    The role of nω-nitro-L-arginine in modulation of pulmonary vascular tone in the maturing newborn pig

    AbstractCurrent therapeutic modalities for treatment of newborn pulmonary hypertensive crisis include but are not limited to the administration of nitric oxide (endothelium-derived relaxing factor). However, few data are available on the role of endogenously produced endothelium-derived relaxing factor in the modulation of pulmonary vascular tone in the neonate. In the current study, we investigated the acute effects of Nω-nitro- L -arginine (a potent competitive inhibitor of endothelium-derived relaxing factor synthase) on the pulmonary vasculature of anesthetized open-chest 48-hour-old ( n = 8) and 2-week-old ( n = 7) Yorkshire pigs. After baseline data were acquired, all animals received a 10 mg/kg per minute infusion of Nω-nitro- L -arginine for 10 minutes. To discern distal and proximal pulmonary arterial vessel changes, input mean and characteristic impedance were respectively determined. Pulmonary vascular resistance was also calculated (units determined in dyne • sec • cm -5 plus or minus the standard errorof the mean). Results showed Nω-nitro- L -arginine infusion did not significantly alter baseline pulmonary arterial pressure (22,370 ±1473 dyne/cm2 ), pulmonary vascular resistance (5171 ±805 dyne • sec • cm -5 ), input impedance (6343 ±806 dyne • sec • cm -5 ), or characteristic impedance (2073 ±418 dyne • sec • cm -5 ) in 48-hour-old pigs. In 2-week-old pigs, infusion of Nω-nitro- L -arginine elevated pulmonary arterial pressure (18,162 ±1415 dyne/cm 2 versus 23,838 ±1810 dyne/cm 2 , p = 0.015), pulmonary vascular resistance (810 ±137 dyne • sec • cm -5 versus 1519 dyne • sec • cm -5 , p = 0.030), and input impedance (2302 ±251 dyne • sec • cm -5 versus 2900 ±255 dyne • sec • cm -5 , p = 0.018). Characteristic impedance was not altered in 2-week-old pigs. These data indicate that Nω-nitro- L -arginine infusion resulted in pulmonary arteriolar vasoconstriction in 2-week-old pigs, but not in 48-hour-old pigs. This finding suggests that endothelium-derived relaxing factor does not modulate basal pulmonary arteriolar tone during the early newborn period, but does play a significant role in 2-week-old pigs. These data also suggest that the functional role for endothelium-derived relaxing factor is confined to the distal arteriolar pulmonary bed and does not extend to the larger proximal arterial vessels.     Ecological Invasion, Roughened Fronts, and a Competitor's Extreme Advance: Integrating Stochastic Spatial-Growth Models

    Both community ecology and conservation biology seek further understanding of factors governing the advance of an invasive species. We model biological invasion as an individual-based, stochastic process on a two-dimensional landscape. An ecologically superior invader and a resident species compete for space preemptively. Our general model includes the basic contact process and a variant of the Eden model as special cases. We employ the concept of a "roughened" front to quantify effects of discreteness and stochasticity on invasion; we emphasize the probability distribution of the front-runner's relative position. That is, we analyze the location of the most advanced invader as the extreme deviation about the front's mean position. We find that a class of models with different assumptions about neighborhood interactions exhibit universal characteristics. That is, key features of the invasion dynamics span a class of models, independently of locally detailed demographic rules. Our results integrate theories of invasive spatial growth and generate novel hypotheses linking habitat or landscape size (length of the invading front) to invasion velocity, and to the relative position of the most advanced invader.Comment: The original publication is available at www.springerlink.com/content/8528v8563r7u2742

    First bromine doped cryogenic implosion at the National Ignition Facility

    We report on the first experiment dedicated to the study of nuclear reactions on dopants in a cryogenic capsule at the National Ignition Facility (NIF). This was accomplished using bromine doping in the inner layers of the CH ablator of a capsule identical to that used in the NIF shot N140520. The capsule was doped with 3×\times1016^{16} bromine atoms. The doped capsule shot, N170730, resulted in a DT yield that was 2.6 times lower than the undoped equivalent. The Radiochemical Analysis of Gaseous Samples (RAGS) system was used to collect and detect 79^{79}Kr atoms resulting from energetic deuteron and proton ion reactions on 79^{79}Br. RAGS was also used to detect 13^{13}N produced dominantly by knock-on deuteron reactions on the 12^{12}C in the ablator. High-energy reaction-in-flight neutrons were detected via the 209^{209}Bi(n,4n)206^{206}Bi reaction, using bismuth activation foils located 50 cm outside of the target capsule. The robustness of the RAGS signals suggest that the use of nuclear reactions on dopants as diagnostics is quite feasible

    The high demand for protein during this period of the calf's life is due to the active growth of muscle tissue, and protein is the structural material of all organs. A lack of protein in the diet of calves contributes to a delay in their growth and an excess to the expenditure of additional energy for the deamination of excess amino acids and the elimination of the corresponding decay products through the excretory system of the body. The younger the calves, the higher the protein level in their diet should be. The work aimed to establish the most effective protein norms in the composition of skim milk substitutes for calves over 65 days of age. The study of the influence of feeding skims milk substitute on calves' physiological state and productivity was carried out on four groups of bulls. All tested skim milk replacers varied in protein content but were almost the same in all nutritional parameters. The main ingredients of skim milk substitutes (ZOM 1) for calves of group I were, %: milk proteins – 70, vegetable proteins (soy + wheat) – 29, vitamin and mineral complex, probiotic culture – 1. For calves of group II (ZOM 2) used,%: milk proteins – 70, vegetable proteins (soy + wheat protein) – 29, vitamin-mineral complex – 1. protein) – 29, vitamin and mineral complex – 1. Studies have shown that skim milk substitutes in calves feeding, containing 20 and 22 % protein in the composition of KR-2 compound feed 10 % by weight, was reflected in the improvement of the morpho-biochemical design of the blood. At the same time, there is a tendency to an increase in the concentration of total protein in the blood serum by 3.1 and 3.3 % with a decrease in the amount of urea by 3.5 and 5.2 %, which made it possible to increase the average daily gain in live weight to 3.1 % while reducing costs feed and its price by 1.5 and 0.9 percent.Высокая потребность в протеине в этот период жизни телёнка обусловлена активным ростом мышечной ткани и тем, что белок является структурным материалом всех органов. Недостаток протеина в рационе телят способствует задержке их роста, а избыток – тратам дополнительной энергии на дезаминирование избыточного количества аминокислот и выведение соответствующих продуктов распада через выделительную систему организма. Чем моложе телята, тем выше должен быть уровень протеина в его рационе. Целью работы было установить наиболее эффективные нормы протеина в составе заменителей обезжиренного молока для телят старше 65-дневного возраста. Изучение влияния скармливания заменителя обезжиренного молока на физиологическое состояние и продуктивность телята проведено на 4-х группах бычков. Все опытные заменители обезжиренного молока были различными по содержанию протеина, но практически одинаковыми по всем показателям питательности. Основными ингредиентами заменителей обезжиренного молока (ЗОМ 1) для телят I группы были, %: молочные белки – 70, растительные белки (соевый + пшеничный) – 29, витаминно-минеральный комплекс, пробиотическая культура – 1. Для телят II группы (ЗОМ 2) использовали, %: молочные белки – 70, растительные белки (соевый + пшеничный протеин) – 29, витаминно-минеральный комплекс – 1. В III опытной группе скармливали (ЗОМ 3) состоящий из, %: молочного белка – 70, растительных белков (соевый протеин) – 29, витаминно-минеральный комплекс – 1. Исследованиями установлено, что использование в кормлении телят заменителей обезжиренного молока, содержащих 20 и 22 % протеина в составе комбикорма КР-2 10 % по массе отразилось в улучшении морфо-биохимического состава крови. При этом наблюдается тенденция к повышению концентрации общего белка в сыворотке крови на 3,1 и 3,3 % при снижении количества мочевины на 3,5 и 5,2 %, что позволило увеличить среднесуточный прирост живой массы до 3,1 % при снижении затрат кормов и его себестоимости на 1,5 и 0,9 процента

    Fetal alcohol spectrum disorder (FASD) is an umbrella term used to describe the craniofacial dysmorphic features, malformations, and disturbances in growth, neurodevelopment and behavior occurring in individuals prenatally exposed to alcohol. Fetal alcohol syndrome (FAS) represents the severe end of this spectrum. Many pathophysiological mechanisms have hitherto been proposed to account for the disrupted growth and morphogenesis seen in FAS. These include impaired cholesterol-modification of the Sonic hedgehog morphogen, retinoic acid deficiency, lipoperoxidative damage due to alcohol-induced reactive oxygen species combined with reduced antioxidant defences, and malfunctioning cell adhesion molecules. In this report, we propose a completely novel concept regarding the pathogenesis of FAS. Based on our observation that transferrin isoelectric focusing (TIEF) – the most widely used screening tool for congenital disorders of glycosylation (CDG) – was transiently abnormal in a newborn with FAS and a confirmed maternal history of gestational alcohol abuse, we came to believe that FAS exemplifies a congenital disorder of glycosylation secondary to alcohol-inflicted disruption of (N-linked) protein glycosylation. Various pieces of evidence were found in the literature to substantiate this hypothesis. This observation implies, among others, that one might need to consider the possibility of maternal alcohol consumption in newborns with transient glycosylation abnormalities. We also present an integrated pathophysiological model of FAS, which incorporates all existing theories mentioned above as well as our novel concept. This model highlights the pivotal role of disrupted isoprenoid metabolism in the origination of FAS

    Reproducibility of shear wave elastography measuresof the Achilles tendon.

    OBJECTIVE To assess the reproducibility of shear wave elastography (SWE) measures in the Achilles tendon (AT) in vivo. MATERIALS AND METHODS Shear wave velocity (SWV) of 14 healthy volunteers [7 males, 7 females; mean age 26.5 ± 3.8 years, mean height 171.6 ± 10.9 cm, mean Victorian Institute of Sports Assessment Achilles questionnaire (VISA-A) score 99.4 ± 1.2] was measured with the foot relaxed and fixed at 90°. Data were collected over five consecutive measures and 5 consecutive days. RESULTS Mean SWV values ranged from 7.91 m/s-9.56 m/s ± 0.27-0.50 m/s. Coefficient of variation (CV), correlations and intra-class correlation coefficient (ICC) scores ranged from 2.9%-6.3%, 0.4-0.7 and 0.54-0.85 respectively. No significant differences were noted for longitudinal or transverse data with respect to protocol or time and no significant differences were noted for foot position in transverse data. Significant differences in SWV values were noted between foot positions for longitudinal scanning (p = <0.05), with a relaxed foot position providing SWV values on average 0.47 m/s faster than a fixed position. Increased reproducibility was obtained with the foot relaxed. ICC between operators was 0.70 for transverse and 0.80 for longitudinal scanning. CONCLUSIONS Reproducible SWE measures were obtained over a 1-h period as well as a period of 5 consecutive days with more reliable measures obtained from a longitudinal plane using a relaxed foot position. SWE also has a high level of agreement between operators making SWE a reproducible technique for quantitatively assessing the mechanical properties of the human AT in vivo

    Actinide covalency measured by pulsed electron paramagnetic resonance spectroscopy

    Our knowledge of actinide chemical bonds lags far behind our understanding of the bonding regimes of any other series of elements. This is a major issue given the technological as well as fundamental importance of f-block elements. Some key chemical differences between actinides and lanthanides—and between different actinides—can be ascribed to minor differences in covalency, that is, the degree to which electrons are shared between the f-block element and coordinated ligands. Yet there are almost no direct measures of such covalency for actinides. Here we report the first pulsed electron paramagnetic resonance spectra of actinide compounds. We apply the hyperfine sublevel correlation technique to quantify the electron-spin density at ligand nuclei (via the weak hyperfine interactions) in molecular thorium(III) and uranium(III) species and therefore the extent of covalency. Such information will be important in developing our understanding of the chemical bonding, and therefore the reactivity, of actinides

    Plants growing in constantly changeable environmental conditions are compelled to evolve regulatory mechanisms to cope with biotic and abiotic stresses. Effective defence to invaders is largely connected with phytohormone regulation, resulting in the production of numerous defensive proteins and specialized metabolites. In our work, we elucidated the role of the Abscisic Acid Insensitive 4 (ABI4) transcription factor in the plant response to the two-spotted spider mite (TSSM). This polyphagous mite is one of the most destructive herbivores, which sucks mesophyll cells of numerous crop and wild plants. Compared to the wild-type (Col-0) Arabidopsis thaliana plants, the abi4 mutant demonstrated increased susceptibility to TSSM, reflected as enhanced female fecundity and greater frequency of mite leaf damage after trypan blue staining. Because ABI4 is regarded as an important player in the plastid-to-nucleus retrograde signalling process, we investigated the plastid envelope membrane dynamics using stroma-associated fluorescent marker. Our results indicated a clear increase in the number of stroma-filled tubular structures deriving from the plastid membrane (stromules) in the close proximity of the site of mite leaf damage, highlighting the importance of chloroplast-derived signals in the response to TSSM feeding activity
