    Cortistain is expressed in a distinct subset of cortical interneurons

    Cortistatin is a presumptive neuropeptide that shares 11 of its 14 amino acids with somatostatin. In contrast to somatostatin, administration of cortistatin into the rat brain ventricles specifically enhances slow wave sleep, apparently by antagonizing the effects of acetylcholine on cortical excitability. Here we show that preprocortistatin mRNA is expressed in a subset of GABAergic cells in the cortex and hippocampus that partially overlap with those containing somatostatin. A significant percentage of cortistatin-positive neurons is also positive for parvalbumin. In contrast, no colocalization was found between cortistatin and calretinin, cholecystokinin, or vasoactive intestinal peptide. During development there is a transient increase in cortistatin-expressing cells in the second postnatal week in all cortical areas and in the dentate gyrus. A transient expression of preprocortistatin mRNA in the hilar region at P16 is paralleled by electrophysiological changes in dentate granule cells. Together, these observations suggest mechanisms by which cortistatin may regulate cortical activity

    Model predictive control of a microgrid with energy-stored quasi-Z-source cascaded H-bridge multilevel inverter and PV systems

    This paper presents a new energy management system (EMS) based on model predictive control (MPC) for a microgrid with solar photovoltaic (PV) power plants and a quasi-Z-source cascaded H-bridge multilevel inverter that integrates an energy storage system (ES-qZS-CHBMLI). The system comprises three modules, each with a PV power plant, quasi-impedance network, battery energy storage system (BESS), and voltage source inverter (VSI). Traditional EMS methods focus on distributing the power among the BESSs to balance their state of charge (SOC), operating in charging or discharging mode. The proposed MPC-EMS carries out a multi-objective control for an ES-qZS-CHBMLI topology, which allows an optimized BESS power distribution while meeting the system operator requirements. It prioritizes the charge of the BESS with the lowest SOC and the discharge of the BESS with the highest SOC. Thus, both modes can coexist simultaneously, while ensuring decoupled power control. The MPC-EMS proposed herein is compared with a proportional sharing algorithm based on SOC (SOC-EMS) that pursues the same objectives. The simulation results show an improvement in the control of the power delivered to the grid. The Integral Time Absolute Error, ITAE, achieved with the MPC-EMS for the active and reactive power is 20 % and 4 %, respectively, lower than that obtained with the SOC-EMS. A 1,3 % higher charge for the BESS with the lowest SOC is also registered.     Clinical aspects of visceral leishmaniasis caused by L. infantum in adults. Ten years of experience of the largest outbreak in Europe: what have we learned?

    BACKGROUND: An outbreak of leishmaniasis caused by Leishmania infantum was declared in the southwest of the Madrid region (Spain) in June 2009. This provided a unique opportunity to compare the management of visceral leishmaniasis (VL) in immunocompetent adults (IC-VL), patients with HIV (HIV-VL) and patients receiving immunosuppressants (IS-VL). METHODS: A cohort of adults with VL, all admitted to the Hospital Universitario de Fuenlabrada between June 2009 and June 2018, were monitored in this observational study, recording their personal, epidemiological, analytical, diagnostic, treatment and outcome variables. RESULTS: The study population was made up of 111 patients with VL (10% HIV-VL, 14% IS-VL, 76% IC-VL). Seventy-one percent of the patients were male; the mean age was 45 years (55 years for the IS-VL patients, P = 0.017). Fifty-four percent of the IC-VL patients were of sub-Saharan origin (P = 0.001). Fever was experienced by 98% of the IC-VL patients vs 73% of the LV-HIV patients (P = 0.003). Plasma ferritin was > 1000 ng/ml in 77% of the IC-VL patients vs 17% of the LV-HIV patients (P = 0.007). Forty-two percent of patients fulfilled the criteria for haemophagocytic lymphohistiocytosis. RDT (rK39-ICT) serological analysis returned sensitivity and specificity values of 45% and 99%, respectively, and ELISA/iIFAT returned 96% and 89%, respectively, with no differences in this respect between patient groups. Fourteen (13.0%) patients with VL experienced treatment failure, eight of whom were in the IC-VL group. Treatment with < 21 mg/kg (total) liposomal amphotericin B (LAB) was associated with treatment failure in the IC-VL patients [P = 0.002 (OR: 14.7; 95% CI: 2.6-83.3)]. CONCLUSIONS: IS-VL was more common than HIV-VL; the lack of experience in dealing with IS-VL is a challenge that needs to be met. The clinical features of the patients in all groups were similar, although the HIV-VL patients experienced less fever and had lower plasma ferritin concentrations. RDT (rK39-ICT) analysis returned a good specificity value but a much poorer sensitivity value than reported in other scenarios. The patients with HIV-VL, IS-VL and IC-VL returned similar serological results.     Cannabis use selectively modulates circulating biomarkers in the blood of schizophrenia patients

    Cannabis use disorder is frequent in schizophrenia patients, and it is associated with an earlier age of onset and poor schizophrenia prognosis. Serotonin 2A receptors (5-HT2AR) have been involved in psychosis and, like Akt kinase, are known to be modulated by THC. Likewise, endocannabinoid system dysregulation has been suggested in schizophrenia. The presence of these molecules in blood makes them interesting targets, as they can be evaluated in patients by a minimally invasive technique. The aim of the present study was to evaluate 5-HT2AR protein expression and the Akt functional status in platelet homogenates of subjects diagnosed with schizophrenia, cannabis use disorder, or both conditions, compared with age- and sex-matched control subjects. Additionally, endocannabinoids and pro-inflammatory interleukin-6 (IL-6) levels were also measured in the plasma of these subjects. Results showed that both platelet 5-HT2AR and the active phospho (Ser473)Akt protein expression were significantly increased in schizophrenia subjects, whereas patients with a dual diagnosis of schizophrenia and cannabis use disorder did not show significant changes. Similarly, plasma concentrations of anandamide and other lipid mediators such as PEA and DEA, as well as the pro-inflammatory IL-6, were significantly increased in schizophrenia, but not in dual subjects. Results demonstrate that schizophrenia subjects show different circulating markers pattern depending on the associated diagnosis of cannabis use disorder, supporting the hypothesis that there could be different underlying mechanisms that may explain clinical differences among these groups.     Referees visual behaviour during offside situations in football

    El estudio analiza el comportamiento visual de 8 árbitros de fútbol durante la percepción del fuera de juego en laboratorio. La tarea consiste en percibir una secuencia de 24 ensayos en una pantalla (5x3m), donde se proyectan situaciones reducidas de juego y que concluyen con una posible acción de fuera de juego. Los participantes deben percibir la secuencia con el ASL Eye Tracking SE5000, y pulsar un puntero laser hacia la pantalla en aquellos ensayos con fuera de juego. Las variables a manipular son la distancia y el ángulo con que se perciben las acciones de fuera de juego. Las variables dependientes son el número y tiempo (media) de fijaciones visuales y el porcentaje de acierto. Los resultados muestran que la distancia y ángulo influyen en el comportamiento visual de los árbitros. Además, son más eficaces detectando el fuera de juego con ángulos pequeños y en distancias cercanas y medias.The study examines the 8 football referees´ visual behavior during the perception of offside actions carried out in a laboratory setting. The task consists on perceiving a rally of 24 trials onto a large screen (5x3m) with reduced play situations and that could conclude with an offside action. Participants perceive the sequence with the ASL Eye Tracking SE5000 and press a laser pointer toward the screen in those trials with offside action. Variables to manipulate are the distance and angle in which the offside trials are perceived. The dependent variables are the number and time (average) of visual fixations and the success rate. The results show that the distance and angle changes the referees´ visual behavior. Moreover, they have higher success rate when they perceive trials with small angles, concretely with short and medium distances.peerReviewe

    Obesity features excessive fat accumulation in several body tissues and induces a state of chronic low-grade inflammation that contributes to the development of diabetes, steatosis, and insulin resistance. Recent research has shown that this chronic inflammation is crucially dependent on p38 pathway activity in macrophages, suggesting p38 inhibition as a possible treatment for obesity comorbidities. Nevertheless, we report here that lack of p38 activation in myeloid cells worsens high-fat diet-induced obesity, diabetes, and steatosis. Deficient p38 activation increases macrophage IL-12 production, leading to inhibition of hepatic FGF21 and reduction of thermogenesis in the brown fat. The implication of FGF21 in the phenotype was confirmed by its specific deletion in hepatocytes. We also found that IL-12 correlates with liver damage in human biopsies, indicating the translational potential of our results. Our findings suggest that myeloid p38 has a dual role in inflammation and that drugs targeting IL-12 might improve the homeostatic regulation of energy balance in response to metabolic stress.S

    Hall effect measurement is one of the most powerful methods for obtaining information about transport mechanisms in polycrystalline semiconductor compounds that constitute the basis for understanding the sensing function of semiconductor gas sensors. The presence of grain boundaries represents the essential difference between single-crystal and polycrystalline semiconductors. The boundaries are important because they generally contain fairly high densities of interface states which trap free carriers from the bulk of the grains. In this paper the grain size of the semiconductor (calculated by the XRGA technique) and Hall effect measurements are used in order to obtain conduction-band profiles. Depending on the preparation method (reactive sputtering, electron beam, serigraphy), three types of conduction control can be distinguished.     Effects of Albumin Treatment on Systemic and Portal Hemodynamics and Systemic Inflammation in Patients With Decompensated Cirrhosis

    BACKGROUND & AIMS: We investigated the effect of albumin treatment (20% solution) on hypoalbuminemia, cardiocirculatory dysfunction, portal hypertension, and systemic inflammation in patients with decompensated cirrhosis with and without bacterial infections. METHODS: We performed a prospective study to assess the effects of long-term (12 weeks) treatment with low doses (1 g/kg body weight every 2 weeks) and high doses (1.5 g/kg every week) of albumin on serum albumin, plasma renin, cardiocirculatory function, portal pressure, and plasma levels of cytokines, collecting data from 18 patients without bacterial infections (the Pilot-PRECIOSA study). We also assessed the effect of short-term (1 week) treatment with antibiotics alone vs the combination of albumin plus antibiotics (1.5 g/kg on day 1 and 1 g/kg on day 3) on plasma levels of cytokines in biobanked samples from 78 patients with bacterial infections included in a randomized controlled trial (INFECIR-2 study). RESULTS: Circulatory dysfunction and systemic inflammation were extremely unstable in many patients included in the Pilot-PRECIOSA study; these patients had intense and reversible peaks in plasma levels of renin and interleukin 6. Long-term high-dose albumin, but not low-dose albumin, was associated with normalization of serum level of albumin, improved stability of the circulation and left ventricular function, and reduced plasma levels of cytokines (interleukin 6, granulocyte colony-stimulating factor, interleukin 1 receptor antagonist, and vascular endothelial growth factor) without significant changes in portal pressure. The immune-modulatory effects of albumin observed in the Pilot-PRECIOSA study were confirmed in the INFECIR-2 study. In this study, patients given albumin had significant reductions in plasma levels of cytokines. CONCLUSIONS: In an analysis of data from 2 trials (Pilot-PRECIOSA study and INFECIR-2 study), we found that albumin treatment reduced systemic inflammation and cardiocirculatory dysfunction in patients with decompensated cirrhosis. These effects might be responsible for the beneficial effects of albumin therapy on outcomes of patients with decompensated cirrhosis. ClinicalTrials.gov, Numbers: NCT00968695 and NCT03451292
