Technical product risk assessment: Standards, integration in the erm model and uncertainty modeling
European Union has accomplished, through introducing New Approach to technical harmonization and standardization, a breakthrough in the field of technical products safety and in assessing their conformity, in such a manner that it integrated products safety requirements into the process of products development. This is achieved by quantifying risk levels with the aim of determining the scope of the required safety measures and systems. The theory of probability is used as a tool for modeling uncertainties in the assessment of that risk. In the last forty years are developed new mathematical theories have proven to be better at modeling uncertainty when we have not enough data about uncertainty events which is usually the case in product development. Bayesian networks based on modeling of subjective probability and Evidence networks based on Dempster-Shafer theory of belief functions proved to be an excellent tool for modeling uncertainty when we do not have enough information about all events aspect
Conformational analysis and in vitro immunomodulatory and insulinotropic properties of the frog skin host-defense peptide rhinophrynin-27 and selected analogs
The study investigates conformational analysis and the in vitro cytokine-mediated immunomodulatory and insulin-releasing activities of rhinophrynin-27 (ELRLPEIARPVPEVLPARLPLPALPRN; RP-27), a proline-arginine-rich peptide first isolated from skin secretions of the Mexican burrowing toad Rhinophrynus dorsalis (Rhinophrynidae). In both water and 50% trifluoroethanol-water, the peptide adopts a polyproline type II helical conformation with a high degree of deviation from the canonical collagen-like folding and a pronounced bend in the molecule at the Glu13 residue. Incubation of mouse peritoneal cells with RP-27 significantly (P < 0.05) inhibited production of the pro-inflammatory cytokines TNF-α and IL-1β and stimulated production of the anti-inflammatory cytokine IL-10. The peptide significantly (P < 0.01) stimulated release of insulin from BRIN-BD11 rat clonal β-cells at concentrations ≥ 1 nM while maintaining the integrity of the plasma membrane and also stimulated insulin release from isolated mouse islets at a concentration of 10−6 M. Increasing the cationicity of RP-27 by substituting glutamic acid residues in the peptide by arginine and increasing hydrophobicity by substituting alanine residues by tryptophan did not result in analogues with increased activity with respect to cytokine production and insulin release. The combination of immunosuppressive and insulinotropic activities together with very low cytotoxicity suggests that RP-27 may represent a template for the development of an agent for use in anti-inflammatory and Type 2 diabetes therapies
Lack of Apoptosis of Infiltrating Cells as the Mechanism of High Susceptibility to EAE in DA Rats
Dark Agouti (DA) rats are highly susceptible to induction of Th-l-mediated autoimmunity
disease, including experimental allergic encephalomyelitis (EAE). In contrast to other susceptible
rat strains in which disease is induced only with encephalitogen emulsified in complete
Freund's adjuvants (CFA), in DA rats EAE develops after injection of encephalitogen in
incomplete Freund's adjuvants (IFA) or Titermax, putative Th-2 directed adjuvant. Lymph
node cells derived from immunized DA rats and stimulated in vitro produce significantly
more Interferon-γ (IFN-γ) than resistant Albino Oxford (AO) rats. However, cells derived
from both strains produce large amounts of IL-10 but not IL-4. Immunized lymph node cells
derived from EAE susceptible (AO × DA) F1rats induce clinical signs of disease in sublethally
irradiated parental DA but not AO rats. The pathohistology of the target tissue in these
recipients clearly demonstrated infiltration of mononuclear cells in both parental strains.
However, the number of CD4+ cells was significantly higher and number of apoptotic cells
significantly lower in DA rats sacrificed 8 days after passive transfer. We postulate that in
addition to higher IFN-γ and TNF-α production, resistance to early apoptosis of the invading
cells in the target tissue possibly due to lack of downregulation by TGF-β leads to exceptional
susceptibility to EAE in DA rats
Energy gap of the bimodal two-dimensional Ising spin glass
An exact algorithm is used to compute the degeneracies of the excited states
of the bimodal Ising spin glass in two dimensions. It is found that the
specific heat at arbitrary low temperature is not a self-averaging quantity and
has a distribution that is neither normal or lognormal. Nevertheless, it is
possible to estimate the most likely value and this is found to scale as L^3
T^(-2) exp(-4J/kT), for a L*L lattice. Our analysis also explains, for the
first time, why a correlation length \xi ~ exp(2J/kT) is consistent with an
energy gap of 2J. Our method allows us to obtain results for up to 10^5
disorder realizations with L <= 64. Distributions of second and third

Pointwise consistency of the kriging predictor with known mean and covariance functions
This paper deals with several issues related to the pointwise consistency of
the kriging predictor when the mean and the covariance functions are known.
These questions are of general importance in the context of computer
experiments. The analysis is based on the properties of approximations in
reproducing kernel Hilbert spaces. We fix an erroneous claim of Yakowitz and
Szidarovszky (J. Multivariate Analysis, 1985) that the kriging predictor is

IL-33/ST2 axis in innate and acquired immunity to tumors
Interleukin-33, a ligand for ST2/T1, has an important role in allergy, autoimmunity and inflammation. The role of IL-33/ST2 axis in cancer is not elucidated. Using metastatic breast cancer model we provide evidence that lack of ST2 signaling led to reduced tumor growth and metastasis and enhanced anti-tumor immunity
Large random correlations in individual mean field spin glass samples
We argue that complex systems must possess long range correlations and
illustrate this idea on the example of the mean field spin glass model. Defined
on the complete graph, this model has no genuine concept of distance, but the
long range character of correlations is translated into a broad distribution of
the spin-spin correlation coefficients for almost all realizations of the
random couplings. When we sample the whole phase space we find that this
distribution is so broad indeed that at low temperatures it essentially becomes
uniform, with all possible correlation values appearing with the same
probability. The distribution of correlations inside a single phase space

Numerical solution to the hermitian Yang-Mills equation on the Fermat quintic
We develop an iterative method for finding solutions to the hermitian
Yang-Mills equation on stable holomorphic vector bundles, following ideas
recently developed by Donaldson. As illustrations, we construct numerically the
hermitian Einstein metrics on the tangent bundle and a rank three vector bundle
on P^2. In addition, we find a hermitian Yang-Mills connection on a stable rank
