    A presynaptic phosphosignaling hub for lasting homeostatic plasticity

    Stable function of networks requires that synapses adapt their strength to levels of neuronal activity, and failure to do so results in cognitive disorders. How such homeostatic regulation may be implemented in mammalian synapses remains poorly understood. Here we show that the phosphorylation status of several positions of the active-zone (AZ) protein RIM1 are relevant for synaptic glutamate release. Position RIMS1045 is necessary and sufficient for expression of silencing-induced homeostatic plasticity and is kept phosphorylated by serine arginine protein kinase 2 (SRPK2). SRPK2-induced upscaling of synaptic release leads to additional RIM1 nanoclusters and docked vesicles at the AZ and is not observed in the absence of RIM1 and occluded by RIMS1045E. Our data suggest that SRPK2 and RIM1 represent a presynaptic phosphosignaling hub that is involved in the homeostatic balance of synaptic coupling of neuronal networks

    Seasonal variation of upwelling in the Alaskan Beaufort Sea : impact of sea ice cover

The definitive version was published in Journal of Geophysical Research 117 (2012): C06022, doi:10.1029/2012JC007985.Data from a mooring array deployed from August 2002 to September 2004 are used to characterize differences in upwelling near the shelf break in the Alaskan Beaufort Sea due to varying sea ice conditions. The record is divided into three ice seasons: open water, partial ice, and full ice. The basic response is the same in each of the seasons. Roughly 8 h after the onset of easterly winds the shelf break jet reverses, followed approximately 10 h later by upwelling of saltier water which is cold near the shelf break (Pacific Winter Water) and warm at depth (Atlantic Water). The secondary circulation at the outer shelf is, to first order, consistent with a two-dimensional Ekman balance of offshore flow in the upper layer and onshore flow at depth. There are, however, important seasonal differences in the upwelling. Overall the response is strongest in the partial ice season and weakest in the full ice season. It is believed that these differences are dictated by the degree to which wind stress is transmitted through the pack-ice, as the strength of the wind-forcing was comparable over the three seasons. An EOF-based upwelling index is constructed using information about the primary flow, secondary flow, and hydrography. The ability to predict upwelling using the wind record alone is explored, which demonstrates that 90% of easterly wind events exceeding 9.5 m s−1 drive significant upwelling.     A time-resolved proteomic and prognostic map of COVID-19

    COVID-19 is highly variable in its clinical presentation, ranging from asymptomatic infection to severe organ damage and death. We characterized the time-dependent progression of the disease in 139 COVID-19 inpatients by measuring 86 accredited diagnostic parameters, such as blood cell counts and enzyme activities, as well as untargeted plasma proteomes at 687 sampling points. We report an initial spike in a systemic inflammatory response, which is gradually alleviated and followed by a protein signature indicative of tissue repair, metabolic reconstitution, and immunomodulation. We identify prognostic marker signatures for devising risk-adapted treatment strategies and use machine learning to classify therapeutic needs. We show that the machine learning models based on the proteome are transferable to an independent cohort. Our study presents a map linking routinely used clinical diagnostic parameters to plasma proteomes and their dynamics in an infectious disease

    The coming decade of digital brain research: a vision for neuroscience at the intersection of technology and computing

    In recent years, brain research has indisputably entered a new epoch, driven by substantial methodological advances and digitally enabled data integration and modelling at multiple scales— from molecules to the whole brain. Major advances are emerging at the intersection of neuroscience with technology and computing. This new science of the brain combines high-quality research, data integration across multiple scales, a new culture of multidisciplinary large-scale collaboration and translation into applications. As pioneered in Europe’s Human Brain Project (HBP), a systematic approach will be essential for meeting the coming decade’s pressing medical and technological challenges. The aims of this paper are to: develop a concept for the coming decade of digital brain research, discuss this new concept with the research community at large, to identify points of convergence, and derive therefrom scientific common goals; provide a scientific framework for the current and future development of EBRAINS, a research infrastructure resulting from the HBP’s work; inform and engage stakeholders, funding organisations and research institutions regarding future digital brain research; identify and address the transformational potential of comprehensive brain models for artificial intelligence, including machine learning and deep learning; outline a collaborative approach that integrates reflection, dialogues and societal engagement on ethical and societal opportunities and challenges as part of future neuroscience research

    Size variation of 0-group plaice: Are earlier influences on growth potential a contributing factor?

    Over a decade of sampling has shown that there are consistent differences in the sizes of 0-group plaice by late summer comparing 21 nursery sites on the Scottish west coast. However, when young fish were collected from two sites which produce particularly small and large fish and reared using a common garden design, growth rates between fish from the two sites were indistinguishable. Either there is little selection for fast or slow growth up to a few weeks post-settlement, or such effects do not persist sufficiently strongly to influence later growth. There were also no significant correlations between the time-series of fish size comparing sites, although within some sites there was evidence of inter-annual density-dependent effects. Any influences of offshore regional scale factors, such as sea temperature or pelagic primary productivity on growth thus appear to be heavily modified by local conditions on the nursery grounds. The field observations combined with the experimental results lead us to conclude that the size 0-group plaice attain in late summer is mainly controlled by post-settlement habitat quality

    Reduction mammoplasty in adolescents and elderly: A ten year case series analyzing age related outcome with focus on safety and complications

    INTRODUCTION Reduction mammoplasty alleviates symptoms of macromastia in various ways. Current study results mainly identify perioperative risk factors for middle aged patients. We investigated a large series of consecutive breast reductions procedures to study whether patients' age at the time of operation is related to the postoperative outcome. METHODS We retrospectively reviewed all non-oncologic breast reduction procedures at a single institution over a ten year time period, analyzing patients (age, BMI, comorbidities, medication) and operation specific characteristics' (pedicle, nipple-to-sternal notch, resection weight, complications) to identify risk factors related to patients' age at the time of operation. Patients were therefore divided into three groups, according to their age (group I ≀ 20 years, group II ≄60 years, group III 21 to 59 years). RESULTS 539 patients were included in the study, in total 1065 reduction mammoplasties were performed over a ten year period. The overall complication rate was 33% (n = 175). Excluding minor complications, the total complication rate was 9.5% (n = 51). High body mass index (≄30 kg/m2) (p = 0.02) could be identified as a statistically significant risk factor for major and minor complications. Smoking (p = 0.09) and age ≄ 60 years (p = 0.08) showed a tendency toward higher risk for major and minor complications. CONCLUSION This study shows an increased risk for complications when performing reduction mammoplasty in older patients, presumably due to the higher prevalence of comorbidities in this patient group as compared to young patients
