Defining Caenorhabditis elegans as a model system to investigate lipoic acid metabolism
Lipoic acid (LA) is a sulfur-containing cofactor that covalently binds to a variety of cognate enzymes that are essential for redox reactions in all three domains of life. Inherited mutations in the enzymes that make LA, namely lipoyl synthase, octanoyltransferase, and amidotransferase, result in devastating human metabolic disorders. Unfortunately, because many aspects of this essential pathway are still obscure, available treatments only serve to alleviate symptoms. We envisioned that the development of an organismal model system might provide new opportunities to interrogate LA biochemistry, biology, and physiology. Here we report our investigations on three Caenorhabditis elegans orthologous proteins involved in this post-translational modification. We established that M01F1.3 is a lipoyl synthase, ZC410.7 an octanoyltransferase, and C45G3.3 an amidotransferase. Worms subjected to RNAi against M01F1.3 and ZC410.7 manifest larval arrest in the second generation. The arrest was not rescued by LA supplementation, indicating that endogenous synthesis of LA is essential for C. elegans development. Expression of the enzymes M01F1.3, ZC410.7, and C45G3.3 completely rescue bacterial or yeast mutants affected in different steps of the lipoylation pathway, indicating functional overlap. Thus, we demonstrate that, similarly to humans, C. elegans is able to synthesize LA de novo via a lipoyl-relay pathway, and suggest that this nematode could be a valuable model to dissect the role of protein mislipoylation and to develop new therapies.Fil: Lavatelli, Antonela. El presente artículo retoma una conferencia dictada en la Alianza Francesa de Buenos Aires en el año 2005, en ocasión del Congreso de Historia del Teatro. La problemática de aplicar ciertas categorías estéticas al análisis de las obras del célebre autor francés constituye el núcleo fundamental del escrito, en particular de las categorías de “clásico” y “moderno”. Se trata de una introducción al conocimiento de la biografía de Bernard-Marie Koltès, a su obra y a la recepción crítica de la misma. Son así comentados, algunos estudios críticos destacados dentro de los estudios teatrales franceses como pueden ser el de Anne Ubersfeld o el de Jean-Pierre Sarrazac
Comunicação e saúde mental: um entrecruzamento poderoso
El texto recupera trayectorias que relatan cómo se incluyó la comunicación dentro del campo de la salud mental en Entre Ríos. Preguntas como: ¿por qué la comunicación en salud mental?, ¿cuáles son los aportes de nuestra (in)disciplina en este escenario? La potencialidad clínica y política guían intervenciones, investigaciones y prácticas. Un boletín institucional, la incorporación de la comunicación en una guardia hospitalaria de urgencias, la radio como un espacio para favorecer la construcción del lazo, la lectura de los medios y el acompañamiento a sus profesionales para lograr coberturas respetuosas sobre la temática, la invención de un personaje para accesibilizar conceptos, la sala de espera como ritual de paso, son algunas de las prácticas que hablan de un camino incipiente, sostenido y potente.O artigo recupera trajetórias que relatam como a comunicação foi inserida no campo da saúde mental em Entre Ríos. Perguntas como: por que comunicação em saúde mental? Quais são as contribuições da nossa (in)disciplina neste cenário? The clinical and political potentiality guide interventions, research and practices. Treatment persistence in patients with schizophrenia treated with lurasidone in Italian clinical practice
Background and rationale: Treatment persistence combines clinician and patient judgment of efficacy, tolerability and safety into a comprehensive measure of effectiveness and is defined as the act of continuing a treatment over time. Studies have reported poor treatment persistence to antipsychotic medications in patients with schizophrenia. This study evaluated treatment persistence to lurasidone (LUR) in patients with schizophrenia in a real-world Italian setting. Methods: This was a retrospective observational study of patients with schizophrenia who started treatment with LUR ≥ 6 months before inclusion. Following informed consent, data were collected starting from the index date (start of LUR treatment) at all visits occurring as per clinical practice. The primary endpoint was treatment persistence during the first 6 months, defined as the time between index date and all-cause discontinuation. Patients treated with LUR > 180 days were considered persistent. As secondary endpoint, treatment persistence was evaluated for a period of ≥ 18 months. Results: Forty-five patients were enrolled and 41 (91.11%) completed the study. Forty-one patients (91.11%) were included in the eligible population as they initiated LUR treatment ≥ 6 months before data collection. Patients were 43.0 ± 15.89 years old and 61% were female. Twenty-two patients (53.66%) started LUR treatment in a hospital setting and 19 (46.34%) in an outpatient setting. Based on Clinical Global Impression-Severity scale (CGI-S) at LUR initiation, 12 patients (29.27%) were severely ill, 17.07% markedly ill, 19.51% moderately ill, 2.44% mildly ill and 4.88% borderline mentally ill. Thirty-two patients (78.05%) were treatment persistent for ≥ 180 days. Among the 19 patients observed for ≥ 18 months, 11 (57.89%) were persistent for ≥ 18 months. Among the 22 study patients observed for < 18 months, 12 (54.54%) were persistent. An improvement in schizophrenia severity according to CGI-S was observed at inclusion (following LUR therapy) compared to the index date. Six patients (14.63%) experienced at least one adverse drug reaction: akathisia (7.32%), extrapyramidal disorder (4.88%), hyperprolactinemia (2.44%), restlessness (2.44%), and galactorrhea (2.44%). None were serious. Conclusions: Persistence to LUR in patients with schizophrenia was relatively high: 78% and 58% of patients were still on LUR after 6 and 18 months of treatment, respectively. This may reflect LUR's relatively favorable balance between efficacy and tolerability, as well as favorable patient satisfaction and acceptance

Este artículo revisa el trabajo realizado por el grupo de investigación Documenta Dramáticas a lo largo de casi veinte años en el campo de las dramaturgias de provincias argentinas. Especializado en análisis dramatúrgico, el grupo amplió su trabajo en dos direcciones, a fin de poder abordar la creación dramatúrgica argentina de posdictadura. En este sentido, discute, entre otros conceptos, el de “nueva dramaturgia”, y expone el carácter político de las dramaturgias de algunas provincias. Por último, adelanta proyectos en curso
