    Evaluation of the epidemiological importance of classical swine fever infected, E2 sub-unit marker vaccinated animals with RT-nPCR positive blood samples

    &lt;p&gt;It has been demonstrated that pigs that have been double vaccinated with an E2 sub-unit marker vaccine and that are infected with classical swine fever virus (CSFV) through a natural contact infection may react positive in a CSFV detecting RT-nPCR test, whereas no virus could be isolated by using the conventional virus isolation (VI) technique. To evaluate whether these vaccinated and infected pigs may spread the virus, three experiments were set up. In the first, susceptible pigs were inoculated with serum originating from vaccinated RT-nPCR positive pigs. In the second, vaccinated RT-nPCR positive pigs were brought into contact with sentinel animals. In the third, vertical transmission was evaluated in RT-nPCR positive vaccinated pregnant gilts. In the first two experiments, no proof of virus transmission was found, whereas in the third vertical transmission was observed. The conclusion is that in vaccinated pigs that are positive in RT-nPCR but negative in VI, the level of circulating virus is probably not high enough for horizontal transmission, whereas vertical transmission of the virus is possible.&lt;/p&gt;</p

    STRUCTURING OF ELECTRODE SURFACES WITH LIGAND-FREE NANOPARTICLES VIA ELECTROPHORETIC DEPOSITION- FUNDAMENTALS AND IN VIVO APPLICATIONS

    Electrodes for neural stimulation and recording are highly relevant in modern medicine, e.g. for the treatment of movement disorders. As these electrodes have to be implanted directly into the patient´s brain, impaired biocompatibility as well as reduced performance due to increased impedance upon tissue contact are serious problems. Strategies to improve the efficiency of electrodes entail the implementation of defined nanoscopic structures to the electrode surface, which increase the surface area and improve the current flow by possible edge effects1. In this context electrophoretic deposition (EPD) of nanoparticles (NP) constitutes an efficient and feasible way for surface structuring as in contrast to e.g. ablative laser machining, electric field lines are naturally ordered perpendicular to the implant´s surface, so that electrophoretic deposition is well compatible to shaped implants and curved surfaces. In this work an EPD process for the structuring of Pt electrode surfaces with NP is systematically investigated. Reference NP from a modern synthesis route named pulsed laser ablation in liquids (PLAL)2 are utilized as they possess a high surface charge density in order to ease their movement in an electric field. The electrophoretic velocity of these NP was examined and found to be linearly-correlated with the electric field strength, while the slope is dictated by the NP´s surface charge density (zeta-potential).3, 4 On the other hand the PLAL-generated NP are, by design, completely free of organic ligand, which significantly affected their deposition in an EPD setup. It was found that the deposited mass linearly increased with process time, yielding a well scalable process, while on the other hand control experiments with ligands showed a saturation of the deposited mass due to electrochemical shielding of the surface by charged ligands.4 It was furthermore demonstrated that the EPD process with ligand-free NP could also be done in a continuous flow-through setup suitable for the parallel structuring of multiple electrodes.5 Interestingly, the deposition velocity was not size dependent as particle size distributions prior to and after EPD were identical.5 In consecutive experiments, the surface properties like coverage, oxidation, wettability6 and impedance of the electrode materials were evaluated and correlated with the EPD process parameters electric field strength, colloid concentration and deposition time. As a result a detailed map was obtained, which allows a defined tuning of Pt surface properties by Pt NP EPD. Finally, the impedance of electrodes coated with ligand-free Pt NP were evaluated in long term stimulation experiments with rats. The NP coating could stabilize the impedance of the electrodes in vivo, while it continuously increased in non-coated controls.7 Furthermore, the coated electrodes exhibited excellent biocompatibility similar to the controls7 while no significant NP desorption from the surface was found upon mechanical tear. 1.     Comparative studies on the pathogenicity and tissue distribution of three virulence variants of classical swine fever virus, two field isolates and one vaccine strain, with special regard to immunohistochemical investigations

    <p>Abstract</p> <p>Background</p> <p>The aim of this study was to compare the tissue distribution and pathogenicity of three virulence variants of classical swine fever virus (CSFV) and to investigate the applicability of various conventional diagnostic procedures.</p> <p>Methods</p> <p>64 pigs were divided into three groups and infected with the highly virulent isolate ISS/60, the moderately virulent isolate Wingene'93 and the live attenuated vaccine strain Riems, respectively. Clinical signs, gross and histopathological changes were compared in relation to time elapsed post infection. Virus spread in various organs was followed by virus isolation, by immunohistochemistry, applying monoclonal antibodies in a two-step method and by <it>in situ </it>hybridisation using a digoxigenin-labelled riboprobe.</p> <p>Results</p> <p>The tissue distribution data are discussed in details, analyzing the results of the various diagnostic approaches. The comparative studies revealed remarkable differences in the onset of clinical signs as well as in the development of the macro- and microscopical changes, and in the tissue distribution of CSFV in the three experimental groups.</p> <p>Conclusion</p> <p>The present study demonstrates that in the case of highly and moderately virulent virus variants the virulence does not affect the pattern of the viral spread, however, it influences the outcome, the duration and the intensity of the disease. Immunohistochemistry has the advantage to allow the rapid detection and localisation of the virus, especially in cases of early infection, when clinical signs are still absent. Compared to virus isolation, the advantage of this method is that no cell culture facilities are required. Thus, immunohistochemistry provides simple and sensitive tools for the prompt detection of newly emerging variants of CSFV, including the viruses of very mild virulence.</p

    A qualitative phenomenological study was conducted to identify and describe the phenomenon of corruption psychology. Two corruptors were interviewed to explore their perceptions on corrupt practices in Central Java. The interview conducted to explore informant perceptions on a real process. Interview data collected resulted in five themes: (1) Corruption is an act of abuse the authority, identical with theft, something that not run correctly, and using public money for personal and group interest intentionally; (2) The motives of the informants in doing corruption are solidarity with the friends\u27 doer, system that enables to corrupt, to earn much more money, and make friends; (3) Process of corruption; budget-making has been done by legislative and executive institution; marking-up the budget, facilities and allowances; reporting the administrative data manipulatively; inter-relating chain in corruption process; and distributing the aspiration fund without a proof of receipts; (4) The impact of corruption is making someone\u27s wiser in life, putting the corruptors in to the jail, humiliating their big family, and also, having a more debt, and (5) The settlement of problems that they employ is by using emotion-focused coping.     Scientific Opinion on peste des petits ruminants

    Peste des petits ruminants (PPR) is a severe viral disease of small ruminants caused by a Morbillivirus closely related to rinderpest virus. It is widespread in Africa and Asia and is currently also found in Turkey and Northern Africa. PPR is transmitted via direct contact, and the disease would mainly be transferred to infection-free areas by transport of infected animals. In the EU, it could only happen through illegal transport of animals. The risk of that depends on the prevalence in the country of origin and the number of animals illegally moved. The extent of the spread would depend mainly on the time during which it is undetected, the farm density, the frequency and distance of travel of animals. PPR has a high within-herd transmission rate, therefore contacts between flocks, e.g. through common grazing areas, should be avoided when PPR is present. If PPR enters EU areas with dense sheep population but low goat density, it may spread rapidly undetected, since goats are considered more susceptible than sheep. Effective measures in limiting the spread of PPR in the EU include prompt culling of infected herds, rapid detection, movement restriction, and disinfection. Live attenuated vaccines against PPR are available, safe and effective, and have been successfully used to control PPR epidemics, but no method exists for differentiating between infected and vaccinated animals; therefore, the development of one is recommended. Awareness-raising campaigns for farmers and veterinary staff to promote recognition of the disease should be considered. The cooperation of the EU with neighbouring countries should be encouraged to prevent the spread 20 of PPR and other transboundary diseases

    Blood-Based Immune Profiling Combined with Machine Learning Discriminates Psoriatic Arthritis from Psoriasis Patients

    Psoriasis (Pso) is a chronic inflammatory skin disease, and up to 30% of Pso patients develop psoriatic arthritis (PsA), which can lead to irreversible joint damage. Early detection of PsA in Pso patients is crucial for timely treatment but difficult for dermatologists to implement. We, therefore, aimed to find disease-specific immune profiles, discriminating Pso from PsA patients, possibly facilitating the correct identification of Pso patients in need of referral to a rheumatology clinic. The phenotypes of peripheral blood immune cells of consecutive Pso and PsA patients were analyzed, and disease-specific immune profiles were identified via a machine learning approach. This approach resulted in a random forest classification model capable of distinguishing PsA from Pso (mean AUC = 0.95). Key PsA-classifying cell subsets selected included increased proportions of differentiated CD4+CD196+CD183-CD194+ and CD4+CD196-CD183-CD194+ T-cells and reduced proportions of CD196+ and CD197+ monocytes, memory CD4+ and CD8+ T-cell subsets and CD4+ regulatory T-cells. Within PsA, joint scores showed an association with memory CD8+CD45RA-CD197- effector T-cells and CD197+ monocytes. To conclude, through the integration of in-depth flow cytometry and machine learning, we identified an immune cell profile discriminating PsA from Pso. This immune profile may aid in timely diagnosing PsA in Pso
