    CD30-positive primary cutaneous lymphoproliferative disorders: molecular alterations and targeted therapies

    Primary cutaneous CD30-positive T-cell lymphoproliferative disorders are the second most common subgroup of cutaneous T-cell lymphomas. They include two clinically different entities with some overlapping features and borderline cases: lymphomatoid papulosis and primary cutaneous anaplastic large cell lymphoma. Molecular studies of primary cutaneous anaplastic large cell lymphoma reveal an increasing level of heterogeneity that is associated with histological and immunophenotypic features of the cases and their response to specific therapies. Here, we review the most significant genetic, epigenetic and molecular alterations described to date in primary cutaneous CD30-positive T-cell lymphoproliferative disorders, and their potential as therapeutic targets

    A feature-rich transmission spectrum for WASP-127b

    WASP-127b is one of the lowest density planets discovered to date. With a sub-Saturn mass (Mp=0.18±0.02MJM_{\rm p}=0.18 \pm 0.02 M_J) and super-Jupiter radius (Rp=1.37±0.04RJR_{\rm p}= 1.37 \pm 0.04 R_J), it orbits a bright G5 star, which is about to leave the main-sequence. We aim to explore WASP-127b's atmosphere in order to retrieve its main atmospheric components, and to find hints for its intriguing inflation and evolutionary history. We used the ALFOSC spectrograph at the NOT telescope to observe a low resolution (R330R\sim330, seeing limited) long-slit spectroscopic time series during a planetary transit, and present here the first transmission spectrum for WASP-127b. We find the presence of a strong Rayleigh slope at blue wavelengths and a hint of Na absorption, although the quality of the data does not allow us to claim a detection. At redder wavelengths the absorption features of TiO and VO are the best explanation to fit the data. Although higher signal-to-noise ratio observations are needed to conclusively confirm the absorption features, WASP-127b seems to posses a cloud-free atmosphere and is one of the best targets to perform further characterization studies in the near future.Comment: Accepted for Publication A&A Letters, May 22nd, 201

    High spatial resolution and high contrast optical speckle imaging with FASTCAM at the ORM

    In this paper, we present an original observational approach, which combines, for the first time, traditional speckle imaging with image post-processing to obtain in the optical domain diffraction-limited images with high contrast (1e-5) within 0.5 to 2 arcseconds around a bright star. The post-processing step is based on wavelet filtering an has analogy with edge enhancement and high-pass filtering. Our I-band on-sky results with the 2.5-m Nordic Telescope (NOT) and the lucky imaging instrument FASTCAM show that we are able to detect L-type brown dwarf companions around a solar-type star with a contrast DI~12 at 2" and with no use of any coronographic capability, which greatly simplifies the instrumental and hardware approach. This object has been detected from the ground in J and H bands so far only with AO-assisted 8-10 m class telescopes (Gemini, Keck), although more recently detected with small-class telescopes in the K band. Discussing the advantage and disadvantage of the optical regime for the detection of faint intrinsic fluxes close to bright stars, we develop some perspectives for other fields, including the study of dense cores in globular clusters.     The ALMA Early Science View of FUor/EXor Objects - V. Continuum Disc Masses and Sizes

    Low-mass stars build a significant fraction of their total mass during short outbursts of enhanced accretion known as FUor and EXor outbursts. FUor objects are characterized by a sudden brightening of ∼5 mag at visible wavelengths within 1 yr and remain bright for decades. EXor objects have lower amplitude outbursts on shorter time-scales. Here we discuss a 1.3 mm Atacama Large Millimeter/submillimeter Array (ALMA) mini-survey of eight outbursting sources (three FUors, four EXors, and the borderline object V1647 Ori) in the Orion Molecular Cloud. While previous papers in this series discuss the remarkable molecular outflows observed in the three FUor objects and V1647 Ori, here we focus on the continuum data and the differences and similarities between the FUor and EXor populations. We find that FUor discs are significantly more massive (∼80–600 MJup) than the EXor objects (∼0.5–40 MJup). We also report that the EXor sources lack the prominent outflows seen in the FUor population. Even though our sample is small, the large differences in disc masses and outflow activity suggest that the two types of objects represent different evolutionary stages. The FUor sources seem to be rather compact (Rc \u3c 20–40 au) and to have a smaller characteristic radius for a given disc mass when compared to T Tauri stars. V1118 Ori, the only known close binary system in our sample, is shown to host a disc around each one of the stellar components. The disc around HBC 494 is asymmetric, hinting at a structure in the outer disc or the presence of a second disc

    Efficacy of DNA amplification in tissue biopsy samples to improve the detection of invasive fungal disease

    AbstractThe performance of a pan-fungal PCR-based technique was evaluated to assess the aetiology of invasive fungal diseases (IFDs). A total of 89 formalin-fixed paraffin-embedded biopsy samples from 84 patients with proven IFD were studied. Culture of tissue was performed in 68 (81%) patients. The sensitivities of the PCR-based technique and microbiological culture of tissues were 89% and 56%, respectively (p <0.01). According to PCR results, Aspergillus species accounted for 67%, Candida species for 13%, zygomycetes for 11%, and rare and emerging fungi for 9%. Aspergillus species were significantly associated with lung samples (79.6%, p <0.01), Mucorales were associated with skin/subcutaneous samples, and Candida species were associated with gastrointestinal samples. Regarding biopsy samples with Aspergillus species, Aspergillus fumigatus DNA was detected in 43 of 50 (86%), and Aspergillus flavus in six of 50 (12%). PCR was positive in 24 of 30 (80%) cases with negative culture. In nine of the 84 patients, the PCR technique failed to amplify the DNA. Six also had negative cultures, and in the remaining three cases culture was positive (Rhizopus microsporus, Rhizopus arrhizus, and Sakseneae vasiformis), suggesting that the PCR technique was not as effective in amplifying the DNA of some species of Zygomycetes. In five cases, there was no correlation between culture results and those obtained with DNA amplification, indicating the possibility of a mixed infection or the presence of colonizer/contaminant microorganisms. In summary, PCR-based techniques for DNA amplification should be implemented in histopathology and microbiology departments, as they appear to be complementary to conventional methods for IFD detection

    We have optimized a synthetic approach to (±)-Ferrugine in 8 steps starting from 5-aminocyclohept-1-ene and using the Brønsted acid-catalyzed pseudotransannular ring-opening of the epoxide derived from this cycloheptene as the key step for the construction of the 8-azabicyclo[3.2.1]octane central core. While attempting the enantioselective synthesis of this natural product from enantiopure 2-hydroxy-8-azabicyclo[3.2.1]octane we have found that this compound shows a pronounced tendency to racemize via an achiral symmetric aziridinium intermediate. This racemization side process has been studied in detail using both experimental and computational methods

    Characterization of a possible nosocomial aspergillosis outbreak

    ObjectiveTo study the epidemiologic aspects of a suspected outbreak of nosocomial invasive aspergillosis.MethodsSixteen Aspergillus fumigatus strains were isolated from bronchoalveolar washings or sputa of 10 patients during a 9-month period. Furthermore, two environmental samples, isolated in a microbiological screening of the hospital, were also available for analysis. Random amplified polymorphic DNA analysis (RAPD) was carried out.ResultsThe analysis performed by RAPD clearly demonstrated substantial genetic variation among the isolates. Both of the two different primers selected for RAPD analysis (R-108 and AP12h) were able to demonstrate that the strains isolated from all patients infected with the same fungal species and the environmental samples were genotypically distinct. The results by RAPD typing demonstrated that this technique could detect variability among isolates of Aspergillus fumigatus from different patients and even from the same patient.ConclusionsRAPD genotyping proved that the outbreak of invasive aspergillosis consisted of a series of events, non-related, and probably not coming from the same source within the hospital. This type of analysis is an easy, quick and highly discriminatory technique that may help in planning epidemiologic studies of aspergillosis
