    Optimal control computation for nonlinear systems with state-dependent stopping criteria

    In this paper, we consider a challenging optimal control problem in which the terminal time is determined by a stopping criterion. This stopping criterion is defined by a smooth surface in the state space; when the state trajectory hits this surface, the governing dynamic system stops. By restricting the controls to piecewise constant functions, we derive a finite-dimensional approximation of the optimal control problem. We then develop an efficient computational method, based on nonlinear programming, for solving the approximate problem. We conclude the paper with four numerical examples

    In this paper, we consider a system that evolves by switching between several subsystems of ordinary differential equations. The switching mechanism is this system induces an instantaneous change in the system's state, which can be controlled through a set of decision parameters. We develop a new computational method, based on nonlinear programming, for optimizing the system parameters and the subsystem switching times. We then successfully apply this method to two interesting examples

    A new exact penalty method for semi-infinite programming problems

    In this paper, we consider a class of nonlinear semi-infinite optimization problems. These problems involve continuous inequality constraints that need to be satisfied at every point in an infinite index set, as well as conventional equality and inequality constraints. By introducing a novel penalty function to penalize constraint violations, we form an approximate optimization problem in which the penalty function is minimized subject to only bound constraints. We then show that this penalty function is exact—that is, when the penalty parameter is sufficiently large, any local solution of the approximate problem can be used to generate a corresponding local solution of the original problem. On this basis, the original problem can be solved as a sequence of approximate nonlinear programming problems. We conclude the paper with some numerical results demonstrating the applicability of our approach to PID control and filter design

    Cloning, expression and purification of squalene synthase from Candida tropicalis in Pichia pastoris

    Squalene synthase (SS) is the key precursor and first committed enzyme of the sterol biosynthesis pathway. In a previous work, SS has been identified as one of the immunogenic proteins that could be a potential diagnostic candidate for the pathogenic fungus Candida tropicalis. In this study, SS from C. tropicalis was cloned and expressed as recombinant protein in Pichia pastoris to investigate its reactivity with serum antibodies. ERG9 gene that encodes for SS was amplified by PCR and cloned in-frame into pPICZB expression vector. The recombinant construct was then transformed into P. pastoris GS115 host strain. Expression of the recombinant protein was confirmed by SDS–PAGE and Western blot analysis using anti-His tag probe. Optimal protein production was achieved by cultivating the culture with 1.0% methanol for 72 h. The recombinant protein was purified to approximately 97% pure in a single step immobilized metal affinity chromatography with a yield of 70.3%. Besides, the purified protein exhibited specific reactivity with immune sera on Western blot. This is the first report on heterologous expression of antigenic SS from C. tropicalis in P. pastoris which can be exploited for large-scale production and further research. The results also suggested that the protein might be of great value as antigen candidate for serodiagnosis of Candida infection

    is article is a case study on health issues in the Orang Asli community. After the outbreak of measles among Orang Asli in Kelantan, the health issue in this community has been questioned. Compared to non-aboriginal communities, the health of the aboriginal people is lagging, health disparity is still obvious. Since the 1950s, the Malaysian Government has tried to carry development and improve the quality of life in Orang Asli communities, along with non-government agencies, but the health status compared to other aboriginal people groups are still lower. There are a lot of factors as to why the health quality for Orang Asli does not meet the recommended standard including poor education environment, superstitions from their ancestors, and limited facilities to receive health information and medical check-ups. As an impact, they were 14 Orang Asli known as Batek tribe died in Kampung Kuala Koh, Gua Musang from the early of May until 6th June. On 17th June 2019, the Health Ministry has confirmed that there was an outbreak of measles among the Orang Asli Batek tribe at Kampung Kuala Koh, Gua Musang

Some sufficient conditions which can guarantee the system's boundedness are obtained. In addition, we work out the region where the solution will remain and furthermore the relationship between the initial function and the bounded region. Based on the new concept of attractor with switching laws, we show that the nonlinear switched delay system is attractive and then obtain the attractive region

    Context-dependent multilingual lexical lookup for under-resourced languages

    Current approaches for word sense disambiguation and translation selection typically require lexical resources or large bilingual corpora with rich information fields and annotations, which are often infeasible for under-resourced languages. We extract translation context knowledge from a bilingual comparable corpora of a richer-resourced language pair, and inject it into a multilingual lexicon. The multilingual lexicon can then be used to perform context-dependent lexical lookup on texts of any language, including under-resourced ones. Evaluations on a prototype lookup tool, trained on a English-Malay bilingual Wikipedia corpus, show a precision score of 0.65 (baseline 0.55) and mean reciprocal rank score of 0.81 (baseline 0.771). Based on the early encouraging results, the context-dependent lexical lookup tool may be developed further into an intelligent reading aid, to help users grasp the gist of a second or foreign language text

    Study of ageing in Al-Mg-Si alloys by positron annihilation spectroscopy

    In many common Al-Mg-Si alloys (6000 series) intermediate storage at or near 'room temperature' after solutionising leads to pronounced changes of the precipitation kinetics during the ensuing artificial ageing step at \approx 180{\deg}C. This is not only an annoyance in production, but also a challenge for researchers. We studied the kinetics of natural 'room temperature' ageing (NA) in Al-Mg-Si alloys by means of various different techniques, namely electrical resistivity and hardness measurement, thermoanalysis and positron lifetime and Doppler broadening (DB) spectroscopy to identify the stages in which the negative effect of NA on artificial ageing might appear. Positron lifetime measurements were carried out in a fast mode, allowing us to measure average lifetimes in below 1 minute. DB measurements were carried out with a single detector and a 68Ge positron source by employing high momentum analysis. The various measurements show that NA is much more complex than anticipated and at least four different stages can be distinguished. The nature of these stages cannot be given with certainty, but a possible sequence includes vacancy diffusion to individual solute atoms, nucleation of solute clusters, Mg agglomeration to clusters and coarsening or ordering of such clusters. Positron lifetime measurements after more complex ageing treatments involving storage at 0{\deg}C, 20{\deg}C and 180{\deg}C have also been carried out and help to understand the mechanisms involved

    Safety Profile and Effects of Pulsed Methylprednisolone on Vital Signs in Thyroid Eye Disease

    Objective. To analyze changes in vital signs (heart rate (HR), systolic (SBP), and diastolic blood pressure (DBP)) during and after intravenous methylprednisolone (IVMP) and any other adverse effects. Methods. Retrospective review of charts of patients who received IVMP as treatment regime for thyroid eye disease. All subjects had vital signs charted during and after infusions. Results. This study included 38 subjects and a total of 242 infusions administered. IVMP resulted in a small but significant percentage drop in mean SBP at 30 min (p<0.001) and 60 min (p=0.03) but no difference at 90 min. There was also small but significant percentage drop in mean DBP and HR (DBP: p<0.001 for 30 min, p=0.001 for 60 min, and p=0.02 for 90 min and HR: p<0.001 for 30 min, 60 min, and 90 min). There were no cumulative effects on change of blood pressure or HR. There were 6 episodes of bradycardia (2.5%) and 12 episodes of moderate to severe hypertension (5%). No significant cardiovascular or hepatic toxicity was found. Conclusion. IVMP is relatively safe and efficacious. IVMP demonstrated mild and noncumulative effects on vital signs. Severe hypertension may occur in susceptible individuals such as those with underlying hypertension and uncontrolled thyroid dysfunction, whereas bradycardia may be more likely in those on beta-blockers

    Lactococcus lactis harbouring Ara h 2.02 alleviates allergen-specific Th2-associated responses in sensitized mice

    Aim: To study the prophylactic effect of recombinant Lactococcus lactis (rLl) harbouring Ara h 2.02 peanut allergen, in sensitized and challenged mice. Methods and results: Ara h 2.02 cDNA was cloned into pNZ8048 for heterologous expression in L. lactis. The purified recombinant allergen showed IgE binding comparable with native Ara h 2. Balb/c mice were fed with either recombinant (rLl), nonrecombinant L. lactis (Ll) or NaHCO3 (Sham) prior to sensitization and challenged with rAra h 2.02, whereas the baseline group was only fed with Ll. Allergen-specific immunoglobulin and splenocyte cytokines responses were determined for each mouse. Mice fed with either Ll or rLl showed significant alleviation of IgE and IgG1 compared to the Sham group. Despite no significant decrease in Th2 (IL-4, IL-13, IL-6) or increase in Th1 (IFN-γ) cytokines, both groups showed lower IL-10 level, while the IL-4 : IFN-γ ratio was significantly lower for rLl compared to Ll group. Conclusions: Oral administration of rLl harbouring Ara h 2.02 demonstrated alleviation of Th2-associated responses in allergen-challenged mice and a possible added allergen-specific prophylactic effect. Significance and impact of the study: Ara h 2.02 coupled with the intrinsic properties of probiotic L. lactis as a delivery vehicle can be explored for the development of a commercially scalable vaccine
