Heavy--light mesons in a bilocal effective theory
Heavy--light mesons are described in an effective quark theory with a
two--body vector--type interaction. The bilocal interaction is taken to be
instantaneous in the rest frame of the bound state, but formulated covariantly
through the use of a boost vector. The chiral symmetry of the light flavor is
broken spontaneously at mean field level. The framework for our discussion of
bound states is the effective bilocal meson action obtained by bosonization of
the quark theory. Mesons are described by 3--dimensional wave functions
satisfying Salpeter equations, which exhibit both Goldstone solutions in the
chiral limit and heavy--quark symmetry for . We present
numerical solutions for pseudoscalar -- and --mesons. Heavy--light meson
spectra and decay constants are seen to be sensitive to the description of
chiral symmetry breaking (dynamically generated vs.\ constant quark mass).Comment: (34 p., standard LaTeX, 7 PostScript figures appended)

Quantitative Estimation of Wheat Phenotyping Traits Using Ground and Aerial Imagery
This study evaluates an aerial and ground imaging platform for assessment of canopy development in a wheat field. The dependence of two canopy traits, height and vigour, on fertilizer treatment was observed in a field trial comprised of ten varieties of spring wheat. A custom-built mobile ground platform (MGP) and an unmanned aerial vehicle (UAV) were deployed at the experimental site for standard red, green and blue (RGB) image collection on five occasions. Meanwhile, reference field measurements of canopy height and vigour were manually recorded during the growing season. Canopy level estimates of height and vigour for each variety and treatment were computed by image analysis. The agreement between estimates from each platform and reference measurements was statistically analysed. Estimates of canopy height derived from MGP imagery were more accurate (RMSE = 3.95 cm, R2 = 0.94) than estimates derived from UAV imagery (RMSE = 6.64 cm, R2 = 0.85). In contrast, vigour was better estimated using the UAV imagery (RMSE = 0.057, R2 = 0.57), compared to MGP imagery (RMSE = 0.063, R2 = 0.42), albeit with a significant fixed and proportional bias. The ability of the platforms to capture differential development of traits as a function of fertilizer treatment was also investigated. Both imaging methodologies observed a higher median canopy height of treated plots compared with untreated plots throughout the season, and a greater median vigour of treated plots compared with untreated plots exhibited in the early growth stages. While the UAV imaging provides a high-throughput method for canopy-level trait determination, the MGP imaging captures subtle canopy structures, potentially useful for fine-grained analyses of plants

The Paleocene/Eocene boundary Global Standard Stratotype-section and Point (GSSP): Criteria for Characterisation and Correlation
The choice of a Paleocene/Eocene (P/E) Global Standard Stratotype-section and Point (GSSP) is complicated by the fact that there exists confusion on the exact denotation of the Paleocene and Eocene Series and their constituent lower rank (stage) units. While we can now resolve this problem by recourse to rigorous historical analysis, actual placement of the GSSP is further exacerbated by an embarrassment of riches (in regards to 7 criteria suitable for characterising and correlating a PIE GSSP but which span a temporal interval of greater than 2 my).
Following the precept that the boundaries between higher level chronostratigraphic units are to be founded upon the boundaries of their lowest constituent stages in a nested hierarchy, we note that one of the criteria providing global correlation potential (a stable isotope excursion in marine and terrestrial stratigraphies) lies at a stratigraphic level more than !my older than the base of the stratotypic Ypresian Stage to which the base of the Eocene Series has been subordinated until now. Lowering a chronostratigraphic unit by this extent risks a significant modification to the original geohistorical denotation of the Ypresian Stage and the Eocene Series.
We discuss here four options that are open to Voting Members of the Paleogene Subcommission. One solution consists in adjusting slightly the base of the Ypresian Stage (and, thus, the Eocene Series) so as to be correlatable on the basis of the lowest occurrence/First Appearance Datum (LO/FAD) of the calcareous nannofossil species Tribrachiatus.digitalis. Another solution would be to decouple series and stages so that the Ypresian Stage remains essentially unaltered but the base of tbe Eocene is relocated so as to be correlated on the basis of the Carbon Isotope Excursion (CIE).
Two (compromise) solutions consist in erecting a new stage for the upper/terminal Paleocene (between the Thanetian [sensu Dollfus] and Ypresian Stages) characterised at its base by the global stable isotope excursion. The P/E GSSP may then be placed at the base of the stratotypic Ypresian Stage (thus preserving historical continuity and conceptual denotation and stability) or at the base of the newly erected stage (facilitating correlation of the base of the Eocene series, at least in principle). Both GSSPs should be placed in suitable marine stratigraphic sections yet to be determined but upon which there is considerable current investigative activity

Antimicrobial Nanoplexes meet Model Bacterial Membranes: the key role of Cardiolipin
Antimicrobial resistance to traditional antibiotics is a crucial challenge of medical research. Oligonucleotide therapeutics, such as antisense or Transcription Factor Decoys (TFDs), have the potential to circumvent current resistance mechanisms by acting on novel targets. However, their full translation into clinical application requires efficient delivery strategies and fundamental comprehension of their interaction with target bacterial cells. To address these points, we employed a novel cationic bolaamphiphile that binds TFDs with high affinity to form self-assembled complexes (nanoplexes). Confocal microscopy revealed that nanoplexes efficiently transfect bacterial cells, consistently with biological efficacy on animal models. To understand the factors affecting the delivery process, liposomes with varying compositions, taken as model synthetic bilayers, were challenged with nanoplexes and investigated with Scattering and Fluorescence techniques. Thanks to the combination of results on bacteria and synthetic membrane models we demonstrate for the first time that the prokaryotic-enriched anionic lipid Cardiolipin (CL) plays a key-role in the TFDs delivery to bacteria. Moreover, we can hypothesize an overall TFD delivery mechanism, where bacterial membrane reorganization with permeability increase and release of the TFD from the nanoplexes are the main factors. These results will be of great benefit to boost the development of oligonucleotides-based antimicrobials of superior efficacy
Re-evaluation of histological diagnoses of malignant mesothelioma by immunohistochemistry
<p>Abstract</p> <p>Background</p> <p>In order to provide reliable tissue material for malignant mesothelioma (MM) studies, we re-evaluated biopsies and autopsy material from 61 patients with a diagnosis of MM from the period of 1980-2002.</p> <p>Methods</p> <p>Basic positive (Calretinin, EMA, Podoplanin, Mesothelin) and negative (CEA, Ber-Ep4) immunohistochemical (IHC) marker reactions were determined. If needed, more markers were used. Histological diagnoses were made by three pathologists. Survival data were calculated.</p> <p>Results</p> <p>49 cases (80%) were considered being MM by a high degree of likelihood, five more cases possible MM. Of the remaining seven cases, three were diagnosed as adenocarcinoma, three as pleomorphic lung carcinoma, in one peritoneal case a clear entity diagnosis could not be given. One of the possible MM cases and two of the lung carcinoma cases had this already as primary diagnoses, but were registered as MM.</p> <p>With a sensitivity of 100%, Calretinin and CEA were the most reliable single markers. The amount of MM cells with positive immunoreactivity (IR) for Podoplanin and Mesothelin showed most reliable inverse relation to the degree of atypia.</p> <p>In the confirmed MM cases, there had been applied either no IHC or between one and 18 markers.</p> <p>The cases not confirmed by us had either lacked IHC (n = 1), non-specific markers were used (n = 4), IR was different (n = 1), or specific markers had not shown positive IR in the right part of the tumour cells (n = 3).</p> <p>46 of the 49 confirmed and three of the not confirmed cases had been diagnosed by us as most likely MM before IHC was carried out.</p> <p>Conclusions</p> <p>In order to use archival tissue material with an earlier MM diagnosis for studies, histopathological re-evaluation is important. In possible sarcomatous MM cases without any positive IR for positive MM markers, radiology and clinical picture are essential parts of diagnostics. IHC based on a panel of two positive and two negative MM markers has to be adapted to the differential diagnostic needs in each single case. New diagnostic tools and techniques are desirable for cases where IHC and other established methods cannot provide a clear entity diagnosis, and in order to improve MM treatment.</p
The silent HIV epidemic among pregnant women within rural Northern Tanzania
BACKGROUND: Many national antenatal clinics (ANC) based HIV surveillance systems in sub-Saharan Africa have limited coverage of remote rural sites, a weakness that compromises adequate estimation, monitoring and development of effective preventive and care programmes. To address this void in rural Manyara and Singida within Northern Tanzania, we conducted antenatal clinic-based sentinel surveillance. METHODS: We consecutively enrolled 1377 counselled and consenting pregnant women attending ANC clinics for the first time during the current pregnancy. The study was conducted in six antenatal clinics, within three divisions of rural Manyara and Singida regions in 2003/2004. Interviews were conducted and blood samples for routine purposes were collected and tested for anti-HIV IgG antibody anonymously, using Bionor HIV-1 & 2 assay (®). RESULTS: Among enrolees, 94% (1296/1377) participated fully. The overall prevalence of HIV was 2.0% (95%CI: 1.34–2.97). The highest HIV prevalence was among women aged between 15–19 years in both rural and remote rural populations. The odds of HIV infection was 4.3 (95%CI: 1.42–12.77) times among women reporting more than one lifetime sexual partners compared with those with one partner. HIV infection was associated with history of genital sores or foul smelling discharge, OR 6.8 (95%CI: 2.78–16.66) and age at first pregnancy (2.5 times higher likelihood of infection if before the age of 18 years versus at a later age). CONCLUSION: Including rural remote sites, as part of the national ANC routine surveillance, is crucial in order to discover imminent silent epidemics such as the one described in this paper. Scaling up HIV prevention efforts is mandatory to prevent the imminent escalation of the HIV epidemic highly associated with a history of sexually transmitted infections (STIs), multiple sexual partners and pregnancies at a younger age. Ignorance of relevant knowledge and low utilisation of condoms underscores the urgency for large-scale preventive efforts. Research to capture a wider representation of the risk factors in the general population should be a priority to enable further customised HIV prevention efforts
A review of the fossil record of turtles of the clade Pan-Carettochelys
Turtles of the total clade Pan-Carettochelys have a relatively poor fossil record that extends from the Early Cretaceous. The clade is only found in Asia during the Cretaceous, but spreads to Europe and North America during the Eocene. Neogene finds are restricted to Europe, Africa and Australia, whereas the only surviving species, Carettochelys insculpta, lives in New Guinea and the Northern Territories of Australia. The ecology of fossil pan-carettochelyids appears similar to that of the extant C. insculpta, although more primitive representatives were likely less adapted to brackish water. Current phylogenies only recognize three internested clades: Pan- Carettochelys, Carettochelyidae and Carettochelyinae. A taxonomic review of the group concludes that of 25 named taxa, 13 are nomina valida, 7 are nomina invalida, 3 are nomina dubia, and 2 are nomina nuda
Lost opportunities in HIV prevention: programmes miss places where exposures are highest
Background: Efforts at HIV prevention that focus on high risk places might be more effective and less stigmatizing than those targeting high risk groups. The objective of the present study was to assess risk behaviour patterns, signs of current preventive interventions and apparent gaps in places where the risk of HIV transmission is high and in communities with high HIV prevalence. Methods: The PLACE method was used to collect data. Inhabitants of selected communities in Lusaka and Livingstone were interviewed about where people met new sexual partners. Signs of HIV preventive activities in these places were recorded. At selected venues, people were interviewed about their sexual behaviour. Peer educators and staff of NGOs were also interviewed. Results: The places identified were mostly bars, restaurants or sherbeens, and fewer than 20% reported any HIV preventive activity such as meetings, pamphlets or posters. In 43% of places in Livingstone and 26% in Lusaka, condoms were never available. There were few active peer educators. Among the 432 persons in Lusaka and 676 in Livingstone who were invited for interview about sexual behaviour, consistent condom use was relatively high in Lusaka (77%) but low in Livingstone (44% of men and 34% of women). Having no condom available was the most common reason for not using one. Condom use in Livingstone was higher among individuals socializing in places where condoms always were available. Conclusion: In the places studied we found a high prevalence of behaviours with a high potential for HIV transmission but few signs of HIV preventive interventions. Covering the gaps in prevention in these high exposure places should be given the highest priority
