    This article aims to present the redesign process of the digital educational material ”Primeiro, Aprender! Digital”, adapted for students who carry hearing impairment, by showing the strategies used in order to make modifications in its interface, so that it meets the specific needs of this public. A pilot version of the material was created and named “Primeiro, Aprender! Digital com Libras”, which is in tests phase, to verify wheter the adjustments contributed to the access of deaf people and, in future, the production of a series of classes based in the original product can be conducted

    Reliability of maximal respiratory nasal pressure tests in healthy young adults

    [Abstract] Introduction: Sniff nasal inspiratory (SNIP) and expiratory pressure (SNEP) may complement the assessment of respiratory muscle strength. Thus, specifying their reliability is relevant to improving the clinical consistency of both tests. Objective: To assess the reliability of SNIP and SNEP in healthy young adults. Methods: This cross-sectional study included self-reported healthy aged 18 to 29 years. SNIP was performed using a plug to occlude one nostril, while SNEP was conducted using a facemask. Participants performed 20 SNIP and SNEP maneuvers with 30-second intervals in between. The intraclass correlation coefficient (ICC), standard error of measurement (SEM), and minimum detectable change (MDC) assessed the reliability of SNIP and SNEP. Analyses were conducted between the highest peak pressure and the first reproducible maneuver in men and women. Results: The total sample comprised 32 participants: 16 men and 16 women. The ICC, SEM, and MDC for SNIP maneuvers were 0.994 (95%CI 0.988 to 0.997), 1.820 cmH2O, and 5.043 cmH2O, respectively. For SNEP, these parameters were 0.950 (95%CI 0.897 to 0.976), 6.03 cmH2O, and 16.716 cmH2O. The SNIP and SNEP in men showed ICC of 0.992 (95%CI 0.977 to 0.997) and 0.877 (95%CI 0.648 to 0.957), SEM of 2.07 and 7.66 cmH2O, and MDC of 5.74 and 21.23 cmH2O. In women, SNIP and SNEP presented ICC of 0.992 (95%CI 0.977 to 0.997) and 0.957 (95%CI 0.878 to 0.985), SEM of 1.15 and 6.11 cmH2O, and MDC of 3.19 and 16.95 cmH2O. Also, 60% of the highest SNIPs occurred among the 11th and 20th maneuvers in men and women. In men, 55% of the highest SNEPs occurred among the 11th and 20th maneuvers; this value was 50% in women. Conclusion: SNIP and SNEP showed excellent reliability. The reliability of SNIP and SNEP in men was good and excellent, respectively, whereas both tests had excellent reliability in women.     Development and characterization of high-absorption microencapsulated organic propolis EPP-AF® extract (i-CAPs)

    The demand for organic and functional food continues to increase yearly. Among the available functional foods, propolis is a bee product that has various beneficial properties, including antimicrobial, antioxidant, and anti-inflammatory activities. However, it generally is only available in ethanol solution, which has poor bioavailability, as it is relatively insoluble in water. The use of such ethanol extracts is often objectionable because of the alcohol content and because they have a strong and striking taste. Development of alternatives that can efficiently and safely increase solubility in water, and that meet organic production specifications, has been a challenge. To address these concerns, microcapsules were developed using spray-dryer technology from an emulsion based on EPP-AF® propolis and gum arabic (i-CAPS). These propolis-loaded microcapsules were characterized using FT-IR, SEM, TGA, HPLC, and spectrophotometric techniques, along with determination of antimicrobial, antioxidant, antitumor, anti-inflammatory, and antihypercholesterolemic activities, as well as permeability in in vitro models. The production system resulted in microcapsules with a spherical shape and an encapsulation efficiency of 93.7 0.7%. They had IC50s of 2.654 0.062 and 7.342 0.058 g/mL by FRAP and DPPH antioxidant methods, respectively. The EPP-AF® i-CAPS also had superior antimicrobial activity against Gram-positive bacteria. Antitumor activity was calculated based on the concentration that inhibited 50% of growth of AGS, Caco-2, and MCF-7 cell strains, giving results of 154.0 1.0, 117 1.0, and 271.0 25 g/mL, respectively. The microcapsule presentation reduced the permeation of cholesterol by 53.7%, demonstrating antihypercholesterolemic activity, and it improved the permeability of p-coumaric acid and artepillin C. The IC50 for NO production in RAW264.7 cells was 59.0 0.1 g/mL. These findings demonstrate the potential of this new propolis product as a food and pharmaceutical ingredient, though additional studies are recommended to validate the safety of proposed dosages.CNPq financially supported this research through the CNPQ/MCTI/SEMPI No. 021/2021 process on RHAE modality, contract number 424727/2021-8.     Lycopene-rich extract from red guava ( Psidium guajava L.) displays cytotoxic effect against human breast adenocarcinoma cell line MCF-7 via an apoptotic-like pathway

    This study investigated a lycopene-rich extract from red guava (LEG) for its chemical composition using spectrophotometry, mass spectrometry, attenuated total reflectance-fourier transform infrared spectroscopy (ATR-FTIR), and computational studies. The cytotoxic activity of LEG and the underlying mechanism was studied in human breast adenocarcinoma cells (MCF-7), murine fibroblast cells (NIH-3T3), BALB/c murine peritoneal macrophages, and sheep blood erythrocytes by evaluating the cell viability with 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method and flow cytometry. Spectrophotometry analysis showed that LEG contained 20% of lycopene per extract dry weight. Experimental and theoretical ATR-FTIR suggests the presence of lycopene, whereas MS/MS spectra obtained after fragmentation of the molecular ion [M]+• of 536.4364 show fragment ions at m/z 269.2259, 375.3034, 444.3788, and 467.3658, corroborating the presence of lycopene mostly related to all-trans configuration. Treatment with LEG (1600 to 6.25μg/mL) for 24 and 72h significantly affected the viability of MCF-7 cells (mean half maximal inhibitory concentration [IC50]=29.85 and 5.964μg/mL, respectively) but not NIH-3T3 cells (IC50=1579 and 911.5μg/mL, respectively). Furthermore LEG at concentrations from 800 to 6.25μg/mL presented low cytotoxicity against BALB/c peritoneal macrophages (IC50≥800μg/mL) and no hemolytic activity. LEG (400 and 800μg/mL) caused reduction in the cell proliferation and induced cell cycle arrest, DNA fragmentation, modifications in the mitochondrial membrane potential, and morphologic changes related to granularity and size in MCF-7 cells; however, it failed to cause any significant damage to the cell membrane or display necrosis or traditional apoptosis. In conclusion, LEG was able to induce cytostatic and cytotoxic effects on breast cancer cells probably via induction of an apoptotic-like pathway.The authors acknowledge the computational time provided by CENAPAD/SP on the project proj697.     INTOXICAÇÃO POR PTERIDIUM ARACHNOIDEUM NO OESTE DE SANTA CATARINA - ESTUDO RETROSPECTIVO (2013-2017)

    Samambaia (Pteridium arachnoideum) é uma das plantas tóxicas mais importantes na criação de bovinos de todo o Brasil. O objetivo desse trabalho foi descrever os diagnósticos de intoxicação por samambaia, em bovinos na região oeste de Santa Catarina, realizados pelo Laboratório de Patologia Veterinária (LPV) do IFC – Campus Concórdia, no período entre 2013 e 2017. No período, foram realizadas 641 necropsias de bovinos, sendo que 22 dessas tiveram o diagnóstico de intoxicação por samambaia, totalizando 3,4% da casuística total de necropsias em bovinos. Foram 13 casos de síndrome hemorrágica aguda, seis casos de carcinomas de células escamosas na base da língua, esôfago ou rúmen, e três casos de hematúria enzoótica bovina. Apesar dos inúmeros trabalhos científicos a respeito da toxicidade do P. arachnoideum, o número de diagnósticos dessa intoxicação é alto na região, e a planta ainda é facilmente encontrada. A melhor forma de diminuir as intoxicações é o adequado manejo das pastagens e a correta adubação do solo, evitando com que a planta esteja disponível ao consumo dos animais, porém essa informação precisa ser difundida aos produtores rurais.  Palavras-chave: diagnóstico, patologia, samambaia, hematúria, carcinoma.

    Defoliation of strawberry mother plants for the production of runner tips

    The objective of this work was to determine the sink-source relationships and their effects on the number and growth of runner tips of 'Camino Real' strawberry stock plants. Three types of sources were evaluated: one defoliation at 96 days after planting (DAP), two defoliations at 50 and 96 DAP, and mother plants without defoliation. Four types of sink were accessed: runner tips collected weekly and monthly, four stolons with rooted runner tips in pots, and four freely-grown stolons. A completely randomized experimental design was used in a split-plot arrangement, with four replicates. The source types were placed in the plots, and sink types in the subplots. The number of runner tips, the crown diameter, and the dry matter mass were determined. Number and growth of tips were higher on plants without defoliation, and decreased 44.7% on twice-defoliated mother plants. The two-defoliation management did not reduce runner tip dry matter mass only on plants with rooted stolons, which produced runner tips 50% heavier. Defoliation of mother plants bearing rooting stolons can be used to reduce their growth, without reducing the emission and growth of runner tips

    Wildfire incidence throughout the Brazilian Pantanal is driven by local climate rather than bovine stocking density

    The Pantanal is the world’s largest and most biodiverse continental sheet-flow wetland. Recently, vast tracts of the Pantanal have succumbed to the occurrence of fires, raising serious concerns over the future integrity of the biodiversity and ecosystem services of this biome, including revenues from ecotourism. These wildfires degrade the baseline of natural ecosystems and the ecotourism economy across the region. Local residents (“Pantaneiros”) anecdotally state that extensive cattle herbivory can solve the contemporary flammability problem of the Pantanal by controlling vegetation biomass, thereby preventing or reducing both fuel loads and fires across the region. Here, we examine the covariation between the presence and density of cattle and the incidence of fires across the Brazilian Pantanal. Variables assessed included bovine cattle density, SPI (Standardized Precipitation Index), GPP (Gross Primary Productivity)/biomass estimate, and fire foci along a 19-year time series (2001 to 2019). Our findings show that fire foci across the Pantanal biome are related to climatic variables, such as lower annual precipitation and higher annual drought indices (SPI) rather than to cattle stocking rates. Therefore, the notion of “cattle firefighting”, a popular concept often discussed in some academic circles, cannot be validated because cattle numbers are unrelated to aboveground phytomass. Gross primary productivity further invalidated the “cattle herbivory” hypothesis because GPP was found to be strongly correlated with cattle density but not with the spatial distribution of fires. Fires throughout the Pantanal are currently aggravated by the presence of livestock and result from a combination of extreme weather events and outdated agricultural practices

    Tele-electrocardiography and bigdata: the CODE (Clinical Outcomes in Digital Electrocardiography) study

    Digital electrocardiographs are now widely available and a large number of digital electrocardiograms (ECGs) have been recorded and stored. The present study describes the development and clinical applications of a large database of such digital ECGs, namely the CODE (Clinical Outcomes in Digital Electrocardiology) study. ECGs obtained by the Telehealth Network of Minas Gerais, Brazil, from 2010 to 17, were organized in a structured database. A hierarchical free-text machine learning algorithm recognized specific ECG diagnoses from cardiologist reports. The Glasgow ECG Analysis Program provided Minnesota Codes and automatic diagnostic statements. The presence of a specific ECG abnormality was considered when both automatic and medical diagnosis were concordant; cases of discordance were decided using heuristisc rules and manual review. The ECG database was linked to the national mortality information system using probabilistic linkage methods. From 2,470,424 ECGs, 1,773,689 patients were identified. After excluding the ECGs with technical problems and patients <16 years-old, 1,558,415 patients were studied. High performance measures were obtained using an end-to-end deep neural network trained to detect 6 types of ECG abnormalities, with F1 scores >80% and specificity >99% in an independent test dataset. We also evaluated the risk of mortality associated with the presence of atrial fibrillation (AF), which showed that AF was a strong predictor of cardiovascular mortality and mortality for all causes, with increased risk in women. In conclusion, a large database that comprises all ECGs performed by a large telehealth network can be useful for further developments in the field of digital electrocardiography, clinical cardiology and cardiovascular epidemiology
