    Experts and evidence in deliberation: scrutinising the role of witnesses and evidence in mini-publics, a case study

    Experts hold a prominent position in guiding and shaping policy-making; however, the nature of expert input to decision-making is a topic of public debate. A key aspect of deliberative processes such as citizens’ juries is the provision of information to participants, usually from expert witnesses. However, there is currently little guidance on some of the challenges that organisers and advocates of citizens’ juries must consider regarding expert involvement, including the role of the witness, issues around witness identification and selection, the format of evidence provision, the evidence itself, and how these factors affect the experience of the participants and the witnesses. Here, we explore these issues through detailed case study of three citizens’ juries on onshore wind farm development in Scotland, including interviews with the witnesses involved. This is complemented by examining a cohort of mini-publics held on energy and the environment topics, and, where possible, discussion with the program organisers. We identify a series of issues and sensitivities that can compromise the effectiveness and fairness of the evidence-giving in mini-publics, for the participants, the witnesses and the organisers. We recommend approaches and areas for future work to address these challenges. This is the first time that the ways of involving witnesses in such processes have been so comprehensively examined, and is timely given the increasing interest in democratic innovations such as mini-publics and the current discourse concerning experts

    Protection against glucolipotoxicity by high density lipoprotein in human PANC-1 hybrid 1.1B4 pancreatic beta cells: the role of microRNA

    High-density lipoproteins provide protection against the damaging effects of glucolipotoxicity in beta cells, a factor which sustains insulin secretion and staves off onset of type 2 diabetes mellitus. This study examines epigenetic changes in small non-coding microRNA sequences induced by high density lipoproteins in a human hybrid beta cell model, and tests the impact of delivery of a single sequence in protecting against glucolipotoxicity. Human PANC-1.1B4 cells were used to establish Bmax and Kd for [3H]cholesterol efflux to high density lipoprotein, and minimum concentrations required to protect cell viability and reduce apoptosis to 30mM glucose and 0.25 mM palmitic acid. Microchip array identified the microRNA signature associated with high density lipoprotein treatment, and one sequence, hsa-miR-21-5p, modulated via delivery of a mimic and inhibitor. The results confirm that low concentrations of high-density lipoprotein can protect against glucolipotoxicity, and report the global microRNA profile associated with this lipoprotein; delivery of miR-21-5p mimic altered gene targets, similar to high density lipoprotein, but could not provide sufficient protection against glucolipotoxicity. We conclude that the complex profile of microRNA changes due to HDL treatment may be difficult to replicate using a single microRNA, findings which may inform current drug strategies focused on this approach

    Low-power synthesis flow for regular processor design

    Flow around an ICE2 high-speed train exiting a tunnel under the influence of a wind gust has been studied using numerical technique called detached eddy simulation. A wind gust boundary condition was derived to approximate previous experimental observations. The body of the train includes most important details including bogies, plugs, inter-car gaps and rotating wheels on the rail. The maximal yawing and rolling moments which possibly can cause a derailment or overturning were found to occur when approximately one third and one half of the train, respectively, has left the tunnel. These are explained by development of a strong vortex trailing along the upper leeward edge of the train. All aerodynamic forces and moments were monitored during the simulation and the underlying flow structures and mechanisms are explained

    Use of a saliva-based diagnostic test to identify tapeworm infection in horses in the UK

    Background: Anthelmintic resistance combined with limited chemotherapeutic options has prompted a change in approaches to control of equine helminth infections. Targeted selective treatment strategies use diagnostics to reduce anthelmintic use by treating individuals with worm burdens or egg shedding levels above a set threshold. While faecal egg count analysis has limitations for informing tapeworm treatment, a commercially available saliva-based diagnostic test accurately diagnoses horses with tapeworm infection. Objectives: Evaluation of a saliva-based diagnostic test to identify horses naturally infected with tapeworm and assess the impact of using the test to inform anthelmintic administration. Study design: Retrospective longitudinal study. Methods: Saliva was collected from horses (n = 237) at a UK welfare charity from autumn 2015 to autumn 2016. Horses diagnosed as positive for tapeworm infection using the EquiSal® Tapeworm test were anthelmintic treated according to weight. The number of horses that received anthelmintic treatment based on the test result was compared with an all-group treatment approach and the reduction in anthelmintic usage calculated. Incoming horses were also tested (n = 143) and the information was used to inform quarantine treatments. Results: In autumn 2015, 85% of 237 horses tested received no anthelmintic and the majority (71%) of these remained below the treatment threshold throughout the study. Of the 69 horses that received treatment, seven required treatment following three subsequent tests, while >50% of horses administered with anthelmintic fell below the treatment threshold at the following test. No increase in tapeworm prevalence within the 237 horses was observed during the study despite a substantial reduction in the application of antitapeworm treatments. A total of 41% of incoming horses required anticestode treatment. Main limitations: Other management practices were not included in the analysis. Conclusions: Compared with an all-group treatment strategy, the diagnostic-led approach used here considerably reduced application of anticestode anthelmintics. This could reduce selection pressure for anthelmintic resistance

    Virtual synchronous-machine control of voltage-source converters in a low-voltage microgrid

    In order to facilitate the further integration of distributed renewable generation into existing power systems, enhanced control schemes for grid-tied power electronic converters are necessary to ensure non-synchronous power sources can provide power and support to the grid. The virtual-synchronous-machine concept proposes the use of control schemes to enable static generators to operate with the dynamics of rotating synchronous generators. In this paper, a control scheme is presented based on the principle of active-power synchronization to regulate the active power of a grid-tied voltage-source converter based on an emulation of the synchronous-machine swing equation. Design of a cascaded inner-loop voltage and resonant current control is presented to regulate the output voltage as specified via the outer-loop virtual-machine control scheme responsible for power regulation. The performance of this control scheme is investigated within the context of microgrid operation for the provision of active and reactive power to the system, and microgrid frequency support. Experimental validation is provided via the use of a 15 kVA three-phase VSC in a 90 kVA 400V microgrid

    Investigations on the occurrence of tapeworm infections in German horse populations with comparison of different antibody detection methods based on saliva and serum samples

    Background Effective and sustainable worm control in horses would benefit from detailed information about the current regional occurrence of tapeworms. Different diagnostic methods are currently available to detect Anoplocephala spp. infections in horses. However, the format as well as the sensitivity and specificity of the methods vary considerably. Methods A coprological, serological and questionnaire study was conducted to investigate the prevalence and risk factors of tapeworm infections on 48 horse farms in the region of Berlin and Brandenburg, Germany. In total, faecal samples of 484 horses were analysed using the double centrifugation/combined sedimentation-flotation and mini-FLOTAC. Serum (n = 481) and saliva (n = 365) samples were analysed by ELISAs to determine antibody levels against Anoplocephala spp. 12/13 kDa excretory/secretory (E/S) antigens. Results Cestode eggs were detected in 0.6% of faecal samples (farm prevalence 6.3%) without differences between the two methods. In contrast, antibodies against Anoplocephala spp. were detected in 16.2% (farm prevalence 52.1%) and in 29.5% (farm prevalence 75.7%) of the serum and saliva samples, respectively. Both ELISA based methods for detection of tapeworms reported a greater number of infected animals requiring treatment than were positively identified by coproscopy. Logistic regression analysis identified permanent pasture access, large pastures and regular pasture changes and high strongyle egg counts as risk factors for positive serum antibody responses to Anoplocephala spp. while last treatment with praziquantel was protective. Other protective factors were the presence of foals and high numbers of horses on the farm. Daily removal of faeces from the pasture and horse age did not have a significant effect. Conclusions The findings of the present serological investigation indicate that tapeworm prevalence in Berlin/Brandenburg horse farms is much higher than would be anticipated by using conventional/coproscopic analyses. Moreover, the majority of tapeworm-positive horses had not received a cestocidal drug at their last treatment. Considering the already known low sensitivity of the coproscopic detection, the equine veterinary diagnostics can be enhanced by the use of antibody detection methods such as the saliva-based ELISA
