    Optimal Inapproximability Results for MAX-CUT and Other 2-Variable CSPs?

    In this paper we show a reduction from the Unique Games problem to the problem of approximating MAX-CUT to within a factor of αGW + ∈, for all ∈ \u3e 0; here αGW ≈ .878567 denotes the approximation ratio achieved by the Goemans-Williamson algorithm [26]. This implies that if the Unique Games Conjecture of Khot [37] holds then the Goemans-Williamson approximation algorithm is optimal. Our result indicates that the geometric nature of the Goemans-Williamson algorithm might be intrinsic to the MAX-CUT problem. Our reduction relies on a theorem we call Majority Is Stablest. This was introduced as a conjecture in the original version of this paper, and was subsequently confirmed in [45]. A stronger version of this conjecture called Plurality Is Stablest is still open, although [45] contains a proof of an asymptotic version of it. Our techniques extend to several other two-variable constraint satisfaction problems. In particular, subject to the Unique Games Conjecture, we show tight or nearly tight hardness results for MAX-2SAT, MAX-q-CUT, and MAX-2LIN(q). For MAX-2SAT we show approximation hardness up to a factor of roughly .943. This nearly matches the .940 approximation algorithm of Lewin, Livnat, and Zwick [41]. Furthermore, we show that our .943... factor is actually tight for a slightly restricted version of MAX-2SAT. For MAX-q-CUT we show a hardness factor which asymptotically (for large q) matches the approximation factor achieved by Frieze and Jerrum [25], namely 1 − 1/q + 2(ln q)/q2 . For MAX-2LIN(q) we show hardness of distinguishing between instances which are (1−∈)-satisfiable and those which are not even, roughly, (q−∈/2)-satisfiable. These parameters almost match those achieved by the recent algorithm of Charikar, Makarychev, and Makarychev [10]. The hardness result holds even for instances in which all equations are of the form xi − xj = c. At a more qualitative level, this result also implies that 1 − ∈ vs. ∈ hardness for MAX-2LIN(q) is equivalent to the Unique Games Conjecture

    Exploring the complex relationships between coping strategies, locus of control and self-esteem with psychopathology: structural equation modeling with a special focus on clinical high-risk of psychosis.

    Abstract Background Coping strategies, competence, and locus of control (LOC) beliefs are important predictors of mental health (MH). However, research into their complex interactions has produced mixed results. Our study investigated them further in the previously unexplored context of clinical high-risk (CHR) of psychosis. Methods We tested six alternative structural equation models in a community sample (N = 523), hypothesizing a mediating role of coping and treating CHR symptoms as (i) an additional mediator or (ii) a specific outcome. Our measurement model included two latent factors of MH: (1) psychopathology (PP), consisting of presence of mental disorders, global and psychosocial functioning, and (2) self-rated health (SRH) status. Results In the model with the best Akaike Information Criterion and the latent factors as outcome variables, maladaptive coping completely mediated the impact of maladaptive LOC on PP and SRH. Additionally, CHR symptoms partially mediated the effect of maladaptive coping on PP and SRH in the community sample, as long as sex was not entered into the model. In the clinical sample (N = 371), the model did not support a mediation by CHR symptoms, despite significant pathways with both coping and MH outcomes; further, competence beliefs directly impacted SRH. Conclusions Coping strategies are an important intervention target for MH promotion, especially in the community. In clinical populations, interventions focusing on coping strategies may improve CHR symptoms, thus potentially supporting better MH, especially SRH. Additionally, due to their mostly cascading effects on MH, improving competence and LOC beliefs may also promote psychological well-being

    Human behavioural adaptations to interglacial lakeshore environments

    The Schöningen 13II-4 “Spear Horizon” is among the most famous lakeshore archaeological sites dating from the Middle Pleistocene in Europe. Multiple well-crafted wooden spears recovered together with a large assemblage of butchered horse bones at Schöningen stimulated a new outlook of the behavioural capabilities of Palaeolithic hunters. Since these discoveries, a wealth of geological and palaeoecological data have been generated to reconstruct the wider Schöningen interglacial lakeshore environment. Yet, the underlying social and economic behaviours of Middle Pleistocene hominins reflected in the archaeological record itself have received less attention. To address these shortcomings, we began a comprehensive zooarchaeological and taphonomic analysis of the entire large mammalian assemblage from the Schöningen 13II-4 “Spear Horizon”, with the goal of re-focusing attention to the “human component” of this important site. In this paper we consider Extended Event-driven Process Chains (eEPCs), a language which is widely used for modeling business processes, documenting industrial reference models and designing workflows. We describe how to translate eEPCs into timed colored Petri nets in order to verify processes given by eEPCs with the CPN Tools

    Burst statistics in an early biofilm quorum sensing model : the role of spatial colony-growth heterogeneity

    Quorum-sensing bacteria in a growing colony of cells send out signalling molecules (so-called "autoinducers") and themselves sense the autoinducer concentration in their vicinity. Once-due to increased local cell density inside a "cluster" of the growing colony-the concentration of autoinducers exceeds a threshold value, cells in this clusters get "induced" into a communal, multi-cell biofilm-forming mode in a cluster-wide burst event. We analyse quantitatively the influence of spatial disorder, the local heterogeneity of the spatial distribution of cells in the colony, and additional physical parameters such as the autoinducer signal range on the induction dynamics of the cell colony. Spatial inhomogeneity with higher local cell concentrations in clusters leads to earlier but more localised induction events, while homogeneous distributions lead to comparatively delayed but more concerted induction of the cell colony, and, thus, a behaviour close to the mean-field dynamics. We quantify the induction dynamics with quantifiers such as the time series of induction events and burst sizes, the grouping into induction families, and the mean autoinducer concentration levels. Consequences for different scenarios of biofilm growth are discussed, providing possible cues for biofilm control in both health care and biotechnology.Peer reviewe

    An Animal Model of Cutaneous Cyst Development Enables the Identification of Three Quantitative Trait Loci, Including the Homologue of a Human Locus (TRICY1)

    Brief Summary Using inbred BN and LE/Stm rats susceptible and resistant, respectively, to chemically induced cutaneous cyst development we were able to further unveil the genetic architecture of inherited multiple cyst formation. N-methyl-N-nitrosourea-treated (BN x LE) F2 intercross rats proved to develop differential numbers of cutaneous cysts, demonstrating epidermal, trichilemmal and verrucous keratinization types. Male rats developed significantly more cysts per animal than females. QTL interval mapping yielded three loci on rat chromosomes 1, 8 and 11 (Ccd1, Ccd2, Ccd3) linked to cutaneous cyst formation. Ccd2 proved to be homologous to the human TRICY1 region which could further be narrowed down by genome comparison in both species. It contains 11 genes with evidence of expression in human keratinocytes.Non peer reviewe

    A Cyanobacteria Enriched Layer of Shark Bay Stromatolites Reveals a New Acaryochloris Strain Living in Near Infrared Light.

    The genus Acaryochloris is unique among phototrophic organisms due to the dominance of chlorophyll d in its photosynthetic reaction centres and light-harvesting proteins. This allows Acaryochloris to capture light energy for photosynthesis over an extended spectrum of up to ~760 nm in the near infra-red (NIR) spectrum. Acaryochloris sp. has been reported in a variety of ecological niches, ranging from polar to tropical shallow aquatic sites. Here, we report a new Acarychloris strain isolated from an NIR-enriched stratified microbial layer 4-6 mm under the surface of stromatolite mats located in the Hamelin Pool of Shark Bay, Western Australia. Pigment analysis by spectrometry/fluorometry, flow cytometry and spectral confocal microscopy identifies unique patterns in pigment content that likely reflect niche adaption. For example, unlike the original A. marina species (type strain MBIC11017), this new strain, Acarychloris LARK001, shows little change in the chlorophyll d/a ratio in response to changes in light wavelength, displays a different Fv/Fm response and lacks detectable levels of phycocyanin. Indeed, 16S rRNA analysis supports the identity of the A. marina LARK001 strain as close to but distinct from from the A. marina HICR111A strain first isolated from Heron Island and previously found on the Great Barrier Reef under coral rubble on the reef flat. Taken together, A. marina LARK001 is a new cyanobacterial strain adapted to the stromatolite mats in Shark Bay

    Pioglitazone Improves Myocardial Blood Flow and Glucose Utilization in Nondiabetic Patients With Combined Hyperlipidemia A Randomized, Double-Blind, Placebo-Controlled Study

    ObjectivesThis study’s aim was to examine whether treatment with pioglitazone, added to conventional lipid-lowering therapy, would improve myocardial glucose utilization (MGU) and blood flow (MBF) in nondiabetic patients with familial combined hyperlipidemia (FCHL).BackgroundThiazolidinediones were found to improve insulin sensitivity and MGU in type 2 diabetes and MBF in Mexican Americans with insulin resistance. Familial combined hyperlipidemia is a complex genetic disorder conferring a high risk of premature coronary artery disease, characterized by high serum cholesterol and/or triglyceride, low high-density lipoprotein (HDL) cholesterol, and insulin resistance.MethodsWe undertook a randomized, double-blind, placebo-controlled study in 26 patients with FCHL, treated with pioglitazone or matching placebo 30 mg daily for 4 weeks, followed by 45 mg daily for 12 weeks. Positron emission tomography was used to measure MBF at rest and during adenosine-induced hyperemia and MGU during euglycemic hyperinsulinemic clamp at baseline and after treatment.ResultsWhereas no change was observed in the placebo group after treatment, patients receiving pioglitazone showed a significant increase in whole body glucose disposal (3.93 ± 1.59 mg/kg/min to 5.24 ± 1.65 mg/kg/min; p = 0.004) and MGU (0.62 ± 0.26 ÎŒmol/g/min to 0.81 ± 0.14 ÎŒmol/g/min; p = 0.0007), accompanied by a significant improvement in resting MBF (1.11 ± 0.20 ml/min/g to 1.25 ± 0.21 ml/min/g; p = 0.008). Furthermore, in the pioglitazone group HDL cholesterol (+28%; p = 0.003) and adiponectin (+156.2%; p = 0.0001) were increased and plasma insulin (−35%; p = 0.017) was reduced.ConclusionsIn patients with FCHL treated with conventional lipid-lowering therapy, the addition of pioglitazone led to significant improvements in MGU and MBF, with a favorable effect on blood lipid and metabolic parameters.     fMRI Reveals Mitigation of Cerebrovascular Dysfunction by Bradykinin Receptors 1 and 2 Inhibitor Noscapine in a Mouse Model of Cerebral Amyloidosis

    Functional magnetic resonance imaging (fMRI) techniques can be used to assess cerebrovascular dysfunction in Alzheimer's disease, an important and early contributor to pathology. We hypothesized that bradykinin receptor inhibition alleviates the vascular dysfunction in a transgenic arcA\u3b2 mouse model of cerebral amyloidosis and that fMRI techniques can be used to monitor the treatment response. Transgenic arcA\u3b2 mice, and non-transgenic littermates of 14 months-of-age were either treated with the bradykinin receptors 1 and 2 blocker noscapine or received normal drinking water as control over 3 months (n = 8-11/group) and all mice were assessed using fMRI at the end of the treatment period. Perfusion MRI using an arterial spin labeling technique showed regional hypoperfusion in arcA\u3b2 compared to non-transgenic controls, which was alleviated by noscapine treatment. Similarly, measuring cerebral blood volume changes upon pharmacological stimulation using vessel dilator acetazolamide revealed recovery of regional impairment of cerebral vascular reactivity in arcA\u3b2 mice upon noscapine treatment. In addition, we assessed with immunohistochemistry beta-amyloid (A\u3b2) and inflammation levels in brain sections. Immunohistological stainings for A\u3b2 deposition (6E10) and related microgliosis (Iba1) in the cortex and hippocampus were found comparable between noscapine-treated and untreated arcA\u3b2 mice. In addition, levels of soluble and insoluble A\u3b238, A\u3b240, A\u3b242 were found to be similar in brain tissue homogenates of noscapine-treated and untreated arcA\u3b2 mice using electro-chemiluminescent based immunoassay. In summary, bradykinin receptors blockade recovered cerebral vascular dysfunction in a mouse model of cerebral amyloidosis. fMRI methods revealed the functional deficit in disease condition and were useful tools to monitor the treatment response
