    Automatic Feeder for Poultry Farm

    This project is to monitor the automatic feeder without the interruption of man power in the poultry sheds. In this project, we have PIC controller and GSM to monitor the system automatically by notifying the owner of poultry by sending a SMS which contains the information regarding the main feeder container. We have also used the ultrasonic sensors to measure the distance between the valve and grains in the feeder trays and the main tank, after measuring the distance it opens the outlet of feeder, if the grains are below the level and close the valve when it reaches to the level

    Materials Cloud, a platform for open computational science

    Materials Cloud is a platform designed to enable open and seamless sharing of resources for computational science, driven by applications in materials modelling. It hosts 1) archival and dissemination services for raw and curated data, together with their provenance graph, 2) modelling services and virtual machines, 3) tools for data analytics, and pre-/post-processing, and 4) educational materials. Data is citable and archived persistently, providing a comprehensive embodiment of the FAIR principles that extends to computational workflows. Materials Cloud leverages the AiiDA framework to record the provenance of entire simulation pipelines (calculations performed, codes used, data generated) in the form of graphs that allow to retrace and reproduce any computed result. When an AiiDA database is shared on Materials Cloud, peers can browse the interconnected record of simulations, download individual files or the full database, and start their research from the results of the original authors. The infrastructure is agnostic to the specific simulation codes used and can support diverse applications in computational science that transcend its initial materials domain.Comment: 19 pages, 8 figure

    Repurposing of neprilysin inhibitor ‘sacubitrilat’ as an anti-cancer drug by modulating epigenetic and apoptotic regulators

    Abstract Modifications in the epigenetic landscape have been considered a hallmark of cancer. Histone deacetylation is one of the crucial epigenetic modulations associated with the aggressive progression of various cancer subtypes. Herein, we have repurposed the neprilysin inhibitor sacubitrilat as a potent anticancer agent using in-silico protein–ligand interaction profiler (PLIP) analysis, molecular docking, and in vitro studies. The screening of PLIP profiles between vorinostat/panobinostat and HDACs/LTA4H followed by molecular docking resulted in five (Sacubitrilat, B65, BDS, BIR, and NPV) FDA-approved, experimental and investigational drugs. Sacubitrilat has demonstrated promising anticancer activity against colorectal cancer (SW-480) and triple-negative breast cancer (MDA-MB-231) cells, with IC50 values of 14.07 μg/mL and 23.02 μg/mL, respectively. FACS analysis revealed that sacubitrilat arrests the cell cycle at the G0/G1 phase and induces apoptotic-mediated cell death in SW-480 cells. In addition, sacubitrilat inhibited HDAC isoforms at the transcriptomic level by 0.7–0.9 fold and at the proteomic level by 0.5–0.6 fold as compared to the control. Sacubitrilat increased the protein expression of tumor-suppressor (p53) and pro-apoptotic makers (Bax and Bid) by 0.2–2.5 fold while decreasing the expression of anti-apoptotic Bcl2 and Nrf2 proteins by 0.2–0.5 fold with respect to control. The observed cleaved PARP product indicates that sacubitrilat induces apoptotic-mediated cell death. This study may pave the way to identify the anticancer potential of sacubitrilat and can be explored in human clinical trials

    Abstract Triple-negative breast cancer (TNBC) is a metastatic disease and a formidable treatment challenge as it does not respond to existing therapies. Epigenetic regulators play a crucial role in the progression and metastasis by modulating the expression of anti-apoptotic, pro-apoptotic markers and related miRNAs in TNBC cells. We have investigated the anti-TNBC potential of dietary flavonoid ‘Apigenin’ and its combination with Vorinostat on MDA-MB-231 cells. At Apigenin generated ROS, inhibited cell migration, arrested the cell cycle at subG0/G1 phases, and induced apoptotic-mediated cell death. Apigenin reduced the expression of the class-I HDACs at the transcriptomic and proteomic levels. In the immunoblotting study, Apigenin has upregulated pro-apoptotic markers and downregulated anti-apoptotic proteins. Apigenin inhibited the enzymatic activity of HDAC/DNMT and increased HAT activity. Apigenin has manifested its effect on miRNA expression by upregulating the tumor-suppressor miR-200b and downregulation oncomiR-21. Combination study reduced the growth of TNBC cells synergistically by modulating the expression of epigenetic and apoptotic regulators. Molecular docking and MD simulations explored the mechanism of catalytic inhibition of HDAC1 and HDAC3 and supported the in-vitro studies. The overall studies demonstrated an anti-TNBC potential of Apigenin and may help to design an effective strategy to treat metastatic phenotype of TNBC

    Design and Fabrication of Advanced Mechanical Tire Stretcher

    A handheld advanced mechanical tire stretcher can be held between two sidewalls of the worn out tire and activated with circular feed to the handle wheel. A bevel gear pair is set to sequentially provide the required speed reduction, followed by development of torque needed to move the tire upper surface in vertical direction with clockwise direction rotation of the handle wheel. Also due to increase in pitch of power screw working get smoother than previous.  Thus the tire gets stretched in axial direction and compressed in radial direction. Thus tire can be placed in hot retreading mould. The principles embodied in the tire stretcher can be manually powered, or preferably motorized and possibly highly automated.     Response surface based optimization of system variables for liquid chromatographic analysis of candesartan cilexetil

    A statistical optimization method was successfully employed to study the effect of system variables on the chromatographic analysis of candesartan cilexetil. The effect of simultaneously varying the flow rate, temperature and concentration of acetonitrile in the mobile phase in water (0.05% O-phosphoric acid (0.05% OPA)) was studied to optimize the method to obtain excellent chromatographic responses. The optimum conditions were determined with the help of response surface methodology (RSM) using Plackett–Burman designs. From the response surface graphs, the optimum regions were selected to be −1, +1 and +1 for flow rate (0.8 ml/min), temperature (25 °C) and concentration of acetonitrile in water (0.05% OPA) (80%, v/v), respectively. Pareto ranking indicated that the most important variable affecting the selected responses was temperature. Linearity was found in the range of 10 of 50 μg/ml, with a significantly high correlation coefficient (r2 = 0.9989). The limits of detection and quantitation were 0.12 and 0.33 μg/ml, respectively. The developed method was validated for accuracy, precision, linearity, range, and specificity. The method was successfully used to analyze a tablet formulation to assess the chromatographic performance, and it was found to be 99.03%, with a standard deviation of ±0.04
