weiterhin wurde in Tieren mit arterieller Hypertonie eine erhöhte Zahl von zirkulierenden Monozyten und Granulozyten beobachtet. Im Hirngewebe von spontan-hypertensiven Ratten nach Schlaganfall wurden auĂerdem eine verminderte Expression des antiinflammatorisch wirksamen Zytokins Interleukin 10, erhöhte Expressionsraten selektiver Leukozyten-rekrutierender Chemokine sowie eine vermehrte Expression des AdhĂ€sionsmolekĂŒls ICAM-1 auf infiltrierenden Leukozyten erfasst. Daneben konnten modellabhĂ€ngige EinflĂŒsse der verschiedenen Induktionsmethoden auf die Immunreaktion identifiziert werden.
Schlussfolgerung: Die Ergebnisse weisen deutlich auf einen Zusammenhang zwischen einem bestehenden arteriellen Hypertonus und einer gesteigerten entzĂŒndlichen Reaktion im Gehirn nach experimentellem Schlaganfall hin und zeigen mögliche zugrundeliegende Mechanismen auf. Gleichzeitig unterstreicht die Arbeit durch eine differenzierte Analyse methodischer und modellabhĂ€ngiger EinflĂŒsse die UnerlĂ€sslichkeit, prĂ€klinische Ergebnisse kritisch zu hinterfragen und in unterschiedlichen Modellen zu ĂŒberprĂŒfen. Auf Grundlage der Untersuchungen kann die spontan-hypertensive Ratte zudem als ein fĂŒr die translationale Schlaganfallforschung geeignetes prĂ€morbides Tiermodell beurteilt werden, in welchem sich der Einfluss des Risikofaktors Bluthochdruck auf die Entwicklung und den Verlauf der postischĂ€mischen EntzĂŒndung gut abbilden lĂ€sst.:Inhaltsverzeichnis

DanksagungIntroduction: Ischemic strokes lead to a sequence of immune responses, including pronounced tissue inflammation in the brain as well as distinct reactions of the peripheral immune system that consistently influence disease process and outcome. The modulation of these immune responses therefore represents a promising experimental approach in stroke therapy. Ischemic stroke is also associated with various comorbidities and risk factors whose effects on the complex postischemic pathology have only been partially elucidated. Being the most important modifiable risk factor this especially applies to arterial hypertension that has a central role in the pathogenesis of ischemic stroke by inducing vascular damage but is also associated with a strong activation of the immune system. However, the specific influence of hypertension on postischemic inflammation has not been sufficiently investigated. Besides, the integration of hypertension and other important concomitant diseases is becoming a more regular tool in preclinical stroke modelling in order to expand the understanding of pathophysiological interactions and overcome the translational gap of new immunomodulatory therapies.
Aim: The main objective of this work is the identification of pathofunctional interations between the immune response after stroke and preexisting arterial hypertension. For this purpose, the central and peripheral inflammatory response after experimental stroke was investigated in a premorbid hypertensive animal model (spontaneously hypertensive rat, SHR) in comparison with normotensive animals by means of flow cytometric, histological and molecular biological methods. In addition, the hypertensive animal model was supposed to be assessed regarding its suitability for the investigation of immunological aspects in translational stroke research.
Material and Methods: For all animal experiments and organ removal, only male rats of the Wistar Kyoto and SHR strains aged 12 to 14 weeks were used. Induction of experimental stroke was performed using either a permanent transcranial stroke model or photochemical thrombosis model. Depending on the study group, animals were killed painlessly 1 or 4 days after infarct induction or sham surgery respectively. In addition to few neuroanatomic and neurofunctional readout parameters, immune cell distribution in the brain and blood was captured as primary variable. Therefore, brain cell isolates and whole blood samples labeled with a maximum of 8 fluorescence-coupled antibodies in different combinations were measured and analyzed in a 3-laser flow cytometer (FACSCanto II). Furthermore, immunofluorescence techniques were applied on cryopreserved brain sections in order to image spatial distribution of relevant immune cell populations and adhesion molecules by means of confocal microscopy (LSM710, Zeiss). In addition, the mRNA and protein expression of selective cytokines and adhesion molecules was determined in dissociated brain tissue. Depending on the targeted parameter statistical analyses were performed by using two-sample t-test, Wilcoxon rank-sum test, Pearson correlation coefficient or analysis of variance. A significance level of p<0.05 was set for all statistical methods.
Results: In addition to an enlarged lesion, in hypertensive animals an increased infiltration of cells of the innate immune system to the ischemic brain area was detected. A shift of the macrophage-granulocyte-ratio further indicated an altered inflammatory profile in hypertensive rats. Furthermore, an increased number of circulating monocytes and granulocytes were observed in animals with hypertension. In brain tissue of SHR after stroke, a decreased expression of the anti-inflammatory cytokine interleukin 10 were recorded along with increased expression levels of selective leukocyte-recruiting chemokines and an increased expression of the adhesion molecule ICAM-1 on infiltrating leukocytes. In addition, caused by different induction methods, model-dependent impact on the immune reaction could be identified.
Conclusion: The results clearly indicate a relationship between existing arterial hypertension and an increased inflammatory response in the brain after experimental stroke and point out potential underlying mechanisms. At the same time, by adopting a differentiated view of methodological and model-dependent influences, the work underscores the need for critical reflection and constant verification of preclinical results in different models. Finally the work validates the SHR strain as a suitable premorbid preclinical system for further translational research since it well models the influence of hypertension on the development and course of postischemic inflammation.:Inhaltsverzeichnis

Molecular Cytogenetic Profiling Reveals Similarities and Differences Between Localized Nodal and Systemic Follicular Lymphomas
Recently, we have developed novel highly promising gene expression (GE) classifiers discriminating localized nodal (LFL) from systemic follicular lymphoma (SFL) with prognostic impact. However, few data are available in LFL especially concerning hotspot genetic alterations that are associated with the pathogenesis and prognosis of SFL. A total of 144 LFL and 527 SFL, enrolled in prospective clinical trials of the German Low Grade Lymphoma Study Group, were analyzed by fluorescence in situ hybridization to detect deletions in chromosomes 1p, 6q, and 17p as well as BCL2 translocations to determine their impact on clinical outcome of LFL patients. The frequency of chromosomal deletions in 1p and 17p was comparable between LFL and SFL, while 6q deletions and BCL2 translocations more frequently occurred in SFL. A higher proportion of 1p deletions was seen in BCL2-translocationâpositive LFL, compared with BCL2-translocationânegative LFL. Deletions in chromosomes 1p, 6q, and 17p predicted clinical outcome of patients with SFL in the entire cohort, while only deletions in chromosome 1p retained its negative prognostic impact in R-CHOPâtreated SFL. In contrast, no deletions in one of the investigated genetic loci predicted clinical outcome in LFL. Likewise, the presence or absence of BCL2 translocations had no prognostic impact in LFL. Despite representing a genetic portfolio closely resembling SFL, LFL showed some differences in deletion frequencies. BCL2 translocation and 6q deletion frequency differs between LFL and SFL and might contribute to distinct genetic profiles in LFL and SFL

Einleitung: IschĂ€mische SchlaganfĂ€lle ziehen ausgeprĂ€gte EntzĂŒndungsprozesse im Gehirn sowie Immunreaktionen in der Körperperipherie nach sich, welche den Erkrankungsverlauf und die Regeneration maĂgeblich beeinflussen. Die Modulation dieser Immunantwort stellt folglich einen vielversprechenden experimentellen Ansatz in der Schlaganfalltherapie dar. Der ischĂ€mische Schlaganfall ist auĂerdem mit verschiedenen KomorbiditĂ€ten und Risikofaktoren assoziiert, deren Auswirkungen auf die komplexen postischĂ€mischen pathophysiologischen Prozesse nur in Teilen aufgeklĂ€rt sind. So hat eine arterielle Hypertonie (Bluthochdruck) als wichtigster modifizierbarer Risikofaktor durch die Induktion von GefĂ€ĂschĂ€den eine zentrale Bedeutung fĂŒr die Pathogenese von ischĂ€mischen SchlaganfĂ€llen und geht auĂerdem mit einer Aktivierung des Immunsystems einher. Der konkrete Einfluss der Hypertonie auf die postischĂ€mische EntzĂŒndungsreaktion wurde bislang nicht hinreichend untersucht. Die Einbeziehung wichtiger Begleiterkrankungen, wie Bluthochdruck, in die prĂ€klinische Schlaganfallforschung gewinnt zunehmend an Bedeutung, da ein erweitertes VerstĂ€ndnis der pathophysiologischen ZusammenhĂ€nge auch eine bessere Ăbertragbarkeit neuer immunmodulatorischer Therapiekonzepte auf den Menschen in Aussicht stellt.
Zielstellung: Das Hauptziel dieser Arbeit ist die Identifizierung pathofunktioneller ZusammenhĂ€nge zwischen der Immunantwort nach Schlaganfall und manifestem Bluthochdruck. DafĂŒr wurde die zentrale und periphere EntzĂŒndungsreaktion nach experimentellem Schlaganfall in einem prĂ€morbiden hypertensiven Tiermodell (spontan-hypertensive Ratte, SHR) im Vergleich mit normotensiven Tieren mithilfe von vorwiegend durchflusszytometrischen, histologischen und molekularbiologischen Methoden analysiert. Daneben sollte das verwendete hypertensive Tiermodell fĂŒr die Untersuchung immunologischer Aspekte der translationalen Schlaganfallforschung evaluiert werden.
Tiere, Material und Methoden: FĂŒr alle Tierversuche und Organentnahmen wurden ausschlieĂlich mĂ€nnliche Ratten der StĂ€mme Wistar-Kyoto und SHR im Alter von 12 bis 14 Wochen verwendet. Die Induktion des ischĂ€mischen Infarkts erfolgte mithilfe eines permanenten, transkraniellen Schlaganfallmodells oder mittels photochemischer Thrombose. In AbhĂ€ngigkeit von der Untersuchungsgruppe wurden die Tiere 1 oder 4 Tage nach Infarktinduktion bzw. Sham-Operation schmerzfrei getötet. Neben wenigen neuroanatomischen und neurofunktionellen Ausleseparametern wurde als HauptzielgröĂe die Immunzellverteilung im Gehirn und im Blut erfasst. DafĂŒr wurden Hirnzellisolate und Vollblutproben mit maximal 8 fluoreszenzgekoppelten Antikörpern in verschiedenen Kombinationen markiert und in einem 3-Laser-Durchflusszytometer (FACSCanto II) gemessen und ausgewertet. Zudem wurden in kryokonservierten Hirnschnitten relevante Immunzellpopulationen und AdhĂ€sionsmolekĂŒle mittels Immunfluorezenztechniken markiert und fĂŒr die Darstellung der rĂ€umlichen Verteilung mit einem Konfokal-Mikroskop (LSM710, Zeiss) analysiert. ZusĂ€tzlich wurde die Gen-und Proteinexpression selektiver Zytokine und AdhĂ€sionsmolekĂŒle in dissoziiertem Hirngewebe ermittelt. Die statistische Auswertung wurde je nach erfasster ZielgröĂe mittels zweiseitigem t-Test, Wilcoxon Rangsummentest, Pearson-Korrelation oder Varianzanalyse-Verfahren durchgefĂŒhrt. Ein Signifikanzniveau von p<0,05 wurde fĂŒr alle statistischen Verfahren festgelegt.
Ergebnisse: Neben einer vergröĂerten LĂ€sion konnte in hypertensiven Tieren insbesondere eine gesteigerte Infiltration von Zellen des angeborenen Immunsystems in das ischĂ€mische Hirn gezeigt werden. Eine Verschiebung des Makrophagen-Granulozyten-VerhĂ€ltnisses wies darĂŒber hinaus auf eine verĂ€nderte EntzĂŒndungskinetik bei Vorliegen von Bluthochdruck hin. Weiterhin wurde in Tieren mit arterieller Hypertonie eine erhöhte Zahl von zirkulierenden Monozyten und Granulozyten beobachtet. Im Hirngewebe von spontan-hypertensiven Ratten nach Schlaganfall wurden auĂerdem eine verminderte Expression des antiinflammatorisch wirksamen Zytokins Interleukin 10, erhöhte Expressionsraten selektiver Leukozyten-rekrutierender Chemokine sowie eine vermehrte Expression des AdhĂ€sionsmolekĂŒls ICAM-1 auf infiltrierenden Leukozyten erfasst. Daneben konnten modellabhĂ€ngige EinflĂŒsse der verschiedenen Induktionsmethoden auf die Immunreaktion identifiziert werden.
Schlussfolgerung: Die Ergebnisse weisen deutlich auf einen Zusammenhang zwischen einem bestehenden arteriellen Hypertonus und einer gesteigerten entzĂŒndlichen Reaktion im Gehirn nach experimentellem Schlaganfall hin und zeigen mögliche zugrundeliegende Mechanismen auf. Gleichzeitig unterstreicht die Arbeit durch eine differenzierte Analyse methodischer und modellabhĂ€ngiger EinflĂŒsse die UnerlĂ€sslichkeit, prĂ€klinische Ergebnisse kritisch zu hinterfragen und in unterschiedlichen Modellen zu ĂŒberprĂŒfen. Auf Grundlage der Untersuchungen kann die spontan-hypertensive Ratte zudem als ein fĂŒr die translationale Schlaganfallforschung geeignetes prĂ€morbides Tiermodell beurteilt werden, in welchem sich der Einfluss des Risikofaktors Bluthochdruck auf die Entwicklung und den Verlauf der postischĂ€mischen EntzĂŒndung gut abbilden lĂ€sst.:Inhaltsverzeichnis

DanksagungIntroduction: Ischemic strokes lead to a sequence of immune responses, including pronounced tissue inflammation in the brain as well as distinct reactions of the peripheral immune system that consistently influence disease process and outcome. The modulation of these immune responses therefore represents a promising experimental approach in stroke therapy. Ischemic stroke is also associated with various comorbidities and risk factors whose effects on the complex postischemic pathology have only been partially elucidated. Being the most important modifiable risk factor this especially applies to arterial hypertension that has a central role in the pathogenesis of ischemic stroke by inducing vascular damage but is also associated with a strong activation of the immune system. However, the specific influence of hypertension on postischemic inflammation has not been sufficiently investigated. Besides, the integration of hypertension and other important concomitant diseases is becoming a more regular tool in preclinical stroke modelling in order to expand the understanding of pathophysiological interactions and overcome the translational gap of new immunomodulatory therapies.
Aim: The main objective of this work is the identification of pathofunctional interations between the immune response after stroke and preexisting arterial hypertension. For this purpose, the central and peripheral inflammatory response after experimental stroke was investigated in a premorbid hypertensive animal model (spontaneously hypertensive rat, SHR) in comparison with normotensive animals by means of flow cytometric, histological and molecular biological methods. In addition, the hypertensive animal model was supposed to be assessed regarding its suitability for the investigation of immunological aspects in translational stroke research.
Material and Methods: For all animal experiments and organ removal, only male rats of the Wistar Kyoto and SHR strains aged 12 to 14 weeks were used. Induction of experimental stroke was performed using either a permanent transcranial stroke model or photochemical thrombosis model. Depending on the study group, animals were killed painlessly 1 or 4 days after infarct induction or sham surgery respectively. In addition to few neuroanatomic and neurofunctional readout parameters, immune cell distribution in the brain and blood was captured as primary variable. Therefore, brain cell isolates and whole blood samples labeled with a maximum of 8 fluorescence-coupled antibodies in different combinations were measured and analyzed in a 3-laser flow cytometer (FACSCanto II). Furthermore, immunofluorescence techniques were applied on cryopreserved brain sections in order to image spatial distribution of relevant immune cell populations and adhesion molecules by means of confocal microscopy (LSM710, Zeiss). In addition, the mRNA and protein expression of selective cytokines and adhesion molecules was determined in dissociated brain tissue. Depending on the targeted parameter statistical analyses were performed by using two-sample t-test, Wilcoxon rank-sum test, Pearson correlation coefficient or analysis of variance. A significance level of p<0.05 was set for all statistical methods.
Results: In addition to an enlarged lesion, in hypertensive animals an increased infiltration of cells of the innate immune system to the ischemic brain area was detected. A shift of the macrophage-granulocyte-ratio further indicated an altered inflammatory profile in hypertensive rats. Furthermore, an increased number of circulating monocytes and granulocytes were observed in animals with hypertension. In brain tissue of SHR after stroke, a decreased expression of the anti-inflammatory cytokine interleukin 10 were recorded along with increased expression levels of selective leukocyte-recruiting chemokines and an increased expression of the adhesion molecule ICAM-1 on infiltrating leukocytes. In addition, caused by different induction methods, model-dependent impact on the immune reaction could be identified.
Conclusion: The results clearly indicate a relationship between existing arterial hypertension and an increased inflammatory response in the brain after experimental stroke and point out potential underlying mechanisms. At the same time, by adopting a differentiated view of methodological and model-dependent influences, the work underscores the need for critical reflection and constant verification of preclinical results in different models. Finally the work validates the SHR strain as a suitable premorbid preclinical system for further translational research since it well models the influence of hypertension on the development and course of postischemic inflammation.:Inhaltsverzeichnis

Sponsorship phenomenon on Instagram : A qualitative study that examines how companies in the health supplement industry communicate with their target group with the help of private individuals
In this essay we have chosen to research how health supplement companies market themselves and in different ways reach out to a vast majority of people with the help of one of the biggest social media channels, Instagram. Itâs common nowadays that companies use social media as a fast, cheap and easy way to market their products to users. Either by promoting it on their own channel or by sponsorship, where they have popular people with a high amount of followers to encourage their viewers to buy their products. Instagram is rapidly growing and today have about 400 million active users every month and 40 % of all Swedish people that uses the Internet are a member of this application. Our research question is: âin which way do health supplement companies work with private individuals to communicate with their target group on Instagram and what is in the medium Instagram that makes it possible to use it for marketing and communication purposes?â By interviewing four individuals who are sponsored by different companies, two experts on the domain and by doing a netnographic observation that examines eight different Instagram accounts we were able to analyse and find the answer to our question. For health supplement companies to successfully communicate with their target group on Instagram it is important that they use the right person as an influencer. This is important to create high credibility towards the consumer. Itâs also important that the influencer describes the product in an interesting way and that the person has a lot of knowledge about the product. We also conclude that the most important function that Instagram can offer is the ability to distribute images and influence other users
Isolation of inflammatory cells from rat brain tissue after stroke
The pathophysiology of sterile inflammation following focal ischemic stroke is complex and not fully understood, but there is growing evidence that it offers several therapeutic options beyond the hitherto existing treatment strategies. The identification and quantification of infiltrating inflammatory cells in animal models of stroke is crucial both for understanding post-stroke inflammation and for drug target identification. Multicolor flow cytometry plays an important role in determining subtypes and quantity of leukocytes that infiltrate the brain tissue after stroke. Until now, most investigations have been performed in mice, most likely due to a significantly broader spectrum of disposable antibodies and available knockout models. High-Sulfated Glycosaminoglycans Prevent Coronavirus Replication
Coronaviruses (CoVs) are common among humans and many animals, causing respiratory or gastrointestinal diseases. Currently, only a few antiviral drugs against CoVs are available. Especially for SARS-CoV-2, new compounds for treatment of COVID-19 are urgently needed. In this study, we characterize the antiviral effects of two high-sulfated glycosaminoglycan (GAG) derivatives against SARS-CoV-2 and bovine coronaviruses (BCoV), which are both members of the Betacoronavirus genus. The investigated compounds are based on hyaluronan (HA) and chondroitin sulfate (CS) and exhibit a strong inhibitory effect against both CoVs. Yield assays were performed using BCoV-infected PT cells in the presence and absence of the compounds. While the high-sulfated HA (sHA3) led to an inhibition of viral growth early after infection, high-sulfated CS (sCS3) had a slightly smaller effect. Time of addition assays, where sHA3 and sCS3 were added to PT cells before, during or after infection, demonstrated an inhibitory effect during all phases of infection, whereas sHA3 showed a stronger effect even after virus absorbance. Furthermore, attachment analyses with prechilled PT cells revealed that virus attachment is not blocked. In addition, sHA3 and sCS3 inactivated BCoV by stable binding. Analysis by quantitative real-time RT PCR underlines the high potency of the inhibitors against BCoV, as well as B.1-lineage, Alpha and Beta SARS-CoV-2 viruses. Taken together, these results demonstrated that the two high-sulfated GAG derivatives exhibit low cytotoxicity and represent promising candidates for an anti-CoV therapy
Sterile inflammation after permanent distal MCA occlusion in hypertensive rats

Hypertension, a major cause and comorbidity of stroke, entails systemic vascular inflammation and may influence poststroke immune responses. This aspect is, however, underestimated in previous studies. Here we aimed to delineate the sequence of cellular inflammation after stroke in spontaneously hypertensive (SH) rats. Spontaneously hypertensive rats were subjected to permanent middle cerebral artery occlusion and killed after 1 or 4 days. Immune cells of the peripheral blood and those which have infiltrated the injured brain were identified and quantified by flow cytometry. The spatial distribution of myeloid cells and T lymphocytes, and the infarct volume were assessed by histology. We observed a concerted infiltration of immune cells into the ischemic brain of SH rats. At day 1, primarily neutrophils, monocytes, macrophages, and myeloid dendritic cells entered the brain, whereas the situation at day 4 was dominated by microglia, macrophages, lymphatic dendritic cells, and T cells. Postischemic inflammation did not cause secondary tissue damage during the subacute stage of experimental stroke in SH rats. Considering the intrinsic vascular pathology of SH rats, our study validates this strain for further translational research in poststroke inflammation
