    Up-regulation of corticotropin releasing hormone is associated with the downregulation of corticotropin releasing hormone binding protein and up-regulation of IL- 33 and IL-8 expression in psoriasis

    Purpose: To determine the expression of corticotropin releasing hormone (CRH) in psoriasis and normal skin biopsy samples, and to correlate the expression of CRH with the expression of CRHBP and inflammatory cytokines IL-8 and IL-33.Methods: Psoriasis and normal skin biopsy samples were obtained from three psoriatic and three normal healthy patients. The mRNA expression was examined by quantitative real-time polymerase chain reaction (RT-PCR). Protein expression analysis was carried out by Western blotting and then further validated by immunohistochemistry.Results: The results of the present study revealed that the expression of CRH was highly significant (p < 0.0001) in psoriatic skin, compared to normal skin. Increase of CRH in psoriatic samples ranged from 2.7 to 3.5-fold. Expression of CRH was associated with the concomitant downregulation of CRHBP in all the psoriatic skin biopsy samples, with expression of CRHBP 3.0 to 6.2-fold lower in psoriatic skin than in normal skin. Analysis of mRNA expression of IL-8 and IL-33, revealed that expression of both IL-8 and IL-33 was significantly (p < 0.0001) upregulated in psoriatic skin samples while the expression of IL-8 and IL-33 was approximately 4.1- and 3.2-fold higher in psoriatic skin than in normal skin. The expression of CRHBP, IL-8 and IL-33 was further confirmed by western blotting and immunohistochemistry results.Conclusion: The results confirm that the expression of CRH is associated with the suppression of CRHBP and upregulation of IL-8 and IL-33.Keywords: Psoriasis, Corticotropin releasing hormone, Inflammatory cytokines, Interleukin, Skin biops

    Anti-Apoptosis Effect of Astragaloside Iv on Alzheimer's Disease Rat Model via Enhancing the Expression of Bcl-2 And Bcl-Xl

    The aim is to explore the protective effect of Astragaloside IV on Alzheimer’s disease (AD) in rats induced  by amyloid-ß peptide (Aß1-42) and its potential therapeutic mechanism. Methods: 50 Male Sprague Dawley  rats were divided into five groups (10 rats for each): control group, model group, treatment groups 1~3.  10μg Aß1-42 was injected bilaterally into the dorsal dentate gyrus of the hippocampus of rats in the model  and treatment groups to prepare the AD models. 24h after modeling, Astragaloside IV administration, with  different drug dosages of 20mg/(kg•day), 40mg/(kg•day) and 60mg/(kg•day), was performed by gastric  perfusion for rats in the treatment group 1~3. Later on, the cognitive ability of rats was examined by a series  of behavioral tests, and the expression of Bcl-2 and Bcl-xl in the hippocampus of rats was detected by the  fluorescein based Quantitative RT-PCR. Results: The spontaneous alternation test in a Y maze and Morris  water maze task have demonstrated that the repeated daily administration of Astragaloside IV at the doses  of 20mg/kg bw/day) (p<0.05), 40mg/kg bw/day) (p<0.01), and 60mg/kg bw/day) (p<0.01) significantly  ameliorated the impairment of performance caused by Aß1–42. Furthermore, Astragaloside IV also enhanced  the expression of Bcl-2 and Bcl-xl in hippocampal neurons of rats in a dosage-dependent manner. Conclusion:  These findings suggest that Astragaloside IV could alleviate cognitive impairment and enhance the  expression of Bcl-2 and Bcl-xl in hippocampus of rat models with AD.

    Intelligent Multi-Modal Sensing-Communication Integration: Synesthesia of Machines

    In the era of sixth-generation (6G) wireless communications, integrated sensing and communications (ISAC) is recognized as a promising solution to upgrade the physical system by endowing wireless communications with sensing capability. Existing ISAC is mainly oriented to static scenarios with radio-frequency (RF) sensors being the primary participants, thus lacking a comprehensive environment feature characterization and facing a severe performance bottleneck in dynamic environments. To date, extensive surveys on ISAC have been conducted but are limited to summarizing RF-based radar sensing. Currently, some research efforts have been devoted to exploring multi-modal sensing-communication integration but still lack a comprehensive review. Therefore, we generalize the concept of ISAC inspired by human synesthesia to establish a unified framework of intelligent multi-modal sensing-communication integration and provide a comprehensive review under such a framework in this paper. The so-termed Synesthesia of Machines (SoM) gives the clearest cognition of such intelligent integration and details its paradigm for the first time. We commence by justifying the necessity of the new paradigm. Subsequently, we offer a definition of SoM and zoom into the detailed paradigm, which is summarized as three operation modes. To facilitate SoM research, we overview the prerequisite of SoM research, i.e., mixed multi-modal (MMM) datasets. Then, we introduce the mapping relationships between multi-modal sensing and communications. Afterward, we cover the technological review on SoM-enhance-based and SoM-concert-based applications. To corroborate the superiority of SoM, we also present simulation results related to dual-function waveform and predictive beamforming design.     A Hyperthermophilic Argonaute From Ferroglobus placidus With Specificity on Guide Binding Pattern

    Argonaute proteins (Agos) from thermophilic archaea are involved in several important processes, such as host defense and DNA replication. The catalytic mechanism of Ago from different microbes with great diversity and genome editing potential is attracting increasing attention. Here, we describe an Argonaute from hyperthermophilic Ferroglobus placidus (FpAgo), with a typical DNA-guided DNA endonuclease activity but adopted with only a short guide 15–20 nt length rather than a broad guide selectivity for reported Agos. FpAgo performed the precise cleavage of phosphodiester bonds between 10 and 11 nt on the target strand (counting from the guide strand) guided strictly by 5′-phosphorylated DNA at temperatures ranging from 75 to 99°C. The cleavage activity was regulated by the divalent cations Mn2+, Mg2+, Co2+, and Ni2+. In addition, FpAgo possesses guide/target mismatch tolerance in the seed region but is sensitive to mismatches in the 3′-guide region. Notably, the EMSA assay revealed that the FpAgo-guide-target ternary complex exhibited a stronger binding affinity for short 15 and 16 nt guide DNAs than longer guides. Moreover, we performed structural modeling analyses that implied the unique PAZ domain of FpAgo for 3′-guide recognition and binding to affect guide length specificity. This study broadens our understanding of thermophilic Agos and paves the way for their use in DNA manipulation

    Influence of laser cutting conditions on electrical characteristics of half-size bifacial silicon solar cells

    Abstract(#br)The half-size bifacial silicon solar cells have garnered significant research attention in photovoltaic (PV) modules because they render enhanced power output. Herein, the influence of cutting surface and scribing iteration times on electrical characteristics of bifacial silicon solar cells is investigated in detail. The results reveal that the cutting process should be carried out from the rear side and scribing iteration times should be twice. Moreover, we have studied the cutting losses of n-type passivated emitter and rear totally diffused (n-PERT) bifacial solar cells and demonstrated that not mechanical breaking but laser scribing is a major source of losses during the cell separation process. In addition, the damage induced by the cut was systematically investigated and it was observed that the heat-affected zone resulted in negligible damage under optimal cutting conditions. Overall, n-PERT half-cell bifacial modules, sectioned under optimal cutting conditions, can maintain high efficiency and excellent reliability

    Cuproptosis-Related lncRNAs are Biomarkers of Prognosis and Immune Microenvironment in Head and Neck Squamous Cell Carcinoma

    Background: Cuproptosis is a new type of cell death that induces protein toxic stress and eventually leads to cell death. Hence, regulating cuproptosis in tumor cells is a new therapeutic approach. However, studies on cuproptosis-related long noncoding RNA (lncRNA) in head and neck squamous cell carcinoma (HNSC) have not been found. This study aimed to explore the cuproptosis-related lncRNAs prognostic marker and their relationship to immune microenvironment in HNSC by using bioinformatics methods.Methods: RNA sequencing, genomic mutations, and clinical data of TCGA_HNSC were downloaded from The Cancer Genome Atlas. HNSC patients were randomly assigned to either a training group or a validation cohort. The least absolute shrinkage and selection operator Cox regression and multivariate Cox regression models were used to determine the prognostic model in the training cohort, and its independent prognostic effect was further confirmed in the validation and entire cohorts.Results: Based on previous literature, we collected 19 genes associated with cuproptosis. Afterward, 783 cuproptosis-related lncRNAs were obtained through coexpression. Cox model revealed and constructed eight cuproptosis-related lncRNAs prognostic marker (AL132800.1, AC090587.1, AC079160.1, AC011462.4, AL157888.1, GRHL3-AS1, SNHG16, and AC021148.2). Patients were divided into high- and low-risk groups based on the median risk score. The Kaplan–Meier survival curve revealed that the overall survival between the high- and low-risk groups was statistically significant. The receiver operating characteristic curve and principal component analysis demonstrated the accurate prognostic ability of the model. Univariate and multivariate Cox regression analysis showed that risk score was an independent prognostic factor. In addition, we used multivariate Cox regression to establish a nomogram of the predictive power of prognostic markers. The tumor mutation burden showed significant differences between the high- and low-risk groups. HNSC patients in the high-risk group responded better to immunotherapy than those in the low-risk group. We also found that risk scores were significantly associated with drug sensitivity in HNSC.Conclusion: In summary, our study identified eight cuprotosis-related lncRNAs signature of HNSC as the prognostic predictor, which may be promising biomarkers for predicting the benefit of HNSC immunotherapy as well as drug sensitivity

    Biocontrol potential and action mechanism of Bacillus amyloliquefaciens DB2 on Bipolaris sorokiniana

    IntroductionBipolaris sorokiniana is the popular pathogenic fungi fungus which lead to common root rot and leaf spot on wheat. Generally, chemical fungicides are used to control diseases. However, the environmental pollution resulting from fungicides should not be ignored. It is important to study the mode of antagonistic action between biocontrol microbes and plant pathogens to design efficient biocontrol strategies.ResultsAn antagonistic bacterium DB2 was isolated and identified as Bacillus amyloliquefaciens. The inhibition rate of cell-free culture filtrate (CF, 20%, v/v) of DB2 against B. sorokiniana reached 92.67%. Light microscopy and scanning electron microscopy (SEM) showed that the CF significantly altered the mycelial morphology of B. sorokiniana and disrupted cellular integrity. Fluorescence microscopy showed that culture filtrate destroyed mycelial cell membrane integrity, decreased the mitochondrial transmembrane potential, induced reactive oxygen species (ROS) accumulation, and nuclear damage which caused cell death in B. sorokiniana. Moreover, the strain exhibited considerable production of protease and amylase, and showed a significant siderophore and indole-3-acetic acid (IAA) production. In the detached leaves and potted plants control assay, B. amyloliquefacien DB2 had remarkable inhibition activity against B. sorokiniana and the pot control efficacy was 75.22%. Furthermore, DB2 suspension had a significant promotion for wheat seedlings growth.ConclusionB. amyloliquefaciens DB2 can be taken as a potential biocontrol agent to inhibit B. sorokiniana on wheat and promote wheat growth

According to the sesult of the research, can be shown as following: price has a correlation and significant determination effort to the increase sale of PT. ROBCAGA Talaud. According to the data analysis, coefficient value moment r = 0,685. That show there is a positive relation, and can be categorize as high and strong, also price coefficient determination to the increase sale is by 46,5% and 53,5% by the rest of it depends on the unknown factors that not been analyze in this research
