    Towards efficient modelling of optical micromanipulation of complex structures

    Computational methods for electromagnetic and light scattering can be used for the calculation of optical forces and torques. Since typical particles that are optically trapped or manipulated are on the order of the wavelength in size, approximate methods such as geometric optics or Rayleigh scattering are inapplicable, and solution or either the Maxwell equations or the vector Helmholtz equation must be resorted to. Traditionally, such solutions were only feasible for the simplest geometries; modern computational power enable the rapid solution of more general--but still simple--geometries such as axisymmetric, homogeneous, and isotropic scatterers. However, optically-driven micromachines necessarily require more complex geometries, and their computational modelling thus remains in the realm of challenging computational problems. We review our progress towards efficient computational modelling of optical tweezers and micromanipulation, including the trapping and manipulation of complex structures such as optical micromachines. In particular, we consider the exploitation of symmetry in the modelling of such devices.Comment: 5 pages, 4 figure

    Improved optically driven microrotors

    Two-photon polymerization of optically curing resins is a powerful method to fabricate micron sized objects which can be used as tools to measure properties at small scales. These microdevices can be driven by means of externally applied focused laser beams (optical tweezers) through angular momentum exchange, giving rise to a net torque. The advantage of the optical drive is that no contact is required, therefore making the microdevices suited to non-invasive biological applications. The fabrication method is versatile and allows building objects of any 3D shape. We discuss the design and modelling of various optically driven rotors. In particular, we consider fabrication of microspheres with an internal shape birefringence in order to obtain rotation in an optical trap. The reason for fabricating this type of object is that they are well-suited for studies of mechanical properties of single biomolecules such as the torsional stiffness of DNA or torque generated by molecular motors. The microspheres fabricated are able to transduce torques of 2000 pNnm with optical powers of 500 mW and could be rotated with frequencies up to 40 Hz in circularly polarized light

    Generalizing and unifying prior results, we solve the subconvexity problem for the LL-functions of \GL_{1} and \GL_{2} automorphic representations over a fixed number field, uniformly in all aspects.     Economic intervention and parenting: a randomized experiment of statewide child development accounts

    Objective: We examine the effects of Child Development Accounts (CDAs) on parenting stress and practices. Methods: We use data from the SEED for Oklahoma Kids (SEED OK) experiment. SEED OK selected caregivers of infants from Oklahoma birth certificates using a probability sampling method, randomly assigned caregivers to the treatment (n = 1,132) or control group (n = 1,098), and provided college savings incentives only to the treatment group. We run regression analyses on a parenting stress scale and six continuous measures of parenting practices. Results: There were no significant differences in parenting outcomes between treatment and control groups, with the exception of frequency of screaming at the child. CDAs have more salient effects on American Indians than on other groups. Conclusions: We present weak evidence for CDAs’ positive impacts on parenting outcomes. Further research is needed to determine whether these initial results change over time

Their safety profile is an important consideration for regulatory bodies, owners and prescribing clinicians.     Inclusive V0V^0 Production Cross Sections from 920 GeV Fixed Target Proton-Nucleus Collisions

    Inclusive differential cross sections dσpA/dxFd\sigma_{pA}/dx_F and dσpA/dpt2d\sigma_{pA}/dp_t^2 for the production of \kzeros, \lambdazero, and \antilambda particles are measured at HERA in proton-induced reactions on C, Al, Ti, and W targets. The incident beam energy is 920 GeV, corresponding to s=41.6\sqrt {s} = 41.6 GeV in the proton-nucleon system. The ratios of differential cross sections \rklpa and \rllpa are measured to be 6.2±0.56.2\pm 0.5 and 0.66±0.070.66\pm 0.07, respectively, for \xf ≈−0.06\approx-0.06. No significant dependence upon the target material is observed. Within errors, the slopes of the transverse momentum distributions dσpA/dpt2d\sigma_{pA}/dp_t^2 also show no significant dependence upon the target material. The dependence of the extrapolated total cross sections σpA\sigma_{pA} on the atomic mass AA of the target material is discussed, and the deduced cross sections per nucleon σpN\sigma_{pN} are compared with results obtained at other energies.Comment: 17 pages, 7 figures, 5 table

    Chemical Proteomics-Based Analysis of Off-target Binding Profiles for Rosiglitazone and Pioglitazone: Clues for Assessing Potential for Cardiotoxicity

    Drugs exert desired and undesired effects based on their binding interactions with protein target(s) and off-target(s), providing evidence for drug efficacy and toxicity. Pioglitazone and rosiglitazone possess a common functional core, glitazone, which is considered a privileged scaffold upon which to build a drug selective for a given target—in this case, PPARγ. Herein, we report a retrospective analysis of two variants of the glitazone scaffold, pioglitazone and rosiglitazone, in an effort to identify off-target binding events in the rat heart to explain recently reported cardiovascular risk associated with these drugs. Our results suggest that glitazone has affinity for dehydrogenases, consistent with known binding preferences for related rhodanine cores. Both drugs bound ion channels and modulators, with implications in congestive heart failure, arrhythmia, and peripheral edema. Additional proteins involved in glucose homeostasis, synaptic transduction, and mitochondrial energy production were detected and potentially contribute to drug efficacy and cardiotoxicity

    Coastal greening of grey infrastructure: an update on the state-of-the-art

    \ua9 2023 Emerald Publishing Limited: All rights reserved.In the marine environment, greening of grey infrastructure (GGI) is a rapidly growing field that attempts to encourage native marine life to colonize marine artificial structures to enhance biodiversity, thereby promoting ecosystem functioning and hence service provision. By designing multifunctional sea defences, breakwaters, port complexes and off-shore renewable energy installations, these structures can yield myriad environmental benefits, in particular, addressing UN SDG 14: Life below water. Whilst GGI has shown great promise and there is a growing evidence base, there remain many criticisms and knowledge gaps, and some feel that there is scope for GGI to be abused by developers to facilitate harmful development. Given the surge of research in this field in recent years, it is timely to review the literature to provide an update update on the state-of-the-art of the field in relation to the many criticisms and identify remaining knowledge gaps. Despite the rapid and significant advances made in this field, there is currently a lack of science and practice outside of academic sectors in the developed world, and there is a collective need for schemes that encourage intersectoral and transsectoral research, knowledge exchange, and capacity building to optimize GGI in the pursuit of contributing to sustainable development

    Ectopic Pregnancy as a Model to Identify Endometrial Genes and Signaling Pathways Important in Decidualization and Regulated by Local Trophoblast

    The endometrium in early pregnancy undergoes decidualization and functional changes induced by local trophoblast, which are not fully understood. We hypothesized that endometrium from tubal ectopic pregnancy (EP) could be interrogated to identify novel genes and pathways involved in these processes. Gestation-matched endometrium was collected from women with EP (n = 11) and intrauterine pregnancies (IUP) (n = 13). RNA was extracted from the tissue. In addition, tissues were prepared for histological analysis for degree of decidualization. We compared a) the samples from EP that were decidualized (n = 6) with non-decidualized samples (n = 5), and b) the decidualized EP (n = 6) with decidualization-matched IUP (n = 6) samples using an Affymetrix gene array platform, with Ingenuity Pathway Analysis, combined with quantitative RT-PCR. Expression of PRL and IGFBP1 was used to confirm the degree of decidualization in each group. There were no differences in PRL or IGFBP1 expression in the decidualization-matched samples but a marked reduction (P<0.001) in the non-decidualized samples. Decidualization was associated with increased expression of 428 genes including SCARA5 (181-fold), DKK1 (71-fold) and PROK1 (32-fold), and decreased expression of 230 genes including MMP-7 (35-fold) and SFRP4 (21-fold). The top canonical pathways associated with these differentially expressed genes were Natural Killer Cell and Wnt/b-Catenin signaling. Local trophoblast was associated with much less alteration of endometrial gene expression with an increase in 56 genes, including CSH1 (8-fold), and a reduction in 29 genes including CRISP3 (8-fold). The top associated canonical pathway was Antigen Presentation. The study of endometrium from tubal EP may promote novel insights into genes involved in decidualization and those influenced by factors from neighboring trophoblast. This has afforded unique information not highlighted by previous studies and adds to our understanding of the endometrium in early pregnancy

    Nitrosylation of Myoglobin and Nitrosation of Cysteine by Nitrite in a Model System Simulating Meat Curing

    Demand is growing for meat products cured without the addition of sodium nitrite. Instead of the direct addition of nitrite to meat in formulation, nitrite is supplied by bacterial reduction of natural nitrate often added as vegetable juice/powder. However, the rate of nitrite formation in this process is relatively slow, and the total ingoing nitrite is typically less than in conventional curing processes. The objective of this study was to determine the impact of the rate of addition of nitrite and the amount of nitrite added on nitrosylation/nitrosation reactions in a model meat curing system. Myoglobin was preferentially nitrosylated as no decrease in sulfhydryl groups was found until maximum nitrosylmyoglobin color was achieved. The cysteine–myoglobin model retained more sulfhydryl groups than the cysteine-only model (p \u3c 0.05). The rate of nitrite addition did not alter nitrosylation/nitrosation reactions (p \u3e 0.05).
