    Effects of lockdown on human sleep and chronotype during the COVID-19 pandemic

    COVID-19 lockdown induced severe changes in light exposure and social cues. Leone et al. show that sleep is later and longer on weekdays with lower social jetlag during lockdown compared with a control condition in the same subjects (n = 1021).     RdDM pathway components differentially modulate Tobamovirus symptom development

    Small RNAs (sRNAs) are important molecules for gene regulation in plants and play an essential role in plantpathogen interactions. Researchers have evaluated the relationship between viral infections as well as the endogenous accumulation of sRNAs and the transcriptional changes associated with the production of symptoms, but little is known about a possible direct role of epigenetics, mediated by 24-nt sRNAs, in the induction of these symptoms. Using diferent RNA directed DNA methylation (RdDM) pathway mutants and a triple demethylase mutant; here we demonstrate that the disruption of RdDM pathway during viral infection produce alterations in the plant transcriptome and in consequence changes in plant symptoms. This study represents the initial step in exposing that DNA methylation directed by endogenous sRNAs has an important role, uncoupled to defense, in the production of symptoms associated with plant-virus interactions.Fil: Leone, Melisa.     Paying the circadian toll: The circadian response to LPS injection is dependent on the Toll-like receptor 4

    Systemic low doses of the endotoxin lipopolysaccharide (LPS) administered at CT15 (circadian time 12 corresponds to locomotor activity onset) induce phase delays of locomotor activity rhythms in mice. To evaluate if this effect was mediated by the Toll-like receptor 4 (TLR4), our present aim was to characterize the circadian behavior and LPS-induced circadian response of TLR4 (LPS receptor)-deficient mice (in C57bl/10 and C3H backgrounds). In mutants, we observed a free-running period and a light-induced phase delay similar to the one observed in their corresponding wild-type (WT) littermates. The LPS-induced phase delay, wheel running inhibition and c-Fos/Per-1 immunoreactivity in the paraventricular nuclei observed in WT mice was absent or significantly decreased in the TLR4-deficient mice.     Laboratory tests on the biocontrol of chestnut insect pests on Etna (Sicily, Italy) by means of entomopathogenic nematodes

    The virulence of seventeen strains of entomopathogenic nematodes, belonging to four species of Steinernema and two of Heterorhabditis, against the chestnut insect parasites Curculio elephas, C. glandium, Pammene castanicola, Cydia splendana and C. fagiglandana, was tested in laboratory. The bioassays were conducted on the insect larvae collected in three different years (2010-2012) in chestnut groves of Etna. Most of the strains tested resulted good biocontrol agents for all the treated insect pests. The strains S. feltiae ESA and S. feltiae EPP were the most effective strains autochthonous of Etna when all the species of insect pests are considered as a whole

    Role of proinflammatory cytokines on lipopolysaccharide-induced phase shifts in locomotor activity circadian rhythm

    We previously reported that early night peripheral bacterial lipopolysaccharide (LPS) injection produces phase delays in the circadian rhythm of locomotor activity in mice. We now assess the effects of proinflammatory cytokines on circadian physiology, including their role in LPS-induced phase shifts. First, we investigated whether differential systemic induction of classic proinflammatory cytokines could explain the time-specific behavioral effects of peripheral LPS. Induction levels for plasma interleukin (IL)-1α, IL-1β, IL-6, or tumor necrosis factor (TNF)-α did not differ between animals receiving a LPS challenge in the early day or early night. We next tested the in vivo effects of central proinflammatory cytokines on circadian physiology. We found that intracerebroventricular (i.c.v.) delivery of TNF-α or interleukin IL-1β induced phase delays on wheel-running activity rhythms. Furthermore, we analyzed if these cytokines mediate the LPS-induced phase shifts and found that i.c.v. administration of soluble TNF-α receptor (but not an IL-1β antagonistic) prior to LPS stimulation inhibited the phase delays. Our work suggests that the suprachiasmatic nucleus (SCN) responds to central proinflammatory cytokines in vivo, producing phase shifts in locomotor activity rhythms. Moreover, we show that the LPS-induced phase delays are mediated through the action of TNF-α at the central level, and that systemic induction of proinflammatory cytokines might be necessary, but not sufficient, for this behavioral outcome.Fil: Leone, Maria Juliana.     Pharmacokinetics of sumatriptan in non-respondent and in adverse drug reaction reporting migraine patients

    Sumatriptan is a selectiveagonist of 5HT1 (1B/1D) receptors,which has proved to be effectiveand safe for the acute treatment ofmigraine attacks. Nevertheless, itsuse by migraine sufferers is stilllimited and some patients consideradverse reactions related to sumatriptan,especially chest symptoms,unacceptable even if not serious.Moreover, in clinical trials, almostone third and one sixth of patients,respectively, fail to experienceheadache relief either after oral orafter subcutaneous sumatriptanadministration. Our aim was to verifywhether differencies in sumatriptanpharmacokinetics couldexplain non-response and/oradverse drug reactions. Sumatriptanlevels were determined by HPLCwith electrochemical detection.Pharmacokinetic parameters werecalculated using a computer program(PK Solutions 2.0; non compartmentalPharmacokinetics DataAnalysis). After oral administration,sumatriptan is rapidly absorbed andsometimes displays multiple peaksof plasma concentration. This “multiplepeaking” gives rise to considerableinter-subject variability inthe time of reaching maximumplasma concentration.Pharmacokinetic parameters ofsumatriptan, both after oral andsubcutaneous administration, weresimilar in the three patient groups.Blood pressure and heart rate didnot show any significant differencesbetween groups. Pharmacokineticparameters and bioavailability ofsumatriptan did not seem to be correlatedeither to the lack of efficacyor the appearance of side effects.These results could depend on thelimited number of patients studied

    Techniques for an image space occlusion culling engine

    In this work we present several techniques applied to implement an Image Space Software Occlusion Culling Engine to increase the speed of rendering general dynamic scenes with high depth complexity. This conservative culling method is based on a tiled Occlusion Map that is updated only when needed, deferring and even avoiding the expensive per pixel rasterization process. We show how the tiles become a useful way to increase the speed of visibility tests.
