    Differential Dynamic Microscopy of Bacterial Motility

    We demonstrate 'differential dynamic microscopy' (DDM) for the fast, high throughput characterization of the dynamics of active particles. Specifically, we characterize the swimming speed distribution and the fraction of motile cells in suspensions of Escherichia coli bacteria. By averaging over ~10^4 cells, our results are highly accurate compared to conventional tracking. The diffusivity of non-motile cells is enhanced by an amount proportional to the concentration of motile cells.Comment: 4 pages, 4 figures. In this updated version we have added simulations to support our interpretation, and changed the model for the swimming speed probability distribution from log-normal to a Schulz distribution.     MLPH Genotype—Melanin Phenotype Correlation in Dilute Dogs

    Coat color dilution in dogs is a specific pigmentation phenotype caused by a defective transport of melanosomes leading to large clumps of pigment. It is inherited as a Mendelian autosomal recessive trait and may be accompanied by hair loss, the so-called color dilution alopecia (CDA), or black hair follicular dysplasia (BHFD). We previously identified the noncoding c.-22G>A transition in the melanophilin gene (MLPH) as a candidate causative mutation for the dilute phenotype. We have now extended our study and genotyped 935 dogs from 20 breeds segregating for dilute coat color. The dilute-associated A allele segregates in many different breeds suggesting an old mutation event. We also investigated skin biopsies of dogs suspected of having either CDA or BHFD, and our data clearly indicate that the dilute mutation is required but not sufficient to develop clinical signs of the disease. The risk to develop CDA/BHFD seems to be breed specific. Interestingly, 22 out of 29 dogs with clinical signs of CDA/BHFD have clumped melanin in the epidermis, the follicular epithelium, and the hair shafts, whereas in dilute dogs without clinical disease, clumped melanin is only found in the follicular epithelium and the hair shafts but not in the epidermi

    The Role of the Mucus Barrier in Digestion

    Mucus forms a protective layer across a variety of epithelial surfaces. In the gastrointestinal (GI) tract, the barrier has to permit the uptake of nutrients, while excluding potential hazards, such as pathogenic bacteria. In this short review article, we look at recent literature on the structure, location, and properties of the mammalian intestinal secreted mucins and the mucus layer they form over a wide range of length scales. In particular, we look at the structure of the gel-forming glycoprotein MUC2, the primary intestinal secreted mucin, and the influence this has on the properties of the mucus layer. We show that, even at the level of the protein backbone, MUC2 is highly heterogeneous and that this is reflected in the networks it forms. It is evident that a combination of charge and pore size determines what can diffuse through the layer to the underlying gut epithelium. This information is important for the targeted delivery of bioactive molecules, including nutrients and pharmaceuticals, and for understanding how GI health is maintained

    Boundary Limitation of Wavenumbers in Taylor-Vortex Flow

    We report experimental results for a boundary-mediated wavenumber-adjustment mechanism and for a boundary-limited wavenumber-band of Taylor-vortex flow (TVF). The system consists of fluid contained between two concentric cylinders with the inner one rotating at an angular frequency Ω\Omega. As observed previously, the Eckhaus instability (a bulk instability) is observed and limits the stable wavenumber band when the system is terminated axially by two rigid, non-rotating plates. The band width is then of order ϵ1/2\epsilon^{1/2} at small ϵ\epsilon (ϵΩ/Ωc1\epsilon \equiv \Omega/\Omega_c - 1) and agrees well with calculations based on the equations of motion over a wide ϵ\epsilon-range. When the cylinder axis is vertical and the upper liquid surface is free (i.e. an air-liquid interface), vortices can be generated or expelled at the free surface because there the phase of the structure is only weakly pinned. The band of wavenumbers over which Taylor-vortex flow exists is then more narrow than the stable band limited by the Eckhaus instability. At small ϵ\epsilon the boundary-mediated band-width is linear in ϵ\epsilon. These results are qualitatively consistent with theoretical predictions, but to our knowledge a quantitative calculation for TVF with a free surface does not exist.Comment: 8 pages incl. 9 eps figures bitmap version of Fig

    YBa2Cu3O7 and Nb NanoSQUIDs for the Investigation of Magnetization Reversal of Individual Magnetic Nanoparticles

    We report on the fabrication, performance and application of sensitive YBa2Cu3O7 (YBCO) and Nb nanoSQUIDs to magnetization reversal measurements of individual magnetic nanoparticles. The YBCO SQUIDs are based on grain boundary Josephson junctions and are patterned in a single layer of epitaxially grown YBCO films by Ga focused ion beam milling. The Nb SQUIDs contain sandwich-type Josephson junctions with normal conducting HfTi barriers; they are fabricated with a multilayer technology that includes patterning by e-beam lithography and a combination of milling techniques and chemical-mechanical polishing. Due to the small inductance of the SQUID loops, ultralow white flux noise at 4.2 K can be achieved, which yields spin sensitivities of down to a few Bohr magnetons per unit bandwidth for a magnetic nanoparticle placed at 10 nm distance to the SQUID loop

    Multiple adhesin proteins on the cell surface of Streptococcus gordonii are involved in adhesion to human fibronectin

    Adhesion of bacterial cells to fibronectin (FN) is thought to be a pivotal step in the pathogenesis of invasive infectious diseases. Viridans group streptococci such as Streptococcus gordonii are considered commensal members of the oral microflora, but are important pathogens in infective endocarditis. S. gordonii expresses a battery of cell-surface adhesins that act alone or in concert to bind host receptors. Here, we employed molecular genetic approaches to determine the relative contributions of five known S. gordonii surface proteins to adherence to human FN. Binding levels to FN by isogenic mutants lacking Hsa glycoprotein were reduced by 70 %, while mutants lacking CshA and CshB fibrillar proteins showed approximately 30 % reduced binding. By contrast, disruption of antigen I/II adhesin genes sspA and sspB in a wild-type background did not result in reduced FN binding. Enzymic removal of sialic acids from FN led to reduced S. gordonii DL1 adhesion (>50 %), but did not affect binding by the hsa mutant, indicating that Hsa interacts with sialic acid moieties on FN. Conversely, desialylation of FN did not affect adherence levels of Lactococcus lactis cells expressing SspA or SspB polypeptides. Complementation of the hsa mutant partially restored adhesion to FN. A model is proposed for FN binding by S. gordonii in which Hsa and CshA/CshB are primary adhesins, and SspA or SspB play secondary roles. Understanding the basis of oral streptococcal interactions with FN will provide a foundation for development of new strategies to control infective endocarditis

    The Hierarchical Age-Period-Cohort model: Why does it find the results that it finds?

    It is claimed the hierarchical-age–period–cohort (HAPC) model solves the age–period–cohort (APC) identification problem. However, this is debateable; simulations show situations where the model produces incorrect results, countered by proponents of the model arguing those simulations are not relevant to real-life scenarios. This paper moves beyond questioning whether the HAPC model works, to why it produces the results it does. We argue HAPC estimates are the result not of the distinctive substantive APC processes occurring in the dataset, but are primarily an artefact of the data structure—that is, the way the data has been collected. Were the data collected differently, the results produced would be different. This is illustrated both with simulations and real data, the latter by taking a variety of samples from the National Health Interview Survey (NHIS) data used by Reither et al. (Soc Sci Med 69(10):1439–1448, 2009) in their HAPC study of obesity. When a sample based on a small range of cohorts is taken, such that the period range is much greater than the cohort range, the results produced are very different to those produced when cohort groups span a much wider range than periods, as is structurally the case with repeated cross-sectional data. The paper also addresses the latest defence of the HAPC model by its proponents (Reither et al. in Soc Sci Med 145:125–128, 2015a). The results lend further support to the view that the HAPC model is not able to accurately discern APC effects, and should be used with caution when there appear to be period or cohort near-linear trends

    Twenty Years After the Velvet Revolution: Shifts in Czech Adolescents’ Perceptions of Family, School, and Society

    Families and schools are traditionally seen as substantial socialization agents forming adolescents’ social values and their views of society. Special attention is paid to the question whether the relative importance of these influences remains stable in times of major social changes. In this study, two different generations of Czech middle adolescents are compared: (a) the “post-totalitarian” generation that grew up in the last decade of the communist regime and entered adolescence during the time of rapid political and socioeconomic changes (data collected in 1995) and (b) the current generation without personal experience with the communist regime, raised in a stable democratic society (data collected in 2010). Both groups of participants (total N = 2,127, aged from 14 to 17 years) were administered an identical questionnaire. First, we examined the changes in adolescents’ perception and evaluation of the society over the last 15 years. Today’s adolescents perceive society more as a community and their future orientations are more focused on materialistic and less on environmental values. While the emotional relationship between the children and parentsremains the same, adolescents learn a somewhat different message in the family, emphasizing self-reliance. School environment is perceived more as positive and engaging than 15 years ago. Second, we predicted adolescents’ social views and values from their assessment of family and school environment. Our results show that the effect of parental values on adolescents’ value orientations is higher in the current generation. Positive school environment contributes to the development of socially responsible orientations despite the changes in society

    Oscillatory surface rheotaxis of swimming E. coli bacteria

    Bacterial contamination of biological conducts, catheters or water resources is a major threat to public health and can be amplified by the ability of bacteria to swim upstream. The mechanisms of this rheotaxis, the reorientation with respect to flow gradients, often in complex and confined environments, are still poorly understood. Here, we follow individual E. coli bacteria swimming at surfaces under shear flow with two complementary experimental assays, based on 3D Lagrangian tracking and fluorescent flagellar labelling and we develop a theoretical model for their rheotactic motion. Three transitions are identified with increasing shear rate: Above a first critical shear rate, bacteria shift to swimming upstream. After a second threshold, we report the discovery of an oscillatory rheotaxis. Beyond a third transition, we further observe coexistence of rheotaxis along the positive and negative vorticity directions. A full theoretical analysis explains these regimes and predicts the corresponding critical shear rates. The predicted transitions as well as the oscillation dynamics are in good agreement with experimental observations. Our results shed new light on bacterial transport and reveal new strategies for contamination prevention.Comment: 12 pages, 5 figure
