Evaluation of cellular proteins binding to the transmembrane envelope protein gp41 of HIV-1
Das von HIV ausgelöste erwobene ImmunschwĂ€che-Syndrom (AIDS) stellt auch heute noch ein Problem dar. Im Jahr 2014 waren laut der Weltgesundheitsorganisation (WHO) 36,9 Millionen Menschen mit HIV infiziert. Heute, 30 Jahre nach der Entdeckung von HIV, gibt es zwar gute therapeutische AnsĂ€tze, eine Heilung ist aber bisher nicht in Aussicht. WĂ€hrend der Lebenszyklus von HIV bereits gut verstanden ist, ist noch unklar wodurch eine Immunsuppression in infizierten Menschen ausgelöst wird. Es gibt Hinweise darauf, dass eine hochkonservierte DomĂ€ne des transmembranen HĂŒllproteins gp41 von HIV dabei eine Rolle spielt. Jedoch konnte fĂŒr gp41 bisher kein putativer Rezeptor mit immunmodulierenden Eigenschaften gefunden werden. Im Rahmen dieser Bachelorarbeit sollte die Bindung von synthetischen Peptiden die der immunsuppressiven DomĂ€ne von gp41 entsprechen (âIsu Peptidâ) und rekombinant hergestelltem gp41 an humane PBMCs untersucht werden. Weiterhin sollte die Bindung des Isu Peptids an Subpopulationen von Lymphozyten und Monozyten analysiert und schlieĂlich putative Interaktionspartner durch ImmunprĂ€zipitation isoliert und mittels Massenspektrometrie charakterisiert werden. Es konnte ein spezifische Bindung des Isu Peptids an Monozyten und Lymphozyten festgestellt werden. Die SpezifitĂ€t der Bindung wurde durch den Einsatz eines Peptids mit randomisierter AminosĂ€uresequenz gezeigt. ZusĂ€tzlich konnte die SpezifitĂ€t der Binding an Monozyten mittels kompetitiver Hemmung demonstriert werden. Als Zielzellen des Isu Peptid wurden B Zellen, sowie klassische und intermediĂ€re Monozyten identifizert. Eine Charakterisierung des rekombinanten gp41 ergab, dass es trimerisiert und N-glykolisiert ist und sich in der six-helix bundle Konformation befindet. Weitere Untersuchung zeigten, dass die Bindung des gp41 zu humanen PBMCs schwach, aber konzentrationsabhĂ€ngig ist. AbschlieĂend wurde eine ImmunprĂ€zipitation mit Isu Peptiden und gp41 an humanen PBMCs durchgefĂŒhrt, die Proben wurden mittels Massenspektrometrie analysiert. Yields and Immunomodulatory Effects of Pneumococcal Membrane Vesicles Differ with the Bacterial Growth Phase
Streptococcus pneumoniae infections are a leading cause of death worldwide. Bacterial membrane vesicles (MVs) are promising vaccine candidates because of the antigenic components of their parent microorganisms. Pneumococcal MVs exhibit low toxicity towards several cell lines, but their clinical translation requires a high yield and strong immunogenic effects without compromising immune cell viability. MVs are isolated during either the stationary phase (24 h) or death phase (48 h), and their yields, immunogenicity and cytotoxicity in human primary macrophages and dendritic cells have been investigated. Death-phase vesicles showed higher yields than stationary-phase vesicles. Both vesicle types displayed acceptable compatibility with primary immune cells and several cell lines. Both vesicle types showed comparable uptake and enhanced release of the inflammatory cytokines, tumor necrosis factor and interleukin-6, from human primary immune cells. Proteomic analysis revealed similarities in vesicular immunogenic proteins such as pneumolysin, pneumococcal surface protein A, and IgA1 protease in both vesicle types, but stationary-phase MVs showed significantly lower autolysin levels than death-phase MVs. Although death-phase vesicles produced higher yields, they lacked superiority to stationary-phase vesicles as vaccine candidates owing to their similar antigenic protein cargo and comparable uptake into primary human immune cells
Assessing the economics of large Energy Storage Plants with an optimisation methodology
Power plants, such as wind farms, that harvest renewable energy are increasing their share of the energy portfolio in several countries, including the United Kingdom. Their inability to match demand power profiles is stimulating an increasing need for large ESP (Energy Storage Plants), capable of balancing their instability and shifting power produced during low demand to peak periods. This paper presents and applies an innovative methodology to assess the economics of ESP utilising UK electricity price data, resulting in three key findings. Firstly the paper provides a methodology to assess the trade-off âreserve capacity vs. profitabilityâ and the possibility of establishing the âoptimum size capacityâ. The optimal reserve size capacity maximizing the NPV (Net Present Value) is smaller than the optimum size capacity minimizing the subsidies. This is not an optimal result since it complicates the incentive scheme to align investors and policy makers' interests. Secondly, without subsidies, none of the existing ESP technologies are economically sustainable. However, subsidies are a relatively small percentage of the average price of electricity in UK. Thirdly, the possibility of operating ESP as both as a reserve and do price arbitrage was identified as a mean of decreasing subsidies for the ESP technologies
Prediction of NO emissions for a hydrogen fueled industrial gas turbine combustor with water injection
Hydrogen is a promising alternative to fossil fuels for future gas turbines since it can be produced using renewable energy sources and uses CO-free combustion. However, due to its higher reactivity (when compared to natural gas) it cannot be used with the state-of-the-art premixed combustors developed for natural gas. Besides the risks of flame flashback and auto ignition, high hydrogen fuels tend to produce significantly higher NO emissions due to a the higher flame temperature. This study observes the effects of the use of a non-premixed combustor and water injection, which can reduce emissions. An industrial, non-premixed gas turbine combustor is measured and characterized at real conditions with a high-pressure combustion test rig. The focus of this study is on the NO emissions as a function of the pressure, the hydrogen content in the fuel, and the amount of directly-injected water. A significant increase in NO emissions for increasing pressure and hydrogen share is shown using the experimental combustor geometry. Furthermore, the emissions increase at elevated air inlet temperature and reduced air inlet velocity. Water directly-injected into the combustor significantly reduces the NO emissions. A numerical investigation of the combustion process within the combustor is performed with a semi-empirical chemical reactor network model that derives the flame temperature and residence time. The results show a good agreement with the experimentally determined NO emissions over the entire range of operating conditions. Besides the effect of the flame temperature, a detailed analysis of the chemical effect of hydrogen and directly-injected water on the combustion process shows a significant impact of the relevant radicals and NO formation mechanisms. Furthermore, a simplified NO model is derived based on the findings. With the help of correlations for flame temperature and fluid residence time in the flame, a correlation is developed that predicts the NO emissions taking the substantial chemical effects of hydrogen and water into account. This opens the possibility to estimate the NO emissions at `real' pressure conditions on the basis of low pressure combustion tests

Grundlage der Leistungssportförderung war das Streben der Gemeinschaft sowie jedes Einzelnen nach , dem unsterblich machenden Ruhm. Verdeutlicht wird dies an polisĂŒbergreifenden Leistungssport fördernden Strukturen sowie am Beispiel der griechischen Kolonie Kroton. Mit Blick auf das Thema der Arbeit werden die leistungssportliche ReprĂ€sentation der griechischen Poleis bei den Olympischen Spielen herausgearbeitet (Festgesandtschaften, Siegerzeremonie, Gestaltung der SchatzhĂ€user, Weihegeschenke u. a.) sowie die vielfĂ€ltigen WertschĂ€tzungen eines Olympiasieges fĂŒr den einzelnen Athleten wie fĂŒr dessen Heimatpolis. Sowohl der Polisegoismus als auch das Streben nach unsterblichem Ruhm erweisen sich als zentrale panhellenische Charakteristika. Die Arbeit beruht auf sporthistorischer und altertumswissenschaftlicher Fachliteratur sowie ausgewĂ€hlten PrimĂ€rquellen (Pindar, Pausanias, Plutarch)
