    Nutritive value of palisadegrass silages affected by year season, dry matter level and bacterial inoculant

    O estudo foi realizado para avaliar o efeito da estação do ano, do teor de matéria seca (MS) e da inoculação bacteriana sobre o valor nutritivo de silagens de Brachiaria brizantha cv. Marandu. O delineamento experimental foi o inteiramente casualizado, disposto em arranjo fatorial (2 × 3 × 3), com duas estações do ano (verão e inverno), três teores de MS (com ou sem emurchecimento ou adição de polpa cítrica peletizada, PCP) e ausência ou presença de inoculante (Lactobacillus plantarum ou Lactobacillus buchneri), perfazendo 18 tratamentos, cada um com quatro repetições. As silagens foram confeccionadas em baldes de plástico de 20 L. Após 90 dias do fechamento, os silos foram abertos para as avaliações. O emurchecimento elevou os teores de matéria seca (MS), fibra em detergente neutro (FDN) e lignina (LIG) em ambas as estações, porém não alterou o coeficiente de digestibilidade verdadeira in vitro da MS (DVIVMS) das silagens. A adição de PCP aumentou os teores de proteína bruta (PB) e carboidratos solúveis (CHO) e reduziu os teores de FDN, o que resultou em silagens com maior DVIVMS em ambas as estações. A inoculação não alterou a fração fibrosa e não aumentou a DVIVMS das silagens. As silagens de verão apresentaram maiores teores de PB e menores constituintes da parede celular, com exceção da hemicelulose (HEMI), o que resultou no maior coeficiente médio de DVIVMS dessas silagens. O baixo valor nutritivo das silagens de inverno contraria os resultados descritos na literatura, principalmente em virtude da maior idade de rebrotação do capim-marandu nesta estação. Desta forma, silagens de verão podem ser recomendadas como volumoso suplementar, principalmente quando acrescidas de PCP.The objectives of this work were to evaluate the effect of year season, dry matter concentration (DM), and bacterial inoculation on the nutritive value of palisadegrass silages. The trial was carried out in a complete randomized experimental design with four replications, in a factorial arrangement (2 × 3 × 3): two year seasons (summer and winter), three dry matter levels (with or without wilting or added with dried citrus pulp), and without microbial additive or with Lactobacillus plantarum or Lactobacillus buchneri, totalizing 18 treatments. Silages were stored in 20 L plastic buckets. After 90-d of fermentation, the silos were opened and the silages were evaluated. Wilting increased DM, neutral detergent fiber (NDF), and lignin (LIG), in both year seasons, and did not change in vitro true dry matter digestibility (IVTDMD). Addition of citrus pulp increased crude protein (CP) concentration and water soluble carbohydrates (WSC) and decreased the NDF concentration. As a result, this additive increased the IVTDMD in both year seasons. The use of microbial inoculants did not change fiber fractions and did not increase the IVTDMD in the silages. Summer-silages had higher CP level and lower cell wall concentrations, except for HEM, resulting in higher IVTDMD. The low nutritive value obtained for the winter silages was not in agreement with the literature, mainly due to the greater regrowth age of palisadegrass in this season. As a result, the overall analysis pointed out that summer season silages are a better option of roughage for animal nutrition, mainly when ensiled with dried and pelleted citrus pulp

    Chemical additive and microbial inoculants effects on the fermentation and on the control of the alcohol production in sugarcane silages

    Avaliou-se a eficácia de aditivo químico e inoculantes microbianos nas alterações fermentativas que inibem a produção de etanol e diminuem as perdas de matéria seca em silagens de cana-de-açúcar. A ensilagem foi realizada em baldes plásticos de 20 litros (minissilos) adaptados com válvulas tipo Bunsen para eliminação dos gases e dispositivo para coleta de efluentes. Utilizou-se uréia (1% na matéria verde) como aditivo e os inoculantes microbianos Lalsil cana® contendo a bactéria heterolática Lactobacillus buchneri (cepa NCIMB 40788) na razão de 3,65 × 10(5) ufc/g massa verde (MV) e um inoculante não comercial contendo a combinação de bactérias homo e heteroláticas Pedioccocus pentosassus e Lactobacillus buchneri (1 × 10(6) ufc/g de massa verde). As amostragens foram realizadas após 110 dias de fermentação. A utilização de uréia e principalmente do aditivo Lactobacillus buchneri + Pediococcus foi eficaz em diminuir a produção de etanol (1,30 e 2,75% vs 8,27% na silagem controle), as perdas de matéria seca (47 e 60%) e o consumo de carboidratos solúveis (22 e 56%) e em aumentar a digestibilidade in vitro da matéria seca (14,7 e 12,4%) quando comparados à silagem controle, respectivamente. Apesar dos altos teores de ácido acético em todas as silagens, principalmente nas silagens acrescidas da combinação de bactérias homo e heteroláticas, não foram verificados efeitos dos inoculantes sobre a população de leveduras. Os valores de pH e ácido lático da silagem com uréia foram maiores que os obtidos na silagem controle. As silagens com L. buchneri apresentaram neste estudo maior produção de etanol (11,53% vs 8,27%, silagem controle), o que refletiu em perdas significativas e baixa recuperação de matéria seca, além de baixa digestibilidade pela perda de carboidratos solúveis e pelo acúmulo de FDN, que foram similares aos das silagens sem aditivo.The objective of this study was to evaluate chemical additive and microbial inoculants effectiveness on fermentative changes that lead to the inhibition of the ethanol production and reduction of dry matter losses in sugarcane silages. Silos were made in 20 liter plastic buckets provided with a Bunsen type valve that allows gas to escape and a device to collect effluents. The applied additives were urea (1% of fresh forage); microbial additive Lalsil cana®, that has heterolactic bacteria Lactobacillus buchneri (NCIMB 40788), 3.65 × 10(5) CFU g-1 of fresh forage, and a non-commercial additive with homolactic bacteria Pedioccocus pentosassus and Lactobacillus buchneri (1 × 10(6) CFU g-1 FM) supplied by Lallemand Inc. (Montreal, Quebec). Sampling occurred after 110 days of fermentation. Urea and mainly the additive Lactobacillus buchneri and Pediococcus were efficient on the reduction of ethanol production (1.30 and 2.75% versus 8.27% in the control treatment), on the reduction of dry matter losses (47 and 60%) and soluble carbohydrates (22 and 56%), and increase in vitro dry matter digestibility (14,7 and 12,3%) when compared to the control silages, respectively. Despite the high values of acetic acid in all silages, mainly in silages inoculated with homo and heterolactic bacteria, it was not verified effects on yeast population. The silage with urea had greater values of pH and lactic acid compared to control silage. Silages with L. buchneri had, in this experiment, the largest ethanol production (11.53% vs 8.27% from control silage), resulting in greater losses and lower dry matter recovery.     Changes in morphological composition according to the maturity stage in corn cultivars for silage production

    Objetivou-se avaliar o teor de matéria seca (MS) e a participação percentual das frações da planta em híbridos de milho colhidos em diversos estádios de maturação. Utilizou-se um delineamento em blocos casualizados, com esquema fatorial 8 × 6 × 2, composto de oito épocas de corte, seis híbridos de milho e duas safras (2001/2002 e 2002/2003). Os híbridos CO 32, AG 5011, P 3041, DKB 333B, AG 1051 e Z 8550 foram colhidos ao atingirem 50% de florescimento masculino, 15 dias após esta data e, posteriormente, a cada semana, totalizando-se oito cortes. As plantas atingiram o teor de MS para ensilagem (32 a 35% MS) 92 a 112 dias após a semeadura (DAS). O avanço da maturação aumentou os teores de MS das frações colmo (de 16,9 para 28%), folha (de 23,2 para 48,8%), sabugo (de 10,0 para 55,5%) e grãos (de 50,4 para 70,9%). Além disso, promoveu aumento das proporções de MS acumulada nos sabugos (de 1,6 para 9,2%) e grãos (de 0 para 43,4%) e redução na proporção de colmo (de 63,4 para 29,5%) e folhas (de 27,5 para 10,8%). A comparação dos híbridos de milho colhidos em estádios fisiológicos diferentes considerando apenas a participação das frações na planta deve ser feita com muito critério, uma vez que a época de corte pode alterar significativamente a composição morfológica da planta.The objective of this experiment was to evaluate the dry matter (DM) content and the percent participation of fractions that compose the plant in corn cultivars harvested at different maturity stages. A randomized block design with an 8 × 6 × 2 factorial scheme composed of eight harvesting times, six corn cultivars and two harvest years (2001/2002 and 2002/2003) was used. Cultivars CO 32, AG 5011, P 3041, DKB 333B, AG 1051 and Z 8550 were harvested when they reached 50% male flowering; 15 days after this date and later, each week, totalizing eight harvesting times. The plants reached the recommended DM content for silage production (32 to 35% DM) between 92 and 112 days after sowing (DAS) at the evaluated years. The maturity advancement resulted in increases in DM content in stem (from 16.9 to 28%), DM content in leaves (from 23.2 to 48.8%), DM content in cob (from 10 to 55.5%), and DM content in grains (from 50.4 to 70.9%). Furthermore, it resulted in increases in cob (from 1.6 to 9.2%) and grain proportion (from 0 to 43.4%), and decrease in stem (from 63.4 to 29.5%) and leaves proportion (from 27.5 to 10.8%). The corn cultivars showed distinct growing pattern for the evaluated variables. Therefore, plants harvested at different physiologic states can only be compared with fractions participation with considerable discernment, once the harvesting time can modify significantly the plant morphological composition

    The use of ensiled sugarcane has been increased lately in Brazil due to the benefits that this technique represents. The objective of this study was to evaluate the effects of chemical additives on the nutritive value of sugarcane silages. The trial was carried out in a completely randomized experimental design with four replicates per treatment. The following additives were applied onto the fresh forage before ensiling: L. buchneri, lime or limestone, 1.0 and 1.5% (wet basis) each, and gypsum 1.0% (wet basis), all of them diluted into 40 L of water per ton of fresh weight of forage. The analyzed variables were: ash, crude protein, neutral detergent fiber, acid detergent fiber and organic and dry matter digestibility. The addition of lime or limestone before ensiling produced silages with higher nutritive value compared to all other treatments, due to the increase of the ash content and dry matter and organic digestibility, and also by reducing the fiber content. The crude protein content range was similar to the values observed in the fresh forage. The treatments containing L. buchneri or gypsum were ineffective in improving the nutritive value of sugarcane silages and became similar to the control silages. Treatment containing lime or limestone improved the nutritive value of the sugarcane silage.A utilização da cana-de-açúcar na forma de silagem constitui-se em um tema que vem se destacando nos últimos anos, e que desperta o interesse de produtores e pesquisadores em função dos benefícios que essa técnica representa. A ensilagem da cana-de-açúcar com L. buchneri ou gesso apresentaram desempenho semelhante ao tratamento controle para as variáveis acima mencionadas.     Tonsils of the soft palate do not mediate the response of pigs to oral vaccination with heat-inactivated Mycobacterium bovis

    Mycobacterium bovis causes animal tuberculosis (TB) in cattle, humans, and other mammalian species, including pigs. The goal of this study was to experimentally assess the responses of pigs with and without a history of tonsillectomy to oral vaccination with heat-inactivated M. bovis and challenge with a virulent M. bovis field strain, to compare pig and wild boar responses using the same vaccination model as previously used in the Eurasian wild boar (Sus scrofa), to evaluate the use of several enzyme-linked immunosorbent assays (ELISAs) and lateral flow tests for in vivo TB diagnosis in pigs, and to verify if these tests are influenced by oral vaccination with inactivated M. bovis. At necropsy, the lesion and culture scores were 20% to 43% higher in the controls than those in the vaccinated pigs. Massive M. bovis growth from thoracic tissue samples was observed in 4 out of 9 controls but in none of the 10 vaccinated pigs. No effect of the presence or absence of tonsils was observed on these scores, suggesting that tonsils are not involved in the protective response to this vaccine in pigs. The serum antibody levels increased significantly only after challenge. At necropsy, the estimated sensitivities of the ELISAs and dual path platform (DPP) assays ranged from 89% to 94%. In the oral mucosa, no differences in gene expression were observed in the control group between the pigs with and without tonsils. In the vaccinated group, the mRNA levels for chemokine (C-C motif) receptor 7 (CCR7), interferon beta (IFN-β), and methylmalonyl coenzyme A mutase (MUT) were higher in pigs with tonsils. Complement component 3 mRNA levels in peripheral blood mononuclear cells (PBMC) increased with vaccination and decreased after M. bovis challenge.     PRODUÇÃO DE BIOHIDROGÊNIO A PARTIR DE ÁGUA RESIDUÁRIA DE FECULARIA EM REATOR ANAERÓBIO DE BATELADA SEQUENCIAL E BIOMASSA IMOBILIZADA

    Este trabalho avaliou a produção de biohidrogênio a partir da água residuária de fecularia, utilizando reator anaeróbio de bateladas sequenciais com biomassa imobilizada. O ensaio utilizou carga orgânica volumétrica de 9gcarboidratostotais.L-1d-1, tempo de ciclo de 4h, e temperatura de 30 °C, com 12 dias de operação. Foram avaliados produtividade molar e rendimento de hidrogênio. O volume de biogás foi quantificado utilizando-se frasco mariotte e sua composição por cromatografia gasosa. Foi possível produzir 27 molH2.m-3.d-1, com rendimento de hidrogênio sobre açúcares totais de 2,35 molH2.Kg-1. Concluiu-se que a produção de biohidrogênio a partir da água residuária de fecularia foi possível, apresentando potencial neste sentido. Palavras Chave: Biogás, Digestão Anaeróbia, Bioenergia

    Persistence and viability of SARS-CoV-2 in primary infection and reinfections

    Since the beginning of the SARS-CoV-2 epidemic, virus isolation in the infected patient was only possible for a short period of time and it was striking that this occurred constantly and did not provide guidance on the clinical course. This fact led to confusion about the efficacy of some of the drugs initially used, which seemed to have a high efficiency in viral clearance and proved ineffective in modifying the course of the disease. The immune response also did not prove to be definitive in terms of evolution, although most of the patients with very mild disease had a weak or no antibody response, and the opposite was true for the most severe patients. With whatever the antibody response, few cases have been re-infected after a first infection and generally, those that have, have not reproduced a spectrum of disease similar to the first infection. Among those re-infected, a large number have been asymptomatic or with very few symptoms, others have had a moderate picture and very few have had a poor evolution. Despite this dynamic of rapid viral clearance, laboratory tests were still able to generate positive results in the recovery of genomic sequences and this occurred in patients who were already symptom-free, in others who were still ill and in those who were very seriously ill. There was also no good correlate. For this reason and with the perspective of this year and the half of pandemic, we compiled what the literature leaves us in these aspects and anticipating that, as always in biology, there are cases that jump the limits of the general behavior of the dynamics of infection in genera

    Actions and attitudes on the immunized patients against SARS-CoV-2

    The access to COVID vaccines by millions of human beings and their high level of protection against the disease, both in its mild and severe forms, together with a plausible decrease in the transmission of the infection from vaccinated patients, has prompted a series of questions from the members of the College of Physicians of Madrid (ICOMEM) and the society. The ICOMEM Scientific Committee on this subject has tried to answer these questions after discussion and consensus among its members. The main answers can be summarized as follows: The occurrence of new SARS-CoV-2 infections in both vaccinated and previously infected patients is very low, in the observation time we already have. When breakthrough infections do occur, they are usually asymptomatic or mild and, purportedly, should have a lower capacity for transmission to other persons. Vaccinated subjects who have contact with a SARS-CoV-2 infected patient can avoid quarantine as long as they are asymptomatic, although this decision depends on variables such as age, occupation, circulating variants, degree of contact and time since vaccination. In countries with a high proportion of the population vaccinated, it is already suggested that fully vaccinated persons could avoid the use of masks and social distancing in most circumstances. Systematic use of diagnostic tests to assess the immune response or the degree of protection against reinfection after natural infection or vaccination is discouraged, since their practical consequences are not known at this time. The existing information precludes any precision regarding a possible need for future revaccination. This Committee considers that when mass vaccination of health care workers and the general population is achieved, SARS-CoV-2 screening tests could be avoided at least in out-patient care and in the case of exploratory procedures that do not require hospitalizatio

    Performance of Holstein heifers fed sugarcane silages treated with urea, sodium benzoate or Lactobacillus buchneri

    O objetivo deste trabalho foi avaliar a performance de novilhas alimentadas com silagens de cana-de-açúcar tratadas com aditivos. Trinta e duas novilhas Holandesas foram aleatoriamente distribuídas em um delineamento em blocos, para avaliar rações (46% silagem; 54% concentrado; 12% proteína bruta) contendo silagem tratada com (base matéria verde) uréia (0,5%), benzoato de sódio (0,1%) ou Lactobacillus buchneri (3,64x105 ufc g-1). A inoculação de L. buchneri melhorou o ganho diário (1,24 vs. 0,94 kg dia-1) e a adição de benzoato melhorou a conversão alimentar (7,6 vs. 9,4 kg de matéria seca por kg de peso vivo), relativamente ao controle (silagem não tratada). Os tratamentos não afetaram o consumo de matéria seca (2,19% do peso vivo). As rações com silagens tratadas com benzoato ou L. buchneri mostraram menor custo por kg de ganho de peso. O tratamento com uréia não melhorou o desempenho animal, mas o custo por kg de ganho de peso foi menor do que na dieta controle.The objective of this work was to evaluate the performance of heifers fed sugarcane silages produced with and without additives. Thirty-two Holstein heifers were randomly assigned, in a block design, to evaluate rations (46% silage; 54% concentrate; 12% crude protein) containing silages treated with (fresh basis) urea (0.5%), sodium benzoate (0.1%) or Lactobacillus buchneri (3.64x105 cfu g-1). Inoculation with L. buchneri improved daily gain (1.24 vs. 0.94 kg day-1), and the addition of benzoate resulted in better feed conversion (7.6 vs. 9.4 kg of dry matter per kg of live weight), in relation to the untreated silage (control). Treatments did not affect dry matter intake (mean of 2.19% of live weight). Rations containing silages treated with benzoate or L. buchneri showed lower cost per kg of weight gain. Treatment with urea did not improve animal performance, but the cost per kg of weight gain was lower than that of the control ration
