    Sampling Plan for Lab Extracted Beverages Data with Multiple Dependent States Using the Odd Log-Logistic Distribution

    In this paper, we propose a Multiple Dependent State Sampling (MDSS) plan for decision-making on lot acceptance/rejection based on properties of current and preceding lots sampled. The plan uses the Odd Log-Logistic Generalized Exponentiated (OLLGE) distribution to determine the median life of the product through a truncated-time life test. Optimal parameters such as sample size, acceptance/rejection numbers, and preceding lots are obtained using the operating characteristic curve (OC-curve). The plan’s performance is compared to that of single sampling (SS) plans using Lab Extracted Beverages data sets on carbon dioxide pressure (MPa)

    BACKGROUND / AIMS : Kisspeptin is the major excitatory regulator of gonadotropin-releasing hormone (GnRH) neurons and is responsible for basal GnRH/LH release and the GnRH/LH surge. Although it is widely assumed, based on mutations in kisspeptin and Kiss1R, that kisspeptin acts to sustain basal GnRH neuronal activity, there have been no studies to investigate whether endogenous basal kisspeptin tone plays a direct role in basal spontaneous GnRH neuronal excitability. It is also of interest to examine possible interactions between endogenous kisspeptin tone and other neuropeptides that have direct effects on GnRH neurons, such as neuropeptide Y (NPY) or cocaine- and amphetamine-regulated transcript (CART), since the activity of all these neuropeptides changes during states of negative energy balance. METHODS : Loose cell-attached and whole-cell current patch-clamp recordings were made from GnRH-GFP neurons in hypothalamic slices from female and male rats. RESULTS : Kisspeptin activated GnRH neurons in a concentration-dependent manner with an EC 50 of 3.32 ± 0.02 n M . Surprisingly, a kisspeptin an-endogenous kisspeptin tone. Furthermore, inhibition of endogenous kisspeptin tone blocked the direct activation of GnRH cells that occurs in response to antagonism of NPY Y5 receptor or by CART. CONCLUSIONS : Our electrophysiology studies suggest that basal endogenous kisspeptin tone is not only essential for spontaneous GnRH neuronal firing, but it is also required for the net excitatory effects of other neuropeptides, such as CART or NPY antagonism, on GnRH neurons.     Early High-Fat Diet Exposure Causes Dysregulation of the Orexin and Dopamine Neuronal Populations in Nonhuman Primates

    Maternal obesity and consumption of a high-fat diet (HFD) during pregnancy has a negative impact on offspring, including an increased risk for the development of obesity in adolescence. The mechanism for this transferred metabolic risk is unclear, but many studies have focused on the brain due to its important role in appetite and body-weight regulation. Two main pathways regulate appetite in the brain; homeostatic regulation that occurs predominantly in hypothalamic circuits and hedonic regulation of feeding that occurs via dopaminergic pathways. The current proposal examined the impact of early HFD exposure on the dopaminergic control of hedonic feeding pathways in a translational nonhuman primate model. Japanese macaque offspring from mothers consuming a control (CTR) or HFD were weaned onto control or HFD at an average 8 months of age yielding four groups: maternal and post-weaning control diet (mCTRpCTR), maternal control diet and post-weaning HFD (mCTRpHFD), maternal HFD and post-weaning control diet (mHFDpCTR) and maternal and post-weaning HFD (mHFDpHFD). Brains from 13-month-old offspring were evaluated for expression of neuropeptides that regulate dopaminergic pathways including orexin, melanin-concentrating hormone (MCH) in the lateral hypothalamus (LH), and tyrosine hydroxylase expression in the ventral tegmental area (VTA). Orexin cell numbers in the LH were significantly increased in animals exposed to a post-weaning HFD, while no difference was observed for orexin mRNA content or MCH cell numbers. Orexin fiber projections to the rostral VTA were significantly reduced in mCTRpHFD, mHFDpCTR, and mHFDpHFD groups, but these differences were not significant in the caudal VTA. There was no difference in the percentage of dopamine neurons receiving close appositions from orexin fibers in either the rostral or caudal VTA, nor was there any difference between groups in the number of orexin contacts per TH cell. In conclusion, the current study finds that prolonged early exposure to HFD during the in utero and postnatal period causes alterations at several levels in the dopaminergic circuits regulating reward

    Cannabinoid modulation of limbic forebrain noradrenergic circuitry

    Both the endocannabinoid and noradrenergic systems have been implicated in neuropsychiatric disorders. Importantly, low levels of norepinephrine are seen in patients with depression, and antagonism of the cannabinoid receptor type 1 (CB1R) is able to induce depressive symptoms in rodents and humans. Whether the interaction between the two systems is important for the regulation of these behaviors is not known. In the present study, adult male Sprague-Dawley rats were acutely or chronically administered the CB1R synthetic agonist WIN 55,212-2, and alpha2A and beta1 adrenergic receptors (AR) were quantified by Western blot. These AR have been shown to be altered in a number of psychiatric disorders and following antidepressant treatment. CB1R agonist treatment induced a differential decrease in alpha2A- and beta1-ARs in the nucleus accumbens (Acb). Moreover, to assess long-lasting changes induced by CB1R activation, some of the chronically treated rats were killed 7 days following the last injection. This revealed a persistent effect on alpha2A-AR levels. Furthermore, the localization of CB1R with respect to noradrenergic profiles was assessed in the Acb and in the nucleus of the solitary tract (NTS). Our results show a significant topographic distribution of CB1R and dopamine beta hydroxylase immunoreactivities (ir) in the Acb, with higher co-localization observed in the NTS. In the Acb, CB1R-ir was found in terminals forming either symmetric or asymmetric synapses. These results suggest that cannabinoids may modulate noradrenergic signaling in the Acb, directly by acting on noradrenergic neurons in the NTS or indirectly by modulating inhibitory and excitatory input in the Acb.This works was supported by PHS grant DA 020129.     Repetitive sampling inspection plan for cancer patients using exponentiated half-logistic distribution under indeterminacy

    Abstract This piece of work deals with a time truncated sampling scheme for cancer patients using exponentiated half-logistic distribution (EHLD) based on indeterminacy. We have studied time truncated schemes like repetitive acceptance sampling plan (RASP) under indeterminacy. We have estimated the projected scheme parameters such as sample size and acceptance and rejection sample numbers for known indeterminacy parameters. In addition to the projected sampling scheme quantities, the corresponding tables are generated for various values of indeterminacy parameters. The results of a sampling scheme show that the average sample number (ASN) decreases as indeterminacy values increase. It leads that the indeterminacy parameter is played a crucial portrayal in ASN. A comparative study is carried out with existing sampling schemes based on indeterminacy and classical sampling schemes. The evaluated sampling schemes are exemplified with the help of cancer data. From tables and exemplification, we wind up that the projected RSP scheme under indeterminacy desired a smaller sample size than the existing schemes

    Testing average traffic fatality using sampling plan for exponentiated half logistic distribution under indeterminacy

    A time-truncated sampling plan for the exponentiated half logistic distribution under the indeterminacy is developed in the present investigation. The proposed design parameters are ascertained by fixing the indeterminacy parameter with the known shape parameter. For different values of indeterminacy parameters at known shape values, the parameters of the proposed plan are determined. The results show that the indeterminacy parameter influences the sample size of the proposed design for exponentiated half-logistic distribution. The results clearly indicate that while indeterminacy parameters increase, the sample size reduces. The relevance of the designed plan is established using data set on traffic fatality on roads in the United States. From the traffic fatality illustration, it is resolved that the proposed plan is helpful to test the aggregate traffic fatality at more modest values of sample size as analogized to the available classical sampling plan

    Application of the multiple dependent state sampling strategy to late adolescent suicide rates

    Abstract A multiple dependent state sampling plan (MDSSP) is designed when the lifetimes of the variables follow New Lomax Rayleigh Distribution (NLRD). The decision to accept or reject a lot in the proposed methodology is based on the quality of the given present or previous lots. A binomial model-based operating characteristic curve (OC curve) for continuous lots of variables under similar settings in healthcare is used in finding the probability of acceptance, acceptance number, rejection number, and the number of preceding (succeeding) lots to consider. Time truncated life test based on the specified median of the NLRD is used in designing the current acceptance sampling plan. For specified values of the parameters of NLRD, quantile ratios, consumer’s risk and producer’s risk, average sampling number (ASN) and probability of acceptance of a lot are reported in tables. Real data on worldwide suicide rates of 15–19 years in the year 2019 from the World Health Organization (WHO) website is considered to illustrate this methodology. The minimum sample size required from the selected data to comment on worldwide suicide rates in late adolescents is explained with MDSSP.
