    Singular boundary behaviour and large solutions for fractional elliptic equations

    We perform a unified analysis for the boundary behaviour of solutions to nonlocal fractional equations posed in bounded domains. Based on previous findings for some models of the fractional Laplacian operator, we show how it strongly differs from the boundary behaviour of solutions to elliptic problems modelled upon the Laplace–Poisson equation with zero boundary data. In the classical case it is known that, at least in a suitable weak sense, solutions of the homogeneous Dirichlet problem with a forcing term tend to zero at the boundary. Limits of these solutions then produce solutions of some non-homogeneous Dirichlet problem as the interior data concentrate suitably to the boundary. Here, we show that, for equations driven by a wide class of nonlocal fractional operators, different blow-up phenomena may occur at the boundary of the domain. We describe such explosive behaviours and obtain precise quantitative estimates depending on simple parameters of the nonlocal operators. Our unifying technique is based on a careful study of the inverse operator in terms of the corresponding Green function

    Proper motions and velocity asymmetries in the RW Aur jet

    We present adaptive optics spectro-imaging observations of the RW Aur jet in optical forbidden lines, at an angular resolution of 0.4 arcsec. Comparison with HST data taken 2 years later shows that proper motions in the blueshifted and redshifted lobes are in the same ratio as their radial velocities, a direct proof that the velocity asymmetry in this jet is real and not an emissivity effect. The inferred jet inclination to the line of sight is i = 46 +/- 3 degrees. The inner knot spacing appears best explained by time variability with at least two modes: one irregular and asymmetric (possibly random) on timescales of <3-10 yr, and another more regular with ~ 20 yr period. We also report indirect evidence for correlated velocity and excitation gradients in the redshifted lobe, possibly related to the blue/red velocity and brightness asymmetry in this system.Comment: 4 pags, 3 figure

    Tissue factor as a novel marker for detection of circulating cancer cells

    Tissue factor (TF) is a molecular marker that is up-regulated in cancer cells and aids tumoral dissemination. Our purpose was to develop a nested RT-PCR strategy against TF for detecting blood-borne tumour cells. Our method detected TF expression in a minimum of 1.5 pg total RNA from MCF7 cells. A preliminary study in blood samples from 16 advanced breast carcinoma patients showed that 80% of patients with high TF load progressed and died, while only 18% with low TF load showed the same behaviour.     Severe Neurological Abnormalities in a Young Boy with Impaired Thyroid Hormone Sensitivity Due to a Novel Mutation in the MCT8 Gene

    Monocarboxylate transporter 8 (MCT8) is an active and specific thyroid hormone transporter into neurons. MCT8 mutations cause an X-linked condition known as Allan-Herndon-Dudley syndrome and are characterized by impaired psychomotor development and typical abnormal thyroid function. We describe a 10-year-old boy with severe cognitive disability, axial hypotonia, spastic quadriplegia and sporadic dyskinetic episodes. He initially presented with thyroid dysfunction (high FT3, low rT3, low FT4 and normal TSH) and generalized retardation of the cerebral and cerebellar myelination in brain magnetic resonance imaging. The clinical and laboratory findings led to sequencing of the SLC16A2/MCT8 gene, which identified a novel missense mutation in exon 5. The study of peripheral markers of thyroid function suggests a paradoxical state of thyrotoxicosis in some peripheral tissues. Our patient had a typical clinical presentation at birth but because of the rarity of his disease his diagnosis was not made until the age of 7. The delay can also be explained by the omission of the free T3 assay in the first thyroid evaluation performed. This case therefore highlights the possible benefit of including the T3 assay in the study of patients with severe psychomotor disability of unknown etiology, thus eliminating extra costs for unnecessary complementary diagnostic tests.info:eu-repo/semantics/publishedVersio

    Seleção de isolados de vírus e determinação da concentração letal média (CL50) para controle de S. Eridania, S. Cosmioides e S. frugiperda.

    Os lepidópteros desfolhadores principalmente do gênero Spodoptera são atualmente um dos mais importantes problemas entomológicos em várias culturas de expressão econômica. Como alternativa ao controle químico, podem ser utilizados microrganismos entomopatogênicos, incluindo bactérias, vírus, fungos e protozoários. Este trabalho teve como objetivo avaliar isolados de vírus para as espécies de S. cosmioides, S. eridania e S. frugiperda. Lagartas de 3º instar (n=50), foram inoculadas com uma suspensão viral [1,0 x 107 corpos poliédricos de inclusão do vírus (CPI/mL)] incorporada na dieta artificial a fim de analisar a mortalidade causada por diferentes isolados de vírus. Para a determinação da CL50 seguiu-se a mesma metodologia, porém utilizando 80 lagartas por concentração (concentrações de 4.000; 8.000; 16.000; 32.000; 64.000 e 128.000 CPI/mL). Os dados de mortalidade foram submetidos à análise Probit, para a estimação CL50, bem como dos parâmetros associados (Intervalo de confiança (IC 95%) e ?2). Para a espécie S. cosmioides o isolado VPN 72 isolado desta mesma espécie resultou em mortalidade de 20%. Os isolados VNP 143, 144 e 152 obtidos de outros hospedeiros, mataram 24, 100 e 80% respectivamente. As lagartas de S. frugiperda apresentaram mortalidade de 100% somente com os isolados obtidos da mesma espécie VPN 76 e VG 89. S. eridania teve 20% de mortalidade com o isolado VPN 144, sendo este isolado o único que provocou mortalidade nas duas espécies. Os valores de ?2 calculados não foram significativos, indicando a homogeneidade dos dados e, principalmente, que os dados adaptaram-se ao modelo de análise Probit. O VPN 144 de A. californica inoculado em S. albula, testado em S. cosmioides apresentou a menor CL50, caracterizando-se como o mais virulento, quando comparado ao VPN 152 que resultou em CL50 de 49,13 CPI/mL ou seja foi duas vezes menos virulento que o VPN 144. Esse mesmo isolado testado em S. eridania foi 67 vezes menos virulento quando comparado com a espécie de S. cosmioides. Esta espécie apresentou-se a mais suscetível a isolados de vírus provenientes de outros hospedeiros.SICONBIOL 2011

    β-Secretase 1’s Targeting Reduces Hyperphosphorilated Tau, Implying Autophagy Actors in 3xTg-AD Mice

    β-site APP cleaving enzyme 1 (BACE1) initiates APP cleavage, which has been reported to be an inducer of tau pathology by altering proteasome functions in Alzheimer's disease (AD). However, the exact relationship between BACE1 and PHF (Paired Helical Filaments) formation is not clear. In this study, we confirm that BACE1 and Hsc70 are upregulated in the brains of AD patients, and we demonstrate that both proteins show enhanced expression in lipid rafts from AD-affected triple transgenic mouse brains. BACE1 targeting increased Hsc70 levels in the membrane and cytoplasm fractions and downregulated Hsp90 and CHIP in the nucleus in the hippocampi of 3xTg-AD mice. However, these observations occurred in a proteasome-independent manner in vitro. The BACE1miR-induced reduction of soluble hyperphosphorylated tau was associated with a decrease in MAPK activity. However, the BACE1 RNAi-mediated reduction of hyperphosphorylated tau was only blocked by 3-MA (3-methyladenine) in vitro, and it resulted in the increase of Hsc70 and LAMP2 in lipid rafts from hippocampi of 3xTg-AD mice, and upregulation of survival and homeostasis signaling. In summary, our findings suggest that BACE1 silencing neuroprotects reducing soluble hyperphosphorylated tau, modulating certain autophagy-related proteins in aged 3xTg-AD mice

    Mechanisms of innate immune activation by gluten peptide p31-43 in mice

    Celiac disease (CD) is an immune-mediated enteropathy triggered by gluten in genetically susceptible individuals. Innate immunity contributes to the pathogenesis of CD, but the mechanisms remain poorly understood. Although previous in vitro work suggests that gliadin peptide p31-43 acts as an innate immune trigger, the underlying pathways are unclear and have not been explored in vivo. Here we show that intraluminal delivery of p31-43 induces morphological changes in the small intestinal mucosa of normal mice consistent with those seen in CD, including increased cell death and expression of inflammatory mediators. The effects of p31-43 were dependent on MyD88 and type I IFNs, but not Toll-like receptor 4 (TLR4), and were enhanced by coadministration of the TLR3 agonist polyinosinic:polycytidylic acid. Together, these results indicate that gliadin peptide p31-43 activates the innate immune pathways in vivo, such as IFN-dependent inflammation, relevant to CD. Our findings also suggest a common mechanism for the potential interaction between dietary gluten and viral infections in the pathogenesis of CD

    Outbreak Of NDM-1-producing Klebsiella Pneumoniae In A Neonatal Unit In Colombia

    Six multiresistant, NDM-1-producing Klebsiella pneumoniae strains were recovered from an outbreak that affected six neonatal patients in a Colombian hospital. Molecular analysis showed that all of the isolates harbored the blaNDM-1, qnrA, and intI1 genes and were clonally related. Multilocus sequence typing showed that the isolates belonged to a new sequence type (ST1043) that was different from the sequence types that had previously been reported. This is the first report of NDM-1-producing isolates in South America

    A theoretical Density Functional study of the trans-3,6-dimethoxy-1,2-dioxane molecule is performed with the purpose to analyze different conformational features.The energetic stability of the different isomers is discussed in terms of several electronic factors. Results are compared with previous semiempirical studies and they are discussed in a comparative fashion. Final results show that ab initio studies allow one to overcome some drawbacks derived from the semiempirical techniques.Centro de Química Inorgánic
