    Überprüfung der Wirksamkeit des Trainings mit sozial unsicheren Kindern

    The aim of the present study was to evaluate the Training mit sozial unsicheren Kindern (Petermann & Petermann, 2006), a cognitive-behavioral treatment program developed for children aged 7 to 12 years suffering from (social) anxiety disorders. The effectiveness of the treatment program was examined with a (wait-list) control group design. The sample consisted of 25 children (13 girls, 12 boys) aged 7 to 12 years who met the ICD-10 diagnostic criteria for an anxiety disorder. The children were randomly assigned to either a treatment (n = 14) or a (wait-list) control group (n = 11). In both conditions social anxiety was assessed by self-, parent- and teacher-report measures before, immediately after and six months after treatment. Comorbid depression and social functioning in school were assessed with further questionnaires. The results of the present study indicate the short- and long-term effectiveness of the treatment program. The parents' report showed a significant reduction of (social) anxiety symptoms after treatment in the treatment group compared to the (wait-list) control group. Furthermore, according to parents' report the treatment group also had lower levels of depressive symptoms at posttreatment, but this pre-post-difference did not exceed the changes in the (wait-list) control group. The parent-reported treatment gains in anxiety and depressive symptoms were maintained at six-month follow-up. According to children's and teachers' ratings the children experienced reduction in anxiety symptoms and improvement in social functioning in school not until six-month follow-up. Sociodemographic variables (gender, age and intelligence) and clinical status (comorbid depression) were not found to be directly related to treatment outcome.     Evaluating embodied conversational agents in multimodal interfaces

    Based on cross-disciplinary approaches to Embodied Conversational Agents, evaluation methods for such human-computer interfaces are structured and presented. An introductory systematisation of evaluation topics from a conversational perspective is followed by an explanation of social-psychological phenomena studied in interaction with Embodied Conversational Agents, and how these can be used for evaluation purposes. Major evaluation concepts and appropriate assessment instruments – established and new ones – are presented, including questionnaires, annotations and log-files. An exemplary evaluation and guidelines provide hands-on information on planning and preparing such endeavours

    Coaggregation of FcεRI with FcγRIIB Inhibits Degranulation but Not Induction of Bcl-2 Family Members A1 and Bim in Mast Cells

    <p/> <p>The aggregation of high-affinity immunoglobulin E (IgE) receptors (FcεRI) on mast cells is a critical event in the initiation of an allergic reaction. Coengagement of FcεRI with immunoglobulin G (IgG) low-affinity receptor FcγRIIB/CD32 inhibits degranulation and the release of inflammatory mediators from mast cells and has therefore been proposed as a new therapeutic approach for the treatment of allergies. In this study, we investigated whether FcγRIIB, besides inhibiting degranulation, negatively regulates other signalling pathways downstream of FcεRI. For this, we determined the phosphorylation and/or expression of proteins involved in the regulation of mast-cell apoptosis. Coaggregation led to an attenuation of Akt phosphorylation but did not inhibit phosphorylation of transcription factor Foxo3a or its proapoptotic target, Bim. Similarly, FcεRI-dependent expression of the prosurvival gene A1 was not affected by coaggregation. Our data demonstrate that coengagement of FcεRI and FcγRIIB inhibits degranulation but not the signalling pathways regulating Bcl-2 family members Bim and A1.</p

    VISION - Vienna survey in Orion. III. Young stellar objects in Orion A

Reproduced with permission from Astronomy & Astrophysics. © 2018 ESOWe extend and refine the existing young stellar object (YSO) catalogs for the Orion A molecular cloud, the closest massive star-forming region to Earth. This updated catalog is driven by the large spatial coverage (18.3 deg^2, ~950 pc^2), seeing limited resolution (~0.7''), and sensitivity (Ks<19 mag) of the ESO-VISTA near-infrared survey of the Orion A cloud (VISION). Combined with archival mid- to far-infrared data, the VISTA data allow for a refined and more robust source selection. We estimate that among previously known protostars and pre-main-sequence stars with disks, source contamination levels (false positives) are at least ∼7% and ∼2.5%, respectively, mostly due to background galaxies and nebulosities. We identify 274 new YSO candidates using VISTA/Spitzer based selections within previously analyzed regions, and VISTA/WISE based selections to add sources in the surroundings, beyond previously analyzed regions. The WISE selection method recovers about 59% of the known YSOs in Orion A's low-mass star-forming part L1641, which shows what can be achieved by the all-sky WISE survey in combination with deep near-infrared data in regions without the influence of massive stars. The new catalog contains 2978 YSOs, which were classified based on the de-reddened mid-infrared spectral index into 188 protostars, 184 flat-spectrum sources, and 2606 pre-main-sequence stars with circumstellar disks. We find a statistically significant difference in the spatial distribution of the three evolutionary classes with respect to regions of high dust column-density, confirming that flat-spectrum sources are at a younger evolutionary phase compared to Class IIs, and are not a sub-sample seen at particular viewing angles.Peer reviewedFinal Accepted Versio

    Demographic and pregnancy‐related predictors of postnatal contraception uptake: A cross‐sectional study

    Objective: To examine the uptake of postnatal contraception (PNC) and experiences of PNC care across a geographical region of England. Design: Cross‐sectional online survey. Setting: The North East and North Cumbria Integrated Care System (ICS). Population: Women who had completed a pregnancy in the previous 3 years. Methods: The uptake of PNC by accessed method(s) and the availability of preferred method(s) is described, and adjusted odds ratios are reported for group differences in uptake by characteristics of interest. Main outcome measures: Uptake of medically prescribed/administered contraception and uptake of long‐acting reversible contraception (LARC) during the postnatal period, and access to preferred PNC methods. Results: Although almost half of respondents (47.1%; n = 1178) reinitiated some form of sexual activity during the postnatal period, only 38.7% (n = 969) of respondents accessed a medically prescribed/administered contraceptive method postnatally, and only 15.5% (n = 389) of respondents accessed a LARC. It is a matter of concern that 18.8% (n = 451) of respondents indicated that they were unable to access their preferred PNC. In multivariate analysis, younger age, lower household income, higher multiparity, operative delivery, unplanned pregnancy and not breastfeeding were significant predictors of higher PNC uptake. Conclusions: The uptake of PNC in this cohort was low, with almost a fifth of women unable to access their preferred method. However, there was some evidence that women belonging to groups perceived to be at risk of rapid repeat pregnancy were more likely to access reliable PNC methods

    Increased pore size of scaffolds improves coating efficiency with sulfated hyaluronan and mineralization capacity of osteoblasts

    Background: Delayed bone regeneration of fractures in osteoporosis patients or of critical-size bone defects after tumor resection are a major medical and socio-economic challenge. Therefore, the development of more effective and osteoinductive biomaterials is crucial. Methods: We examined the osteogenic potential of macroporous scaffolds with varying pore sizes after biofunctionalization with a collagen/high-sulfated hyaluronan (sHA3) coating in vitro. The three-dimensional scaffolds were made up from a biodegradable three-armed lactic acid-based macromer (TriLA) by cross-polymerization. Templating with solid lipid particles that melt during fabrication generates a continuous pore network. Human mesenchymal stem cells (hMSC) cultivated on the functionalized scaffolds in vitro were investigated for cell viability, production of alkaline phosphatase (ALP) and bone matrix formation. Statistical analysis was performed using student's t-test or two-way ANOVA. Results: We succeeded in generating scaffolds that feature a significantly higher average pore size and a broader distribution of individual pore sizes (HiPo) by modifying composition and relative amount of lipid particles, macromer concentration and temperature for cross-polymerization during scaffold fabrication. Overall porosity was retained, while the scaffolds showed a 25% decrease in compressive modulus compared to the initial TriLA scaffolds with a lower pore size (LoPo). These HiPo scaffolds were more readily coated as shown by higher amounts of immobilized collagen (+ 44%) and sHA3 (+ 25%) compared to LoPo scaffolds. In vitro, culture of hMSCs on collagen and/or sHA3-coated HiPo scaffolds demonstrated unaltered cell viability. Furthermore, the production of ALP, an early marker of osteogenesis (+ 3-fold), and formation of new bone matrix (+ 2.5-fold) was enhanced by the functionalization with sHA3 of both scaffold types. Nevertheless, effects were more pronounced on HiPo scaffolds about 112%. Conclusion: In summary, we showed that the improvement of scaffold pore sizes enhanced the coating efficiency with collagen and sHA3, which had a significant positive effect on bone formation markers, underlining the promise of using this material approach for in vivo studies. © 2019 The Author(s)

    Children’s mental health problems and their relation to parental stress in foster mothers and fathers

These children often show adverse mental health outcomes reflected in increased externalizing and internalizing problems. It is expected that these adverse outcomes are associated with increased parental stress levels experienced by foster mothers as well as foster fathers. Methods The study sample included 79 children living in foster families and 140 children living in biological families as comparison group. The age of the children ranged from 2 to 7 years. Mental health problems were assessed with the Child Behavior Checklist, while parenting stress was measured with a parenting stress questionnaire including subscales on the amount of experienced stress and the amount of perceived support. The Child Behavior Checklist assessments were based mainly on maternal reports, while the parental stress assessments were based on maternal as well as paternal reports. Results As expected the results showed increased externalizing and internalizing scores for the foster children accompanied by increased parental stress experiences in the foster family sample (however only in the maternal, but not in the paternal stress reports). The stress differences between the foster and biological family groups disappeared, when the children’s mental health problem scores were included as covariates. Moreover, especially the externalizing scores were strong predictors of parental stress in both, the groups of foster and biological parents. The amount of perceived social support was associated with reduced parental stress, but only in the group of biological fathers. Conclusion The emergence of parental stress in biological as well as foster parents is closely related to child characteristics (mainly externalizing child problems). Possible implications for the reduction of parental stress are discussed as a consequence of the present results

    Polymorphisms in the tyrosine kinase 2 and interferon regulatory factor 5 genes are associated with systemic lupus erythematosus

Please click on the hyperlink in Additional Links fieldSystemic lupus erythematosus (SLE) is a complex systemic autoimmune disease caused by both genetic and environmental factors. Genome scans in families with SLE point to multiple potential chromosomal regions that harbor SLE susceptibility genes, and association studies in different populations have suggested several susceptibility alleles for SLE. Increased production of type I interferon (IFN) and expression of IFN-inducible genes is commonly observed in SLE and may be pivotal in the molecular pathogenesis of the disease. We analyzed 44 single-nucleotide polymorphisms (SNPs) in 13 genes from the type I IFN pathway in 679 Swedish, Finnish, and Icelandic patients with SLE, in 798 unaffected family members, and in 438 unrelated control individuals for joint linkage and association with SLE. In two of the genes--the tyrosine kinase 2 (TYK2) and IFN regulatory factor 5 (IRF5) genes--we identified SNPs that displayed strong signals in joint analysis of linkage and association (unadjusted P<10(-7)) with SLE. TYK2 binds to the type I IFN receptor complex and IRF5 is a regulator of type I IFN gene expression. Thus, our results support a disease mechanism in SLE that involves key components of the type I IFN system
