    Conjugative plasmid mediated inducible nickel resistance in Hafnia alvei 5-5

    Hafnia alvei 5-5, isolated from a soil-litter mixture underneath the canopy of the nickel-hyperaccumulating tree Sebertia acuminata (Sapotaceae) in New Caledonia, was found to be resistant to 30 mM Ni2+ or 2 mM Co2+. The 70-kb plasmid, pEJH 501, was transferred by conjugation to Escherichia coli, Serratia marcescens, and Klebsiella oxytoca. Transconjugant strains expressed inducible nickel resistance to between 5 and 17 mM Ni2+, and cobalt resistance to 2 mM Co2+. A 4.8-kb Sal–EcoRI fragment containing the nickel resistance determinant was subcloned, and the hybrid plasmid was found to confer a moderate level of resistance to nickel (7 mM Ni2+) even to E. coli. The expression of nickel resistance was inducible by exposure to nickel chloride at a concentration as low as 0.5 mM Ni2+. By random TnphoA´-1 insertion mutagenesis, the fragment was shown to have structural genes as well as regulatory regions for nickel resistance. Southern hybridization studies showed that the nickel-resistance determinant from pEJH501 of H. alvei 5-5 was homologous to that of pTOM9 from Alcaligenes xylosoxydans 31A

    Video-Assisted Thoracoscopic Surgery for Correction of Adolescent Idiopatic Scoliosis: Comparison of 4.5 mm versus 5.5 mm Rod Constructs

    PURPOSE: The purpose of this study is to report the comparative results of thoracoscopic correction achieved via cantilever technique using a 4.5 mm thin rod and the poly-axial reduction screw technique using a 5.5 mm thick rod in Lenke type 1 adolescent idiopathic scoliosis (AIS). MATERIALS AND METHODS: Radiographic data, Scoliosis Research Society (SRS) patient-based outcome questionnaires, and operative records were reviewed for forty-nine patients undergoing surgical treatment of scoliosis. The study group was divided into a 4.5 mm thin rod group (n = 24) and a 5.5 mm thick rod group (n = 25). The radiographic parameters that were analyzed included coronal curve correction, the most caudal instrumented vertebra tilt angle correction, coronal balance, and thoracic kyphosis. RESULTS: The major curve was corrected from 49.8 degrees and 47.2 degrees pre-operatively to 24.5 degrees and 18.8 degrees at the final follow-up for the thin and thick rod groups, respectively (50.8% vs. 60.2% correction). There were no significant differences between the two groups in terms of kyphosis, coronal balance, or tilt angle at the time of the final follow-up. The mean number of levels fused was 6.2 in the thin rod group, compared with 5.9 levels in the thick rod group. There were no major intraoperative complications in either group. CONCLUSION: Significant correction loss was observed in the thin rod system at the final follow-up though both groups had comparable correction immediately post-operative.     Intravenous KITENIN shRNA Injection Suppresses Hepatic Metastasis and Recurrence of Colon Cancer in an Orthotopic Mouse Model

    KITENIN (KAI1 C-terminal interacting tetraspanin) promotes invasion and metastasis in mouse colon cancer models. In the present study, we evaluated the effects of KITENIN knockdown by intravenous administration of short hairpin RNAs (shRNAs) in an orthotopic mouse colon cancer model, simulating a primary or adjuvant treatment setting. We established orthotopic models for colon cancer using BALB/c mice and firefly luciferase-expressing CT-26 (CT26/Fluc) cells. Tumor progression and response to therapy were monitored by bioluminescence imaging (BLI). In the primary therapy model, treatment with KITENIN shRNA substantially delayed tumor growth (P = 0.028) and reduced the incidence of hepatic metastasis (P = 0.046). In the adjuvant therapy model, KITENIN shRNA significantly reduced the extent of tumor recurrence (P = 0.044). Mice treated with KITENIN shRNA showed a better survival tendency than the control mice (P = 0.074). Our results suggest that shRNA targeting KITENIN has the potential to be an effective tool for the treatment of colon cancer in both adjuvant and metastatic setting

    Bio-anthropological Studies on Human Skeletons from the 6th Century Tomb of Ancient Silla Kingdom in South Korea

    In November and December 2013, unidentified human skeletal remains buried in a mokgwakmyo (a traditional wooden coffin) were unearthed while conducting an archaeological investigation near Gyeongju, which was the capital of the Silla Kingdom (57 BCE– 660 CE) of ancient Korea. The human skeletal remains were preserved in relatively intact condition. In an attempt to obtain biological information on the skeleton, physical anthropological, mitochondrial DNA, stable isotope and craniofacial analyses were carried out. The results indicated that the individual was a female from the Silla period, of 155 ± 5 cm height, who died in her late thirties. The maternal lineage belonged to the haplogroup F1b1a, typical for East Asia, and the diet had been more C3- (wheat, rice and potatoes) than C4-based (maize, millet and other tropical grains). Finally, the face of the individual was reconstructed utilizing the skull (restored from osseous fragments) and three-dimensional computerized modelling system. This study, applying multi-dimensional approaches within an overall bio-anthropological analysis, was the first attempt to collect holistic biological information on human skeletal remains dating to the Silla Kingdom period of ancient Korea

    Determination of Phthalate Metabolites in Human Serum and Urine as Biomarkers for Phthalate Exposure Using Column-Switching LC-MS/MS

    Objectives:Although phthalates like dibutyl phthalate (DBP) and di-2-ethylhexyl phthalate (DEHP) are commonly used as plasticizers and their metabolites are especially suspected of reproductive toxicity, little is known about occupational exposure to those phthalates. The aim of this study was to assess the utility of measuring the metabolite concentrations of DBP and DEHP in serum and urine samples as an indicator of occupational exposure to those phthalates. Methods:Phthalate metabolites were analyzed by using column-switching high-performance liquid chromatography tandem mass spectrometry (LC-MS/MS). Results:We detected phthalate metabolites in serum and urine matrices at approximately 10-fold lower than the limit of detection of those metabolites in the same matrix by LC-MS/MS without column switching, which was sufficient to evaluate concentrations of phthalate metabolites for industrial workers and the general population. Conclusion:The accuracy and precision of the analytical method indicate that urinary metabolite determination can be a more acceptable biomarker for studying phthalate exposure and adverse health outcomes

    Impact of Glaucoma on Quality of Life and Activities of Daily Living

    Objective/Background: This study was performed to assess the associations of glaucoma with health-related quality of life (HRQOL) and activities of daily living (ADL). Methods: Using data from the Korean National Health and Nutrition Examination Survey III in 2005, we analysed 9,047 participants (≥ 50 years old) and 3,518 participants (≥ 65 years old) to assess the associations of glaucoma with HRQOL and ADL, respectively. The diagnosis of glaucoma was based on questionnaires. HRQOL was evaluated by the EuroQol five-dimension descriptive system (EQ-5D) and the EuroQol visual analogue scale. The ADL was evaluated by the Korean-optimised ADL questionnaire. Logistic-regression analyses were used to assess the associations of glaucoma with HRQOL and ADL. Results: The glaucoma patients showed lower HRQOL and ADL scores compared to the healthy participants. The glaucoma patients showed significantly higher prevalence of impaired health status in all dimensions of EQ-5D, as well as in the “bathing” dimension of ADL as compared with the healthy participants. Impaired HRQOL and ADL were significantly associated with the risk of glaucomatous status (odds ratio [95% confidence interval] of EQ-5D: 1.65 [1.33, 2.05], EuroQol visual analogue scale: 1.75 [1.43, 2.15], and ADL: 2.04 [1.12, 3.74]). Conclusion: In line with previous studies conducted in other countries, Koreans with glaucoma had impaired HRQOL and ADL as compared with healthy Koreans

    Evaluating the Efficiency of Transit-Oriented Development Using Network Slacks-Based Data Envelopment Analysis

    The purpose of this research is to evaluate transit-oriented development (TOD) efficiency in Seoul using the network slacks-based measure data envelopment analysis (NSBM DEA) model. The smartcard data and socio-economic data are used to evaluate the transit efficiency of 352 subway station areas in Seoul. To measure the TOD efficiency, the two-stage network is designed with the transit design stage and the transit efficiency stage. The overall efficiency score of each station area is estimated through each score of the stage. The results of the efficiency evaluation by station area indicate that the overall efficiency score average is 0.349, with the transit design score and efficiency score estimated to be 0.453 and 0.245, respectively. The results indicate that the balance of each stage is crucial to achieve an efficient station in the concept of transit efficiency. With the efficiency scores of the 352 subway station areas, the TOD efficiency is also evaluated by the administrative units in Seoul. The results of district analysis reveal that the top 10 efficient administrative units are characterized by both residential and commercial land use. The results indicate that efficiency is found to be good in areas having both residential and commercial characteristics

    A Travel Behavior-Based Skip-Stop Strategy Considering Train Choice Behaviors Based on Smartcard Data

    This study analyzes a skip-stop strategy considering four types of train choice behavior with smartcard data. The proposed model aims to minimize total travel time with realistic constraints such as facility condition, operational condition, and travel behavior. The travel time from smartcard data is decomposed by two distributions of the express trains and the local trains using a Gaussian mixture model. The utility parameters of the train choice model are estimated with the decomposed distribution using the multinomial logit model. The optimal solution is derived by a genetic algorithm to designate the express stations of the Bundang line in the Seoul metropolitan area. The results indicate the travel times of the transfer-based strategy and the high ridership-based strategy are estimated to be 21.2 and 19.7 min/person, respectively. Compared to the travel time of the current system, the transfer-based strategy has a 5.8% reduction and the high ridership-based strategy has a 12.2% reduction. For the travel behavior-based strategy, the travel time was estimated to be 18.7 minutes, the ratio of the saved travel time is 17.9%, and the energy consumption shows that the travel behavior-based strategy consumes 305,437 (kWh) of electricity, which is about 12.7% lower compared to the current system

    Estimating Express Train Preference of Urban Railway Passengers Based on Extreme Gradient Boosting (XGBoost) using Smart Card Data

    As the share of public transport increases, the express strategy of the urban railway is regarded as one of the solutions that allow the public transportation system to operate efficiently. It is crucial to express the urban railway’s express strategy to balance a passenger load between the two types of trains, that is, local and express trains. This research aims to estimate passengers’ preference between local and express trains based on a machine learning technique. Extreme gradient boosting (XGBoost) is trained to model express train preference using smart card and train log data. The passengers are categorized into four types according to their preference for the local and express trains. The smart card data and train log data of Metro Line 9 in Seoul are combined to generate the individual trip chain alternatives for each passenger. With the dataset, the train preference is estimated by XGBoost, and Shapley additive explanations (SHAP) is used to interpret and analyze the importance of individual features. The overall F1 score of the model is estimated to be 0.982. The results of feature analysis show that the total travel time of the local train feature is found to substantially affect the probability of express train preference with a 1.871 SHAP value. As a result, the probability of the express train preference increases with longer total travel time, shorter in-vehicle time, shorter waiting time, and few transfers on the passenger’s route. The model shows notable performance in accuracy and provided an understanding of the estimation results

    Exploring for Route Preferences of Subway Passengers Using Smart Card and Train Log Data

    As the mode share of the subway in Seoul has increased, the estimation of passenger travel routes has become a crucial issue to identify the congestion sections in the subway network. This paper aims to estimate the travel train of subway passengers in Seoul. The alternative routes are generated based on the train log data. The travel route is then estimated by the empirical cumulative distribution functions (ECDFs) of access time, egress time, and transfer time. The train choice probability is estimated for alternative train combinations and the train combination with the highest probability is assigned to the subway passenger. The estimated result is validated using the transfer gate data which are recorded on private subway lines. The result showed that the accuracy of the estimated travel train is shown to be 95.6%. The choice ratios for no-transfer, one-transfer, two-transfer, three-transfer, and four-transfer trips are estimated to be 53.9%, 37.7%, 6.5%, 1.5%, and 0.4%, respectively. Regarding the practical application, the passenger kilometers by lines are estimated with the travel route estimation of the whole network. As results of the passenger kilometer calculation, the passenger kilometer of the proposed algorithm is estimated to be 88,314 million passenger kilometer. The proposed algorithm estimates the passenger kilometer about 13% higher than the shortest path algorithm. This result implies that the passengers do not always prefer the shortest path and detour about 13% for their convenience
