    Multicomponent bioactive extract from red stage Haematococcus pluvialis wet paste: avoiding the drying step and toxic solvents

    info:eu-repo/grantAgreement/AEI/Plan Estatal de Investigación Científica y Técnica y de Innovación 2013-2016/EQC2018-005011-P/ESThis study provides an efficient alternative by extracting bioactive compounds from Haematococcus pluvialis via matrix solid-phase dispersion (MSPD) from its wet form, reducing one of the process steps with the greatest economic impact, the drying of the microalga. To obtain a suitable extract for nutraceutical purposes, solvents with the generally recognised as safe (GRAS) designation (ethanol, ethyl lactate, and ethyl acetate) with limitations of use (acetone) and extractants with higher toxicity such as methanol and methyl-tert-butyl ether (MTBE) are contrasted. Through the optimisation of the extractive process, ethanol, a GRAS solvent, presents the best overall recovery for carotenoid compounds and fatty acids, showing an antioxidant activity of 1.58 mmolTE g−1 DW, comparable to its synthetic alternative of petrochemical origin without the drawback of having limitations in its food use. In addition, the identification of the phenolic compounds, phloroglucinol, p-coumaric acid, gallic acid, and catechin, not previously characterised in red stage H. pluvialis, provides a response to the phenolic activity present in the extract (24.65 mmolGAE g−1 DW). Comparison of the extractive efficiency obtained with the main methods for the extraction of carotenoids and fatty acids in H. pluvialis, in contrast to the proposed method, shows a positive feasibility of this approachOpen Access funding provided thanks to the CRUE-CSIC agreement with Springer Nature.     El anonimato y el altruismo en la donación de gametos: la producción de biocapital en la industria reproductiva

    En este artículo analizamos de qué modo el anonimato y el altruismo de la donación de gametos desempeña una función primordial en el sostenimiento de una industria reproductiva pujante en el contexto español. A partir de nuestra experiencia etnográfica, y apoyándonos en otros datos extraídos de fuentes secundarias (estadísticas, directrices de organismos oficiales, legislación), retomamos conceptos de la teoría clásica de Marx (trabajo productivo/reproductivo, plusvalía, alienación, entre otros) para dar cuenta de la producción de biocapital a partir de los circuitos de las donaciones reproductivas. Consideramos el anonimato y el altruismo como instrumentos legales que favorecen la apropiación privada y privativa de los gametos “donados” en cantidad suficiente para el desarrollo del mercado y de la industria reproductiva. Estos instrumentos legales establecen las condiciones que hacen posible esa apropiación por parte de los centros médicos, así como la fijación de los precios y la manera en que esos gametos son “donados”

    The Main Clinical Indicators of Sarcopenia in Patients with Chronic Respiratory Disease: Skeletal Muscle Dysfunction Approach

    Patients with chronic respiratory diseases (CRDs) have a disorder in muscle structure and function, but their function increases with physical progress and decreases the risk of general, and muscular weakness are more likely to develop sarcopenia. We randomly selected patients (N = 38) with mean age of 72 ± 1.0 years old men and women elderly with chronic respiratory diseases such as asthma, COPD, bronchiectasis and obesity with dyspnea score ≥ 2 in MRC index. All patients after receiving research information and signing informed consent have gone through performing clinical assessments. They performed femur bone mineral density (FBMD) and ultrasound on the rectus femoris muscle mid-tight cross-sectional area (RFMTCSA) in the quadriceps muscle. The significant changes in BMI were seen in all patients, pre-rehabilitation, BMI = 30 ± 1.06 kg/m2 and post-rehabilitation, BMI = 29 ± 1.00 kg/m2. In Pearson’s correlation of r = 0.607 between T-score and Z-score in FBMD and RFMTCSA in pre-rehabilitation, there is a little bit significant correlation between the variables than in the Pearson’s correlation of r = 0.910 in post-rehabilitation, P < 0.00. Comparing femur bone and rectus femoris muscle parameters as indicators for diagnosis of sarcopenia in chronic respiratory patients, we observed that in rectus femoris muscle, ultrasound is the most effective foot muscle detector

This manuscript version is made available under the CC-BY-NC-ND 4.0 licenseThis study presents a new multi-residue analytical method for the simultaneous determination of 38 psychoactive drugs (including benzodiazepines, antidepressants and drugs of abuse) and related metabolites in raw wastewater. Potential analyte losses during sample filtration and stability in wastewater were evaluated. Analyte losses, especially for 12 compounds, were observed during filtration, indicating a strong sorption onto the filter material. In order to overcome this effect, filtered water samples were combined with methanolic washes of the corresponding filters and the resulting solutions were solid-phase extracted on mixed-mode (reverse-phase plus cation-exchange) sorbents. Extracts were analyzed by ultra-high performance liquid chromatography-tandem mass spectrometry. Quantification was performed by the internal standard method with isotopic labeled analogs. Recovery percentages varied between 65% and 137%; method quantification limits ranged between 0.2 and 22 ng/L in ultrapure water and between 0.3 and 30 ng/L in wastewater for all the analytes but three (for which they were ∼60–80 ng/L). The analysis of 24 h-composite samples collected during one week in the city of Santiago de Compostela demonstrated the ubiquity of 31 analytes, which were positively quantified in all samples. The highest concentrations were found for some of the antidepressants, with mean and maximum levels exceeding, in some cases, the levels previously reported in literature. This fact could be related to the additional washing step of the filters using methanol, which allowed to desorb retained analytes highlighting the importance of this step during the sample preparation protocolS

    Vertical preparation: a new technique for analogical and digital impressions

    The BOPT technique (Biologically Oriented Preparation Technique) was proposed by Loi et al. (1) and has become a popular form of vertical preparation for complete crowns with promising results. In this procedure, the clinician can operate on the gingival tissue during preparation and modify its structure in thickness and height by adding modifications on the provisional restoration. However, one of the main challenges in this technique is the transference of information about the gingival tissue to the laboratory technician, who will place the margin of the restoration randomly in a space determined by two marks on the working cast.The technique proposed enables the accurate transmission of the exact point where the margin of the restoration needs to be placed. Furthermore, it facilitates the recording of conventional impression materials and intraoral scanners due to the compression of polytetrafluoroethylene (PTFE) tape into the sulcus, which allows to arrest bleeding or intracrevicular liquid and is easily registered irrespective of the method of impression used

    Use of Petiveria alliacea Linn as palliative treatment of pulpal pain

    Introduction: among the Cuban flora plants with medicinal properties is Petiveria alliacea Linn (anamú), a natural plant from tropical Americas that contains numerous active ingredients and to which multiple properties are attributed. The vast majority of reports refer specifically to its analgesic action, coming from its leaves and roots. Objective: to evaluate the therapeutic effect of 20% anamú tincture on the remission of pulpal pain in reversible serous acute pulpitis, during the period from September 2020 to September 2021. Method: a quasi-experimental study was carried out in patients with pain from the stomatological emergency room of the Policlínico Comunitario "Hermanos Martínez Tamayo". The population was made up of all the patients who attended the consultation for pain (No. 42) and were diagnosed with reversible serous acute pulpitis. They were included in the study using the intentional non-probabilistic method according to the order of reception in the dental office. The variables taken into account were: age, pain refraction time and therapeutic effect. The statistical processing of the data was carried out using descriptive statistics and the percentage was used as a summary measure. Results: in 64.3% of the cases studied, pain remission could be achieved in the first 5 minutes of applying the phytopharmaceutical. Conclusion: the tincture of Petiveria alliacea Linn (anamu) at 20% has a favorable effect on the remission of pain caused by reversible serous acute pulpitis

    Electrocatalytic Palladium Nanoclusters as Versatile Indicators of Bioassays: Rapid Electroanalytical Detection of SARS-CoV-2 by Reverse Transcription Loop-Mediated Isothermal Amplification

    Quantitative polymerase chain reaction (qPCR) is considered the gold standard for pathogen detection. However, improvement is still required, especially regarding the possibilities of decentralization. Apart from other reasons, infectious diseases demand on-site analysis to avoid pathogen spreading and increase treatment efficacy. In this paper, the detection of SARS-CoV-2 is carried out by reverse transcription loop-mediated isothermal amplification, which has the advantage of requiring simple equipment, easily adaptable to decentralized analysis. It is proposed, for the first time, the use of palladium nanoclusters (PdNCs) as indicators of the amplification reaction at end point. The pH of the medium decreases during the reaction and, in turn, a variation in the catalytic activity of PdNCs on the oxygen reduction reaction (ORR) can be electrochemically observed. For the detection, flexible and small-size screen-printed electrodes can be premodified with PdNCs, which together with the use of a simple and small electrochemical equipment would greatly facilitates their integration in field-deployable devices. This would allow a faster detection of SARS-CoV-2 as well as of other future microbial threats after an easy adaptation
