    Synthesis of Polysubstituted 3-Methylisoquinolines through the 6Ď€-Electron Cyclization/Elimination of 1-Azatrienes derived from 1,1-Dimethylhydrazine

    A convenient one pot microwave-assisted 6π-electron cyclization/aromatization approach toward 3-methylisoquinolines is reported. The starting 1-azatriene derivatives were prepared in situ by reaction of 2-propenylbenzaldehydes with 1,1-dimethylhydrazine, which exhibited superior performance when compared with other hydrazine derivatives. Minor amounts of the related 3,4-dihydro isoquinolines were formed concomitantly with the isoquinolines, and a mechanism for their generation was proposed. The reaction conditions were optimized, and its scope and limitations were explored. In general, the transformation proceeded in moderate to good yields.Fil: Vargas Vargas, Didier Farley.     Differential Proteome Analysis of Bone Marrow Mesenchymal Stem Cells from Adolescent Idiopathic Scoliosis Patients

    Adolescent idiopathic scoliosis (AIS) is a complex three-dimensional deformity of the spine. The cause and pathogenesis of scoliosis and the accompanying generalized osteopenia remain unclear despite decades of extensive research. In this study, we utilized two-dimensional fluorescence difference gel electrophoresis (2D-DIGE) coupled with mass spectrometry (MS) to analyze the differential proteome of bone marrow mesenchymal stem cells (BM-MSCs) from AIS patients. In total, 41 significantly altered protein spots were detected, of which 34 spots were identified by MALDI-TOF/TOF analysis and found to represent 25 distinct gene products. Among these proteins, five related to bone growth and development, including pyruvate kinase M2, annexin A2, heat shock 27 kDa protein, γ-actin, and β-actin, were found to be dysregulated and therefore selected for further validation by Western blot analysis. At the protein level, our results supported the previous hypothesis that decreased osteogenic differentiation ability of MSCs is one of the mechanisms leading to osteopenia in AIS. In summary, we analyzed the differential BM-MSCs proteome of AIS patients for the first time, which may help to elucidate the underlying molecular mechanisms of bone loss in AIS and also increase understanding of the etiology and pathogenesis of AIS

    Discovery of new Givetian (Devonian) brachiopods from Boulonnais (N France)

    peer reviewedFour brachiopod species from the upper Givetian of Ferques are described of which two are new: Cupularostrum minimum n. sp. (Rhynchonellida) and Cleiothyridina blacourti n. sp. (Athyridida). The third, which is assigned to Stringocephalus aff. fontanus Veevers, 1959, corresponds after revision to S. sp. (in Brice & Loones 2002) pro parte. The fourth is Stringodiscus giganteus (Sowerby, 1840) which is recorded for the first time in France. Material originates mainly from the Couderousse Member of the Blacourt Formation, dated by conodonts from the “Middle-Upper varcus Zone”, but exclusively for S. aff. fontanus and St. giganteus. Some rare specimens of C. minimum and Cl. blacourti have been collected in the lower part of the Bastien Member, upper part of the Blacourt Formation. Cl. blacourti is very common in the upper Givetian of Ferques, so the genus Cleiothyridina appears as soon as the upper Givetian as in Spain and not in the Famennian as it is reported in the “Treatise revised” (Alvarez & Rong, 2002).Quatre espèces de brachiopodes sont décrites, provenant du Givétien supérieur de Ferques, dont deux sont nouvelles : Cupularostrum minimum n. sp. (Rhynchonellida) et Cleiothyridina blacourti n. sp. (Athyridida). La troisième, attribuée à Stringocephalus aff. fontanus Veevers, 1959, correspond après révision à S. sp. (in Brice & Loones 2002) pro parte et à laquelle s’ajoute Stringodiscus giganteus (Sowerby, 1840) qui est reconnu pour la première fois en France. Le matériel provient essentiellement du Membre de Couderousse (Formation de Blacourt), daté par conodontes de la Zone à varcus moyenne à supérieure, mais exclusivement pour S. aff. fontanus et St. giganteus. De rares spécimens de C. minimum et Cl. blacourti proviennent de la partie inférieure du Membre Bastien (partie supérieure de la Formation de Blacourt). Le genre Cleiothyridina, abondamment représenté par Cl. blacourti, apparaît dès le Givétien supérieur comme en Espagne et non au Famennien comme il est mentionné dans le « Treatise revised » (Alvarez & Rong, 2002)
