    En este artículo se hace la presentación de un modelo de detección de congestión el cual se implementó en la red LAN de Universidad Nacional sede Bogotá. La idea de este modelo es que con ayuda de los usuarios se haga una detección de los problemas de congestión en la red, de modo que se hace también el diseño de un sistema multiagente que es la base para el modelo de detección. En el proceso se han definido cinco variables de interés las cuales definen el comportamiento de la red LAN y son presentadas en los resultados obtenidos del proceso de detección

    Validating subcellular localization prediction tools with mycobacterial proteins

    <p>Abstract</p> <p>Background</p> <p>The computational prediction of mycobacterial proteins' subcellular localization is of key importance for proteome annotation and for the identification of new drug targets and vaccine candidates. Several subcellular localization classifiers have been developed over the past few years, which have comprised both general localization and feature-based classifiers. Here, we have validated the ability of different bioinformatics approaches, through the use of SignalP 2.0, TatP 1.0, LipoP 1.0, Phobius, PA-SUB 2.5, PSORTb v.2.0.4 and Gpos-PLoc, to predict secreted bacterial proteins. These computational tools were compared in terms of sensitivity, specificity and Matthew's correlation coefficient (MCC) using a set of mycobacterial proteins having less than 40% identity, none of which are included in the training data sets of the validated tools and whose subcellular localization have been experimentally confirmed. These proteins belong to the TBpred training data set, a computational tool specifically designed to predict mycobacterial proteins.</p> <p>Results</p> <p>A final validation set of 272 mycobacterial proteins was obtained from the initial set of 852 mycobacterial proteins. According to the results of the validation metrics, all tools presented specificity above 0.90, while dispersion sensitivity and MCC values were above 0.22. PA-SUB 2.5 presented the highest values; however, these results might be biased due to the methodology used by this tool. PSORTb v.2.0.4 left 56 proteins out of the classification, while Gpos-PLoc left just one protein out.</p> <p>Conclusion</p> <p>Both subcellular localization approaches had high predictive specificity and high recognition of true negatives for the tested data set. Among those tools whose predictions are not based on homology searches against SWISS-PROT, Gpos-PLoc was the general localization tool with the best predictive performance, while SignalP 2.0 was the best tool among the ones using a feature-based approach. Even though PA-SUB 2.5 presented the highest metrics, it should be taken into account that this tool was trained using all proteins reported in SWISS-PROT, which includes the protein set tested in this study, either as a BLAST search or as a training model.</p

    Evaluation of conidia production and mycelial growth in solid culture media from native strains of entomopathogenic fungi isolated from citrus-growing areas of México

    It is important to know the ability of native strains to carry out the process of sporulation and growth in different conditions and to determine their possible potential as biological control of pests of agricultural importance, mainly in citrus areas. The objective of this study was to evaluate five different solid culture media for the determination of the production of conidia and mycelial growth of twenty-five native isolates from the Mexican states of Sinaloa, San Luis Potosí, Nuevo León and Tamaulipas, and three collection strains using five different solid culture media. The results showed no statistical significant difference between the five media tested for the production of conidia per milliliter and mycelial growth among different isolates. However between isolates a significant difference was found. For conidia per milliliter, the isolate with the highest production of conidia was HIB-4 with an average of 4.85 x 109 (spores/ml). With respect to mycelial growth strain Ma presented the highest value with 8.06 cm on average.Key words: Entomopathogenic fungi, esporulation, conidia, mycelial growth

    Evaluation of the quality of the Voluntary Geographic Information for the road network in Bogotá D.C

    The production of Voluntary Geographic Information has been growing considerably and continues to be an active area of research. However, the lack of knowledge about the quality of information generated on a voluntary and participatory basis raises challenges and questions about its use. In the review carried out for the Colombian case, no studies related to the subject were identified; consequently, this study is presented on the evaluation of the quality of this type of information on the road network of Bogotá with respect to completeness, positional accuracy and thematic accuracy. This evaluation was carried out by means of a semi-automatic process that uses a mobile buffer and the centroid of the roads to make the corresponding comparisons between two data sources. The results found reveal that the method used allowed to compare up to 85.0% of the data, and that the OpenStreetMap mesh has a completeness of 85.4%, over the entire area of Bogotá. A positional accuracy of 3.98 m and a thematic accuracy related to the percentage of error in the attributes: Road hierarchy, direction of flow and road naming of 35.8%, 15.0% and 34.6% respectively. The quality evaluated through completeness, positional and thematic accuracy in synergistic terms is deficient with respect to the minimum quality levels established in the standard data model, however, the evaluation for each of the attributes shows an acceptable quality in terms of completeness and thematic accuracy

    Efecto de las intervenciones con ingesta de leguminosas y/o ejercicio supervisado sobre el perfil lipídico de mujeres jóvenes, sanas y sedentarias

    Objective: To contribute to the knowledge of some aspects of the Healthy Life Style by studying the effects of including legumes in the diet and exercise at two intensity levels, along with the lipid profile of young sedentary women living at 2640 meters above sea level. Materials and methods: The study included a non-randomized clinical trial with four intervention groups: exercise at 45% VO2 peak plus legumes in diet, exercise at 65% VO2 peak plus legumes in diet, only exercise at 65% VO2 peak, and only inclusion of legumes in diet. In each group, 20 to 23 sedentary women were included. The intervention was carried out for four weeks, three days a week. Exercise prescription was based on measurement of VO2 peak by ergospirometry; and the current intervention was monitored with heart-rate monitors. The outcome variables were total serum cholesterol, LDL cholesterol, HDL cholesterol, and triacylglycerols measured at baseline and after two and four weeks of intervention. Results: The measurements taken of participants in the group of exercise at 65% VO2 peak plus legumes in diet showed a reduction of 19.8 mg/dl in total cholesterol, of 21.8 mg/dl of LDL cholesterol, of 20.7 mg/dl of tracylglycerol, and an increase of 6.2 mg/dl of HDL cholesterol. The serum levels of HDL also increased in the group of only exercise at 65% VO2 peak. No significant changes in serum levels were documented for participants in the group with only dietary modifications. Conclusions: The results suggest that different interventions that meet some of the criteria for healthy eating and life style show different effects with regards to the level of change in the lipid profile components. Objetivo: Contribuir al conocimiento de algunos aspectos prácticos para llevar a cabo una alimentación y estilo de vida saludable, mediante el estudio de los efectos en el perfil lipídico de la inclusión de leguminosas (fríjol, lenteja y garbanzos) en la dieta de mujeres sanas pero sedentarias, acompañada de ejercicio con dos niveles de intensidad, moderada y alta, efectuados a una altura de 2,640 metros sobre el nivel del mar (msnm) Material y métodos: Estudio de intervención clínica no aleatorizada con cuatro grupos de intervención: ejercicio a 45% VO2 pico + leguminosas, ejercicio a 65% VO2 pico + leguminosas, sólo ejercicio a 45% VO2 picoy sólo inclusión de leguminosas en la dieta. En cada grupo se incluyeron de 20 a 23 mujeres jóvenes sedentarias evaluadas mediante el International Physical Activity Questionnaire (IPAQ), y la intervención se llevó a cabo durante 4 semanas. La prescripción del ejercicio se hizo previa determinación del VO2 pico por ergo espirometría, y su realización se monitoreó con sensores de frecuencia cardíaca. El efecto de la intervención se evaluó mediante los cambios en los niveles séricos de colesterol total (CT), colesterol LDL (cLDL), colesterol HDL (cHDL) y triacilgliceroles (TAG), tomados a las semanas cero, dos y cuatro. Resultados: En el grupo «ejercicio a 65% del VO2 pico, más inclusión de leguminosas en la dieta» se obtuvieron los siguientes resultados significantes: una reducción promedio del colesterol total de 19.8 mg/dl (p=0.0015), del colesterol LDL de 21.8 mg/dl (p=0.0001), un aumento de colesterol HDL de 6.2 mg/dl (p=0.0001) y una disminución de los triacilgliceroles de 20.7 mg/dl (p=0.0001). En el grupo de sólo ejercicio a 65% del VO2 pico se obtuvo un aumento del HDL. No hubo modificaciones al perfil lipídico en el grupo correspondiente a sólo inclusión de leguminosas en la alimentación. Conclusiones: Los resultados sugieren que en mujeres jóvenes sedentarias que viven a 2,640 metros sobre el nivel del mar, la combinación de ejercicio de intensidad alta e inclusión de leguminosas (fríjol, lenteja o garbanzos) producen un mayor cambio benéfico en el perfil lipídico que una intervención con sólo ejercicio de intensidad alta o sólo inclusión de leguminosas en la alimentación. Los cambios benéficos significantes se obtuvieron luego de 4 semanas de intervención en el colesterol total, colesterol LDL, colesterol HDL y triacilgliceroles, lo que pone de presente la bondad de esta intervención, así como la utilidad del perfil lipídico como indicador temprano de cambios en los estilos de vida. Las intervenciones estudiadas, compatibles con alimentación y estilo de vida saludables, producen efectos diferentes en los niveles del perfil lipídico estudiados, y al poner de presente las debilidades de un diseño no aleatorizado, se puede recomendar que se insista en las intervenciones de salud pública que contemplen cambios en la dieta y cambios en la actividad física

    Identification of plasmodium vivax proteins with potential role in invasion using sequence redundancy reduction and profile hidden markov models

    Background: This study describes a bioinformatics approach designed to identify Plasmodium vivax proteins potentially involved in reticulocyte invasion. Specifically, different protein training sets were built and tuned based on different biological parameters, such as experimental evidence of secretion and/or involvement in invasion-related processes. A profile-based sequence method supported by hidden Markov models (HMMs) was then used to build classifiers to search for biologically-related proteins. The transcriptional profile of the P. vivax intra-erythrocyte developmental cycle was then screened using these classifiers. Results: A bioinformatics methodology for identifying potentially secreted P. vivax proteins was designed using sequence redundancy reduction and probabilistic profiles. This methodology led to identifying a set of 45 proteins that are potentially secreted during the P. vivax intra-erythrocyte development cycle and could be involved in cell invasion. Thirteen of the 45 proteins have already been described as vaccine candidates; there is experimental evidence of protein expression for 7 of the 32 remaining ones, while no previous studies of expression, function or immunology have been carried out for the additional 25. Conclusions: The results support the idea that probabilistic techniques like profile HMMs improve similarity searches. Also, different adjustments such as sequence redundancy reduction using Pisces or Cd-Hit allowed data clustering based on rational reproducible measurements. This kind of approach for selecting proteins with specific functions is highly important for supporting large-scale analyses that could aid in the identification of genes encoding potential new target antigens for vaccine development and drug design. The present study has led to targeting 32 proteins for further testing regarding their ability to induce protective immune responses against P. vivax malaria

    Emergence of the Stoner-Wohlfarth astroid in thin films at dynamic regime

    The Stoner-Wohlfarth (SW) model is the simplest model that describes adequately the magnetization reversal of nanoscale systems that are small enough to contain single magnetic domains. However for larger sizes where multi-domain effects are present, e.g., in thin films, this simple macrospin approximation fails and the experimental critical curve, referred as SW astroid, is far from its predictions. Here we show that this discrepancy could vanish also in extended system. We present a detailed angular-dependent study of magnetization reversal dynamics of a thin film with well-defined uniaxial magnetic anisotropy, performed over 9 decades of applied field sweep rate (dH/dt). The angular-dependent properties display a gradual transition from domain wall pinning and motion-like behaviour to a nucleative single-particle one, as dH/dt increases. Remarkably, in the high dynamic regime, where nucleation of reversed domains is the dominant mechanism of the magnetization reversal (nucleative regime), the magnetic properties including the astroid become closer to the ones predicted by SW model.     RICORS2040 : The need for collaborative research in chronic kidney disease

    Chronic kidney disease (CKD) is a silent and poorly known killer. The current concept of CKD is relatively young and uptake by the public, physicians and health authorities is not widespread. Physicians still confuse CKD with chronic kidney insufficiency or failure. For the wider public and health authorities, CKD evokes kidney replacement therapy (KRT). In Spain, the prevalence of KRT is 0.13%. Thus health authorities may consider CKD a non-issue: very few persons eventually need KRT and, for those in whom kidneys fail, the problem is 'solved' by dialysis or kidney transplantation. However, KRT is the tip of the iceberg in the burden of CKD. The main burden of CKD is accelerated ageing and premature death. The cut-off points for kidney function and kidney damage indexes that define CKD also mark an increased risk for all-cause premature death. CKD is the most prevalent risk factor for lethal coronavirus disease 2019 (COVID-19) and the factor that most increases the risk of death in COVID-19, after old age. Men and women undergoing KRT still have an annual mortality that is 10- to 100-fold higher than similar-age peers, and life expectancy is shortened by ~40 years for young persons on dialysis and by 15 years for young persons with a functioning kidney graft. CKD is expected to become the fifth greatest global cause of death by 2040 and the second greatest cause of death in Spain before the end of the century, a time when one in four Spaniards will have CKD. However, by 2022, CKD will become the only top-15 global predicted cause of death that is not supported by a dedicated well-funded Centres for Biomedical Research (CIBER) network structure in Spain. Realizing the underestimation of the CKD burden of disease by health authorities, the Decade of the Kidney initiative for 2020-2030 was launched by the American Association of Kidney Patients and the European Kidney Health Alliance. Leading Spanish kidney researchers grouped in the kidney collaborative research network Red de Investigación Renal have now applied for the Redes de Investigación Cooperativa Orientadas a Resultados en Salud (RICORS) call for collaborative research in Spain with the support of the Spanish Society of Nephrology, Federación Nacional de Asociaciones para la Lucha Contra las Enfermedades del Riñón and ONT: RICORS2040 aims to prevent the dire predictions for the global 2040 burden of CKD from becoming true
