    The geodesic rule for higher codimensional global defects

    We generalize the geodesic rule to the case of formation of higher codimensional global defects. Relying on energetic arguments, we argue that, for such defects, the geometric structures of interest are the totally geodesic submanifolds. On the other hand, stochastic arguments lead to a diffusion equation approach, from which the geodesic rule is deduced. It turns out that the most appropriate geometric structure that one should consider is the convex hull of the values of the order parameter on the causal volumes whose collision gives rise to the defect. We explain why these two approaches lead to similar results when calculating the density of global defects by using a theorem of Cheeger and Gromoll. We present a computation of the probability of formation of strings/vortices in the case of a system, such as nematic liquid crystals, whose vacuum is RP2\mathbb{R}P^2.Comment: 17 pages, no figures.     Comparison of patients undergoing switching versus augmentation of antipsychotic medications during treatment for schizophrenia

    It is often difficult to determine whether a patient may best benefit by augmenting their current medication or switching them to another. This post-hoc analysis compares patients’ clinical and functional profiles at the time their antipsychotic medications were either switched or augmented. Adult outpatients receiving oral antipsychotic treatment for schizophrenia were assessed during a 12-month international observational study. Clinical and functional measures were assessed at the time of first treatment switch/augmentation (0–14 days prior) and compared between Switched and Augmented patient groups. Due to low numbers of patients providing such data, interpretations are based on effect sizes. Data at the time of change were available for 87 patients: 53 Switched and 34 Augmented. Inadequate response was the primary reason for treatment change in both groups, whereas lack of adherence was more prevalent in the Switched group (26.4% vs 8.8%). Changes in clinical severity from study initiation to medication change were similar, as indicated by Clinical Global Impressions–Severity scores. However, physical and mental component scores of the 12-item Short-Form Health Survey improved in the Augmented group, but worsened in the Switched group. These findings suggest that the patient’s worsening or lack of meaningful improvement prompts clinicians to switch antipsychotic medications, whereas when patients show some improvement, clinicians may be more likely to try bolstering the improvements through augmentation. Current findings are consistent with physicians’ stated reasons for switching versus augmenting antipsychotics in the treatment of schizophrenia. Confirmation of these findings requires further research

    Photometric variability of the T Tauri star TW Hya on time scales of hours to years

    MOST (Microvariability & Oscillations of STars) and ASAS (All Sky Automated Survey) observations have been used to characterize photometric variability of TW Hya on time scales from a fraction of a day to 7.5 weeks and from a few days to 8 years, respectively. The two data sets have very different uncertainties and temporal coverage properties and cannot be directly combined, nevertheless, they suggests a global variability spectrum with "flicker noise" properties, i.e. with amplitudes a ~ 1/sqrt(f), over >4 decades in frequency, in the range f = 0.0003 to 10 cycles per day (c/d). A 3.7 d period is clearly present in the continuous 11 day, 0.07 d time resolution, observations by MOST in 2007. Brightness extrema coincide with zero-velocity crossings in periodic (3.56 d) radial velocity variability detected in contemporaneous spectroscopic observations of Setiawan et al. (2008) and interpreted as caused by a planet. The 3.56/3.7 d periodicity was entirely absent in the second, four times longer MOST run in 2008, casting doubt on the planetary explanation. Instead, a spectrum of unstable single periods within the range of 2 - 9 days was observed; the tendency of the periods to progressively shorten was well traced using the wavelet analysis. The evolving periodicities and the overall flicker-noise characteristics of the TW Hya variability suggest a combination of several mechanisms, with the dominant ones probably related to the accretion processes from the disk around the star.Comment: MNRAS submitte

    Structural and mechanistic analysis of ATPase inhibitors targeting mycobacterial DNA gyrase

    Objectives To evaluate the efficacy of two novel compounds against mycobacteria and determine the molecular basis of their action on DNA gyrase using structural and mechanistic approaches. Methods Redx03863 and Redx04739 were tested in antibacterial assays, and also against their target, DNA gyrase, using DNA supercoiling and ATPase assays. X-ray crystallography was used to determine the structure of the gyrase B protein ATPase sub-domain from Mycobacterium smegmatis complexed with the aminocoumarin drug novobiocin, and structures of the same domain from Mycobacterium thermoresistibile complexed with novobiocin, and also with Redx03863. Results Both compounds, Redx03863 and Redx04739, were active against selected Gram-positive and Gram-negative species, with Redx03863 being the more potent, and Redx04739 showing selectivity against M. smegmatis. Both compounds were potent inhibitors of the supercoiling and ATPase reactions of DNA gyrase, but did not appreciably affect the ATP-independent relaxation reaction. The structure of Redx03863 bound to the gyrase B protein ATPase sub-domain from M. thermoresistibile shows that it binds at a site adjacent to the ATP- and novobiocin-binding sites. We found that most of the mutations that we made in the Redx03863-binding pocket, based on the structure, rendered gyrase inactive. Conclusions Redx03863 and Redx04739 inhibit gyrase by preventing the binding of ATP. The fact that the Redx03863-binding pocket is distinct from that of novobiocin, coupled with the lack of activity of resistant mutants, suggests that such compounds could have potential to be further exploited as antibiotics

    Using Real-World Data to Guide Ustekinumab Dosing Strategies for Psoriasis: A Prospective Pharmacokinetic-Pharmacodynamic Study.

    Variation in response to biologic therapy for inflammatory diseases, such as psoriasis, is partly driven by variation in drug exposure. Real-world psoriasis data were used to develop a pharmacokinetic/pharmacodynamic (PK/PD) model for the first-line therapeutic antibody ustekinumab. The impact of differing dosing strategies on response was explored. Data were collected from a UK prospective multicenter observational cohort (491 patients on ustekinumab monotherapy, drug levels, and anti-drug antibody measurements on 797 serum samples, 1,590 measurements of Psoriasis Area Severity Index (PASI)). Ustekinumab PKs were described with a linear one-compartment model. A maximum effect (Emax ) model inhibited progression of psoriatic skin lesions in the turnover PD mechanism describing PASI evolution while on treatment. A mixture model on half-maximal effective concentration identified a potential nonresponder group, with simulations suggesting that, in future, the model could be incorporated into a Bayesian therapeutic drug monitoring "dashboard" to individualize dosing and improve treatment outcomes

    Design, construction and characterization of a set of insulated bacterial promoters

    We have generated a series of variable-strength, constitutive, bacterial promoters that act predictably in different sequence contexts, span two orders of magnitude in strength and contain convenient sites for cloning and the introduction of downstream open-reading frames. Importantly, their design insulates these promoters from the stimulatory or repressive effects of many 5′- or 3′-sequence elements. We show that different promoters from our library produce constant relative levels of two different proteins in multiple genetic contexts. This set of promoters should be a useful resource for the synthetic-biology community
