    Dignity Norfolk: How One Tidewater Group Enabled Gay and Lesbian Catholics to Form Long Lasting Friendships and Chosen Families

    In the past decade, research has been conducted to look into the history of the Queer Community of the Tidewater Region. Students and community volunteers have conducted interviews and gathered documents connected to the queer community to grow the study and breadth of available information for future researchers. However, more in-depth knowledge of community connections and familial ties within the queer community in the Tidewater Region was needed. Through research into Our Own Newspaper, local historical background information, and in-person interviews, I pieced together first-hand accounts of life through the eyes of a gay man or lesbian woman living in the Tidewater Region between 1970 - the present day. I found that the connections and emotional bonds made years ago have stayed strong, similar to those of a biological family. This research opens up the possibilities for future researchers to find more information, connect the lives of many others within the Tidewater Community, and save the history of the queer community who lived and thrived in the Tidewater Area before the primary sources have passed on

    The Development of an Evaluational Process for the Group Home Project of the Northeast Multnomah District of Childrens\u27 Service Division

    Childrens\u27 Services Division of the State of Oregon is involved in a group home project. They have to date nine group homes in operation within Multnomah County. Their staff consists of a director, three social workers and the group home provider staff. The primary goal for this practicum was the development of an evaluative system that would establish descriptions of the type of client who entered the Group Home Program and some criteria for success in that program. In developing this system, it was necessary to design instruments to measure behavioral changes and attitudes and also to collect descriptive data. Such a system should be as comprehensive as possible while remaining fairly simple and easy to implement. A secondary goal was to pre-test the evaluative system and gather data for the use of the Group Home Project. The instruments could be used continuously in an on-going evaluative process with pre-test data as a standard by which to set more definite objectives and evaluation of the program.     Enumeration of islets by nuclei counting and light microscopic analysis

    Author Manuscript 2011 May 1.Islet enumeration in impure preparations by conventional dithizone staining and visual counting is inaccurate and operator dependent. We examined nuclei counting for measuring the total number of cells in islet preparations, and we combined it with morphological analysis by light microscopy (LM) for estimating the volume fraction of islets in impure preparations. Cells and islets were disrupted with lysis solution and shear, and accuracy of counting successively diluted nuclei suspensions was verified with (1) visual counting in a hemocytometer after staining with crystal violet, and automatic counting by (2) aperture electrical resistance measurement and (3) flow cytometer measurement after staining with 7-aminoactinomycin-D. DNA content averaged 6.5 and 6.9 pg of DNA per cell for rat and human islets, respectively, in agreement with literature estimates. With pure rat islet preparations, precision improved with increasing counts, and samples with about greater than or equal to 160 islets provided a coefficient of variation of about 6%. Aliquots of human islet preparations were processed for LM analysis by stereological point counting. Total nuclei counts and islet volume fraction from LM analysis were combined to obtain the number of islet equivalents (IEs). Total number of IE by the standard method of dithizone staining/manual counting was overestimated by about 90% compared with LM/nuclei counting for 12 freshly isolated human islet research preparations.     Rheological and biological properties of a hydrogel support for cells intended for intervertebral disc repair

    <p>Abstract</p> <p>Background</p> <p>Cell-based approaches towards restoration of prolapsed or degenerated intervertebral discs are hampered by a lack of measures for safe administration and placement of cell suspensions within a treated disc. In order to overcome these risks, a serum albumin-based hydrogel has been developed that polymerizes after injection and anchors the administered cell suspension within the tissue.</p> <p>Methods</p> <p>A hydrogel composed of chemically activated albumin crosslinked by polyethylene glycol spacers was produced. The visco-elastic gel properties were determined by rheological measurement. Human intervertebral disc cells were cultured <it>in vitro </it>and <it>in vivo </it>in the hydrogel and their phenotype was tested by reverse-transcriptase polymerase chain reaction. Matrix production and deposition was monitored by immuno-histology and by biochemical analysis of collagen and glycosaminoglycan deposition. Species specific <it>in situ </it>hybridization was performed to discriminate between cells of human and murine origin in xenotransplants.</p> <p>Results</p> <p>The reproducibility of the gel formation process could be demonstrated. The visco-elastic properties were not influenced by storage of gel components. <it>In vitro </it>and <it>in vivo </it>(subcutaneous implants in mice) evidence is presented for cellular differentiation and matrix deposition within the hydrogel for human intervertebral disc cells even for donor cells that have been expanded in primary monolayer culture, stored in liquid nitrogen and re-activated in secondary monolayer culture. Upon injection into the animals, gels formed spheres that lasted for the duration of the experiments (14 days). The expression of cartilage- and disc-specific mRNAs was maintained in hydrogels <it>in vitro </it>and <it>in vivo</it>, demonstrating the maintenance of a stable specific cellular phenotype, compared to monolayer cells. Significantly higher levels of hyaluronan synthase isozymes-2 and -3 mRNA suggest cell functionalities towards those needed for the support of the regeneration of the intervertebral disc. Moreover, mouse implanted hydrogels accumulated 5 times more glycosaminoglycans and 50 times more collagen than the <it>in vitro </it>cultured gels, the latter instead releasing equivalent quantities of glycosaminoglycans and collagen into the culture medium. Matrix deposition could be specified by immunohistology for collagen types I and II, and aggrecan and was found only in areas where predominantly cells of human origin were detected by species specific <it>in situ </it>hybridization.</p> <p>Conclusions</p> <p>The data demonstrate that the hydrogels form stable implants capable to contain a specifically functional cell population within a physiological environment.</p

    What does it mean to be a pariah? : assimilation, depersonalization and uniqueness in the thought of Hannah Arendt

    Esta tese pergunta pelo sentido do pária na obra de Hannah Arendt, buscando compreende-lo dentro do sentido mais amplo do pensamento político da autora. A pesquisa teve como ponto de partida artigos reunidos na coletânea Escritos Judaicos, em que a autora tratou do pária pelo viés de experiências dos judeus europeus, sobretudo entre o século 19 e o século 20; passou por Origens do Totalitarismo (1951), texto em que Arendt definiu o movimento de exclusão e destruição de párias europeus, em especial os judeus, como o “agente catalítico” do colapso da Europa; chegando a textos como A Condição Humana (1958), em que Arendt apresentou a pluralidade e a unicidade como novos princípios requeridos pela reconstrução da dignidade humana num contexto póstotalitário. Articulando-se de forma não-monística, este texto tem, como eixos, temas da obra de Arendt como a assimilação, tentativa de absorção de judeus e outros párias pelo “social”; a despersonalização, movimento radical de dissolução da personalidade e alienação do “eu” em favor de forças históricas; e a unicidade, como condição básica de seres humanos plurais e insubstituíveis. O trabalho sustenta, de forma geral, que a experiência do pária, levando em conta sua exclusão, desaparecimento e tentativa de reaparecimento, é um exemplo fundamental para a ação e o pensamento políticos na contemporaneidade.This dissertation questions the meaning of the pariah in Hannah Arendt’s work, aiming at its comprehension within the broader context of Arendt’s political thought. The research departed from articles published in the anthology The Jewish Writings, in which Arendt approached the pariah relying on experiences of European Jews, mainly between the 19th and the 20th centuries; dealt with The Origins of Totalitarianism, text in which she defined the exclusion and destruction of European pariahs, specially Jews, as the “catalytic agent” of Europe’s broader collapse; arrived at texts such as The Human Condition (1958), in which Arendt presented plurality and uniqueness as new principles required by the reestablishment of human dignity in a post-totalitarian world. Developed in non-monistic lines, this text deals with topics of Arendt’s work, as assimilation, the attempt of absorption of Jews and other pariahs by the “social”; depersonalization, the radical movement of dissolution of personality and alienation of the ego towards historical forces; and uniqueness, as a basic condition of plural and irreplaceable human beings. It holds, in broad terms, that the pariah’s experience, its exclusion, disappearance and attempt of reappearance, is a fundamental example for acting and thinking politically in the present world
