    Protein O-Fucosyltransferase 1 Undergoes Interdomain Flexibility in Solution

    Protein O-fucosyltransferase 1 (PoFUT1) is a GT-B fold enzyme that fucosylates proteins containing EGF-like repeats. GT-B glycosyltransferases have shown a remarkable grade of plasticity adopting closed and open conformations as a way of tuning their catalytic cycle, a feature that has not been observed for PoFUT1. Here, we analyzed Caenorhabditis elegans PoFUT1 (CePoFUT1) conformational behavior in solution by atomic force microscopy (AFM) and single-molecule fluorescence resonance energy transfer (SMF-FRET). Our results show that this enzyme is very flexible and adopts mainly compact conformations and to a lesser extend a highly dynamic population that oscillates between compact and highly extended conformations.     Estudio electrocardiografía en estación y de cúbito lateral derecho en crías de alpaca (Vicugna pacos)

    The aim of the study was to determine the electrocardiographic parameters in both station and right lateral decubitus positions in baby alpacas. The work was done in 30 alpacas (17 males and 13 females) of 60 days old, from the communities alpaqueras Maranganí, Canchis in Cusco. The electrocardiographic values in station and right lateral decubitus were: heart rate 124.83 and 114.16, P wave duration 0.04 and 0.04 s, P wave amplitude 0.180 and 0.173 mV, duration and PQ 0.101 s 0.102, 0.039 and 0.038 QRS duration s, QRS complex 0.643 and 0.557 mV, duration and QT 0.225 0.2370.040 s, T wave amplitude 0.393 and 0.413 mV. The values of ECG parameters showed no significant differences between the two positions. We conclude that is possible to use any positions for the clinic examen in alpacas.El presente estudio tuvo por objetivo determinar los parámetros electrocardiográficos en las posiciones estación y decúbito lateral derecha en crías de alpacas. El trabajo se hizo en 30 alpacas (17 machos y 13 hembras) menores de 60 días de edad, provenientes de las comunidades alpaqueras de Maranganí, Canchis, en Cusco. Los valores electrocardiográficos en estación y en decúbito lateral derecho fueron: Frecuencia cardiaca 124.83 y 114.16, Duración onda P 0.04 y 0.04 s, amplitud onda P 0.180 y 0.173 mV, duración PQ 0.102 y 0.101 s, duración QRS 0.039 y 0.038 s, complejo QRS 0.643 y 0.557 mV, duración QT 0.225 y 0.2370.040 s, amplitud onda T 0.393 y 0.413 mV, los valores no mostraron diferencias significativas al comparalos entre ambas posiciones.     Structural Insights into the Mechanism of Protein O-Fucosylation

    Protein O-fucosylation is an essential post-translational modification, involved in the folding of target proteins and in the role of these target proteins during embryonic development and adult tissue homeostasis, among other things. Two different enzymes are responsible for this modification, Protein O-fucosyltransferase 1 and 2 (POFUT1 and POFUT2, respectively). Both proteins have been characterised biologically and enzymatically but nothing is known at the molecular or structural level. Here we describe the first crystal structure of a catalytically functional POFUT1 in an apo-form and in complex with GDP-fucose and GDP. The enzyme belongs to the GT-B family and is not dependent on manganese for activity. GDP-fucose/GDP is localised in a conserved cavity connected to a large solvent exposed pocket, which we show is the binding site of epidermal growth factor (EGF) repeats in the extracellular domain of the Notch Receptor. Through both mutational and kinetic studies we have identified which residues are involved in binding and catalysis and have determined that the Arg240 residue is a key catalytic residue. We also propose a novel SN1-like catalytic mechanism with formation of an intimate ion pair, in which the glycosidic bond is cleaved before the nucleophilic attack; and theoretical calculations at a DFT (B3LYP/6-31+G(d,p) support this mechanism. Thus, the crystal structure together with our mutagenesis studies explain the molecular mechanism of POFUT1 and provide a new starting point for the design of functional inhibitors to this critical enzyme in the future

    CO2 adsorption on agricultural biomass combustion ashes

    Carbon capture and storage has become an alternative means of confronting global warming. Further research and development into adequate and low-cost materials is required for CO2 adsorption technologies.Samples of fly ash, bottom ash and their respective pellets, produced from wheat bran combustion, were characterized and tested to assess their capacity for CO2 adsorption at different temperatures. Neither the ashes nor their pellets were subject to prior thermochemical activation.The bottom ash sample and its pellets showed a higher adsorption capacity for the majority of the temperatures studied. The pelletized bottom ash reached the maximum adsorption capacity (0,07 mmol CO2/g), followed by the non-pelletized bottom ash (0,06 mmol CO2/g); both at an adsorption temperature of 25°C.CO2 adsorption of bottom ash, from the combustion of wheat bran (agricultural biomass), by a physical adsorption mechanism was demonstrated whereas with the fly ash sample, CO2 adsorption by both physical and chemical adsorption mechanisms was identified

    A Native Ternary Complex Trapped in a Crystal Reveals the Catalytic Mechanism of a Retaining Glycosyltransferase

    Glycosyltransferases (GTs) comprise a prominent family of enzymes that play critical roles in a variety of cellular processes, including cell signaling, cell development, and host–pathogen interactions. Glycosyl transfer can proceed with either inversion or retention of the anomeric configuration with respect to the reaction substrates and products. The elucidation of the catalytic mechanism of retaining GTs remains a major challenge. A native ternary complex of a GT in a productive mode for catalysis is reported, that of the retaining glucosyl-3-phosphoglycerate synthase GpgS from M. tuberculosis in the presence of the sugar donor UDP-Glc, the acceptor substrate phosphoglycerate, and the divalent cation cofactor. Through a combination of structural, chemical, enzymatic, molecular dynamics, and quantum-mechanics/molecular-mechanics (QM/MM) calculations, the catalytic mechanism was unraveled, thereby providing a strong experimental support for a front–side substrate-assisted SNi-type reaction

    Environmental disasters, ranging from catastrophic floods to extreme temperatures, have caused more than 30,000 deaths per year and more than US$ 250–300 billion a year in economic losses, globally, between 1995 and 2015. Improved infrastructure and planning for extreme events is essential in urban areas, where an increasingly greater fraction of the world’s inhabitants reside. In response, international governmental and private initiatives have placed the goal of resilience at the center stage of urban planning. [For example, The 100 Resilient Cities Initiative (www.100resilientcities.org/); the Global Covenant of Mayors (https://www.compactofmayors.org/globalcovenantofmayors/); and the recent UN Habitat III (https://habitat3.org/the-new-urban-agenda)].     The aim of this study was to determine the levels of serum potassium and electrocardiographic parameters in young alpacas. Thirty alpacas (17 males and 13 females) from 1 to 60 days of age from various alpaca farms in Maranganí, Cusco were used. The levels of serum potassium was 5.91 ± 1.48 mmol/L and without statistical differences between sexes. The electrocardiographic values were: P wave time, 0.04 ± 0.0s; P wave amplitude, 0.173 ± 0.058 mV; PQ time, 0.101 ± 0.02 s; QRS time, 0.038 ± 0.006 s; QRS complex, 0.557 ± 0.412 mV; QT time, 0.237 ± 0.040 s; T wave amplitude, 0.413 ± 0.266 mV. Moreover, animals with increased T wave had potassium values above 5.5 mmol/L but without signs of heart disease. In addition, values of serum potassium and electrocardiographic paramters showed similarity to the values of other species.El presente estudio tuvo por objetivo determinar los niveles de potasio sérico y los parámetros electrocardiográficos. El trabajo se hizo en 30 alpacas (17 machos y 13 hembras) menores de 60 días de edad, provenientes de las comunidades alpaqueras de Maranganí, Canchis, en Cusco. El nivel de potasio sérico fue de 5.91 ± 1.48 mmol/L, sin diferencias estadísticas entre sexos. Los valores electrocardiográficos fueron: Duración onda P, 0.04 ± 0.0 s; amplitud onda P, 0.173 ± 0.058 mV; duración PQ, 0.101 ± 0.020 s; duración QRS, 0.038 ± 0.006 s; complejo QRS, 0.557 ± 0.412 mV; duración QT, 0.237 ± 0.040 s; amplitud onda T, 0.413 ± 0.266 mV.     El objetivo de este estudio fue determinar los valores hematológicos y de bioquímica sérica de una población de monos choros (Lagothrix lagotricha) que se encontraban en semicautiverio en el Centro de Rescate y Rehabilitación Ikama peru, en Lagunas, Loreto. Se trabajó con 40 monos, 12 machos y 28 hembras entre juveniles, sub-adultos y adultos en aparente buen estado de salud. Los valores hematológicos fueron similares, los niveles de triglicéridos y de AST estuvieron elevados y los de proteínas totales yALP estuvieron bajos en relación a estudios realizados en zoológicos. El grupo etarioinfluenció los niveles de eritrocitos, proteínas totales, bilirrubina total, colesterol yfosfatasa alcalina; asimismo, no hubo diferencias por efecto del sexo en la serie eritrocítica, conteo de plaquetas y bioquímica sérica, con excepción del número de monocitos.The objective of this study was to determine the haematological and serum biochemistry values in a population of woolly monkeys choros (Lagothrix lagotricha)reared in semi-captivity at the Centro de Rescate y Rehabilitación Ikamaperu in Lagunas, Loreto, Peru. Forty primates, 12 males and 28 females, which included juveniles, subadults and adults in apparent good health conditions were sampled. The haematological values were similar, triglyceride and AST values were elevated, and total protein and ALP values were low as compared to other studies in zoos. The age group influenced erythrocyte counts, total protein, total bilirubin, cholesterol and alkaline phosphatase. No sex effect was found on erythrocytes, leukocytes and platelet counts and in serum chemistry values, except for the number of monocytes
