    Evolution of carbon fluxes during initial soil formation along the forefield of Damma glacier, Switzerland

    Soil carbon (C) fluxes, soil respiration and dissolved organic carbon (DOC) leaching were explored along the young Damma glacier forefield chronosequence (7-128years) over a three-year period. To gain insight into the sources of soil CO2 effluxes, radiocarbon signatures of respired CO2 were measured and a vegetation-clipping experiment was performed. Our results showed a clear increase in soil CO2 effluxes with increasing site age from 9±1 to 160±67gCO2-Cm−2year−1, which was linked to soil C accumulation and development of vegetation cover. Seasonal variations of soil respiration were mainly driven by temperature; between 62 and 70% of annual CO2 effluxes were respired during the 4-month long summer season. Sources of soil CO2 effluxes changed along the glacier forefield. For most recently deglaciated sites, radiocarbon-based age estimates indicated ancient C to be the dominant source of soil-respired CO2. At intermediate site age (58-78years), the contribution of new plant-fixed C via rhizosphere respiration amounted up to 90%, while with further soil formation, heterotrophically respired C probably from accumulated ‘older' soil organic carbon (SOC) became increasingly important. In comparison with soil respiration, DOC leaching at 10cm depth was small, but increased similarly from 0.4±0.02 to 7.4±1.6gDOCm−2year−1 over the chronosequence. A strong rise of the ratio of SOC to secondary iron and aluminium oxides strongly suggests that increasing DOC leaching with site age results from a faster increase of the DOC source, SOC, than of the DOC sink, reactive mineral surfaces. Overall, C losses from soil by soil respiration and DOC leaching increased from 9±1 to 70±17 and further to 168±68gCm−2year−1 at the <10, 58-78, and 110-128year old sites. By comparison, total ecosystem C stocks increased from 0.2 to 1.1 and to 3.1kgCm−2 from the young to intermediate and old sites. Therefore, the ecosystem evolved from a dominance of C accumulation in the initial phase to a high throughput system. We suggest that the relatively strong increase in soil C stocks compared to C fluxes is a characteristic feature of initial soil formation on freshly exposed rock

    Separated Oscillatory Fields for High-Precision Penning Trap Mass Spectrometry

    Ramsey's method of separated oscillatory fields is applied to the excitation of the cyclotron motion of short-lived ions in a Penning trap to improve the precision of their measured mass. The theoretical description of the extracted ion-cyclotron-resonance line shape is derived out and its correctness demonstrated experimentally by measuring the mass of the short-lived 38^{38}Ca nuclide with an uncertainty of 1.61081.6\cdot 10^{-8} using the ISOLTRAP Penning trap mass spectrometer at CERN. The mass value of the superallowed beta-emitter 38^{38}Ca is an important contribution for testing the conserved-vector-current hypothesis of the electroweak interaction. It is shown that the Ramsey method applied to mass measurements yields a statistical uncertainty similar to that obtained by the conventional technique ten times faster.Comment: 5 pages, 4 figures, 0 table

    Potent Sensitisation of Cancer Cells to Anticancer Drugs by a Quadruple Mutant of the Human Deoxycytidine Kinase.

    Identifying enzymes that, once introduced in cancer cells, lead to an increased efficiency of treatment constitutes an important goal for biomedical applications. Using an original procedure whereby mutant genes are generated based on the use of conditional lentivector genome mobilisation, we recently described, for the first time, the identification of a human deoxycytidine kinase (dCK) mutant (G12) that sensitises a panel of cancer cell lines to treatment with the dCK analogue gemcitabine. Here, starting from the G12 variant itself, we generated a new library and identified a mutant (M36) that triggers even greater sensitisation to gemcitabine than G12. With respect to G12, M36 presents an additional mutation located in the region that constitutes the interface of the dCK dimer. The simple presence of this mutation halves both the IC50 and the proportion of residual cells resistant to the treatment. Furthermore, the use of vectors with self-inactivating LTRs leads to an increased sensitivity to treatment, a result compatible with a relief of the transcriptional interference exerted by the U3 promoter on the internal promoter that drives the expression of M36. Importantly, a remarkable effect is also observed in treatments with the anticancer compound cytarabine (AraC), for which a 10,000 fold decrease in IC50 occurred. By triggering the sensitisation of various cancer cell types with poor prognosis to two commonly used anticancer compounds M36 is a promising candidate for suicide gene approaches

    The Ramsey method in high-precision mass spectrometry with Penning traps: Experimental results

    The highest precision in direct mass measurements is obtained with Penning trap mass spectrometry. Most experiments use the interconversion of the magnetron and cyclotron motional modes of the stored ion due to excitation by external radiofrequency-quadrupole fields. In this work a new excitation scheme, Ramsey's method of time-separated oscillatory fields, has been successfully tested. It has been shown to reduce significantly the uncertainty in the determination of the cyclotron frequency and thus of the ion mass of interest. The theoretical description of the ion motion excited with Ramsey's method in a Penning trap and subsequently the calculation of the resonance line shapes for different excitation times, pulse structures, and detunings of the quadrupole field has been carried out in a quantum mechanical framework and is discussed in detail in the preceding article in this journal by M. Kretzschmar. Here, the new excitation technique has been applied with the ISOLTRAP mass spectrometer at ISOLDE/CERN for mass measurements on stable as well as short-lived nuclides. The experimental resonances are in agreement with the theoretical predictions and a precision gain close to a factor of four was achieved compared to the use of the conventional excitation technique.Comment: 12 pages, 14 figures, 2 table

    Electronic detection of charged particle effects in a Penning trap

    We present a thorough analysis of the electronic detection of charged particles, confined in a Penning trap, via image charges induced in the trap electrodes. Trapping of charged particles in an electrode structure leads to frequency shifts, which are due to image charge and space charge effects. These effects are of importance for Penning trap experiments which involve high charge densities or require high precision in the motional frequencies. Our analysis of image charges shows that only (higher order) odd powers of the particle displacement lead to induced charge differences, giving rise to a signal. This implies that, besides the centre-of-mass frequency of a trapped particle cloud, also higher order individual particle frequencies induce a signal, which can be picked up by an electronic detection circuit attached to the trap electrodes. We also derive analytic expressions for the image charge and space charge induced frequency shifts and perform simulations of space charge effects. In relation to this, we discuss the consequences of the shifted particle frequencies for resistive cooling of the particle motion.Comment: 16 pages, 4 figure

    Interleukin-6-dependent survival of multiple myeloma cells involves the Stat3-mediated induction of micro-RNA-21 through a highly conserved enhancer

    Signal transducer and activator of transcription 3 (Stat3) is implicated in the pathogenesis of many malignancies and essential for IL-6–dependent survival and growth of multiple myeloma cells. Here, we demonstrate that the gene encoding oncogenic microRNA-21 (miR-21) is controlled by an upstream enhancer containing 2 Stat3 binding sites strictly conserved since the first observed evolutionary appearance of miR-21 and Stat3. MiR-21 induction by IL-6 was strictly Stat3 dependent. Ectopically raising miR-21 expression in myeloma cells in the absence of IL-6 significantly reduced their apoptosis levels. These data provide strong evidence that miR-21 induction contributes to the oncogenic potential of Stat3

    Barriers and opportunities for evidence-based health service planning: the example of developing a Decision Analytic Model to plan services for sexually transmitted infections in the UK

    Decision Analytic Models (DAMs) are established means of evidence-synthesis to differentiate between health interventions. They have mainly been used to inform clinical decisions and health technology assessment at the national level, yet could also inform local health service planning. For this, a DAM must take into account the needs of the local population, but also the needs of those planning its services. Drawing on our experiences from stakeholder consultations, where we presented the potential utility of a DAM for planning local health services for sexually transmitted infections (STIs) in the UK, and the evidence it could use to inform decisions regarding different combinations of service provision, in terms of their costs, cost-effectiveness, and public health outcomes, we discuss the barriers perceived by stakeholders to the use of DAMs to inform service planning for local populations, including (1) a tension between individual and population perspectives; (2) reductionism; and (3) a lack of transparency regarding models, their assumptions, and the motivations of those generating models

    Tau protein, A beta 42 and S-100B protein in cerebrospinal fluid of patients with dementia with Lewy bodies

    The intra vitam diagnosis of dementia with Lewy bodies (DLB) is still based on clinical grounds. So far no technical investigations have been available to support this diagnosis. As for tau protein and beta-amyloid((1-42)) (Abeta42), promising results for the diagnosis of Alzheimer's disease ( AD) have been reported; we evaluated these markers and S-100B protein in cerebrospinal fluid (CSF), using a set of commercially available assays, of 71 patients with DLB, 67 patients with AD and 41 nondemented controls (NDC) for their differential diagnostic relevance. Patients with DLB showed significantly lower tau protein values compared to AD but with a high overlap of values. More prominent differences were observed in the comparison of DLB patients with all three clinical core features and AD patients. Abeta42 levels were decreased in the DLB and AD groups versus NDC, without significant subgroup differences. S-100B levels were not significantly different between the groups. Tau protein levels in CSF may contribute to the clinical distinction between DLB and AD, but the value of the markers is still limited especially due to mixed pathology.
