Surface Plasmon Resonance kinetic analysis of the interaction between G-quadruplex nucleic acids and an anti-G-quadruplex monoclonal antibody
Background
G-quadruplexes (G4s) are nucleic acids secondary structures formed in guanine-rich sequences. Anti-G4 antibodies represent a tool for the direct investigation of G4s in cells. Surface Plasmon Resonance (SPR) is a highly sensitive technology, suitable for assessing the affinity between biomolecules. We here aimed at improving the orientation of an anti-G4 antibody on the SPR sensor chip to optimize detection of binding antigens.
Methods
SPR was employed to characterize the anti-G4 antibody interaction with G4 and non-G4 oligonucleotides. Dextran-functionalized sensor chips were used both in covalent coupling and capturing procedures.
Results
The use of two leading molecule for orienting the antibody of interest allowed to improve its activity from completely non-functional to 65% active. The specificity of the anti-G4 antobody for G4 structures could thus be assessed with high sensitivity and reliability.
Conclusions
Optimization of the immobilization protocol for SPR biosensing, allowed us to determine the anti-G4 antibody affinity and specificity for G4 antigens with higher sensitivity with respect to other in vitro assays such as ELISA. Anti-G4 antibody specificity is a fundamental assumption for the future utilization of this kind of antibodies for monitoring G4s directly in cells.
General significance
The heterogeneous orientation of amine-coupling immobilized ligands is a general problem that often leads to partial or complete inactivation of the molecules. Here we describe a new strategy for improving ligand orientation: driving it from two sides. This principle can be virtually applied to every molecule that loses its activity or is poorly immobilized after standard coupling to the SPR chip surface
The Role of Consumer's Risk Aversion on Price Rigidity
This paper aims to contribute to the research agenda on the sources of price rigidity. Based on broadly accepted assumptions on the behavior of economic agents, we show that firms’ competition can lead to the adoption of sticky prices as a sub-game perfect equilibrium strategy to optimally deal with consumers’ risk aversion, even if firms have no adjustment costs. To this end, we build a model economy based on consumption centers with several complete markets and relax some traditional assumptions used in standard monetary policy models by assuming that households have imperfect information about the inefficient time-varying cost shocks faced by the .rms. Furthermore, we assume that the timing of events is such that, at every period, consumers have access to the actual prices prevailing in the market only after choosing a particular consumption center. Since such choices under uncertainty may decrease the expected utilities of risk-averse consumers, competitive firms adopt some degree of price stickiness in order to minimize the price uncertainty and "attract more customers".
The geometry of Bayesian programming
We give two geometry of interaction models for a typed λ-calculus with recursion endowed with operators for sampling from a continuous uniform distribution and soft conditioning, namely a paradigmatic calculus for higher-order Bayesian programming. The models are based on the category of measurable spaces and partial measurable functions, and the category of measurable spaces and s-finite kernels, respectively. The former is proved adequate with respect to both a distribution-based and a sampling-based operational semantics, while the latter is proved adequate with respect to a sampling-based operational semantics
Classes of behavior of small-world networks
Small-world networks are the focus of recent interest because they appear to
circumvent many of the limitations of either random networks or regular
lattices as frameworks for the study of interaction networks of complex
systems. Here, we report an empirical study of the statistical properties of a
variety of diverse real-world networks. We present evidence of the occurrence
of three classes of small-world networks: (a) scale-free networks,
characterized by a vertex connectivity distribution that decays as a power law;
(b) broad-scale networks, characterized by a connectivity distribution that has
a power-law regime followed by a sharp cut-off; (c) single-scale networks,
characterized by a connectivity distribution with a fast decaying tail.
Moreover, we note for the classes of broad-scale and single-scale networks that
there are constraints limiting the addition of new links. Our results suggest
that the nature of such constraints may be the controlling factor for the
emergence of different classes of networks
Color space distortions in patients with type 2 diabetes mellitus
Color vision impairment was examined in patients with type 2 diabetes mellitus (DM2) without retinopathy. We assessed the type and degree of distortions of individual color spaces. DM2 patients (n = 32), and age-matched controls (n = 20)were tested using the Farnsworth D-15 and the Lanthony D-15d tests. In addition, subsets of caps
from both tests were employed in a triadic procedure (Bimler & Kirkland, 2004). Matrices of inter-cap subjective dissimilarities were estimated from each subject’s “odd-one-out” choices, and processed using non-metric
multidimensional scaling. Two-dimensional color spaces, individual and group (DM2 patients; controls), were reconstructed, with the axes interpreted as the R0G and B0Y perceptual opponent systems. Compared to controls, patient results were not significant for the D-15 and D-15d. In contrast, in the triadic procedure the residual distances were significantly different compared to controls: right eye, P 0.021, and left eye, P 0.022. Color
space configurations for the DM2 patients were compressed along the B0Y and R0G dimensions. The present findings agree with earlier studies demonstrating diffuse losses in early stages of DM2. The proposed method of testing uses color spaces to represent discrimination and provides more differentiated quantitative diagnosis, which
may be interpreted as the perceptual color system affected. In addition, it enables the detection of very mild color vision impairment that is not captured by the D-15d test. Along with fundoscopy, individual color spaces may serve for monitoring early functional changes and thereby to support a treatment strategy
The Additive Effects of Cell Phone Use and Dental Hygiene Practice on Finger Muscle Strength: A Pilot Study
Purpose: The purpose of this study was to determine strength of muscles involved with instrumentation (scaling) by dental hygienists and the additive effects of cellular (mobile) phone usage, as indicated by measurements of muscular force generation. Methods: A convenience sample of licensed dental hygienists currently in clinical practice (n=16) and an equal number of individuals not currently using devices/tools repetitively for work (n=16), agreed to participate in this pilot study. All participants completed a modified cell phone usage questionnaire to determine their use pattern and frequency. Upon completion of the questionnaire, participants\u27 force production in six muscle groups was measured using a hand-held dynamometer. Descriptive statistics were used to analyze the data. Results: A total of 16 licensed dental hygienists (n=16) and 16 participants with no history of using tools/devices repetitively for work (n=16), comprised the experimental and control groups, repectively. The control group generated greater muscle force than the experimental group for the abductor pollicis longus (p=0.045). Significant differences were identified when comparing the low mobile phone users in the experimental group to the control group for the flexor pollicis brevis (p=0.031), abductor pollicis longus (p=0.031), and flexor digitorum (p=0.006), with the control group demonstrating higher muscle force. Years in clinical practice and mobile phone use was shown to have a significant effect on muscular force generation for the flexor pollicis brevis (F=3.645, df=3, p=0.020) and flexor digitorum (F=3.560, df=3, p=0.022); subjects who practiced dental hygiene the longest produced the least amount of muscle force. Conclusion: Results from this pilot study indicate there are no significant additive effects of cell phone use and dental hygiene practice on finger muscles used for instrumentation. However, results indicate that dental hygiene practice demonstrated significant effects on muscular strength as compared to individuals who do not use tools/devices repetitively for work. The small sample size may have impacted results and the study should be repeated with a larger sample
The Power of Non-Determinism in Higher-Order Implicit Complexity
We investigate the power of non-determinism in purely functional programming
languages with higher-order types. Specifically, we consider cons-free programs
of varying data orders, equipped with explicit non-deterministic choice.
Cons-freeness roughly means that data constructors cannot occur in function
bodies and all manipulation of storage space thus has to happen indirectly
using the call stack.
While cons-free programs have previously been used by several authors to
characterise complexity classes, the work on non-deterministic programs has
almost exclusively considered programs of data order 0. Previous work has shown
that adding explicit non-determinism to cons-free programs taking data of order
0 does not increase expressivity; we prove that this - dramatically - is not
the case for higher data orders: adding non-determinism to programs with data
order at least 1 allows for a characterisation of the entire class of
elementary-time decidable sets.
Finally we show how, even with non-deterministic choice, the original
hierarchy of characterisations is restored by imposing different restrictions.Comment: pre-edition version of a paper accepted for publication at ESOP'1

Space Weather Application Using Projected Velocity Asymmetry of Halo CMEs
Halo coronal mass ejections (HCMEs) originating from regions close to the
center of the Sun are likely to be responsible for severe geomagnetic storms.
It is important to predict geo-effectiveness of HCMEs using observations when
they are still near the Sun. Unfortunately, coronagraphic observations do not
provide true speeds of CMEs due to the projection effects. In the present
paper, we present a new technique allowing estimate the space speed and
approximate source location using projected speeds measured at different
position angles for a given HCME (velocity asymmetry). We apply this technique
to HCMEs observed during 2001-2002 and find that the improved speeds are better
correlated with the travel times of HCMEs to Earth and with the magnitudes
