    Thermal energy storage in a confined aquifer: Experimental results

Copyright 1979 American Geophysical UnionTo aid in testing the idea of storing thermal energy in aquifers, an experiment was performed by Auburn University in which 54,784 m3 of water was pumped from a shallow supply aquifer, heated to an average temperature of 55°C, and injected into a deeper confined aquifer where the ambient temperature was 20°C. After a storage period of 51 days, 55,345 m3 of water were produced from the confined aquifer. Throughout the experiment, which lasted approximately 6 months, groundwater temperatures were recorded at six depths in each of 10 observation wells, and hydraulic heads were recorded in five observation wells. In order to prevent errors due to thermal convection, most of the observation wells recording temperature had to be backfilled with sand. During the 41-day production period, the temperature of the produced water varied from 55° to 33°C, and 65% of the injected thermal energy was recovered. At no time was an appreciable amount of free thermal convection observed in the storage formation. The dominant heat dissipation mechanisms appeared to be hydrodynamic thermal dispersion and possible mixing of cold and hot water induced by clogging and unclogging of the injection-production well. On the basis of laboratory and field studies, it was concluded that clogging of the injection well, which constituted the major technical problem during the experiment, was caused by the freshwater-sensitive nature of the storage aquifer. Due to the relatively low concentration of cations in the supply water, clay particles would swell, disperse, and migrate until they became trapped in the relatively small pores connecting the larger pores. Surging the pump and back washing the injection well would dislodge the clogging particles and temporarily improve the storage formation permeability. The phenomenon seems largely independent of temperature because it was reproduced in the laboratory with unheated water. It may, however, depend on pore velocity. Future research should be directed toward procedures for selecting storage aquifers that will have minimal susceptibility to clogging and other geochemical problems. Procedures for overcoming such difficulties are needed also because clogging and related phenomena will be more the rule than the exception. Designing an aquifer thermal storage system for maximum energy recovery would involve selecting an appropriate aquifer, analyzing the effects of hydrodynamic thermal dispersion and thermal convection if it is predicted to occur, anticipating geochemical problems, designing the optimum supply-injection-production well configuration and injecting a sufficiently large volume of heated water to realize economies of scale related to increasing volume-surface area ratio

    MEDLAB Sicilia Le occasioni per l\u2019innovazione sociale e territoriale MEDLAB in Sicily An opportunity for social and territorial innovation

    The volume contains written in italian and english language on the reflections and the final results of international partnership experience by Medlab (project promoted by European Commission, MED Program) concerning the case of Living Labs in Sicily. Gelardi and Salemi showed the action of the regional innovation instruments of urban and regional planning at institutional regional level and in particular the role of experience of international cooperation in the comparison of experiences of different kind of territorial innovation in the Euro-Mediterranean area. Marsh shows as the MedLab project has been to examine the current and potential role of Living Labs within regional development policy and the questions thrown up by considering Living Labs as a policy tool. This means looking at issues of development policy and linking their relevance to broader territorial innovation strategies. In addition, governance issues arise for coordinating different Living Lab initiatives within a territorial domain, with the aim of maximising benefits not only to ICT-based innovation and the local knowledge economy, but also to the fields of application that Living Labs address: the environment, the economy, social and government services, etc. The process has thus involved first looking at how these questions can be addressed from the standpoints of different types of actors and how they can engage in reciprocal learning processes. The idea is to develop a model of \u201cinnovation literacy\u201d for local authorities and policy makers, namely the capacity to structure the demand for and supply of ICT-based innovation in order to maximise the concrete benefits to specific policies and initiatives. The ultimate vision is of a virtuous circle whereby regional development authorities apply the Living Lab model in an increasing array of fields and the ICT industry increasingly recognises the value proposition of engaging in co-design processes in concrete local and regional development initiatives. In the following pages, we explore the experiences developed in the MedLab project according to three issues: a) a first exploration of the concept of \u201cterritorial innovation\u201d at the basis of the MedLab hypothesis, and its potential impact on policy and governance; b) The case story of the formation of the original TLL-Sicily partnership in 2007 and how that experience shaped the MedLab workplan; c) The summative conclusions of the MedLab project, exploring the concept of an emergent macro-regional Living Lab. Giambalvo e Lucido involved the analysis of urban change in a social and economic context particularly full of difficulties as the town of Favara. Their contribution tries to define the processes of social innovation when they born and move the first steps, this contribution aims to collect the challenge born inside the Medlab project \u2013 Mediterranean Living Lab for the Territorial Innovation to reflect on some of the outcomes of the Living Lab approach to support territorial innovation in the geographical areas of Sicily concerned by the pilot cases of MedLab (the spatial planning of the province of Ragusa and the strategic plan of the municipality of Favara). More specifically, the remark starts from a work of research on the field and facilitation of communicative processes aimed to identify what dynamics of social innovation, such as transactions among the social actors and what new micro-economic ecosystems are developing in the municipality of Favara, whose background has been explored and treated as territorial Living Lab. Di Bono e Parisi describe the way of the Province of Ragusa tried to initiate arrangements to create a Living Lab from experience of local planning and addressing to the involvement of the local firm to obtain a dinamic digital mapping of local creativity. In the contribution the autors try to show that the concept of innovation is still evolving and the market is not disappearing from this innovation concept, but it is one of its components; the social dimension is increasingly relevant, as well as the institutional innovation. In fact, public boards, dealing with the themes of territorial innovation, can re-design their role starting from networking open innovation approaches and building up public-private partnerships able to activate technological and social innovation processes in a participatory way. Trapani present a cotribution on theme about the report of launching a citizen initiative for the mobilization of the social capital in the second Constituency of Palermo; it includes the Brancaccio neighborhood very famous for the murder of Father Puglisi. The initiative is designed as an integrated program of architectural, urban, cultural, social, environmental and economic qualification for the development (also for touristic aims) of Castle Maredolce and (oncoming) park close the monument and the nearby gardens. The proposal which is maturing in this period, falls within the framework of infrastructural transformations taking place in the districts of Brancaccio and Bandita and compared to the new role of the metropolitan city in the new economies of the Mediterranean, namely in a context in which Sicily seems to remain more at the edge of Europe and see, in the cautious use of its cultural resources, a way of survival. The proposal initially moves from the definition of projects and visions for the architectural setting and the urban and social redefinition of a new square in front of an important Moorish Castle undergoing restoration. Furthermore, the interest is moved to the second municipal constituency. The integrated urban program consists of interventions which specify the contents of the strategic plan of Palermo for the second constituency. The urban part, specifically, is characterized by the presence of important infrastructure and services for the production of small and medium-sized enterprises. It has been possible to examine the characteristics of the citizens, their expectations in terms of their objectives and concerning the concrete possibilities of integrated planning and design within participatory and urban sphere on the background of the failure of urban traditional planning tools

    Spinach, a cyclops, and the search for a cure for prostate cancer

    Abstract only availableOther than skin cancer, prostate cancer is the most prevalent form of cancer in men. The likelihood of developing prostate cancer increases with age; so that nearly every male will die with some form of prostate cancer though most likely not from the cancer itself. Prostate cancer is regulated by endocrine and dietary factors, as well as genetic predisposition. In the male reproductive tract, Sonic Hedgehog (Shh) signaling is necessary for the development of the prostate. It has also been found to be extremely important in the growth of a number of tumor types including prostate cancer, basal cell carcinoma (skin cancer), medulloblastoma, glioma, sarcoma, tumors of the digestive tract, small cell lung cancer and pancreatic carcinoma. The Lubahn lab has been studying the roles of Estrogen Receptors and estrogenic compounds on prostate cancer, and has recently used a series of phytoestrogens, natural estrogens found in plants, to inhibit the hedgehog-signaling pathway. One natural product, cyclopamine, is able to inhibit the pathway and has been shown to inhibit prostate cancer cell growth both in vitro and in vivo xenograft models. Additionally, some flavanoid compounds in spinach have been found to have a similar chemical structure to several other phytoestrogens that have been shown to have preventative effects on prostate cancer. A competitive binding assay was performed using various doses of cyclopamine and an unpurified spinach extract to determine Kd for both compounds to ER alpha and ER beta. The compounds were also tested in the NIH-3T3 Shh Light II cells, which have a stably transfected Gli-Luciferase reporter, to see if they downregulated the hedgehog signaling pathway.Food for the 21st Century Undergraduate Research Program in Nutritional Science

    Substrate Micropatterning as a New in Vitro Cell Culture System to Study Myelination

    Artículo de publicación ISIMyelination is a highly regulated developmental process whereby oligodendrocytes in the central nervous system and Schwann cells in the peripheral nervous system ensheathe axons with a multilayered concentric membrane. Axonal myelination increases the velocity of nerve impulse propagation. In this work, we present a novel in vitro system for coculturing primary dorsal root ganglia neurons along with myelinating cells on a highly restrictive and micropatterned substrate. In this new coculture system, neurons survive for several weeks, extending long axons on defined Matrigel tracks. On these axons, myelinating cells can achieve robust myelination, as demonstrated by the distribution of compact myelin and nodal markers.     Validation of clinical acceptability of deep-learning-based automated segmentation of organs-at-risk for head-and-neck radiotherapy treatment planning

    IntroductionOrgan-at-risk segmentation for head and neck cancer radiation therapy is a complex and time-consuming process (requiring up to 42 individual structure, and may delay start of treatment or even limit access to function-preserving care. Feasibility of using a deep learning (DL) based autosegmentation model to reduce contouring time without compromising contour accuracy is assessed through a blinded randomized trial of radiation oncologists (ROs) using retrospective, de-identified patient data.MethodsTwo head and neck expert ROs used dedicated time to create gold standard (GS) contours on computed tomography (CT) images. 445 CTs were used to train a custom 3D U-Net DL model covering 42 organs-at-risk, with an additional 20 CTs were held out for the randomized trial. For each held-out patient dataset, one of the eight participant ROs was randomly allocated to review and revise the contours produced by the DL model, while another reviewed contours produced by a medical dosimetry assistant (MDA), both blinded to their origin. Time required for MDAs and ROs to contour was recorded, and the unrevised DL contours, as well as the RO-revised contours by the MDAs and DL model were compared to the GS for that patient.ResultsMean time for initial MDA contouring was 2.3 hours (range 1.6-3.8 hours) and RO-revision took 1.1 hours (range, 0.4-4.4 hours), compared to 0.7 hours (range 0.1-2.0 hours) for the RO-revisions to DL contours. Total time reduced by 76% (95%-Confidence Interval: 65%-88%) and RO-revision time reduced by 35% (95%-CI,-39%-91%). All geometric and dosimetric metrics computed, agreement with GS was equivalent or significantly greater (p<0.05) for RO-revised DL contours compared to the RO-revised MDA contours, including volumetric Dice similarity coefficient (VDSC), surface DSC, added path length, and the 95%-Hausdorff distance. 32 OARs (76%) had mean VDSC greater than 0.8 for the RO-revised DL contours, compared to 20 (48%) for RO-revised MDA contours, and 34 (81%) for the unrevised DL OARs.ConclusionDL autosegmentation demonstrated significant time-savings for organ-at-risk contouring while improving agreement with the institutional GS, indicating comparable accuracy of DL model. Integration into the clinical practice with a prospective evaluation is currently underway

    Development and validation of a unifying pre-treatment decision tool for intracranial and extracranial metastasis-directed radiotherapy

    BackgroundThough metastasis-directed therapy (MDT) has the potential to improve overall survival (OS), appropriate patient selection remains challenging. We aimed to develop a model predictive of OS to refine patient selection for clinical trials and MDT.Patients and methodsWe assembled a multi-institutional cohort of patients treated with MDT (stereotactic body radiation therapy, radiosurgery, and whole brain radiation therapy). Candidate variables for recursive partitioning analysis were selected per prior studies: ECOG performance status, time from primary diagnosis, number of additional non-target organ systems involved (NOS), and intracranial metastases.ResultsA database of 1,362 patients was assembled with 424 intracranial, 352 lung, and 607 spinal treatments (n=1,383). Treatments were split into training (TC) (70%, n=968) and internal validation (IVC) (30%, n=415) cohorts. The TC had median ECOG of 0 (interquartile range [IQR]: 0-1), NOS of 1 (IQR: 0-1), and OS of 18 months (IQR: 7-35). The resulting model components and weights were: ECOG = 0, 1, and > 1 (0, 1, and 2); 0, 1, and > 1 NOS (0, 1, and 2); and intracranial target (2), with lower scores indicating more favorable OS. The model demonstrated high concordance in the TC (0.72) and IVC (0.72). The score also demonstrated high concordance for each target site (spine, brain, and lung).ConclusionThis pre-treatment decision tool represents a unifying model for both intracranial and extracranial disease and identifies patients with the longest survival after MDT who may benefit most from aggressive local therapy. Carefully selected patients may benefit from MDT even in the presence of intracranial disease, and this model may help guide patient selection for MDT

    Rapid and Long-Lasting Increase in Sites for Synapse Assembly during Late-Phase Potentiation in Rat Hippocampal Neurons

    Long-term potentiation in hippocampal neurons has stages that correspond to the stages of learning and memory. Early-phase (10–30 min) potentiation is accompanied by rapid increases in clusters or puncta of presynaptic and postsynaptic proteins, which depend on actin polymerization but not on protein synthesis. We have now examined changes in pre- and postsynaptic puncta and structures during glutamate-induced late-phase (3 hr) potentiation in cultured hippocampal neurons. We find that (1) the potentiation is accompanied by long-lasting maintenance of the increases in puncta, which depends on protein synthesis, (2) most of the puncta and synaptic structures are very dynamic, continually assembling and disassembling at sites that are more stable than the puncta or structures themselves, (3) the increase in presynaptic puncta appears to be due to both rapid and more gradual increases in the number of sites where the puncta may form, and also to the stabilization of existing puncta, (4) under control conditions, puncta of postsynaptic proteins behave similarly to puncta of presynaptic proteins and share sites with them, and (5) the increase in presynaptic puncta is accompanied by a similar increase in presumably presynaptic structures, which may form at distinct as well as shared sites. The new sites could contribute to the transition between the early and late phase mechanisms of plasticity by serving as seeds for the formation and maintenance of new synapses, thus acting as local “tags” for protein synthesis-dependent synaptic growth during late-phase plasticity

    The proteasome controls presynaptic differentiation through modulation of an on-site pool of polyubiquitinated conjugates

    Differentiation of the presynaptic terminal is a complex and rapid event that normally occurs in spatially specific axonal regions distant from the soma; thus, it is believed to be dependent on intra-axonal mechanisms. However, the full nature of the local events governing presynaptic assembly remains unknown. Herein, we investigated the involvement of the ubiquitin-proteasome system (UPS), the major degradative pathway, in the local modulation of presynaptic differentiation. We found that proteasome inhibition has a synaptogenic effect on isolated axons. In addition, formation of a stable cluster of synaptic vesicles onto a postsynaptic partner occurs in parallel to an on-site decrease in proteasome degradation. Accumulation of ubiquitinated proteins at nascent sites is a local trigger for presynaptic clustering. Finally, proteasome-related ubiquitin chains (K11 and K48) function as signals for the assembly of presynaptic terminals. Collectively, we propose a new axon-intrinsic mechanism for presynaptic assembly through local UPS inhibition.     Effects of Neurofeedback on Neuropsychological Functioning in an Adult with Autism

    Autism spectrum condition (ASC) is a complex neurodevelopmental disorder that impacts physiological processes, cognition, functional behaviors, social-communication, and often has comorbidities. One approach gaining empirical support for ASC treatment is neurofeedback. Neurofeedback uses operant conditioning to normalize cerebral activity through auditory and visual reinforcement. Live Z-score Training (LZT) has become the latest advancement in neurofeedback. There is no published research to date on LZT neurofeedback in adulthood ASC. The purpose of this study was to evaluate LZT’s impact on neuropsychological measures in an adult with ASC. A multiple baseline single-case research design was used with a convenience sample of one adult with ASC to evaluate the effects of 20 LZT sessions using the Conservative Dual Criterion visual inspection method as the primary form analysis. ADHD, mood stability, anxiety, depression, and ASC symptoms were significantly reduced according to the Neuropsych Questionnaire. The participant improved significantly on the CNS Vital Signs (CNVS) Neurocognitive measures of executive function, cognitive flexibility, reaction time, and complex attention. Also, the participant increased intelligence as measured by the Test of Nonverbal Intelligence. Lastly, the participant had changes in brain function according to quantitative electroencephalography and low-resolution brain electromagnetic tomography. CNVS processing speed was the only measure that did not significantly change. No adverse effects were reported. This study may lead to positive social change by providing a technologically advanced intervention for adults with ASC, which may improve their overall quality of life and promote self-sufficiency through adulthood
