    Trace-metaldynamics in response of increase CO<sub>2</sub> and iron availability in a coastal mesocosm experiment

    A mesocosm experiment was performed in the Raunefjord (Norway) to study changes in dissolved Cu (dCu) and Fe (dFe), and in the elemental composition of particles during an Emiliania huxleyi dominated bloom. The CO2 treatments consisted of present (LC; 390 ppmV) and predicted levels (HC; 900 ppmV) and iron conditions were created with the addition of the siderophore desferoxamine B (DFB). Our results showed the DFB addition enhanced the solubility of Fe in this fjord environment. Initially, dFe was comparable among treatments but after the addition, the HC and/or +DFB treatments presented higher levels and finally, the only ones maintaining high dFe were the +DFB treatments. Unlike dCu presented indistinguishable levels in all mesocosms over time. Particulate metals were normalised to P and Al to evaluate the relative influence of biotic and abiotic sources. The Fe:P ratios decreased with time and compared to published phytoplankton ratios suggest Fe storage. On the other hand, Fe:Al ratios were relatively closer to the crustal ratios suggesting that the abiotic source was more important for this metal.     The effects of ocean acidification and siderophore additions on trace-metal dynamics during an Emiliania Huxleyi- dominated bloom in a coastal mesocosm experiment

    The combined impacts of high CO2 and changes in trace metal availability on plankton remain largely unknown. A 22 days mesocosm experiment was carried out in the Raunefjord, off Bergen, Norway (June 2012) to investigate the interactive effects of increased CO2 and changes in iron availability in the pelagic community. Twelve mesocosms covered by PAR and UVR transparent lids were used. The CO2 treatments consisted of present (LC; 390 ppmV) and predicted levels for year 2100 (HC; 900 ppmV). On day 7, half of the mesocoms were amended with the siderophore desferoxamine B (DFB) at 70 nM (final concentration). We studied changes in dissolved Cu (dCu) and Fe (dFe), and in the elemental composition of particles. Dissolved Fe and dCu were measured by chemiluminescence flow injection analysis, while particulate metals were determined by high-resolution inductively coupled mass spectrometry. The metals were normalised to P and Al to evaluate the relative influence of biotic and abiotic sources. Samples were taken throughout a bloom of the coccolithophore E. huxleyi.     Effect of increased CO2 and dissolved iron on primary productivity and inorganic carbon uptake during a bloom of the coccolithophore Emiliania huxleyi

    Net primary production (NPP) and inorganic carbon uptake were analysed during a bloom of Emiliania huxleyi in mesocosms. The experiment lasted 25 days and was carried out in the Raunefjord off Bergen (Norway, June 2012) in order to investigate the interactive effects of increased CO2 and iron availability on the phytoplanktonic community. For this purpose two different partial pressures of CO2 corresponding to present (390 ppmV) and predicted levels for year 2100 (900ppmV) (LC and HC respectively) were used. The siderophore desferoxamine B (DFB) was added to half of the mesocosms to create two different iron conditions (+DFB and –DFB). Three mesocosm bags were used for each treatment. NPP was estimated using short-term 14C uptake assays. The isotope disequilibrium assay (ID) was used to determine the relative fraction of HCO3- and CO2 uptake in concentrated cell suspensions. Short-term cellular 14C fixation was monitored during a transient disequilibrium between 14CO2 and H14 CO3- in solution, and so, the fraction of HCO3- uptake (f) and the rate of extracellular CO2: HCO3- interconversion (α), as a measure of extracellular carbonic anhydrase activity (eCA), were calculated.     Volatile composition of wines elaborated from organic and non-organic grapes

    The aim of this work was to study the evolution of volatile compounds during the alcoholic fermentation of organic and non-organic grapes. To do this, grapes were cultivated using organic and non-organic grapes; their tasting showed some differences between both types of Monastrell grapes. Throughout the alcoholic fermentation, the samples of organic grapes had higher concentration of total alcohols but lower concentration of esters and acids than the samples of conventional ones. Therefore, the volatile composition of wines from two different cultivated grapes using both agronomic practices was different. Moreover, regarding to the volatile compounds that contributed directly to wine aroma, generally organic wine had more chemicals and floral aromas, while the wines from conventional practices had more fruity aromas. Principal component analysis (PCA) showed that it is possible to differentiate between both types of wines in terms of concentration of volatile compounds formed during the alcoholic fermentation. Consequently, agronomic practices affected the grape taste, the wine volatile composition and its quality.Peer Reviewe

    Report of a case of discoid lupus erythematosus localised to the oral cavity: immunofluorescence findings.

    Discoid Lupus Erythematosus (DLE) is a chronic disease with a typical cutaneous involvement. This pathology rarely involves mucosa: oral cavity is interested in 20% of DLE patients. We describe a case of oral DLE in a 50-year-old woman with an anamnesis for autoimmune disorders. This study shows the helpful role of immunofluorescence in the diagnosis of autoimmune diseases. The first diagnostic step was the clinical observation of the oral mucosa: the lesion area was erythematous, athrophic and hyperkeratotic. The patient then underwent laboratory examination. We utilized human epithelial cells (Hep-2010) for Indirect Immuno-Fluorescence (IIF). Moreover, the biopsy site for Direct Immuno-Fluorescence (DIF) and histopathological analysis was the untreated oral lesion. IIF detected an increase of Anti-Nuclear Antibody (ANA) and positivity for SSA-RO. By DIF, we observed IgG/IgA/fibrinogen along basal layer. Multiple biopsies reported signs of chronic basal damage. Steroid systemic therapy induced a considerable lesion regression. We suggest the use of immunofluorescence with the integration of further data to improve diagnosis of rare diseases and to establish a suitable therapy

    Update on gingival overgrowth by cyclosporine A in renal transplants

    Severe gingival overgrowth is one of the most frequent side effects in renal transplant patients associated with assumption of cyclosporine A. Several associations with age, sex, dosage, duration of therapy or interval since transplantation have been hypothesized. The introduction of alternative immunosuppressant drugs have been suggested to permit better long-term transplant outcomes and a decrease in incidence of gingival overgrowth.     Preferences, use of and satisfaction with mental health services among a sample of Puerto Rican cancer patients

    OBJECTIVE: The purpose of this study was to describe the preferences, the use, satisfaction of mental health services (MHS) among a sample of Puerto Rican patients with cancer undergoing oncology treatment. METHODS: A convenience sample of 120 patients diagnosed with cancer was recruited. Self-report questionnaires assessed socio-demographic and background questions, and the Mental Health Service Preference, Utilization and Satisfaction Questionnaire (MHSPUS). The Socio-demographic and Background Questionnaire inquired about participants\u27 demographic and socioeconomic characteristics, and included questions such as history of psychiatric diagnosis and spiritual practices. Univariate and Chi square analyses were used for descriptive purposes. Logistic regressions were used to explore associations between sociodemographic factors and MHS preferences and use. RESULTS: The majority of the sample were females (53.8%), 61 and older (53.8%), and married or living with partner (57.1%), and reported an income equal to or less than $12,000 per year (44.4%), which places them under the US federal poverty line. Most of the participants (66.7%) reported being receptive to seeking services. Findings showed a significant association between living situation and past (p \u3c .05) and lifetime use (p \u3c .05) of MHS and past use of MHS. Participants living alone were more like to have used MHS in the past and during their lifetime. Adjusted logistic regression analyses revealed that living with someone was a protective factor for not using MHS in their lifetime (OR = 0.28; C1 = 0.08-0.95). Participants preferred to receive MHS at the oncology clinic, preferably on the date of their oncology appointments and during morning hours. CONCLUSION: Findings support the integration of mental health services within the oncology practice setting

    Bioinformatics approach to mRNA markers discovery for detection of circulating tumor cells in patients with gastrointestinal cancer

    [Abstract] Background: Detection of tumor cells in the blood, or minimal deposits in distant organs as bone marrow, could be important to identify cancer patients at high risk of relapse or disease progression. Quantitative polymerase chain reaction (PCR) amplification of tissue or tumor selective mRNA is the most powerful tool for the detection of this circulating or occult metastatic cells. Our study aims to identify novel gastrointestinal cancer-specific markers for circulating tumor cell detection. Method: Phase I preclinical study was performed by means of computational tools for expression analysis. In silico data were used to identify and prioritize molecular markers highly expressed in gastrointestinal cancers but absent in hematopoietic-derived libraries. Selected genes were evaluated by means of qRT-PCR in gastrointestinal cancer and hematopoietic cell-lines, normal human bone marrows and bloods, tumor tissue, and blood from cancer patients. Results: Novel and known mRNA markers for circulating tumor cell detection in gastrointestinal cancer have been identified. Among all the genes assessed, PKP3, AGR2, S100A16, S100A6, LGALS4, and CLDN3 were selected and assays based on blood qRT-PCR were developed. Reliably qRT-PCR assays for the novel targets plakophilin 3 (PKP3) and anterior gradient-2 (AGR2) to identify blood-borne cells in cancer patients were developed. Conclusions: Novel and known gastrointestinal-specific mRNA markers for circulating tumor cells have been identified through in silico analysis and validated in clinical material. qRT-PCR assay targeted to PKP3 and AGR2 mRNAs might be helpful to detect circulating tumor cells in patients with gastrointestinal cancer

    Effects of increased co2 in the carbon budget and the photosynthetic yield of the arctic seaweeds alaria esculenta and desmarestia aculeata

    The physiological acclimation to increased pCO2 (1300 ppm) of two common Arctic seaweeds from Kongsfjord (Svalbard) was analysed under laboratory conditions after 7 days of incubation. Growth rate changed in both species as a result of a reorganization of the carbon budget of the cell. Since increased CO2 have the potential to modify physiological mechanisms in different ways for each species, it is expected that it may lead to changes at the seaweeds community level that could alter the whole food web.Universidad de Málaga.
