Microbial−mammalian cometabolites dominate the age-associated urinary metabolic phenotype in Taiwanese and American populations
Understanding the metabolic processes associated with aging is key to developing effective management and treatment strategies for age-related diseases. We investigated the metabolic profiles associated with age in a Taiwanese and an American population. 1H NMR spectral profiles were generated for urine specimens collected from the Taiwanese Social Environment and Biomarkers of Aging Study (SEBAS; n = 857; age 54–91 years) and the Mid-Life in the USA study (MIDUS II; n = 1148; age 35–86 years). Multivariate and univariate linear projection methods revealed some common age-related characteristics in urinary metabolite profiles in the American and Taiwanese populations, as well as some distinctive features. In both cases, two metabolites—4-cresyl sulfate (4CS) and phenylacetylglutamine (PAG)—were positively associated with age. In addition, creatine and β-hydroxy-β-methylbutyrate (HMB) were negatively correlated with age in both populations (p < 4 × 10–6). These age-associated gradients in creatine and HMB reflect decreasing muscle mass with age. The systematic increase in PAG and 4CS was confirmed using ultraperformance liquid chromatography–mass spectrometry (UPLC–MS). Both are products of concerted microbial–mammalian host cometabolism and indicate an age-related association with the balance of host–microbiome metabolism
Metabolic profiling of biological fluids and tissues in pre-clinical toxicological studies
Metabonomics involves the application of advanced analytical tools (NMR, MS) to profile the diverse of biological fluids or tissue. Multivariate statistical analysis of the complex spectral profiles enables discrimination between phenotypes of interest and identifies discriminatory metabolites that represent candidate biomarkers. This thesis aims to evaluate metabolic profiling UPLC-MS approach as a tool in pre-clinical toxicological studies. In order to address this aim UPLC-MS was used to investigate the metabolic effects of the hepatotoxin galactosamine (galN) and the mechanism by which glycine protects against such toxicity. Histopathological and clinical chemistry analyses confirmed the protective effect of glycine and revealed interindividual variability in the toxic response at the cohort of rats treated with galN enabling the classification of ‘responder’ and ‘non-responder’ phenotypes.In order to enhance the metabolome coverage a hydrophilic interaction chromatography (HILIC) UPLC-MS method for explaratory urinary metabolic profiling was developed and can be used complementary to the traditionally used reversed-phese (RP) UPLC-MS method. Moreover an efficient and reproducible protocol was developed and optimized for fecal untargeted metabolic profiling by UPLC-MS. In both methods the within-run repeatability and stability over large sample sets was assessed through the repeat analysis of a quality control sample. Then the metabolic profiles of urine, serum, feces and liver tissue were determined in the studies of the hepatotoxic action of galN using both RP and HILIC UPLC-MS methods in positive and negative electrospray (ESI) modes. The data revealed excellent classification of biofluids and tissues metabolic profiles according to treatment and inter-animal variability in response to galN with significant changes observed in specific endogenous metabolites after galN exposure. This metabonomic approach enabled simultaneous characterization of the xenobiotic and endogenous metabolic pertubations at the toxicity study of galN.Τα τελευταία χρόνια η αλματώδης ανάπτυξη των φασματοσκοπικών τεχνικών και της υπολογιστικής ισχύος έχει κάνει εφικτή την ολιστική προσέγγιση των βιολογικών δειγμάτων μέσω της μεταβονομικής/ μεταβολομικής (metabonomics/ metabolomics). Η μεταβονομική είναι ένα νέο διεπιστημονικό πεδίο που εφαρμόζει υψηλής-αποδοτικότητας (high-throughput) τεχνολογίες στον προσδιορισμό μικρού μοριακού βάρους βιομορίων που εμπλέκονται σε διάφορα βιοχημικά μονοπάτια. Με την εφαρμογή της μεταβονομικής στην τοξικολογική ανάλυση πραγματοποιείται διερεύνηση του συνόλου των ενδογενών μεταβολιτών ενός βιολογικού δείγματος (μεταβολικό προφίλ) με απώτερο σκοπό τον εντοπισμό αλλαγών στον οργανισμό που προκαλούνται από την έκθεση του οργανισμού στην τοξική ουσία ή φάρμακο. Το αντικείμενο της διατριβής εντάσσεται στο σύγχρονο ερευνητικό πεδίο της μεταβονομικής στην τοξικολογία. Στόχος μας ήταν η ανάπτυξη μεθόδων ανάλυσης ολικού μεταβολικού προφίλ χρησιμοποιώντας την αναλυτική τεχνική της υγρής χρωματογραφίας υπερ-υψηλής απόδοσης σε συνδυασμό με φασματομετρία μαζών (UPLC-MS) και η εφαρμογή αυτών των μεθόδων στη μελέτη της τοξικής δράσης της γαλακτοζαμίνης (galN) για την περαιτέρω κατανόηση του μηχανισμού ηπατοξικότητας που προκαλεί. Αρχικά αναπτύχθηκε μέθοδος HILIC-UPLC-MS για την ανάλυση μεταβολικού προφίλ ούρων, η οποία μπορεί να ενισχύσει την ολιστική προσέγγιση του προφίλ μεταβολιτών στα υπό εξέταση δείγματα και μπορεί να εφαρμοστεί ως συμπληρωματική τεχνική της RP-UPLC-MS αυξάνοντας τον αριθμό των μεταβολιτών που μπορούν να προσδιοριστούν σε ένα βιολογικό δείγμα. Στη συνέχεια, αναπτύχθηκε μέθοδος προκατεργασίας δειγμάτων κοπράνων για την ανάλυση μεταβολικού προφίλ η οποία αξιολογήθηκε όσον αφορά την επαναληψιμότητά της και την αξιοπιστία της. Τέλος, οι μέθοδοι αυτές χρησιμοποιήθηκαν στις πειραματικές μελέτες του μηχανισμού ηπατοξικότητας της γαλακτοζαμίνης. Η αξιολόγηση των δεδομένων φανέρωσε υψηλή σταθερότητα του συστήματος UPLC-MS που χρησιμοποιήθηκε, καθ’ όλη τη διάρκεια των πειραμάτων, δίνοντας έτσι δεδομένα άριστης ποιότητας για την περαιτέρω στατιστική ανάλυση.Στην πρώτη πειραματική μελέτη χορηγήθηκε galN σε επίμυες και μελετήσαμε τις αλλαγές στα μεταβολικά προφίλ των βιολογικών δειγμάτων ούρων, ορού αίματος, ήπατος και κοπράνων. Στη δεύτερη μελέτη χορηγήθηκε ταυτόχρονα galN και γλυκίνη σε επίμυες και μελετήθηκε η προστατευτική δράση της γλυκίνης όταν χορηγείται σε συνδυασμό με τη galN. Τα αποτελέσματα από την ανάλυση των βιολογικών δειγμάτων και στις δύο πειραματικές μελέτες κατέδειξαν ότι τα μεταβολικά προφίλ αυτών διαφέρουν σημαντικά λόγω της ανάπτυξης ηπατοτοξικότητας στους επίμυες. Τα αποτελέσματα αυτά συμφωνούν με τα αποτελέσματα των ιστοπαθολογικών εξετάσεων και των αναλύσεων κλινικής χημείας. Η ανταπόκριση κάθε πειραματόζωου στην galN ποικίλει κατατάσσοντας τους επίμυες σε responders και non-responders. Τα μεταβολικά προφίλ των δειγμάτων της ομάδας ελέγχου βρέθηκαν να διαφέρουν από αυτά των πειραματόζωων που χαρακτηρίζονται ως non-responders. Συγκρίνοντας την ομάδα επίμυων στους οποίους χορηγήθηκε ταυτόχρονα galN+gly με την ομάδα ελέγχου δεν βρέθηκαν στατιστικώς σημαντικές διαφορές στα μεταβολικά προφίλ των βιολογικών τους δειγμάτων. Η ολιστική ανάλυση μεταβολικού προφίλ κατέδειξε διαφορές σε ενδογενείς μεταβολίτες που εμπλέκονται σε διάφορα σημαντικά μεταβολικά μονοπάτια. Η τάση αύξησης ή μείωσης των μεταβολιτών αυτών αναστέλλεται όταν χορηγείται ταυτόχρονα gly με την galN. Συγκεκριμένα, τα αποτελέσματα έδειξαν ότι: Η απέκκριση της glcNAc, της galN και των πυραζινών της σχετίζονται με το βαθμό της ηπατικής βλάβης των επίμυων που ανταποκρίθηκαν στην τοξικότητα και αποτελούν δείκτες εμφάνισης ηπατοτοξικότητας της galN. Επίσης, η ανάλυση ολικού μεταβολικού προφίλ ούρων, ήπατος και ορού αίματος κατέδειξε διαταραχή στη βιοσύνθεση και στο μεταβολισμό των αμινοξέων στο ήπαρ, στη βιοσύνθεση των πυριμιδινών και των πουρινών, στο μεταβολικό μονοπάτι της μεθειονίνης και της ιστιδίνης, στη μεταβολική δραστηριότητα της εντερικής μικροβιακής χλωρίδας, στα επίπεδα των ενδιάμεσων προϊόντων του κύκλου του Krebs και στην παραγωγή των χολικών οξέων. Τέλος, τα αποτελέσματα που προέκυψαν από την ολιστική ανάλυση των δειγμάτων ούρων ανέδειξαν αυξημένα επίπεδα οροτικού οξέος και ουριδίνης στην ομάδα χορήγησης γλυκίνης σε σχέση με την ομάδα ελέγχου. Τα αυξημένα επίπεδα οροτικού οξέος συνηγορούν στην αυξημένη παραγωγή UMP και κατ’ επέκταση αύξηση των μορίων που αποτελούν δότες ουριδίνης γεγονός που συμβάλει στο μηχανισμό προστασίας από την εμφάνιση τοξικότητας. Τα αποτελέσματα των πειραματικών μελετών επιβεβαιώνουν και ενισχύουν τα ευρήματα των προηγούμενων μελετών ανάλυσης ολικού μεταβολικού προφίλ στη μελέτη της ηπατοτοξίνης galN με την τεχνική NMR. Με την παρούσα διατριβή βελτιώνεται η γνώση μας όσον αφορά τις αλλαγές που υφίστανται στα μεταβολικά προφίλ των βιολογικών δειγμάτων επίμυων που έχουν αναπτύξει ηπατοτοξικότητα εξαιτίας της χαρήγησης galN σε σχέση με την ομάδα ελέγχου. Τα αποτελέσματα καταδεικνύουν τη σημαντικότητα της εφαρμογής πολλαπλών αναλυτικών τεχνικών (π.χ. NMR και MS) για τη συλλογή όσο το δυνατόν περισσότερης πληροφορίας στην ανάλυση μεταβολικού προφίλ

The same reactants in a copper-catalyzed reaction afford mainly the corresponding N-arylo compounds. Arylhydrazines are mainly oxidized by the ylide and arylation occurs only in a low yield
Non-conditioned solid-phase extraction of amphetamine from post-mortem specimens
Solid-phase extraction (SPE) procedures are being used increasingly in all types of drug testing laboratories, including those, which carry out forensic toxicological analysis. Extraction techniques are used to isolate chemical substances from biological and post-mortem substrates in order to be identified by chromatographic methods. The advantages of such procedures include decreased operation time, reduced solvent volumes and increased extraction efficiency.The present work describes a novel and faster SPE technique, without column preconditioning, for the extraction of amphetamine from spiked biological and post-mortem specimens. Abselut NEXUS columns were used for the extraction from spiked blood, bile, vitreous humour, pericardial fluid, cerebrospinal fluid, liver, brain, bone and bone marrow.The analysis of amphetamine was accomplished by gas chromatography after derivatisation with heptafluorobutyric anhydride (HFBA). The recoveries of amphetamine ranged from 78.5% (pericardial fluid) to 87.5% (bile), with relative standard deviations less than 8.0
Immunoassay thechnologies for drugs of abuse testing - General principles - Recognized advantages and disadvantages
The multiple application of immunoassay technology as the «initial test» to screen for drug abuse in various biological fluids and forensic matrices is well established and regularly practiced. Immunoassay methods refer to extremely convenient and quick analytical systems that rely on specific antigen antibody reactions for drug monitoring analysis. The evolution of development and commercialization of these technologies have significantly contributed to the overall efforts in deterring and detecting the abuse of illicit substances and the misuse of various prescription medications. Generally speaking, immunoassays serve as an economic and efficient screening tool to eliminate negative specimens from further analysis and to identify the class of drugs that requires the second confirmatory step.The present article provides an overview of the fundamental principles of immunoassay methods, their general characteristics, specific limitations, various and recognized advantages and disadvantages. It describes clearly the most commonly used immunoassays for drug abuse screening, such as EMIT, RIA, FPIA and CEDIA
Workplace drug testing in Northern Greece during the period 2000-2006
The aim of the present study is to present information concerning Workplace Drug Testing in Northern Greece for the period of 2000-2006, which is in continuation of previous testing. Cases included: professional drivers, individuals at pre-employment stage for security services, prostitute houses and housekeepers at prostitute houses. During the period 2000 - 2006, we handled a total of 2665 cases, each of which was examined for the presence of 5 groups of controlled substances (cannabinoids, opiates, benzodiazepines, cocaine and amphetamines). Screening was performed with Enzyme Multiplied Immunoassay Technique (EMIT), while verification of positive results and identification of substances present was achieved through chromatographic techniques.Of the 2665 cases handled in this period of 7 years, 67% concerned candidates for employment at security services, 18% concerned professional drivers, 13% concerned individuals pursuing employment at prostitute houses and 2% concerned individuals pursuing employment as housekeepers at prostitute houses.Of the total 2665 samples tested, 105 samples were positive, corresponding to 4% of the total cases. Of the controlled substances abused, cannabinoids seem to be the most popular abused drug representing 72.81% of the positive samples. Opiate abuse corresponds to 16.27% of the positive samples, cocaine abuse corresponds to 5.19% of the positive samples and benzodiazepine abuse corresponds to 5.73% of the positive samples. Finally, we noticed no cases of amphetamine abuse

Optimizing the Use of Quality Control Samples for Signal Drift Correction in Large-Scale Urine Metabolic Profiling Studies
The evident importance of metabolic profiling for biomarker
discovery
and hypothesis generation has led to interest in incorporating this
technique into large-scale studies, e.g., clinical and molecular phenotyping
studies. Nevertheless, these lengthy studies mandate the use of analytical
methods with proven reproducibility. An integrated experimental plan
for LC–MS profiling of urine, involving sample sequence design
and postacquisition correction routines, has been developed. This
plan is based on the optimization of the frequency of analyzing identical
quality control (QC) specimen injections and using the QC intensities
of each metabolite feature to construct a correction trace for all
the samples. The QC-based methods were tested against other current
correction practices, such as total intensity normalization. The evaluation
was based on the reproducibility obtained from technical replicates
of 46 samples and showed the feature-based signal correction (FBSC)
methods to be superior to other methods, resulting in ∼1000
and 600 metabolite features with coefficient of variation (CV) <
15% within and between two blocks, respectively. Additionally, the
required frequency of QC sample injection was investigated and the
best signal correction results were achieved with at least one QC
injection every 2 h of urine sample injections (<i>n</i> = 10). Higher rates of QC injections (1 QC/h) resulted in slightly
better correction but at the expense of longer total analysis time
